    • Latency is a fundamental problem for all distributed systems including digital libraries. To reduce user perceived delays both caching -- keeping accessed objects for future use -- and prefetching -- transferring objects ahead of access time -- can be used. In a previous paper we have reported that caching is not worthwhile for digital libraries due to low re-access frequencies. In this paper we evaluate our previous findings that prefetching can be used instead. To do this we have set up an experimental prefetching proxy which is able to retrieve documents from remote fulltext archives before the user demands them. Using a simple prediction to keep the overhead of unnecessarily transfered data limited, we find that it is possible to cut the user perceived average delay a factor of two.

  • Common variants in HNF-1 alpha and risk of type 2 diabetes.

    • Mutations in the hepatocyte nuclear factor 1-alpha gene (HNF-1 alpha, now known as the transcription factor 1 gene [TCF1]) cause the most common monogenic form of diabetes, MODY3, but it is not known if common variants in HNF-1a are associated with decreased transcriptional activity or phenotypes related to type 2 diabetes, or whether they predict future type 2 diabetes. We studied the effect of four common polymorphisms (rs1920792, I27L, A98V and S487N) in and upstream of the HNF-1 alpha gene on transcriptional activity in vitro, and their possible association with type 2 diabetes and insulin secretion in vivo. Certain combinations of the I27L and A98V polymorphisms in the HNF-1 alpha gene showed decreased transcriptional activity on the target promoters glucose transporter 2 (now known as solute carrier family 2 [facilitated glucose transporter], member 2) and albumin in both HeLa and INS-1 cells. In vivo, these polymorphisms were associated with a modest but significant impairment in insulin secretion in response to oral glucose. Insulin secretion deteriorated over time in individuals carrying the V allele of the A98V polymorphism (n=2,293; p=0.003). In a new case-control (=1,511 and n=2,225 respectively) data set, the I27L polymorphism was associated with increased risk of type 2 diabetes, odds ratio (OR)=1.5 (p=0.002; multiple logistic regression), particularly in elderly (age > 60 years) and overweight (BMI > 25 kg/m(2)) patients (OR=2.3, p=0.002). This study provides in vitro and in vivo evidence that common variants in the MODY3 gene, HNF-1 alpha, influence transcriptional activity and insulin secretion in vivo. These variants are associated with a modestly increased risk of late-onset type 2 diabetes in subsets of elderly overweight individuals.

  • The potential to use QSAR to populate ecotoxicity characterisation factors for simplified LCIA and chemical prioritisation

    • Purpose: Today’s chemical society use and emit an enormous number of different, potentially ecotoxic, chemicals to the environment. The vast majority of substances do not have characterisation factors describing their ecotoxicity potential. A first stage, high throughput, screening tool is needed for prioritisation of which substances need further measures. Methods: USEtox characterisation factors were calculated in this work based on data generated by quantitative structure-activity relationship (QSAR) models to expand substance coverage where characterisation factors were missing. Existing QSAR models for physico-chemical data and ecotoxicity were used, and to further fill data gaps, an algae QSAR model was developed. The existing USEtox characterisation factors were used as reference to evaluate the impact from the use of QSARs to generate input data to USEtox, with focus on ecotoxicity data. An inventory of chemicals that make up the Swedish societal stock of plastic additives, and their associated predicted emissions, was used as a case study to rank chemicals according to their ecotoxicity potential. Results and discussion: For the 210 chemicals in the inventory, only 41 had characterisation factors in the USEtox database. With the use of QSAR generated substance data, an additional 89 characterisation factors could be calculated, substantially improving substance coverage in the ranking. The choice of QSAR model was shown to be important for the reliability of the results, but also with the best correlated model results, the discrepancies between characterisation factors based on estimated data and experimental data were very large. Conclusions: The use of QSAR estimated data as basis for calculation of characterisation factors, and the further use of those factors for ranking based on ecotoxicity potential, was assessed as a feasible way to gather substance data for large datasets. However, further research and development of the guidance on how to make use of estimated data is needed to achieve improvement of the accuracy of the results.

    • Maintenance of transportation infrastructure assets can be relatively expensive, since it does not only include the direct cost of interventions, but also the indirect consequences of traffic disruptions. To make optimal decisions about maintenance actions, including rehabilitation and upgrading, reliable information about the performance of existing structures is needed. However, obtaining such information might require significant efforts and can be done in various ways. The purpose of an ongoing Swedish research project BIG BRO is to develop a framework for a decision support methodology that can be used for implementing maintenance strategies for bridges on a rational basis. The present paper provides a brief overview about the project as well as describes some of the ongoing work.

  • Modelling of bending creep of low- and high-temperature-dried spruce timber

    • In the current project, a finite element model is developed to analyse the long-term behaviour of timber beams. The time-dependent response of wood subjected to bending and moisture changes is investigated in terms of strains and stresses. A rheological model is implemented to capture the effects of creep, mechano-sorption and hygroexpansion. The model is validated against test results from Bengtsson and Kliger (Holzforschung 57:95–100, 2003). The results of the analysis showed that the mechano-sorptive creep of low- (LT) and high-temperature-dried (HT) timber beams can be sufficiently modelled with a spring and a single Kelvin body. The different mechano-sorptive behaviour of LT- and HT-dried specimens is considered with different mechano-sorptive and shrinkage–swelling parameters. The presented model could be used to derive general mechano-sorptive parameters: (1) for better prediction of creep over the service life and (2) to provide a basis of time-dependent probabilistic calculations for structural-sized timber in serviceability limit state.

    • Context. The light curves of tidally locked hot Jupiters transiting fast-rotating, early-type stars are a rich source of information about both the planet and star, with full-phase coverage enabling a detailed atmospheric characterisation of the planet. Although it is possible to determine the true spin-orbit angle ψ-a notoriously difficult parameter to measure-from any transit asymmetry resulting from gravity darkening induced by the stellar rotation, the correlations that exist between the transit parameters have led to large disagreements in published values of ψ for some systems. Aims. We aimed to study these phenomena in the light curves of the ultra-hot Jupiter MASCARA-1 b, which is characteristically similar to well-studied contemporaries such as KELT-9 b and WASP-33 b. Methods. We obtained optical CHaracterising ExOPlanet Satellite (CHEOPS) transit and occultation light curves of MASCARA-1 b, and analysed them jointly with a Spitzer/IRAC 4.5 μm full-phase curve to model the asymmetric transits, occultations, and phase-dependent flux modulation. For the latter, we employed a novel physics-driven approach to jointly fit the phase modulation by generating a single 2D temperature map and integrating it over the two bandpasses as a function of phase to account for the differing planet-star flux contrasts. The reflected light component was modelled using the general ab initio solution for a semi-infinite atmosphere. Results. When fitting the CHEOPS and Spitzer transits together, the degeneracies are greatly diminished and return results consistent with previously published Doppler tomography. Placing priors informed by the tomography achieves even better precision, allowing a determination of ψ = 72.1-2.4+2.5 deg. From the occultations and phase variations, we derived dayside and nightside temperatures of 3062-68+66 K and 1720 ± 330 K, respectively.Our retrieval suggests that the dayside emission spectrum closely follows that of a blackbody. As the CHEOPS occultation is too deep to be attributed to blackbody flux alone, we could separately derive geometric albedo Ag = 0.171-0.068+0.066 and spherical albedo As = 0.266-0.100+0.097 from the CHEOPS data, and Bond albedoAB = 0.057-0.101+0.083 from the Spitzer phase curve.Although small, the Ag and As indicate that MASCARA-1 b is more reflective than most other ultra-hot Jupiters, where H- absorption is expected to dominate. Conclusions. Where possible, priors informed by Doppler tomography should be used when fitting transits of fast-rotating stars, though multi-colour photometry may also unlock an accurate measurement of ψ. Our approach to modelling the phase variations at different wavelengths provides a template for how to separate thermal emission from reflected light in spectrally resolved James Webb Space Telescope phase curve data.

  • The Discovery and Follow-up of Four Transiting Short-period Sub-Neptunes Orbiting M Dwarfs

    • Sub-Neptunes with radii of 2-3 R ⊕ are intermediate in size between rocky planets and Neptune-sized planets. The orbital properties and bulk compositions of transiting sub-Neptunes provide clues to the formation and evolution of close-in small planets. In this paper, we present the discovery and follow-up of four sub-Neptunes orbiting M dwarfs (TOI-782, TOI-1448, TOI-2120, and TOI-2406), three of which were newly validated by ground-based follow-up observations and statistical analyses. TOI-782 b, TOI-1448 b, TOI-2120 b, and TOI-2406 b have radii of R p = 2.740 − 0.079 + 0.082 R ⊕ , 2.769 − 0.068 + 0.073 R ⊕ , 2.120 ± 0.067 R ⊕, and 2.830 − 0.066 + 0.068 R ⊕ and orbital periods of P = 8.02, 8.11, 5.80, and 3.08 days, respectively. Doppler monitoring with the Subaru/InfraRed Doppler instrument led to 2σ upper limits on the masses of <19.1 M ⊕, <19.5 M ⊕, <6.8 M ⊕, and <15.6 M ⊕ for TOI-782 b, TOI-1448 b, TOI-2120 b, and TOI-2406 b, respectively. The mass-radius relationship of these four sub-Neptunes testifies to the existence of volatile material in their interiors. These four sub-Neptunes, which are located above the so-called “radius valley,” are likely to retain a significant atmosphere and/or an icy mantle on the core, such as a water world. We find that at least three of the four sub-Neptunes (TOI-782 b, TOI-2120 b, and TOI-2406 b), orbiting M dwarfs older than 1 Gyr, are likely to have eccentricities of e ∼ 0.2-0.3. The fact that tidal circularization of their orbits is not achieved over 1 Gyr suggests inefficient tidal dissipation in their interiors.

    • Human aquaporin 5 (HsAQP5)facilitates the transport of water across plasma membranes and has been identified within cells of the stomach, duodenum, pancreas, airways, lungs, salivary glands, sweat glands, eyes, lacrimal glands, and the inner ear. AQP5, like AQP2, is subject to posttranslational regulation by phosphorylation, at which point it is trafficked between intracellular storage compartments and the plasma membrane. Details concerning the molecular mechanism of membrane trafficking are unknown. Here we report the x-ray structure of HsAQP5 to 2.0-angstrom resolution and highlight structural similarities and differences relative to other eukaryotic aquaporins. A lipid occludes the putative central pore, preventing the passage of gas or ions through the center of the tetramer. Multiple consensus phosphorylation sites are observed in the structure and their potential regulatory role is discussed. We postulate that a change in the conformation of the C terminus may arise from the phosphorylation of AQP5 and thereby signal trafficking.

  • The influence of microstructure and mechanical properties on the machinability of martensitic and bainitic prehardened mould steels

    • The machinability of two prehardened mould steels, one continuously-cooled and one quenched and tempered, is compared in terms of cutting temperature, cutting force and tool life. Both materials have a hardness of similar to 40 HRC which is a typical hardness for prehardened mould steels. The results of machinability tests are related to mechanical properties and microstructural features, and the material removal rate is estimated for the materials. The continuously-cooled steel with bainitic structure shows higher machinability and possesses superior impact toughness and ductility compared to the quenched and tempered steel with martensitic structure. These properties are very important for plastic mould steels. The continuously-cooled bainitic steel also has the advantage of not requiring any costly quenching and tempering processes in connection with its production.

  • High-resolution impact assessment of climate change on building energy performance considering extreme weather events and microclimate – Investigating variations in indoor thermal comfort and degree-days

    • Climate change and urbanization are two major challenges when planning for sustainable energy transition in cities. The common approach for energy demand estimation is using only typical meso-scale weather data in building energy models (BEMs), which underestimates the impacts of extreme climate and microclimate variations. To quantify the impacts of such underestimation on assessing the future energy performance of buildings, this study simulates a high spatiotemporal resolution BEM for two representative residential buildings located in a 600 × 600 m2 urban area in Southeast Sweden while accounting for both climate change and microclimate. Future climate data are synthesized using 13 future climate scenarios over 2010-2099, divided into three 30-year periods, and microclimate data are generated considering the urban morphology of the area. It is revealed that microclimate can cause 17% rise in cooling degree-day (CDD) and 7% reduction in heating degree-day (HDD) on average compared to mesoclimate. Considering typical weather conditions, CDD increases by 45% and HDD decreases by 8% from one 30-year period to another. Differences can become much larger during extreme weather conditions. For example, CDD can increase by 500% in an extreme warm July compared to a typical one. Results also indicate that annual cooling demand becomes four and five times bigger than 2010-2039 in 2040-2069 and 2070-2099, respectively. The daily peak cooling load can increase up to 25% in an extreme warm day when accounting for microclimate. In the absence of cooling systems during extreme warm days, the indoor temperature stays above 26°C continuously over a week and reaches above 29.2°C. Moreover, the annual overheating hours can increase up to 140% in the future. These all indicate that not accounting for influencing climate variations can result in maladaptation or insufficient adaptation of urban areas to climate change.
