  • A protein deep sequencing evaluation of metastatic melanoma tissues.

    • Malignant melanoma has the highest increase of incidence of malignancies in the western world. In early stages, front line therapy is surgical excision of the primary tumor. Metastatic disease has very limited possibilities for cure. Recently, several protein kinase inhibitors and immune modifiers have shown promising clinical results but drug resistance in metastasized melanoma remains a major problem. The need for routine clinical biomarkers to follow disease progression and treatment efficacy is high. The aim of the present study was to build a protein sequence database in metastatic melanoma, searching for novel, relevant biomarkers. Ten lymph node metastases (South-Swedish Malignant Melanoma Biobank) were subjected to global protein expression analysis using two proteomics approaches (with/without orthogonal fractionation). Fractionation produced higher numbers of protein identifications (4284). Combining both methods, 5326 unique proteins were identified (2641 proteins overlapping). Deep mining proteomics may contribute to the discovery of novel biomarkers for metastatic melanoma, for example dividing the samples into two metastatic melanoma "genomic subtypes", ("pigmentation" and "high immune") revealed several proteins showing differential levels of expression. In conclusion, the present study provides an initial version of a metastatic melanoma protein sequence database producing a total of more than 5000 unique protein identifications. The raw data have been deposited to the ProteomeXchange with identifiers PXD001724 and PXD001725.

  • Goal Framing as a Tool for Changing People’s Car Travel Behavior in Sweden

    • This paper reports on a study of car drivers’ assessment of a sustainability policy involving increased car parking fees in Swedish city centers. The aim of the study was to investigate how framing of information in text and pictures influences acceptance of increasing car parking fees and how values, general beliefs and norms as well as measure-specific beliefs influence the acceptability of the measure. Drawing on Goal Framing Theory, the acceptance of a parking fee policy was tested using three different goal frames (hedonic, gain and normative); the frames were compared with each other and a control message. The study was based on a survey directed to residents (18 to 75 years of age) in 51 larger municipalities in Sweden. The survey had an experimental design. Respondents were presented with a scenario of an increase in parking fees to promote environmental sustainability. The scenario was presented in three ways (manipulations), highlighting hedonic (e.g., emotional), gain, and normative aspects, respectively, in text and pictures. The results showed that the three message frames had different effects and were overall more effective than the control message in engendering the desired reduction in private car use and, thus, the intended environmental impact. Further, the degree of acceptability of the increased parking fee influenced the expected behavioral change in the groups receiving a goal framed message in relation to the parking fee measure. Implications from a sustainability perspective concern the importance of how environmental policies are framed when communicated to the public in order to increase acceptance and support.

  • The importance of socio-demographic characteristics, geographic setting, and attitudes for adoption of electric vehicles in Sweden

    • Although the number of different types of EVs is increasing, they still constitute only a small share of the total vehicle market. There are a number of barriers to car owners’ adoption of an EV: travel needs, charging infrastructure, the individual car owner’s socio-economic characteristics, attitudinal factors, and environmental concern. In this study, the characteristics and geographic location of all private car owners in Sweden (N = 4,447,118) are charted. Through analysis of survey data (N = 1192), the importance of socio-demographic attributes, geographic conditions, car interest, personal and social norms, and environmental concern is estimated. Mapping EV ownership shows that, so far, EV adoption has mainly occurred in metropolitan areas and also to some extent in hotspots outside the metropolitan areas, and that these hotspots are tourist regions that may be exposed to EVs via, for example, Norwegian tourists in the Swedish case. Logistic regression analyses show that age and education level have positive impacts on EV ownership. Residential area also has an influence to some extent, pointing to a slight neighborhood effect in EV adoption. However, the most important factor influencing EV ownership is the individual’s personal norms. In addition to showcasing EV adoption patterns in Sweden, the contribution of this study is to point to the importance of the attitudinal factor of personal norm even when geographical conditions and socio-demographic characteristics are controlled for. Implications of the findings are discussed.

    • A temporal study of the bacterial utilisation of allochthonous dissolved organic carbon (DOC) was performed in a lake and estuary situated in the same boreal watershed. A greater utilisation of allochthonous DOC was observed under estuarine as compared to Limnic conditions. However, the bacterial biomass yield was not significantly different between the environments, indicating a lower growth efficiency on allochthonous DOC by the estuarine bacterioplankton. The largest fraction of the allochthonous DOC utilised (20 %) was observed under estuarine conditions and coincided with the spring flood. The utilisation of allochthonous DOC and bacterial biomass yield in the experimental bottles was Limited by the availability of nitrogen or phosphorus in both environments. Our results showed that the major bacterial transformation of DOC occurred in the estuarine rather than the Limnic environment, due to the shift in physicochemical milieu and bacterial assemblage. However, the estuarine bacteria also respired a greater fraction of the DOC utilised than their limnic counterparts.

    • An method for fabrication of long silica nano wires is presented. The nano structured material is an integral part of the inner surface of narrow bore fused silica capillary tubing. The wire preparation method is based on a hydrothermal action and decomposition of 2-chloro-1,1,2-trifluoroethyl methyl ether at elevated temperature. In our previous work, reproducible nano wire outgrowth could only be accomplished in capillaries with an inner diameter less than 10 mum [1], and the centre of the capillary lumen remained free of wires. In the present article we report on nano wire outgrowth in capillaries with larger inner diameters. These wires are entangled via carbon nucleating particles and stretch across the entire lumen of the capillary. The long nano wire outgrowth was induced by a time dependent, non isothermal etching of the capillaries. Suggested mechanisms for the growth process are discussed.

    • A method for preparation of silica nanowires with dimensions of d = 10-100 nm, 5-500 nm, is described. The nanostructured material is an integral part of the inner surface of narrow bore fused-silica capillary tubing. The wire preparation method is based on a decomposition of 2-chloro-1,1,2-trifluoroethyl methyl ether at elevated temperature and pressure. The silica bulk material is rearranged via a sustained silica-hydrogen fluoride chemistry, and reaction mechanisms for this process are proposed. The method is suitable for preparing long lengths of tubing with the modified surface. It is our belief that the texture of the capillary wall with its increased surface area is useful for applications such as microreactions, catalysis, and high-resolution pressure and/or electrodriven open-tubular liquid chromatography.

  • Large meta-analysis of genome-wide association studies identifies five loci for lean body mass

    • Lean body mass, consisting mostly of skeletal muscle, is important for healthy aging. We performed a genome-wide association study for whole body (20 cohorts of European ancestry with n = 38,292) and appendicular (arms and legs) lean body mass (n = 28,330) measured using dual energy X-ray absorptiometry or bioelectrical impedance analysis, adjusted for sex, age, height, and fat mass. Twenty-one single-nucleotide polymorphisms were significantly associated with lean body mass either genome wide (p < 5 x 10(-8)) or suggestively genome wide (p < 2.3 x 10(-6)). Replication in 63,475 (47,227 of European ancestry) individuals from 33 cohorts for whole body lean body mass and in 45,090 (42,360 of European ancestry) subjects from 25 cohorts for appendicular lean body mass was successful for five single-nucleotide polymorphisms in/ near HSD17B11, VCAN, ADAMTSL3, IRS1, and FTO for total lean body mass and for three single-nucleotide polymorphisms in/ near VCAN, ADAMTSL3, and IRS1 for appendicular lean body mass. Our findings provide new insight into the genetics of lean body mass.
