  • Slipdisc : A versatile sample preparation platform for point of care diagnostics

    • We report a microfluidic sample preparation platform called "Slipdisc" based on slipchip technology. Slipdisc is a rotational slipchip that uses a unique hand-wound clockwork mechanism for precise movement of specially fabricated polycarbonate discs. In operation, the microchannels and microchambers carved on the closely aligned microfluidic discs convert from continuous filled paths to defined compartments using the slip movement. The clockwork mechanism introduced here is characterised by a food dye experiment and a conventional HRP TMB reaction before measuring lactate dehydrogenase (LDH) enzyme levels, which is a crucial biomarker for neonatal diagnostics. The colorimetry based detection of LDH was performed with an unmodified camera and an image analysis procedure based on normalising images and observing changes in red channel intensity. The analysis showed a close to unity coefficient of determination (R2 = 0.96) in detecting the LDH concentration when compared with a standard Chemical Analyser, demonstrating the excellent performance of the slipdisc platform with colorimetric detection. The versatile point of care sample preparation platform should ideally be suited for a multitude of applications at resource-limited settings.

  • Three dimensional Slipdisc aimed at HIV viral load detection

    • In this study, we introduce a three dimensional Slipdisc1 based on slipchip technology, aimed to be used at detection of HIV-1 viral load in resource limited settings. This method is based on magnetic bead based isolation of RNA from sample, followed by release of RNA in an elution buffer, followed by amplification of the initial concentration of RNA in an elution buffer. The novelty is the extraction method which is a one step method and involves using magnetic beads attached to sample DNA to pass through an immiscible oil phase2 for one step washing.

  • Meru couples planar cell polarity with apical-basal polarity during asymmetric cell division

    • Polarity is a shared feature of most cells. In epithelia, apical-basal polarity often coexists, and sometimes intersects with planar cell polarity (PCP), which orients cells in the epithelial plane. From a limited set of core building blocks (e.g. the Par complexes for apical-basal polarity and the Frizzled/Dishevelled complex for PCP), a diverse array of polarized cells and tissues are generated. This suggests the existence of little-studied tissue-specific factors that rewire the core polarity modules to the appropriate conformation. In Drosophila sensory organ precursors (SOPs), the core PCP components initiate the planar polarization of apical-basal determinants, ensuring asymmetric division into daughter cells of different fates. We show that Meru, a RASSF9/RASSF10 homologue, is expressed specifically in SOPs, recruited to the posterior cortex by Frizzled/Dishevelled, and in turn polarizes the apical-basal polarity factor Bazooka (Par3). Thus, Meru belongs to a class of proteins that act cell/tissue-specifically to remodel the core polarity machinery.

  • The spectrum of acute central nervous system symptoms during the treatment of childhood acute lymphoblastic leukaemia

    • Background: Children with central nervous system (CNS) toxicity during therapy for acute lymphoblastic leukaemia (ALL) are at risk for treatment modifications, long-term sequelae and even higher mortality. A better understanding of CNS symptoms and their complications improves the potential to prevent and treat them.Methods: Patient files from 649 children treated with Nordic Society of Pediatric Hematology and Oncology ALL92 and ALL2000 protocols in Finland were reviewed retrospectively for any acute CNS symptom. Detailed data on symptoms, examinations and treatment of the underlying CNS complications were collected from the medical records. Disease-related and outcome data were retrieved from the Nordic leukaemia registry.Results: Altogether, 13% (86) of patients with ALL had acute CNS symptoms. Most symptoms (64%) occurred during the first 2 months of therapy. Posterior reversible encephalopathy syndrome was the most frequent complication (4.5%). Cerebrovascular events were diagnosed in 10 cases (1.6%), while methotrexate-related stroke-like syndrome (SLS) was observed in only one patient (0.2%). CNS symptoms due to systemic or unclear conditions, especially sepsis, were important for differential diagnosis. CNS leukaemia was associated with CNS symptoms (hazard ratio [HR] = 4.03; P = .003), and epilepsy was a common sequel of CNS complications (19%).Conclusions: Acute CNS symptoms are common during ALL therapy, occurring mainly during the first 2 months of treatment. Patients with CNS leukaemia at diagnosis are at a higher risk for CNS toxicity. Despite intensive CNS-directed methotrexate treatment, SLS was diagnosed extremely rarely in our series.

  • Influence of discrete delay on pattern formation in a ratio-dependent prey-predator model

    • In this paper we explore how the two mechanisms, Turing instability and Hopf bifurcation, interact to determine the formation of spatial patterns in a ratio-dependent prey predator model with discrete time delay. We conduct both rigorous analysis and extensive numerical simulations. Results show that four types of patterns, cold spot, labyrinthine, chaotic as well as mixture of spots and labyrinthine can be observed with and without time delay. However, in the absence of time delay, the two aforementioned mechanisms have a significant impact on the emergence of spatial patterns, whereas only Hopf bifurcation threshold is derived by considering the discrete time delay as the bifurcation parameter. Moreover, time delay promotes the emergence of spatial patterns via spatio-temporal Hopf bifurcation compared to the non-delayed counterpart, implying the destabilizing role of time delay. In addition, the destabilizing role is prominent when the magnitude of time delay and the ratio of diffusivity are comparatively large.

  • Magnetism in FeNiW disordered alloys : Experiment and theory

    • We report here an experimental study of magnetization of FeNiW alloys at different compositions. We have studied variation of magnetization with temperature (at low external fields) and magnetic field (at low temperatures). The alloy shows para to ferromagnetic transitions across the composition range. We do not find any indication of the spin-glass phase. We have supplemented the experimental work with theoretical analysis using the first-principles tight-binding linear muffin-tin orbitals based augmented space recursion method. Our theoretical estimates of magnetic moment and Curie temperatures agree well with experiment. Our mean-field phase analysis also does not indicate the possibility of a spin-glass.

We analyze the data using a fist-principles density functional based electronic structure method and a mean-field phase diagram study.
