    • Ett verk för självspelande piano, dator och musiker. Ett komplext system av fyra virtuella musiker kontrolleras gestiskt från 16 trycksensorer, med hjälp av en mappningsteknik som jag utvecklat under de senaste åren. Det musikaliska resultatet är en komplex improvisativ pianomusik, som på makronivå kontrolleras i ögonblicket av tonsättaren, samtidigt som dess detaljer är komplexa och oförutsägbara.

  • Full circle : Rise and fate of genetic variation in Marasmius oreades fairy rings

    • Genetic variation is a prerequisite for evolution. The degree of variability within a species is governed by forces including mutation, recombination and selection. In the kingdom of fungi, where periodic sexual reproduction may be interleaved with extended vegetative phases, generators of variability are not restricted to act only during sexual cycles. Such generators may be in the form of mutations to the genome, affecting single base pairs up to large-scale rearrangements, movement of transposable elements, or non-meiotic shuffling of genetic variants by mitotic recombination or parasexuality. Particularly in mushroom-forming fungi, where mycelia may become large and old, the evolutionary potential of variation acquired over vegetative growth is expected to be large. In this thesis, I have studied the rise and fate of variation gained during vegetative growth in the mushroom-forming fungus Marasmius oreades: a non-model species known for growing in ‘fairy rings’. By taking advantage of state-of-the-art genome sequencing technology and developing new bioinformatics methods, the genome sequence of M. oreades was successfully reconstructed. This resource was combined with genome re-sequencing to identify different types of mutations in M. oreades fairy rings, and to investigate the transmission of such mutations into the next generation through sexual spores. The results presented in this thesis reveal that the M. oreades genome is extremely stable at all levels during vegetative growth in its natural environment. Furthermore, the few mutations that arise do not seem to be transferred to the sexual spores. A significant amount of transposon movement was however revealed in monokaryotic strains when separated from dikaryons and grown in the laboratory. The combination of these results suggests that fungi possess an unknown system to suppress the accumulation of mutations during growth in nature, and that the apparent lack of a segregated germline in fungi potentially has to be reconsidered. Thus, contrary to expectations, the vegetative life stage in long-lived mushroom-forming fungi does not contribute much genetic variation, making these organisms more similar to animals and plants than previously considered. Further studies are needed to reveal how fungi control mutation accumulation, and elucidate if transposon activity is high also in naturally derived monokaryons in the form of meiotic progeny. The findings in this thesis add to what is known about how genetic variation is introduced into natural populations, how fungi deal with mutations, and highlight the complexity of genetic systems in mushroom-forming fungi.

    • Let C be the unit circle in R2. We can view C as a plane graph whose vertices are all the points on C, and the distance between any two points on C is the length of the smaller arc between them. We consider a graph augmentation problem on C, where we want to place k ≥ 1 shortcuts on C such that the diameter of the resulting graph is minimized. We analyze for each k with 1 ≤ k ≤ 7 what the optimal set of shortcuts is. Interestingly, the minimum diameter one can obtain is not a strictly decreasing function of k. For example, with seven shortcuts one cannot obtain a smaller diameter than with six shortcuts. Finally, we prove that the optimal diameter is 2 + ⊖ (1/k 2 3 ) for any k.

  • Scattering by an anisotropic circle

    • The scattering by a circle is considered when the outside medium is isotropic and the inside medium is anisotropic (orthotropic). The problem is a scalar one and is phrased as a scattering problem for elastic waves with polarization out of the plane of the circle (SH wave), but the solution is with minor modifications valid also for scattering of electromagnetic waves. The equation inside the circle is first transformed to polar coordinates and it then explicitly contains the azimuthal angle through trigonometric functions. Making an expansion in a trigonometric series in the azimuthal coordinate then gives a coupled system of ordinary differential equations in the radial coordinate that is solved by power series expansions. With the solution inside the circle complete the scattering problem is solved essentially as in the classical case. Some numerical examples are given showing the influence of anisotropy, and it is noted that the effects of anisotropy are generally strong except at low frequencies where the dominating scattering only depends on the mean stiffness and not on the degree of anisotropy.

  • Initiation of bacteriophage P2 rolling circle replication

    • Viruses are very simple entities. In its most simple form, the virus consists of a protein shell containing genetic material. To propagate the virus needs a host to parasitize, in which it can replicate its genetic material and assembly new virus particles. Viruses that parasitize bacteria are termed bacteriophages. In this study the DNA replication of bacteriophage P2 has been investigated. Bacteriophage P2 replicates by a rolling circle mechanism, which is a replication strategy used by many replicons, ranging from bacterial plasmids to animal viruses. Two phage encoded proteins are necessary for P2 rolling circle replication; A and B. The A protein initiates rolling circle replication by a single-stranded, site-specific nick at the origin of replication. This exposes a free 3' end which serves as a primer for DNA polymerisation. Upon nicking, P2 A links itself covalently to the 5' end of the DNA through one of its two active-site tyrosine residues. The A protein can also catalyse the reverse reaction. This event releases the displaced strand simultaneously as a new round of replication is initiated. The B protein has been shown to be required for lagging strand synthesis and has been suggested to be an E. coli DnaB helicase loader.In this work, the interplay between the two tyrosine residues located in the presumed catalytic site of P2 A has been closely examined. We found that the two tyrosine residues, 450 and 454, play non-equivalent functional roles in their catalytic activity. We propose that tyrosine 454 initiates replication and that tyrosine 450 only cleaves when tyrosine 454 is covalently joined to DNA, thereby reinitiating replication. Also, data from the catalytic activity of P2 A, involving different divalent cations, suggests that conformational changes occur within the protein upon binding DNA. Another intriguing phenomenon of P2 A is that it acts in cis in vivo, i.e. it can only act upon the molecule from which it was transcribed. Here, we show that the cis-activity is retained in vitro during coupled transcription and translation in a S30 extract and that the information required for the cis-activity in vivo and in vitro is contained entirely in the coding region for the A protein. An elegant model for the mechanism of the P2 A cis-activity is also presented. Further, in this work in vivo and in vitro evidence for a physical association between P2 B and E. coli DnaB are provided supporting the view that P2 B is a helicase loader. Presumably, the B protein plays an important role as a molecular matchmaker for the whole P2 replisome assembly. Here, also presented are the enigma of the inverted palindrome nature of the P2 origin, the P2 A localisation within the bacterial cell, other RCR initiating enzymes, a compilation of all known cis-acting proteins, evolutionary aspects of cis-action, the outline for exploiting P2 A as a biotechnology tool and much much more.

    • Implementation of e-Government means cultural changes involving changes of attitudes and competences, apart from technological changes. There is a need for competence development of employees and citizens using relevant educational forms and pedagogical approaches. An interview study of a web-based course for issues related to implementation of e-Government is described and analysed. The course was developed with a study circle pedagogical approach. Interviews were made with eighteen respondents from different authorities, different geographical locations and different professional roles. The respondents were especially positive to the group discussions of the studied course. The discussions were relevant for support of behaviour changes and competence development related to implementation of e-Government. A study circle approach could be relevant in order to encourage development-oriented learning needed for implementation of e-Government. There is also a need for pedagogical learning of producers and organisers of web-based education in public organisation, in order to make them conscious about the importance of different pedagogical approaches for different education objectives.

  • Threads without ends : a mobile sewing circle

    • In this paper the exhibition Threads – a Mobile Sewing Circle is used as an example of a design that travels. To tell the story of how Threads travels we use the concepts of immutable mobile (Latour 1990) and fluidity (de Laet and Mol 2000) – concepts that invite us to think of standardisation and stability on one hand and changes and adaptability on the other. Since Threads is continuously assembled, disassembled and reassembled in different contexts and by different actors, we argue that Threads needs to be able to deal with changes and local conditions and cannot strive for stability in the sense of ‘no change’. On the contrary, Threads is dependent on local actors’ engagement, which partly is done through adding, replacing and altering parts and practices of Threads which also redraws its boundaries. We further argue that it is through what has been called design-after-design (Ehn 2008) that Threads can become entangled in the local setting and thereby matter. Through examples from Threads it is also shown that, what we call, a fluid designer role is helpful when making fluid designs travel.

    • We consider spin-polarized electrons in a single Landau level on a cylinder as the circumference of the cylinder goes to infinity. This gives a model of interacting electrons on a circle where the momenta of the particles are restricted and there is no kinetic energy. Quantum Hall states are exact ground states for appropriate short range interactions, and there is a gap to excitations. These states develop adiabatically from this one-dimensional quantum Hall circle to the bulk quantum Hall states and further on into the Tao–Thouless states as the circumference goes to zero. For low filling fractions a gapless state is formed which we suggest is connected to the Wigner crystal expected in the bulk.
