  • The ubiquitous hyaluronan: Functionally implicated in the oviduct?

    • Hyaluronan (hyaluronic acid) is a simple, nonantigenic, nonsulfated glycosaminoglycan present everywhere in the extracellular compartments of the body. Noteworthy, it is highly conserved phylogenetically, from sauropsida to mammals; and plays a plethora of roles from embryonic/fetal development to adult physiological and pathological events, including tumor development. In reproduction, hyaluronan has proven related to initial events as sperm survival, buildup of the sperm reservoir in the oviduct, regulation of sperm capacitation, and prefertilization to later participate in embryo, fetal, and placental development. Synthesis, binding (via the CD44 membrane receptor), and degradation of hyaluronan occur in male and female genital organs, the oviduct being no exception. This review discusses our current knowledge on roles of this ubiquitous glycosaminoglycan on the survival of immunologically foreign spermatozoa in the pig oviduct, a relevant event for fertility. During preovulatory storage in the functional tubal sperm reservoir, spermatozoa are entrapped in a mucus-like tubal fluid. This fluid contains fluctuating levels of hyaluronan, which is synthesized by the lining epithelium by hyaluronan synthase 3. Both hyaluronan and its CD44 receptor are particularly evident in the deep mucosal furrows of the sperm reservoir, in which most spermatozoa are embedded in; kept alive, uncapacitated but also undetected by the immune system of the female. Hyaluronan is also present in the seminal plasma, and evidence points toward an involvement of hyaluronan and its receptor in the local (tubal and possibly uterine) production of antiinflammatory cytokines, such as interleulcin-10, pertaining maternal immune tolerance of these foreign cells.   • Microscale immiscible phase magnetic processing for bioanalytical applications

    • Bioanalytical processes such as ligand-binding assays, nucleic acid amplification tests and cancer cell identification methods require multiple steps including sampling, transport, sample preparation, chemical reaction/s, detection and interpretation. These necessitate trained personnel and laboratory infrastructure, and are often time-consuming and labor-intensive. Integrated platforms capable of combining the aforementioned steps are generally challenging to realize. Approaches employing immiscible phases constrained in the microscale and paramagnetic particles have emerged as pathways to extract proteins, nucleic acids and whole cells from complex matrices via affinity purification and magnetic actuation. In recent years, further steps for detection and readout of these bioanalytes have been incorporated, providing versatile lab-on-a-chip platforms for multiple integrated bioanalytical workflows and with great potential as point-of-need devices. In this manuscript, we review and summarize the different strategies employed and lay out further avenues for development and implementation.

  • Applications of artificial intelligence, machine learning, big data and the internet of things to the COVID-19 pandemic : A scientometric review using text mining

    • The COVID-19 pandemic has wreaked havoc in every country in the world, with serious health-related, economic, and social consequences. Since its outbreak in March 2020, many researchers from different fields have joined forces to provide a wide range of solutions, and the support for this work from artificial intelligence (AI) and other emerging concepts linked to intelligent data analysis has been decisive. The enormous amount of research and the high number of publications during this period makes it difficult to obtain an overall view of the different applications of AI to the management of COVID-19 and an understanding of how research in this field has been evolving. Therefore, in this paper, we carry out a scientometric analysis of this area supported by text mining, including a review of 18,955 publications related to AI and COVID-19 from the Scopus database from March 2020 to June 2021 inclusive. For this purpose, we used VOSviewer software, which was developed by researchers at Leiden University in the Netherlands. This allowed us to examine the exponential growth in research on this issue and its distribution by country, and to highlight the clear hegemony of the United States (USA) and China in this respect. We used an automatic process to extract topics of research interest and observed that the most important current lines of research focused on patient-based solutions. We also identified the most relevant journals in terms of the COVID-19 pandemic, demonstrated the growing value of open-access publication, and highlighted the most influential authors by means of an analysis of citations and co-citations. This study provides an overview of the current status of research on the application of AI to the pandemic.

  • Monitoring internal load parameters during competitive synchronized swimming duet routines in elite athletes

    • The aim of the study is to compare the heart rate (HR) and rate of perceived exertion (RPE) responses as internal load indicators while performing duet routines during training and competition, both in the technical and free programs of synchronized swimming (SS). Participants were 10 SS Olympic medalists (age, 17.4 ± 3.0 years; height, 164.0 ± 6.1 cm; body mass, 52.0 ± 6.4 kg; training, 36.3 ± 6.2 h·wk; experience, 9.2 ± 2.6 years). They were monitored while performing the same technical duet or free duet, during a training session (T) and during an official competition (C). Heart rate was continuously monitored. Rate of perceived exertion was assessed using the Borg CR10 scale. Heart rate responses during T and C were almost identical: pre-exercise mean HR (b·min) was 130.5 ± 13.9 (T) and 133.6 ± 7.7 (C) and quickly increased yielding mean peak values of 184.8 ± 5.8 (T) and 184.8 ± 6.6 (C), with interspersed bradycardic events down to 86.6 ± 4 (T) and 86.3 ± 5 (C). Routines were perceived as "hard" to "extremely hard" by the swimmers in both conditions, and mean RPE scores (0-10+) were equally high during C (7.9 ± 1.2) and T (7.5 ± 1.2) (p = 0.223). Rate of perceived exertion inversely correlated with minimum (R = -0.545; p = 0.008) and mean HR (R = -0.452; p = 0.026) and positively correlated with HRrange (R = 0.520; p = 0.011). The internal load imposed by SS duets performed during training is virtually identical to that elicited in a real competitive situation. Therefore, practicing competitive routines is suitable for developing and maintaining the cardiovascular fitness that is needed for specific conditioning in elite synchronized swimmers, with the added value of favoring exercise automaticity, interindividual coordination, and artistic expression simultaneously.

  • Elemental stable isotope assessment of groundwater contamination: Recent developments

    • Anthropogenic contamination of water resources remains a severe environmental concern on a global scale. Radiogenic and stable isotope measurements (especially for light elements) constitute well-known tools for the identification of pollution sources and thus have considerable potential in prevention and remediation efforts. Recent instrumental and methodological advances have extended the isotope ‘toolbox’ to include a significant number of new stable isotope systems, which in turn resulted in rapid growth of studies using these novel tracers in the field of environmental forensics. Isotopic fractionation, occurring during post-release transformation of contaminants, offers an additional benefit of possibility to study the fate of pollutants in aquatic systems. This review will focus on selected relevant studies in the field and present future trends and development.

  • MRI-detected subchondral bone marrow signal alterations of the knee joint: terminology, imaging appearance, relevance and radiological differential diagnosis

    • Objective: To discuss terminology, radiological differential diagnoses and significance of magnetic resonance imaging (MRI)-detected subchondral bone marrow lesions (BMLs) of the knee joint. Methods: An overview of the published literature is presented. In addition, the radiological appearance and differential diagnosis of subchondral signal alterations of the knee joint are discussed based on expert consensus. A recommendation for terminology is provided and the relevance of these imaging findings for osteoarthritis (OA) research is emphasized. Results: A multitude of differential diagnoses of subchondral BMLs may present with a similar aspect and signal characteristics. For this reason it is crucial to clearly and specifically define the type of BML that is being assessed and to use terminology that is appropriate to the condition and the pathology. In light of the currently used terminology, supported by histology, it seems appropriate to apply the widely used term "bone marrow lesion" to the different entities of subchondral signal alterations and in addition to specifically and precisely define the analyzed type of BML. Water sensitive sequences such as fat suppressed T2-weighted, proton density-weighted, intermediate-weighted fast spin echo or short tau inversion recovery (STIR) sequences should be applied to assess non-cystic BMLs as only these sequences depict the lesions to their maximum extent. Assessment of subchondral non-cystic ill-defined BMLs on gradient echo-type sequences should be avoided as they will underestimate the size of the lesion. Differential diagnoses of CA related BMLs include traumatic bone contusions and fractures with or without disruption of the articular surface. Osteonecrosis and bone infarcts, inflammation, tumor, transient idiopathic bone marrow edema, red marrow and post-surgical alterations should also be considered. Conclusion: Different entities of subchondral BMLs that are of relevance in the context of OA research may be distinguished by specific imaging findings, patient characteristics, symptoms, and history and are discussed in this review.   • The association between meniscal damage of the posterior horns and localized posterior synovitis detected on T1-weighted contrast-enhanced MRI-The MOST study

    • Objective: Synovitis is thought to be a secondary phenomenon in the osteoarthritis (OA) process and the menisci might be triggers of localized synovitis. The aim was to assess the cross-sectional associations of posterior horn meniscal damage with perimeniscal synovitis, and with synovitis posterior to the posterior cruciate ligament (PCL) using contrast enhanced (CE) MRI. Design: The Multicenter Osteoarthritis (MOST) Study is a longitudinal observational study of subjects with or at risk for knee OA. Subjects are a subset of MOST who were examined with 1.5 T CE MRI and had semiquantitative synovitis (scored from 0 to 2 at 11 locations) and meniscal readings (scored with WORMS from 0 to 4) available. Logistic regression was used to assess the association of posterior meniscal damage and perimeniscal synovitis in the same compartment, and between posterior meniscal damage and synovitis posterior to the PCL. Results: Three hundred and seventy seven knees were included (mean age 61.1 years +/- 6.9, mean BMI 29.6 +/- 4.9, 44.3% women). The odds for ipsi-compartmental perimeniscal synovitis were increased for knees with medial posterior horn meniscal damage (adjusted odds ratio [aOR] 2.5, 95% confidence intervals [95% CI] 1.3,4.8), but not for lateral damage (aOR 1.7, 95% CI 0.4,6.6). No positive associations were found for meniscal damage and presence of synovitis posterior to the PCL (aOR 0.9, 95% CI 0.6,1.5). Conclusions: Meniscal damage of the posterior horns is associated with ipsi-compartmental perimensical synovitis. No associations were found for posterior horn meniscal damage with synovitis posterior to the PCL, which suggests that synovitis posterior to the PCL is likely to be triggered by different pathomechanisms.   • The physiology and genetics of bacterial responses to antibiotic combinations

    • Combining several antibiotics, either in mixtures or sequential order, is proposed to increase treatment efficacy and reduce resistance evolution. In this Review, Andersson and colleagues discuss the effects of antibiotic combinations, the directional effects of previous antibiotic treatments and the role of stress-response systems as well as the interactions between drugs and resistance mutations. Several promising strategies based on combining or cycling different antibiotics have been proposed to increase efficacy and counteract resistance evolution, but we still lack a deep understanding of the physiological responses and genetic mechanisms that underlie antibiotic interactions and the clinical applicability of these strategies. In antibiotic-exposed bacteria, the combined effects of physiological stress responses and emerging resistance mutations (occurring at different time scales) generate complex and often unpredictable dynamics. In this Review, we present our current understanding of bacterial cell physiology and genetics of responses to antibiotics. We emphasize recently discovered mechanisms of synergistic and antagonistic drug interactions, hysteresis in temporal interactions between antibiotics that arise from microbial physiology and interactions between antibiotics and resistance mutations that can cause collateral sensitivity or cross-resistance. We discuss possible connections between the different phenomena and indicate relevant research directions. A better and more unified understanding of drug and genetic interactions is likely to advance antibiotic therapy.

  • A global view of aspen : conservation science for widespread keystone systems

    • Across the northern hemisphere, six species of aspen (Populus spp.) play a disproportionately important role in promoting biodiversity, sequestering carbon, limiting forest disturbances, and providing other ecosystem services. These species are illustrative of efforts to move beyond single-species conservation because they facilitate hundreds of plants and animals worldwide. This review is intended to place aspen in a global conservation context by focusing on the many scientific advances taking place in such biologically diverse systems. In this manner, aspen may serve as a model for other widespread keystone systems where science-based practice may have world implications for biodiversity conservation. In many regions, aspen can maintain canopy dominance for decades to centuries as the sole major broadleaf trees in forested landscapes otherwise dominated by conifers. Aspen ecosystems are valued for many reasons, but here we highlight their potential as key contributors to regional and global biodiversity. We present global trends in research priorities, strengths, and weaknesses based on, 1) a qualitative survey, 2) a systematic literature analysis, and 3) regional syntheses of leading research topics. These regional syntheses explore important aspen uses, threats, and research priorities with the ultimate intent of research sharing focused on sound conservation practice. In all regions, we found that aspen enhance biodiversity, facilitate rapid (re)colonization in natural and damaged settings (e.g., abandoned mines), and provide adaptability in changing environments. Common threats to aspen ecosystems in many, but not all, regions include effects of herbivory, land clearing, logging practices favoring conifer species, and projected climate warming. We also highlight regional research gaps that emerged from the three survey approaches above. We believe multi-scale research is needed that examines disturbance processes in the context of dynamic climates where ecological, physiological, and genetic variability will ultimately determine widespread aspen sustainability. Based on this global review of aspen research, we argue for the advancement of the "mega-conservation" strategy, centered on the idea of sustaining a set of common keystone communities (aspen) that support wide arrays of obligate species. This approach contrasts with conventional preservation which focuses limited resources on individual species residing in narrow niches.

  • Does low-energy sweetener consumption affect energy intake and body weight? : A systematic review, including meta-analyses, of the evidence from human and animal studies

    • By reducing energy density, low-energy sweeteners (LES) might be expected to reduce energy intake (EI) and body weight (BW). To assess the totality of the evidence testing the null hypothesis that LES exposure (versus sugars or unsweetened alternatives) has no effect on EI or BW, we conducted a systematic review of relevant studies in animals and humans consuming LES with ad libitum access to food energy. In 62 of 90 animal studies exposure to LES did not affect or decreased BW. Of 28 reporting increased BW, 19 compared LES with glucose exposure using a specific 'learning' paradigm. Twelve prospective cohort studies in humans reported inconsistent associations between LES use and body mass index (-0.002 kg m(-2) per year, 95% confidence interval (CI) -0.009 to 0.005). Meta-analysis of short-term randomized controlled trials (129 comparisons) showed reduced total EI for LES versus sugar-sweetened food or beverage consumption before an ad libitum meal (-94 kcal, 95% CI -122 to -66), with no difference versus water (-2 kcal, 95% CI -30 to 26). This was consistent with EI results from sustained intervention randomized controlled trials (10 comparisons). Meta-analysis of sustained intervention randomized controlled trials (4 weeks to 40 months) showed that consumption of LES versus sugar led to relatively reduced BW (nine comparisons; -1.35 kg, 95% CI -2.28 to -0.42), and a similar relative reduction in BW versus water (three comparisons; -1.24 kg, 95% CI -2.22 to -0.26). Most animal studies did not mimic LES consumption by humans, and reverse causation may influence the results of prospective cohort studies. The preponderance of evidence from all human randomized controlled trials indicates that LES do not increase EI or BW, whether compared with caloric or non-caloric (for example, water) control conditions. Overall, the balance of evidence indicates that use of LES in place of sugar, in children and adults, leads to reduced EI and BW, and possibly also when compared with water.
