    • LifeTime aims to track, understand and target human cells during the onset and progression of complex diseases and their response to therapy at single-cell resolution. This mission will be implemented through the development and integration of single-cell multi-omics and imaging, artificial intelligence and patient-derived experimental disease models during progression from health to disease. Analysis of such large molecular and clinical datasets will discover molecular mechanisms, create predictive computational models of disease progression, and reveal new drug targets and therapies. Timely detection and interception of disease embedded in an ethical and patient-centered vision will be achieved through interactions across academia, hospitals, patient-associations, health data management systems and industry. Applying this strategy to key medical challenges in cancer, neurological, infectious, chronic inflammatory and cardiovascular diseases at the single-cell level will usher in cell-based interceptive medicine in Europe over the next decade.

    • Plant traits-the morphological, anatomical, physiological, biochemical and phenological characteristics of plants-determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits-almost complete coverage for 'plant growth form'. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait-environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. This can only be achieved in collaboration with other initiatives.

  • Interhemispheric Comparisons of Large Nighttime Magnetic Perturbation Events Relevant to GICs

    • Nearly all studies of impulsive magnetic perturbation events (MPEs) with large magnetic field variability (dB/dt) that can produce dangerous geomagnetically induced currents (GICs) have used data from the Northern Hemisphere. Here we present details of four large‐amplitude MPE events (|ΔBx| > 900 nT and |dB/dt| > 10 nT/s in at least one component) observed between 2015 and 2018 in conjugate high‐latitude regions (65–80° corrected geomagnetic latitude), using magnetometer data from (1) Pangnirtung and Iqaluit in eastern Arctic Canada and the magnetically conjugate South Pole Station in Antarctica and (2) the Greenland West Coast Chain and two magnetically conjugate chains in Antarctica, AAL‐PIP and BAS LPM. From one to three different isolated MPEs localized in corrected geomagnetic latitude were observed during three premidnight events; many were simultaneous within 3 min in both hemispheres. Their conjugate latitudinal amplitude profiles, however, matched qualitatively at best. During an extended postmidnight interval, which we associate with an interval of omega bands, multiple highly localized MPEs occurred independently in time at each station in both hemispheres. These nighttime MPEs occurred under a wide range of geomagnetic conditions, but common to each was a negative interplanetary magnetic field Bz that exhibited at least a modest increase at or near the time of the event. A comparison of perturbation amplitudes to modeled ionospheric conductances in conjugate hemispheres clearly favored a current generator model over a voltage generator model for three of the four events; neither model provided a good fit for the premidnight event that occurred near vernal equinox.

  • Loneliness, Not Social Support, Is Associated with Cognitive Decline and Dementia Across Two Longitudinal Population-Based Cohorts

    • Background: Poor social health is likely associated with cognitive decline and risk of dementia; however, studies show inconsistent results. Additionally, few studies separate social health components or control for mental health.Objective: To investigate whether loneliness and social support are independently associated with cognitive decline and risk of dementia, and whether depressive symptoms confound the association.Methods: We included 4,514 participants from the population-based Rotterdam Study (RS; aged 71±7SD years) followed up to 14 years (median 10.8, interquartile range 7.4-11.6), and 2,112 participants from the Swedish National Study on Aging and Care in Kungsholmen (SNAC-K; aged 72±10SD years) followed up to 10 years (mean 5.9±1.6SD). At baseline, participants were free of major depression and scored on the Mini-Mental State Examination (MMSE) ≥26 for RS and ≥25 for SNAC-K. We investigated loneliness, perceived social support, and structural social support (specifically marital status and number of children). In both cohorts, dementia was diagnosed and cognitive function was repeatedly assessed with MMSE and a global cognitive factor (g-factor).Results: Loneliness was prospectively associated with a decline in the MMSE in both cohorts. Consistently, persons who were lonely had an increased risk of developing dementia (RS: HR 1.34, 95%CI 1.08-1.67; SNAC-K: HR 2.16, 95%CI 1.12-4.17). Adjustment for depressive symptoms and exclusion of the first 5 years of follow-up did not alter results. Neither perceived or structural social support was associated with cognitive decline or dementia risk.Conclusion: Loneliness, not social support, predicted cognitive decline and incident dementia independently of depressive symptoms.

  • Selecting tree species with high transpiration and drought avoidance to optimise runoff reduction in passive irrigation systems

    • Rainfall in cities can generate large volumes of stormwater runoff which degrades receiving waterways. Irrigating trees with runoff (passive irrigation) has the potential to increase transpiration and contribute to stormwater management by reducing runoff received by downstream waterways, but the stochastic nature of rainfall may expose trees with high transpiration to drought stress. We hypothesized that for success in passive irrigation systems, tree species should exhibit i) high maximum transpiration rates under well-watered conditions, ii) drought avoidance between rainfall events, and iii) high recovery of transpiration with rainfall following a drought. We assessed 13 commonly planted urban tree species in Melbourne, Australia against three metrics representing these behaviours (crop factor, hydroscape area, and transpiration recovery, respectively) in a glasshouse experiment. To aid species selection, we also investigated the relationships between these three metrics and commonly measured plant traits, including leaf turgor loss point, wood density, and sapwood to leaf area ratio (Huber value). Only one species (Tristaniopsis laurina) exhibited a combination of high crop factor (>1.1 mm mm−1 d−1) indicating high transpiration, small hydroscape area (<3 MPa2) indicating drought avoidance, and high transpiration recovery (>85%) following water deficit. Hence, of the species measured, it had the greatest potential to reduce runoff from passive irrigation systems while avoiding drought stress. Nevertheless, several other species showed moderate transpiration, hydroscape areas and transpiration recovery, indicating a balanced strategy likely suitable for passive irrigation systems. Huber values were negatively related to crop factor and transpiration recovery and may therefore be a useful tool to aid species selection. We propose that selecting tree species with high transpiration rates that can avoid drought and recover well could greatly reduce stormwater runoff, while supporting broader environmental benefits such as urban cooling in cities.

    • The nucleation of Alzheimer-associated Aβ peptide monomers can be catalyzed by preexisting Aβ fibrils. This leads to autocatalytic amplification of aggregate mass and underlies self-replication and generation of toxic oligomers associated with several neurodegenerative diseases. However, the nature of the interactions between the monomeric species and the fibrils during this key process, and indeed the ultrastructural localization of the interaction sites have remained elusive. Here we used NMR and optical spectroscopy to identify conditions that enable the capture of transient species during the aggregation and secondary nucleation of the Aβ42 peptide. Cryo-electron microscopy (cryo-EM) images show that new aggregates protrude from the entire length of the progenitor fibril. These protrusions are morphologically distinct from the wellordered fibrils dominating at the end of the aggregation process. The data provide direct evidence that self-replication through secondary nucleation occurs along the sides of fibrils, which become heavily decorated under the current solution conditions (14 μM Aβ42, 20 mM sodium phosphate, 200 μM EDTA, pH 6.8).
