  • The autoantibody response to cyclic citrullinated collagen type II peptides in rheumatoid arthritis

    • OBJECTIVES: The detection of anti-citrullinated peptide antibodies (ACPAs) is a serological hallmark of RA. Autoantibodies reactive with collagen type II (CII) are present in RA sera and synovial fluid and are potentially pathogenic. Here, we investigate the prevalence and specificity of the autoantibody responses to defined citrullinated cyclic peptides derived from CII in a China RA cohort. METHODS: Using bead-based multiplex assay, we examined the presence of autoantibodies binding to 54 cyclic 17-mer citrullinated CII peptides, encompassing all citrullinate epitopes in CII, and the corresponding unmodified peptides in 415 RA patients, in addition to 304 patients with OA. Furthermore, the autoantibody responses to a selected set of 10 cyclic citrullinated peptides were also examined in 203 healthy individuals. RESULTS: Autoantibody responses to cyclic citrullinated CII peptides were higher in RA patients as compared with OA patients or healthy individuals, whereas little or negligible antibody responses to cyclic unmodified CII peptides were observed. Interestingly, several novel citrullinated CII epitopes were identified. Antibodies to these novel citrullinated CII epitopes showed not only substantial overlapping reactivities but also had unique specificities. CONCLUSION: We found a high prevalence of autoantibodies against cyclic citrullinated CII in the sera of patients in a China RA cohort. The present study revealed heterogeneous binding patterns against novel citrullinated CII epitopes, which may help to stratify RA patients into different subgroups.

  • Semantics-aware Dynamic Graph Convolutional Network for Traffic Flow Forecasting

    • Traffic flow forecasting is a challenging task due to its spatio-temporal nature and the stochastic features underlying complex traffic situations. Currently, Graph Convolutional Network (GCN) methods are among the most successful and promising approaches. However, most GCNs methods rely on a static graph structure, which is generally unable to extract the dynamic spatio-temporal relationships of traffic data and to interpret trip patterns or motivation behind traffic flows. In this paper, we propose a novel Semantics-aware Dynamic Graph Convolutional Network (SDGCN) for traffic flow forecasting. A sparse, state-sharing, hidden Markov model is applied to capture the patterns of traffic flows from sparse trajectory data; this way, latent states, as well as transition matrices that govern the observed trajectory, can be learned. Consequently, we can build dynamic Laplacian matrices adaptively by jointly considering the trip pattern and motivation of traffic flows. Moreover, high-order Laplacian matrices can be obtained by a newly designed forward algorithm of low time complexity. GCN is then employed to exploit spatial features, and Gated Recurrent Unit (GRU) is applied to exploit temporal features. We conduct extensive experiments on three real-world traffic datasets. Experimental results demonstrate that the prediction accuracy of SDGCN outperforms existing traffic flow forecasting methods. In addition, it provides better explanations of the generative Laplace matrices, making it suitable for traffic flow forecasting in large cities and providing insight into the causes of various phenomena such as traffic congestion.   • Characterization of chronic relapsing antibody mediated arthritis in mice with a mutation in Ncf1 causing reduced oxidative burst

    • Rheumatoid arthritis (RA) is a chronic autoimmune disorder affecting joints with a hallmark of autoantibody production. Mannan-enhanced collagen type II (COL2) antibody induced arthritis (mCAIA) in neutrophil cytosolic factor 1(Ncf1) mutation mouse is a chronic disease model imitating RA in mice. In this study, we characterize the chronic phase of mCAIA in Ncf1 mutated (BQ.Ncf1m1j/m1j) mice. Arthritis was induced by an intravenous injection of anti-COL2 monoclonal antibodies on day 0 followed by intra-peritoneal injections of mannan (from Saccharomyces cerevisiae) on days 3 and 65 in BQ.Ncf1 m1j/m1j and BQ mice. Bone erosion was analysed by computed tomography (CT) and blood cell phenotypes by flow cytometry. Cytokines and anti-COL2 antibodies were analyzed with multiplex bead-based assays. The arthritis in the Ncf1m1j/m1j mice developed with a chronic and relapsing disease course, which was followed for 200 days and bone erosions of articular joints were evaluated. An increased number of circulating CD11b+ Ly6G+ neutrophils were observed during the chronic phase, together with a higher level of G-CSF (granulocyte colony-stimulating factor) and TNF-α. In conclusion, the chronic relapsing arthritis of mCAIA in the Ncf1m1j/m1j mice develop bone erosions associated with a sustained neutrophil type of inflammatory responses.

    • Proteolysis-targeting chimeras (PROTACs), the hetero-bifunctional compounds containing a specific ligand to bind the target protein, a suitable linker, and an E3 ubiquitin ligase substrate, are being developed for therapeutic applications. PROTACs hijack the catalytic activity of ubiquitin E3 ligases to mediate proteasome dependent degradation of selected protein of interest (POI), by bringing the ligase and POI into close spatial proximity and initiating the poly-ubiquitination process. Compared to the traditional small-molecule drugs, PROTACs reduce the problems of dosage, drug resistance, side effects and undruggable targets that could not be targeted pharmacologically. In this review, all the POIs, and peptide to small-molecule based PROTACs developed during the past two decades are summarized and directions for future development are discussed. © 2020 Elsevier Inc.

  • Familial risk and heritability of intellectual disability : a population-based cohort study in Sweden

    • Background: Intellectual disability (ID) aggregates in families, but factors affecting individual risk and heritability estimates remain unknown. Methods: A population-based family cohort study of 4,165,785 individuals born 1973–2013 in Sweden, including 37,787 ID individuals and their relatives. The relative risks (RR) of ID with 95% confidence intervals (95% CI) were obtained from stratified Cox proportional-hazards models. Relatives of ID individuals were compared to relatives of unaffected individuals. Structural equation modeling was used to estimate heritability. Results: Relatives of ID individuals were at increased risk of ID compared to individuals with unaffected relatives. The RR of ID among relatives increased proportionally to the degree of genetic relatedness with ID probands; 256.70(95% CI 161.30–408.53) for monozygotic twins, 16.47(13.32–20.38) for parents, 14.88(12.19–18.16) for children, 7.04(4.67–10.61) for dizygotic twins, 8.38(7.97–8.83) for full siblings, 4.56(4.02–5.16) for maternal, 2.90(2.49–3.37) for paternal half-siblings, 3.03(2.61–3.50) for nephews/nieces, 2.84(2.45–3.29) for uncles/aunts, and 2.04(1.91–2.20) for cousins. Lower RRs were observed for siblings of probands with chromosomal abnormalities (RR 5.53, 4.74–6.46) and more severe ID (mild RR 9.15, 8.55–9.78, moderate RR 8.13, 7.28–9.08, severe RR 6.80, 5.74–8.07, and profound RR 5.88, 4.52–7.65). Male sex of relative and maternal line of relationship with proband was related to higher risk (RR 1.33, 1.25–1.41 for brothers vs. sisters and RR 1.49, 1.34–1.68 for maternal vs. paternal half-siblings). ID was substantially heritable with 0.95(95% CI 0.93–0.98) of the variance in liability attributed to genetic influences. Conclusions: The risk estimates will benefit researchers, clinicians, families in understanding the risk of ID in the family and the whole population. The higher risk of ID related to male sex and maternal linage will be of value for planning and interpreting etiological studies in ID. © 2021 The Authors.   • Report on decarbonisation design-approaches based on urban typologies : Deliverable D2.5

    • This report identifies different typology-based approaches and methods for decarbonising the energy sector of cities. Respectively, typologies were evaluated, and design approaches were developed. In a first step, already existing typologies were evaluated, including a study by the Technical University of Darmstadt and examples from the City of Vienna. In a next step, conceivable structuring criteria and decarbonisation approaches from existing work within the DCP project were identified and summarised. These include structuring criteria such as heat demand density, renewable energy sources or types of refurbishment activities. On this basis, a new typology was developed. Five highly weighted criteria could be derived from the results of the expert survey, including structural energy efficiency, coverage of district heating, potential for renewable sources, potential for waste heat and heat demand density. These criteria formed the basis for the development of the novel typology. The first typology represents areas with high compatibility with highly weighted criteria, the third typology represents areas with comparably low compatibility, while the second typology is associated in between. Based on the developed typology, six design approaches were presented in this report. One short-term and one long-term approach for each typology include recommendations as well as concrete measures for strategic decision-making.

    • The aim of this study was to evaluate professional development and promotion as well as whether the new competence was requested after completion of a one-year master programme (MP) in Cardiovascular (CV) nursing. In Sweden the first one-year MP within the CV area took place between 2003 and 2005 at Halmstad University with a follow-up in 2007. The sample consisted of seventeen former students who had completed the MP. A questionnaire was developed comprising twenty items grouped into three main sections. Most of the nurses stated that their knowledge was requested and that they had developed their professional attitude, skills and knowledge. Seven of the nurses had a new position and five of them had advanced in terms of leadership. In three cases, the master exam was critical for the employers’ decision. Six of the nurses who had a new post had obtained an increase in salary. In conclusion, the MP led to increased competence as well as improved career prospects. Information about the MP should be available at clinics, and collaboration between clinical practice and education is necessary in order to ensure relevant use of master educated nurses’ competence.

    • The aim of the present study was to illustrate how a patient with a myocardial infarction history may experience current anxiety. We conducted the assessment using a phenomenological method. The participant was a 62-year old man, chronically ill due to two myocardial infarctions and heart failure. Five categories emerged from the data: distress, worthlessness, insecurity, indifference, and lack of strength, illustrating feelings embedded in the current anxiety. These feelings relate to past and present experiences as well as to an insecure future. The findings are not generalizable in a statistical manner, but they are an illustration of the importance of identifying a cardiac patient’s current anxiety from a holistic perspective, in order to understand what is occurring emotionally and to understand the need for emotional support. Qualities in the caring relationship may create possibilities for this patient to receive and perceive emotional support. © 2019 Springer Nature Switzerland AG. Part of Springer Nature.

    • Theoretically, one part of supervision is the direct object i.e. what can be learnt, another part is the indirect object aiming at how the learning process is constituted and influenced by both the doctoral student and his/her supervisor. Emphasising both the what and the how object of learning as well as their interrelationship may be an important factor for the development of the role of supervisor in research. The aim of this study was to describe supervision of doctoral students from the perspective of a research supervisor. Interview data were analysed in accordance with content analysis. The result showed that research supervision had a latent content of awareness of underlying structures and the preconditions for learning that became visible through self-appraisal, orientation towards a goal, in the interaction and the performance of an art. Awareness was a cornerstone in research supervision that enabled both the what and the how aspect of learning.

    • The aim of this paper was to describe family support in terms of the sources as well as the consequences of burden for caregivers, patients and family interaction. Caregivers' sources of burden were related to difficulties in coping with patients' emotional responses, physical complaints and altered life style behaviour as well as future oriented factors. Consequences in terms of caregivers' perceptions of emotional distress and personal losses are elucidated. In terms of family support, when focusing on sources of burden for patients' concerning family support, emotional support is emphasised and related to overprotection. Two different definitions of the overprotection concept are used in order to elucidate different consequences of the phenomenon. Sources and consequences of burden are also related to family interaction. The family is regarded as a system in which a cardiac disease may cause an imbalance. Poor marital quality or poor family functioning is described as a hindrance to a new balance. To summarise, the reviewed literature unanimously shows that the interactive support between family members when dealing with cardiac disease constitutes a considerable emotional burden. Therefore the support for emotional communication within the family should take priority. The knowledge described can provide a foundation for the development of family support in the cardiac area.
