  • Profile of and risk factors for poststroke cognitive impairment in diverse ethnoregional groups

    • Objective To address the variability in prevalence estimates and inconsistencies in potential risk factors for poststroke cognitive impairment (PSCI) using a standardized approach and individual participant data (IPD) from international cohorts in the Stroke and Cognition Consortium (STROKOG) consortium. Methods We harmonized data from 13 studies based in 8 countries. Neuropsychological test scores 2 to 6 months after stroke or TIA and appropriate normative data were used to calculate standardized cognitive domain scores. Domain-specific impairment was based on percentile cutoffs from normative groups, and associations between domain scores and risk factors were examined with 1-stage IPD meta-analysis. Results In a combined sample of 3,146 participants admitted to hospital for stroke (97%) or TIA (3%), 44% were impaired in global cognition and 30% to 35% were impaired in individual domains 2 to 6 months after the index event. Diabetes mellitus and a history of stroke were strongly associated with poorer cognitive function after covariate adjustments; hypertension, smoking, and atrial fibrillation had weaker domain-specific associations. While there were no significant differences in domain impairment among ethnoracial groups, some interethnic differences were found in the effects of risk factors on cognition. Conclusions This study confirms the high prevalence of PSCI in diverse populations, highlights common risk factors, in particular diabetes mellitus, and points to ethnoracial differences that warrant attention in the development of prevention strategies.

  • Land set-up systems in Italy: A long tradition of soil and water conservation sewed up to a variety of pedo-climatic environments

    • Land set-up systems in Italy were and, in a few cases, still are integral parts of agricultural landscapes. The soils of Italy mirror a wide variety of climates and morphologies and derive from a great diversity of parent materials influenced by different soil temperature and moisture regimes. Furthermore, their development was influenced by several anthropogenic activities over time including land settings. Land set-up systems have been widely used throughout the Italian soilscape to regulate surface and underground water and to improve the physical soil features in a way that ensures mixed herbaceous and tree-crops cultivation. In recent times, new technologies have been introduced in agroforestry exploitation of the land, while the ancient but tested hydraulic soil systems have been abandoned. As a result, a great part of Italian soilscapes is prone to water erosion and floods, an unforeseen impact, which can be counterbalanced by good soil management practices. Moreover, new pedotechniques have been recently adopted, transforming soils and landscapes, but generating sometimes soil degradation issues. After presenting the main features characterizing Italian soils and landscapes, we describe all the historical land set-up systems adopted over time, highlighting their positive functions in terms of both soil conservation and agricultural production. We also emphasise the importance of maintaining such land settings both as distinctive parts of cultural landscapes and strategies of soil conservation, and the need of developing new systems based on the principles of environmental sustainability.

  • Initial Evolution of Passivation Layers in Non-Aqueous Aluminium Batteries

    • Aluminium batteries (AlBs) have gathered considerable attention, primarily due to the high capacity, the low cost, the large abundance in the Earth's crust, and the recyclability of the Al metal anode. However, several hurdles must be surpassed to make AlBs a feasible energy storage technology and two of them are interconnected; the presence of an ionic and electronically insulating native oxide layer on the Al metal anode that calls for special non-aqueous, most often ionic liquid based acidic electrolytes, to enable reversible plating and stripping of Al. We here find the passivation layer initially formed in contact with an ionic liquid electrolyte (ILE) to have a porous and very complex nature, i.e. an outer inorganic/organic layer and an inner oxide-rich layer. Furthermore, it grows under open circuit voltage conditions by simultaneous dissolution and re-deposition of dissolved products, while during galvanostatic cycling this is exacerbated by an electrochemical etching that causes pitting corrosion of the Al metal itself. All of this leads to unstable interfaces being formed and the co-existence of several species at the Al metal anode surface, of which a proper understanding and mitigation are crucial to make AlBs a reality.

  • Ecosystem-specific microbiota and microbiome databases in the era of big data

    • The rapid development of sequencing methods over the past decades has accelerated both the potential scope and depth of microbiota and microbiome studies. Recent developments in the field have been marked by an expansion away from purely categorical studies towards a greater investigation of community functionality. As in-depth genomic and environmental coverage is often distributed unequally across major taxa and ecosystems, it can be difficult to identify or substantiate relationships within microbial communities. Generic databases containing datasets from diverse ecosystems have opened a new era of data accessibility despite costs in terms of data quality and heterogeneity. This challenge is readily embodied in the integration of meta-omics data alongside habitat-specific standards which help contextualise datasets both in terms of sample processing and background within the ecosystem. A special case of large genomic repositories, ecosystem-specific databases (ES-DB's), have emerged to consolidate and better standardise sample processing and analysis protocols around individual ecosystems under study, allowing independent studies to produce comparable datasets. Here, we provide a comprehensive review of this emerging tool for microbial community analysis in relation to current trends in the field. We focus on the factors leading to the formation of ES-DB's, their comparison to traditional microbial databases, the potential for ES-DB integration with meta-omics platforms, as well as inherent limitations in the applicability of ES-DB's.

  • Improving Plant Health Through Nutrient Remineralization in Aquaponic Systems

    • The exploitation of readily bioavailable fish excreta as a source of plant nutrients lies at the cornerstone of aquaponics farming. Research on nutrient cycling in aquaponic systems has devoted considerable attention to the plant uptake of dissolved nutrients in fish excreta, however, the integration of particulate-bound nutrients into downstream hydroponic farming has remained elusive. The high amount of organic carbon present in fish sludge may lead to biofouling if directly incorporated into hydroponic circulation systems, reducing the utility of incorporating fish solids on a large scale. In this study, we implemented a novel treatment system capable of reducing the carbon and nitrogen load of fish solids to produce a liquid fertilizer for a downstream hydroponics unit. Lettuce (Lactuca sativa) fertilized with exclusively a commercial nutrient solution, the biofilter effluent (coupled aquaponic system), effluent from the solids treatment system, or the latter two combined were grown in nutrient flow technique gutters downstream of a recirculating aquaculture system stocked with rainbow trout (Oncorhynchus mykiss). While crop yields were lower for the aquaponic treatments compared to lettuce grown in a commercial nutrient solution, plant sap analysis demonstrated a contrasting picture with respect to internal nutrient concentrations. Lettuce grown in the commercial hydroponic solution were deficient in several mineral nutrients (Mg, Ca, Na, and Si) nor did they have higher iron concentrations despite the significantly higher EDTA-chelated aqueous iron (460 x greater than other treatments) in the nutrient solution. Nutrient uptake in the rhizosphere was not investigated on a molecular level, although stunted rhizosphere growth in the commercial nutrient solution control suggests a weakened capacity for nutrient uptake in comparison to other treatments. Alongside the remineralization of micronutrients, the solids treatment system addressed the common issue of excess carbon leading to biofouling via a total suspended solids reduction of 87.27% +/- 9.95 during the coupled aquaponics cultivation period. Ultimately, these data lead to two important conclusions. Firstly, optimizing nutrient bioavailability is not synonymous to increasing the presence of a nutrient in the water column. Secondly, estimating ideal nutrient solution concentrations involves both preventing nutrient blocking and improving bioavailability.

  • Simultaneous biomethane production and nutrient remineralization from aquaculture solids

    • The rapid expansion of the aquaculture industry has brought about a heightened focus on the waste produced by high intensity fish farming. In closed-containment, recirculating aquaculture systems (RAS), fish solids are mechanically separated and/or coagulated before being disposed as waste. Subsequent revalorization is typically limited to the direct dispersal of aquaculture solids onto agricultural fields. Here, we developed a novel, continuous flow, low-cost solids waste treatment system for freshwater and saline RAS. Rotating drum filter backwash was collected as the primary feedstock for anaerobic digestion. A laboratory scale set up was used to monitor the conversion of the solids into a methane-rich (60-80% purity) biogas stream. Iron supplementation (ferric iron at 100 mg/L and 1000 mg/L) improved salt tolerance of the methanogenic community, leading to higher methane yields in a supplemented (FeCl3 at 1000 mg/L) saline treatment than the saline control. The application of iron additionally improves pH stability and volatile fatty acid utilization. The methane yield ranged from 0.1 to 0.4 NL CH4/ g VS across the three freshwater treatments and the iron-supplemented saline treatment, however, it was significantly lower for the saltwater control: ranging between 0.08 and 0.25 NL CH4/ g VS. These values correspond to a percentage yield of 57-86% of the total biomethane potential. Overall, implementing anaerobic digestion for RAS waste valorization may generate significant amounts of biomethane to be used in electricity and heating for large-scale aquaculture facilities, while even for smaller facilities it may off-set costs and mitigate environmental impacts of the waste streams.

  • Plants Dictate Root Microbial Composition in Hydroponics and Aquaponics

    • The role of the microbial community in mediating fish and plant co-culture is often considered the black box of aquaponics. Despite widespread recognition regarding the dependency of plants on their rhizosphere, the extent to which upstream aquaculture influences downstream hydroponic root communities has been poorly described in the literature. In this study we performed a taxonomic survey (16S rRNA metabarcoding) of microbial communities originating in the facility water source, hydroponic nutrient solution (HNS) sump, nutrient supplemented biofilter effluent (BF) sump, and recirculating aquaculture system tanks stocked with Nile tilapia (Oreochromis niloticus). Lettuce (Lactuca sativa) was then grown using the HNS and BF effluent under sterilized or mature (prior aquaponics/hydroponics lettuce culture water) conditions, likewise, the influence of probiotic addition or inoculation with soil-grown lettuce rhizosphere was assessed. Compositional similarities across treatments suggest that under soil-less conditions, plants are able to exert a stronger discriminatory influence on their rhizosphere composition than is done by colonization from upstream sources. Furthermore, cluster dendrograms grouped the sterilized and unsterilized treatments more consistently together than hydroponics and aquaponics treatments. These findings contradict conventional beliefs that microbial communities in the water column colonize roots based on their presence alone, ignoring the role that plants play in rhizosphere community selection.

  • Association of Autoimmune Addison's Disease with Alleles of STAT4 and GATA3 in European Cohorts

    • Background: Gene variants known to contribute to Autoimmune Addison's disease (AAD) susceptibility include those at the MHC, MICA, CIITA, CTLA4, PTPN22, CYP27B1, NLRP-1 and CD274 loci. The majority of the genetic component to disease susceptibility has yet to be accounted for. Aim: To investigate the role of 19 candidate genes in AAD susceptibility in six European case-control cohorts. Methods: A sequential association study design was employed with genotyping using Sequenom iPlex technology. In phase one, 85 SNPs in 19 genes were genotyped in UK and Norwegian AAD cohorts (691 AAD, 715 controls). In phase two, 21 SNPs in 11 genes were genotyped in German, Swedish, Italian and Polish cohorts (1264 AAD, 1221 controls). In phase three, to explore association of GATA3 polymorphisms with AAD and to determine if this association extended to other autoimmune conditions, 15 SNPs in GATA3 were studied in UK and Norwegian AAD cohorts, 1195 type 1 diabetes patients from Norway, 650 rheumatoid arthritis patients from New Zealand and in 283 UK Graves' disease patients. Meta-analysis was used to compare genotype frequencies between the participating centres, allowing for heterogeneity. Results: We report significant association with alleles of two STAT4 markers in AAD cohorts (rs4274624: P = 0.00016; rs10931481: P = 0.0007). In addition, nominal association of AAD with alleles at GATA3 was found in 3 patient cohorts and supported by meta-analysis. Association of AAD with CYP27B1 alleles was also confirmed, which replicates previous published data. Finally, nominal association was found at SNPs in both the NF-kappa B1 and IL23A genes in the UK and Italian cohorts respectively. Conclusions: Variants in the STAT4 gene, previously associated with other autoimmune conditions, confer susceptibility to AAD. Additionally, we report association of GATA3 variants with AAD: this adds to the recent report of association of GATA3 variants with rheumatoid arthritis.

  • Psychosocial characteristics differentiate non-distressing and distressing voices in 10,346 adolescents

    • Adolescents hearing non-existent voices may be at risk for psychosis, but the prevalence of voice-hearing (VH) in the general population complicates clinical interpretations. Differentiating between VH with and without distress may aid treatment decisions in psychosis services, but understanding the differences between these two phenomena as they present in the normal adolescent population is necessary to validate this differentiation. The present study compared VH with and without distress in 10,346 adolescents in relation to clinical characteristics, known risk factors, predictors and psychosocial moderators of psychosis. A population-based cohort of Norwegian 16-19 years old adolescents completed a comprehensive web-based questionnaire, including two questions from the extended Launay-Slade Hallucinations Scale: (1) I often hear a voice speaking my thoughts aloud and (2) I have been troubled by hearing voices in my head. Adolescents reporting no VH, non-distressing VH or distressing VH were compared on 14 psychosocial and clinical variables. A multinomial regression model showed that non-disturbing voices were predicted by better school grades, social dysfunction, distractibility, affective symptoms and experience of trauma, while the disturbing voices were predicted by the experience of bullying and trauma, perceived negative self-worth and self-efficacy, less family support, dysregulation of activation, distractibility, self-harm and anxiety. Hearing voices without distress versus being distressed by the voices is related to different constellations of psychosocial variables, suggesting that they represent two separate groups of adolescents. The findings validate the emphasis on distress in clinical practice.

    • In the present study, the correlation between sperm number, sperm quality (speed, viability, longevity and length), sperm bundles quality (size and dissolving rate) and male body size has been tested in the eastern mosquitofish Gambusia holbrooki a poecilid species characterized by coercive mating tactics where males do not possess obvious ornaments, and the body size is the key determinant of pre-copulatory male mating success. The results do not tally with theoretical predictions. Indeed, no correlation between male body size and either sperm or sperm-bundle traits has been found, evidencing the lack of the theoretically expected trade-off between the investment in characters involved in mate acquisition and the investment in ejaculate quality. An explanation for the observed pattern comes from the extremely dynamic mating system of G. holbrooki, characterized by variable size-related male mating success and strong post-copulatory selective pressure, with all males facing a similar high level of sperm competition. In this situation, a higher investment in growth and maintenance at the expense of ejaculate quality is not expected. These results underscore the necessity to comprehend detailed information on species' reproductive biology and reproductive environment to understand both the evolution of ejaculate characteristics and possible deviations from theoretical predictions.
