    • Sperm samples may be used for assisted reproductive technologies (e.g., farmed or endangered species) or as a source of haploid DNA or sperm-specific RNA. When ejaculated spermatozoa are not available or are very difficult to obtain, as is the case for most wild endangered species, the epididymides of dead animals (e.g., animals that have been found dead, shot by hunters or poachers, or that that require euthanasia in zoological collections) can be used as a source of sperm. Such epididymal sperm samples are usually contaminated with cellular debris, erythrocytes, leukocytes, and sometimes also bacteria. These contaminants may be sources of reactive oxygen species that damage spermatozoa during freezing or contribute undesired genetic material from diploid cells. We used single-layer centrifugation through a colloid formulation, Androcoll-C, to successfully separate wolf epididymal spermatozoa from contaminating cells and cellular debris in epididymal samples harvested from carcasses. Such a procedure may potentially be applied to epididymal sperm samples from other species.

  • Djurhållning och antibiotika: en gyllene triangel!

    • För ett par veckor sedan var jag i Amsterdam och talade om etiska dilemman och utmaningar i arbetet med antibiotikaresistens i djurhållning och veterinärmedicin på en konferens. ”Från fabrik till gödselstack” var devisen för mitt bidrag, för att framhålla att djurhållningens användning av antibiotika – precis som sjukvårdens – är en del av en större helhet, där även produktionssystemen för läkemedel och mat (och friska djur och människor), och naturmiljön ingår. Varje del både bidrar till utmaningen med resistens – att vår användning och hantering av dessa viktiga mediciner gör dem obrukbara – och utgör löften för att sätta in åtgärder som kan mildra problemet. Alla sådana åtgärder kommer samtidigt ha nackdelar och kostnader – därav behovet att analysera etiken. På konferensen fick jag också chans att förkovra mig i olika slags forskning om hur jordbruk och annan djurhållning kan hantera antibiotikaresistensen bättre. Jag lärde mig då att området utgör en riktig gyllene triangel – på gott och ont.

  • Completeness of metabolic disease recordings in Nordic national databases for dairy cows

    • The four Nordic countries Denmark (DK), Finland (FI), Norway (NO) and Sweden (SE) all have national databases where diagnostic events in dairy cows are recorded. Comparing and looking at differences in disease occurrence between countries may give information on factors that influence disease occurrence, optimal diseases control and treatment strategies. For such comparisons to be valid, the data in these databases should be standardised and of good quality.The objective of the study presented here was to assess the quality of metabolic disease recordings, primarily milk fever and ketosis, in four Nordic national databases. Completeness of recording figures of database registrations at two different levels was chosen as a measure of data quality. Firstly, completeness of recording of all disease events on a farm regardless of veterinary involvement, called 'Farmer observed completeness', was determined. Secondly, completeness of recording of veterinary treated disease events only, called 'Veterinary treated completeness', was determined. To collect data for calculating these completeness levels a simple random sample of herds was obtained in each country. Farmers who were willing to participate, recorded for 4 months in 2008, on a purpose made registration form, any observed illness in cows, regardless of veterinary involvement. The number of participating herds was 105, 167, 179 and 129 in DK, FI, NO and SE respectively. In total these herds registered 247, 248, 177 and 218 metabolic events for analysis in DK, FI, NO and SE, respectively. Data from national databases were subsequently extracted, and the two sources of data were matched to find the proportion, or completeness, of diagnostic events registered by farmers that also existed in national databases. Matching was done using a common diagnostic code system and allowed for a discrepancy of 7 days for registered date of the event. For milk fever, the Farmer observed completeness was 77%, 67%, 79% and 79%, and for Veterinary treated completeness 88%, 71%, 80% and 82%, in DK, FI, NO and SE, respectively. For ketosis, the Farmer observed completeness was 77%, 55%, 70% and 46%, and for Veterinary treated completeness 84%, 75%, 79% and 56%, in DK, FI, NO and SE, respectively. The differences observed in some of these completeness figures indicate that metabolic disease frequency measures when calculated from these four national databases do not accurately reflect the true difference in metabolic disease occurrence in dairy cows between countries. Further standardisation of disease registrations is necessary for valid between-country assessments.

  • Farmer awareness of cow longevity and implications for decision-making at farm level

    • The aim of this study was to investigate if farmer awareness of longevity can explain why the longevity of dairy cows does not increase in Sweden, despite the fact that the genetic trend for longevity in Swedish dairy cattle is positive. In this study, farmers' decisions were put in the forefront through a qualitative study with semi-structured interviews. The results showed that farmers seemed well aware of biological factors related to cow longevity, that they think the demands on dairy cows have increased over time and that they correlate the higher demands to shorter longevity. However, the farmers had not worked explicitly with longevity of their own herd. There are subjective management decisions behind a culling of a dairy cow, and the reporting of culling reasons does not mirror the farmer's reasoning behind the decisions, which is a key factor determining longevity.

    • This study investigated the relationship of the health of the newborn foal and (1) number of polymorphonuclear leukocytes (PMNLs) in the amniotic fluid, (2) bacteria present in the amniotic fluid and the venous umbilical blood, and (3) bacteria present in the uterus of the newly foaled mare. A further aim was to investigate relationships between the bacteriologic findings in the amniotic fluid, umbilical blood, and uterus postpartum. Samples were taken from 50 Standardbred trotter foaling mares from a well-managed stud in Sweden. Parturition was spontaneous in all cases. Length of pregnancy, parturition and postpartum complications, health status of the foal, the time between foaling and the expulsion of the placenta, and the number of postfoaling mares becoming pregnant after insemination were recorded. Amniotic fluid was collected when the amniotic vesicle was clearly visible; it was analyzed for bacteriology and occurrence of PMNLs. Umbilical blood was analyzed for the presence of bacteria and the concentration of serum amyloid A. The uterus of themarewas swabbed for bacteriology 6 to 17 hours postpartum. A blood samplewas taken from the foal before administering plasma. The foals were divided into two groups: group 1 required up to 2 hours to rise after birth (2 hours; 31 foals) and group 2 requiredmore than two hours (>2 hours; 19 foals). The length of gestation varied between 332 and 356 days; there was no significant difference in gestation length between the two foal groups. Partus and postpartum complications occurred in a significantly higher proportion of mares giving birth to group 2 foals than group 1 foals (P ¼ 0.02), although uterine culture postpartum and the subsequent pregnancy rate per season were not different between the groups. Compromised health status was significantly higher among foals belonging to group 2 than group 1 (P ¼ 0.001). Most of the amniotic samples contained 5% or less PMNLs. Only three samples contained more than 30% PMNLs; group 2 foals had the highest percentage of PMNLs. Bacterial growth was found in both amniotic fluid (57%) and umbilical blood (35%) in mares irrespective of whether their foals were healthy or compromised. Coagulase-negative staphylococci were the most frequent bacteria. There were no differences in bacterial occurrence in amniotic fluid or in umbilical blood between the two foal groups

    • A farmer or a dairy consultant's ability to identify and properly treat lame dairy cows is key to managing lameness on farms. However, this ability is dependent on their knowledge and perceptions regarding lameness. To date these topics are poorly understood in all dairy systems. To our knowledge, this is the first study that uses a qualitative approach to describe the perception of lameness in Brazilian pasture-based dairy farms. The aim of this study was firstly, to contextualize farmers and dairy consultants' culture and knowledge on the topic of lameness and, secondly, to understand how these factors may influence lameness management on the farms. Indepth face-to-face semi-structured interviews were done with participating farm owners (n = 21) and dairy consultants (n = 13). Thematic analyses of the interview content provided evidence that the dairy community's overarching culture around lameness acts as a barrier preventing positive actions targeting lameness control and prevention. The emerging themes identified indicate that lameness is a vague, ill-defined concept among both farmers and dairy consultants working in this region. There was a shared belief that pasture-based systems have a low risk for lameness. Furthermore, the prevailing culture for tackling lameness was reactive, particularly in the case of lame cows affected by physical trauma or environmental factors. Treatment was frequently delayed and the misuse of antibiotics was evident. Described community culture was rationalised via the cognitive dissonance and health belief models. We suggest that increased dialogue, including educational efforts within the community, may lead to increased sensitivity of risk perception of lameness within the community and in turn trigger appropriate diagnosis and treatment of lame cows.

  • Decabromobiphenyl, Polybrominated Diphenyl Ethers, and Brominated Phenolic Compounds in Serum of Cats Diagnosed With the Endocrine Disease Feline Hyperthyroidism

    • The incidence of cats being diagnosed with feline hyperthyroidism (FH) has increased greatly since it was first described in 1979. The cause of FH has not been established. Hypothetically, there is a link between increasing FH and exposure to brominated flame retardants. Much greater polybrominated diphenyl ethers (PBDE) concentrations have been reported in cat serum compared with human serum, likely due to cat licking behaviour. This study aimed to extend the present identification of brominated compounds in cat serum, with a focus on hydroxylated metabolites of PBDE, to improve the understanding of feline metabolism of PBDEs. A pooled serum sample from 30 Swedish pet cats with FH was analysed, and brominated species were identified. The results showed exposure to the discontinued flame retardant decabromobiphenyl (BB-209) and technical penta- and octa-BDEs. Altogether 12 PBDE congeners were identified along with 2'-MeO-BDE68. Furthermore, 2,4-dibromophenol, 2,4,6-, 2,4,5- and 2,3,4-tribromophenol plus 2'-OH-BDE68, 6-OH-BDE47, 5-OH-BDE47, 4'-OH-BDE49 were identified. 2,4,6-tribromophenol and 6-OH-BDE47 were the most prominent species in cat serum. Considering that these are natural products, it can be concluded that metabolism of PBDEs to OH-PBDEs is not a major route of PBDE elimination in cats. It is notable that BB-209, 6-OH-BDE47, and 2,4,6-tribromophenol all suggested that endocrine-disrupting chemicals were present in high concentrations in cat serum.
