  • Imunoreactivity of zinc transporter 7 (ZNT7) in mouse dorsal root ganglia.

    • In the present study, we showed for the first time the localization of ZNT7 immunoreactivity in the mouse dorsal root ganglion (DRG) by means of immunohistochemistry and confocal laser scanning microscopy. Our results revealed that ZNT7 immunoreactivity was abundantly expressed in the nerve cells of the mouse DRG. Strong ZNT7 immunoreactivity was predominantly distributed in the perinuclear region of positive cells, while the nuclei were devoid of staining. Double immunofluorescence labeling of ZNT7 and TGN38 revealed a colocalization of the two antigens in the Golgi apparatus. In addition, the presence of labile zinc ions was detected with in vivo zinc selenium autometallography (AMG). AMG observations showed that the zinc staining pattern was also predominately located in the perinuclear Golgi area, like the ZNT7 immunostaining pattern in the DRG. These observations strongly suggest that ZNT7 may play an important role in facilitating zinc transport into the Golgi apparatus from the cytosol in the mouse DRG.

  • Filamin B deficiency in mice results in skeletal malformations and impaired microvascular development

    • Mutations in filamin B (FLNB), a gene encoding a cytoplasmic actin-binding protein, have been found in human skeletal disorders, including boomerang dysplasia, spondylocarpotarsal syndrome, Larsen syndrome, and atelosteogenesis phenotypes I and III. To examine the role of FLNB in vivo, we generated mice with a targeted disruption of Flnb. Fewer than 3% of homozygous embryos reached term, indicating that Flnb is important in embryonic development. Heterozygous mutant mice were indistinguishable from their wild-type siblings. Flnb was ubiquitously expressed; strong expression was found in endothelial cells and chondrocytes. Flnb-deficient fibroblasts exhibited more disorganized formation of actin filaments and reduced ability to migrate compared with wild-type controls. Flnb-deficient embryos exhibited impaired development of the microvasculature and skeletal system. The few Flnb-deficient mice that were born were very small and had severe skeletal malformations, including scoliotic and kyphotic spines, lack of intervertebral discs, fusion of vertebral bodies, and reduced hyaline matrix in extremities, thorax, and vertebrae. These mice died or had to be euthanized before 4 weeks of age. Thus, the phenotypes of Flnb-deficient mice closely resemble those of human skeletal disorders with mutations in FLNB.

  • Filamins in cardiovascular development.

    • Filamins are classically recognized as large cytoplasmic proteins that cross-link cortical actin into dynamic 3-dimensional structures and transmit extracellular signals through integrin receptors into the cytoplasm. However, recent reports indicate that filamins interact with a large number of other proteins with diverse functions, including transcriptional factors and cellular molecules involved in signaling, adhesion, and cellular motility, and are also present in the cell nucleus. In addition, genetic mutations in filamins have been linked to a wide range of human genetic disorders, including skeletal, central nervous system, and cardiovascular malformations, highlighting distinct filamin interactions. Here, we update the cardiovascular phenotypes of patients with mutations in filamin genes and mice deficient in filamins and filamin-interacting proteins.

    • We serendipitously identified a single nucleotide polymorphism (SNP), 8636C>A (rs1804197) in the 3'-untranslated region of the adenomatous polyposis coli (APC) gene to be associated with autism spectrum disorder (ASD). In order to gain further evidence for the association between the APC locus and ASD, we genotyped four additional adjacent common SNPs (rs2229992, rs42427, rs459552, and rs465899) in the coding regions within the APC gene in a set of Swedish ASDs and controls. One common haplotype TGAG was found to be associated with ASD after haplotype analysis using both Haploview v3.1.1 (P = 0.006) and COCAPHASE v2.403 (P = 0.030). This result is the first to suggest that the genomic locus at APC is associated with ASD, and that the APC gene itself is a good predisposing candidate to be evaluated in future studies due to its important role in neuronal development and function.

  • Impact of Magnetoresistance and Anisotropy on Synchronized Spin Torque Oscillators

    • Synchronization of pairs of serially connected Spin Torque Oscillators (STO) was studied using Landau-Lifshitz-Gilbert-Slonczewski magnetodynamic simulations. The impact of realistic process variations on STO synchronization was estimated by allowing for different in-plane anisotropy fields (H(k)) in the two STOs. We find that the synchronized state is extremely sensitive to STO process variations and can only sustain up to 4% H(k) variation. However, the addition of a small time delay dramatically improves the robustness of the synchronization and allows as much as 145% H(k) variation in the entire out-of-plane precession regime.
