  • Detoxified synthetic bacterial membrane vesicles as a vaccine platform against bacteria and SARS-CoV-2

    • The development of vaccines based on outer membrane vesicles (OMV) that naturally bud off from bacteria is an evolving field in infectious diseases. However, the inherent inflammatory nature of OMV limits their use as human vaccines. This study employed an engineered vesicle technology to develop synthetic bacterial vesicles (SyBV) that activate the immune system without the severe immunotoxicity of OMV. SyBV were generated from bacterial membranes through treatment with detergent and ionic stress. SyBV induced less inflammatory responses in macrophages and in mice compared to natural OMV. Immunization with SyBV or OMV induced comparable antigen-specific adaptive immunity. Specifically, immunization with Pseudomonas aeruginosa-derived SyBV protected mice against bacterial challenge, and this was accompanied by significant reduction in lung cell infiltration and inflammatory cytokines. Further, immunization with Escherichia coli-derived SyBV protected mice against E. coli sepsis, comparable to OMV-immunized group. The protective activity of SyBV was driven by the stimulation of B-cell and T-cell immunity. Also, SyBV were engineered to display the SARS-CoV-2 S1 protein on their surface, and these vesicles induced specific S1 protein antibody and T-cell responses. Collectively, these results demonstrate that SyBV may be a safe and efficient vaccine platform for the prevention of bacterial and viral infections

  • Bringing the Nature Futures Framework to life : creating a set of illustrative narratives of nature futures

    • To halt further destruction of the biosphere, most people and societies around the globe need to transform their relationships with nature. The internationally agreed vision under the Convention of Biological Diversity—Living in harmony with nature—is that “By 2050, biodiversity is valued, conserved, restored and wisely used, maintaining ecosystem services, sustaining a healthy planet and delivering benefits essential for all people”. In this context, there are a variety of debates between alternative perspectives on how to achieve this vision. Yet, scenarios and models that are able to explore these debates in the context of “living in harmony with nature” have not been widely developed. To address this gap, the Nature Futures Framework has been developed to catalyse the development of new scenarios and models that embrace a plurality of perspectives on desirable futures for nature and people. In this paper, members of the IPBES task force on scenarios and models provide an example of how the Nature Futures Framework can be implemented for the development of illustrative narratives representing a diversity of desirable nature futures: information that can be used to assess and develop scenarios and models whilst acknowledging the underpinning value perspectives on nature. Here, the term illustrative reflects the multiple ways in which desired nature futures can be captured by these narratives. In addition, to explore the interdependence between narratives, and therefore their potential to be translated into scenarios and models, the six narratives developed here were assessed around three areas of the transformative change debate, specifically, (1) land sparing vs. land sharing, (2) Half Earth vs. Whole Earth conservation, and (3) green growth vs. post-growth economic development. The paper concludes with an assessment of how the Nature Futures Framework could be used to assist in developing and articulating transformative pathways towards desirable nature futures.

    • The 4th International Workshop in Vaccine Adjuvants and Parasitic Vaccines (Adjuvant 2008), hosted by the Cuban Society for Immunology, attracted approximately 70 scientists from 22 countries. The meeting goal was mainly to share recent progress and discuss future challenges regarding vaccine adjuvants for the development of mucosal vaccines, as well as antiparasitic vaccines. Five keynote addresses, 21 oral presentations and 28 posters were presented, and the meeting was ended with a 'hot-topic' session discussing future challenges. This article highlights the most important issues discussed.

  • Lubrication Properties of Bottle-Brush Polyelectrolytes : An AFM Study on the Effect of Side Chain and Charge Density

    • The effect of side chain to charge ratio on the frictional properties of adsorbed layers formed by bottle-brush polyelectrolytes with poly(ethylene oxide) side chains has been investigated. The brush polyelectrolytes were preadsorbed from 0.1 mM NaNO3 solutions onto mica and silica surfaces; the interfacial friction was then measured in polyelectrolyte-free solutions via AFM (with the silica surface acting as the colloidal probe). It was concluded that the decisive factor for achieving favorable lubrication properties is the concentration of nonadsorbing poly(ethylene oxide) side chains in the interfacial region. However, contrary to what may be expected, the results showed that an ideal brush layer structure with the adsorbed polymers adopting comb-like conformation is not necessary for achieving a low coefficient of friction in the asymmetric mica-silica system. In fact, the lowest coefficient of friction (< 0.01) under applied pressures as high as 30 MPa was observed for a system with a side chain to charge ratio of 9: 1, incapable of forming brush-like layers.

    • In this paper we present research results from the multi-disciplinary EU research project USEMP (USEMP is a project funded from EU research framework, additional information about project scope and deliverables are available at project’s public website at: http://​www.​usemp-project.​eu/​). In particular, we look at the legal aspects of personal data licensing and profile transparency, the development of a personal data value model in Online Social Networks (OSNs) and the development of disclosure scoring and personal data value frameworks. In the first part of the paper we show how personal data usage licensing and profile transparency for OSN activities provides for Data Protection by Design (DPbD). We also present an overview of the existing personal data monetization ecosystem in OSNs and its possible evolutions for increasing privacy and transparency for consumers about their OSN presence. In the last part of the paper, we describe the USEMP scoring framework for personal information disclosure and data value that can assist users to better perceive how their privacy is affected by their OSN presence and what the value of their OSN activities is.

  • A global point prevalence survey of antimicrobial use in neonatal intensive care units : The no-more-antibiotics and resistance (NO-MAS-R) study

    • Background: Global assessment of antimicrobial agents prescribed to infants in the neonatal intensive care unit (NICU) may inform antimicrobial stewardship efforts.Methods: We conducted a one-day global point prevalence study of all antimicrobials provided to NICU infants. Demographic, clinical, and microbiologic data were obtained including NICU level, census, birth weight, gestational/chronologic age, diagnoses, antimicrobial therapy (reason for use; length of therapy), antimicrobial stewardship program (ASP), and 30-day in-hospital mortality.Findings: On July 1, 2019, 26% of infants (580/2,265; range, 0-100%; median gestational age, 33 weeks; median birth weight, 1800 g) in 84 NICUs (51, high-income; 33, low-to-middle income) from 29 countries (14, high-income; 15, low-to-middle income) in five continents received >= 1 antimicrobial agent (92%, antibacterial; 19%, antifungal; 4%, antiviral). The most common reasons for antibiotic therapy were "rule-out" sepsis (32%) and "culture-negative" sepsis (16%) with ampicillin (40%), gentamicin (35%), amikacin (19%), vancomycin (15%), and meropenem (9%) used most frequently. For definitive treatment of presumed/confirmed infection, vancomycin (26%), amikacin (20%), and meropenem (16%) were the most prescribed agents. Length of therapy for culture-positive and "culture-negative" infections was 12 days (median; IQR, 8-14) and 7 days (median; IQR, 5-10), respectively. Mortality was 6% (42%, infection-related). An NICU ASP was associated with lower rate of antibiotic utilization (p = 0.02).Interpretation: Global NICU antibiotic use was frequent and prolonged regardless of culture results. NICU-specific ASPs were associated with lower antibiotic utilization rates, suggesting the need for their implementation worldwide.

  • Potential of prompt γ -ray emission studies in fast-neutron induced fission : a first step

    • Prompt γ-ray spectra emitted in fast-neutron induced fission of 239Pu have been recently measured by using the LICORNE directional neutron source at En = 1.8 MeV. The results are used in parallel with the measurements of fast-neutron induced fission of 238U and spontaneous fission of 252Cf to assess the potential of such reactions and observables, in contributing to the understanding of fission. The γ-ray spectra were measured and analyzed under similar conditions, allowing a consistent and robust comparison between the three systems. They are further compared to Monte–Carlo simulations based on two widely-used semi-empirical codes, FREYA and GEF. Differences in the low and high energy portions of the spectrum are interpreted based on simple arguments involving nuclear structure and evaporation effects. The significance and potential of experimental campaigns of this kind, as well as current limitations, are highlighted, together with straightforward but mandatory extensions.

    • Prompt fission gamma-ray spectra (PFGS) have been measured for the Pu-239(n,f) reaction using fast neutrons at E-n =1.81 MeV produced by the LICORNE directional neutron source. The setup makes use of LaBr3 scintillation detectors and PARIS phoswich detectors to measure the emitted prompt fission gamma rays (PFG). The mean multiplicity, average total energy release per fission and average energy of photons are extracted from the unfolded PFGS. These new measurements provide complementary information to other recent work on thermal neutron induced fission of Pu-239 and spontaneous fission of Cf-252.

    • Fire and smoke monitoring systems are useful in different industry such as military, social security and economical. The recent methods for fire and smoke detection are used only motion and color characteristics thus many wrong alarms are happening and this is decrease the performance of the systems. This research presents a new method for fire and smoke detection through image processing. In this algorithm all objects in an image is considered and then check them to figure out which objects are smoke and fire. The color, motion and disorder are useful characteristics in fire and smoke detection algorithm. Smoke of fire will blur the whole or part of the images. Thus by processing of the video frames, different objects will detect. Due to evaluate the features of objects, the goal objects (fire and smoke) can be defined easily. Two-dimensional wavelet analysis is used in the presented method. The results of this research present the proposed features that can reduce the wrong alarms and increase the system performances.
