  • Alexandrium catenella and Alexandrium minutum blooms in the Mediterranean Sea: Toward the identification of ecological niches

    • Annual recurrent blooms of the toxic dinoflagellates Alexandrium catenella and Alexandrium minutum were detected from 2000 to 2003 in harbours along the Catalan coast. The interrelation study between the occurrence of the blooms and specific external conditions at the study sites demonstrated that different factors are required for the bloom of each Alexandrium species. Concentrations higher than 105 cells l−1 of A. catenella were only detected in Tarragona harbour. These blooms were associated with water surface temperature between 21 and 25 °C and salinities of around 34 psu or higher than 37 psu. A. minutum appeared widely spread along the Catalan coast, though the most intensive and recurrent blooms of this species were observed in Arenys de Mar harbour. Concentrations of millions of cells per litre of A. minutum were associated with water temperatures below 14 °C and salinities of around 34–36 psu. A. minutum cell densities showed a positive significant correlation with NO3 but a negative correlation with NH4. On the other hand, A. catenella blooms dominated when both NO3 and NH4 levels were high. The prevailing inorganic nitrogen form (NO3 vs. NH4) could explain why these two species rarely coincide in the same harbours. Accumulation of cysts in the sediment was found to be an important potential factor for the recurrence of these species. The 4.3 × 103 A. catenella cysts cm−3 of wet sediment in Tarragona harbour and the 3.02 × 103 A. minutum cysts cm−3 of wet sediment in Vilanova harbour were the highest concentrations observed from the cyst study. Confined waters such as harbours play an important role as reservoirs for the accumulation of cysts and vegetative cells, which contributes to the expansion of these dinoflagellates in the region. However, the particular environmental conditions are also decisive factors of bloom intensity.

  • Genetic associations of LYN with systemic lupus erythematosus

    • We targeted LYN, a src-tyosine kinase involved in B-cell activation, in case-control association studies using populations of European-American, African-American and Korean subjects. Our combined European-derived population, consisting of 2463 independent cases and 3131 unrelated controls, shows significant association with rs6983130 in a female-only analysis with 2254 cases and 2228 controls (P=1.1 x 10(-4), odds ratio (OR)=0.81 (95% confidence interval: 0.73-0.90)). This single nucleotide polymorphism (SNP) is located in the 5' untranslated region within the first intron near the transcription initiation site of LYN. In addition, SNPs upstream of the first exon also show weak and sporadic association in subsets of the total European-American population. Multivariate logistic regression analysis implicates rs6983130 as a protective factor for systemic lupus erythematosus (SLE) susceptibility when anti-dsDNA, anti-chromatin, anti-52 kDa Ro or anti-Sm autoantibody status were used as covariates. Subset analysis of the European-American female cases by American College of Rheumatology classification criteria shows a reduction in the risk of hematological disorder with rs6983130 compared with cases without hematological disorders (P=1.5 x 10(-3), OR=0.75 (95% CI: 0.62-0.89)). None of the 90 SNPs tested show significant association with SLE in the African American or Korean populations. These results support an association of LYN with European-derived individuals with SLE, especially within autoantibody or clinical subsets.

    • The Finnish wolf population (Canis lupus) was sampled during three different periods (1996-1998, 1999-2001 and 2002-2004), and 118 individuals were genotyped with 10 microsatellite markers. Large genetic variation was found in the population despite a recent demographic bottleneck. No spatial population subdivision was found even though a significant negative relationship between genetic relatedness and geographic distance suggested isolation by distance. Very few individuals did not belong to the local wolf population as determined by assignment analyses, suggesting a low level of immigration in the population. We used the temporal approach and several statistical methods to estimate the variance effective size of the population. All methods gave similar estimates of effective population size, approximately 40 wolves. These estimates were slightly larger than the estimated census size of breeding individuals. A Bayesian model based on Markov chain Monte Carlo simulations indicated strong evidence for a long-term population decline. These results suggest that the contemporary wolf population size is roughly 8% of its historical size, and that the population decline dates back to late 19th century or early 20th century. Despite an increase of over 50% in the census size of the population during the whole study period, there was only weak evidence that the effective population size during the last period was higher than during the first. This may be caused by increased inbreeding, diminished dispersal within the population, and decreased immigration to the population during the last study period.

    • In this work we present a micromachined twin sensor that can distinguish between carbon monoxide (CO) and O2 gas taking advantage of the high sensitivity of SnO2 to these gases. The SnO2 nanoparticles of both sensors are catalytically modified with different Pd loadings that act as active filters. In this way, one sensor response is turned to present a higher sensitivity to CO than to O2, whereas the other sensor is mainly turned for detecting O2 variations. The twin sensor has two membranes in the same die of micromachined silicon. Each membrane works independently from the other without any cross talk of temperature. The resistance data obtained from this twin sensor as a function of CO and O 2 concentrations is parametrized. Then, two functions are calculated for the quantification of CO/O2 gas mixtures.   • Evaluation of Limited-Area Models for the Representation of the Diurnal Cycle and Contrasting Nights in CASES-99

    • This study evaluates the ability of three limited-area models [the fifth-generation Pennsylvania State University–National Center for Atmospheric Research Mesoscale Model (MM5), the Coupled Ocean–Atmosphere Mesoscale Prediction System (COAMPS), and the High-Resolution Limited-Area Model (HIRLAM)] to predict the diurnal cycle of the atmospheric boundary layer (ABL) during the Cooperative Atmosphere–Surface Exchange Study (CASES-99) experimental campaign. Special attention is paid to the stable ABL. Limited-area model results for different ABL parameterizations and different radiation transfer parameterizations are compared with the in situ observations. Model forecasts were found to be sensitive to the choice of the ABL parameterization both during the day and at night. At night, forecasts are particularly sensitive to the radiation scheme. All three models underestimate the amplitude of the diurnal temperature cycle (DTR) and the near-surface wind speed. Furthermore, they overestimate the stable boundary layer height for windy conditions and underestimate the stratification of nighttime surface inversions. Favorable parameterizations for the stable boundary layer enable rapid surface cooling, and they have limited turbulent mixing. It was also found that a relatively large model domain is required to model the Great Plains low-level jet. A new scheme is implemented for the stable boundary layer in the Medium-Range Forecast Model (MRF). This scheme introduces a vegetation layer, a new formulation for the soil heat flux, and turbulent mixing based on the local scaling hypothesis. The new scheme improves the representation of surface temperature (especially for weak winds) and the stable boundary layer structure.
