  • Increasing CSF phospho-tau levels during cognitive decline and progression to dementia
  • 2008
  • Ingår i: Neurobiology of Aging. - : Elsevier BV. - 0197-4580 .- 1558-1497. ; 29:10, s. 1466-1473
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: Little is known about longitudinal changes of cerebrospinal fluid (CSF) biomarkers during cognitive decline in neurodegenerative disease progression. OBJECTIVE: To investigate longitudinal changes in CSF biomarkers--total-tau (T-tau), phospho-tau (P-tau) and beta-amyloid (Abeta42)--during cognitive decline. METHODS: Forty memory clinic patients (47.5% females), aged 61.3+/-7.6 (S.D.) years, non-demented at baseline, underwent lumbar puncture and neuropsychological testing at two occasions. Baseline mean MMSE-score was 28.3+/-1.8. Patients were divided into three groups based on baseline memory functioning; severely impaired (SIM), moderately impaired (MIM) and no impairment (NIM). RESULTS: There was a significant increase in P-tau in the SIM-group during follow-up, while P-tau in MIM and NIM did not change. Eighty-three percent of the SIM-patients converted to dementia (80% AD), while most MIM- and NIM-patients remained non-demented. T-tau- and Abeta42-levels did not change in any of the memory groups during follow-up. CONCLUSION: Increasing P-tau levels during cognitive decline and conversion to dementia suggest that P-tau may be useful as a longitudinal marker of the neurodegenerative process.

  • Exploring Perceptions of Route Environments in Relation to Walking
  • 2024
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Background: Every walk takes place in a route environment, which can play an important role in deterring or facilitating walking. The focus of this thesis is on the perceptions of environmental variables, and how they relate to appraisals of route environments as hindering – stimulating for walking and unsafe – safe for reasons of traffic, in two metropolitan environments. Another focus is to expand the state of knowledge concerning the criterion-related validity of the Active Commuting Route Environment Scale (ACRES).Methods: Commuting pedestrians in Greater Stockholm, Sweden, were recruited via advertisements. From the inner urban area there were 294 participants (77% women), aged 49.5 years, and from the suburban areas there were 233 participants (82% women), aged 50.0 years. 77 individuals walked in both areas. The participants evaluated their commuting route environments using the ACRES. Correlation, multiple regression, and mediation analyses were used to explore the relationships between the variables. Comparisons of environmental ratings between groups and settings were performed with t-tests. Studies 1 and 2 focused on the inner urban area and studies 3 and 4 on the suburban areas. Studies 1 and 3 focused exclusively on the relations between the four motorized traffic variables (vehicle speed, vehicle flow, noise, and exhaust fumes), and their relations to the outcome variables (hinders – stimulates walking and unsafe – safe traffic). Studies 2 and 4 utilised proxies from studies 1 and 3 and combined them with other environmental variables to further the understanding of route environmental variables in relation to walking. Results: In both areas, aesthetics and greenery were positively related to stimulating walking, whereas noise, a proxy for motorized traffic, was negatively related. Aesthetics was also positively related to unsafe – safe traffic in the inner urban area, whereas greenery had the corresponding role in suburbia. Another important finding was that greenery also influenced aesthetics positively in both areas. Thus, greenery had both a direct and an indirect positive effect. On the other hand, noise influenced aesthetics negatively in the inner urban area, whereas vehicle flow had the corresponding role in suburbia. A number of variables conjointly influenced the outcome unsafe – safe traffic negatively in both areas (speeds of motor vehicles, noise, conflicts, congestion: pedestrians, red lights, and course of the route). The route environment profiles differed distinctly between the two areas. Conclusions: Several route environmental variables appear to be particularly influential in relation to pedestrian commuting, e.g., aesthetics, greenery, and noise. An important finding is that both positive and negative interactions, between certain predictor variables, were disclosed. The contrasting route environment profiles in the different settings strengthen the criterion-related validity of the ACRES. The findings expands the state of knowledge concerning the relations between the environment and walking. If implemented, these findings can influence public health positively.

  • Surveillance of indeterminate pulmonary nodules detected with CT in a Swedish population-based study (SCAPIS) : Psychosocial consequences and impact on health-related quality of life - A multicentre prospective cross-sectional study
  • 2021
  • Ingår i: BMJ Open. - : BMJ Publishing Group Ltd. - 2044-6055. ; 11:9
  • Tidskriftsartikel (refereegranskat)abstract
    • Objectives: To investigate whether surveillance of pulmonary nodules detected with low-dose CT (LDCT) impacted health-related quality of life and psychosocial consequences in the Swedish population-based study, Swedish CArdioPulmonary bioImage Study (SCAPIS).Design: A prospective cross-sectional study.Settings and participants: This multicentre (five sites) observational study, which included a cohort from SCAPIS, consisted of 632 participants with indeterminate pulmonary nodules detected with LDCT. These participants continued surveillance for up to 36 months, during which lung cancer was not detected (surveillance group). Additionally, 972 participants with a negative pulmonary LDCT scan were included as a control group. Matching criteria were LDCT date (±2 weeks), gender and site.Outcome measures: All participants completed a health-related quality of life questionnaire (RAND-36) and the Consequences of Screening (COS) questionnaire, an average of 3 years after LDCT was conducted at entry into SCAPIS.Results: Participants were 51-70 years old at study commencement. Overall, the two groups did not differ in demographic or psychosocial variables, smoking habits or pulmonary medical history. Individuals from countries other than Sweden and those with low socioeconomic status were less likely to participate (p<0.001). No effects on health-related quality of life were observed via RAND-36. In COS, the surveillance group demonstrated a higher OR for anxiety about lung cancer (OR 3.96, 95% CI 2.35 to 6.66, p<0.001), experiencing a sense of dejection (OR 1.35, 95% CI 1.06 to 1.72, p=0.015) and thoughts about existential values (OR 1.30, 95% CI 1.04 to 1.60, p=0.018).Conclusions: Lung surveillance with LDCT contributed to significant experiences of sense of dejection, anxiety about lung cancer and development of thoughts about existential values among participants in the surveillance group compared with the controls. The risk of side effects should be communicated for informed decision-making about (non-)attendance in lung cancer screening.

  • Apoptotic neutrophils augment the inflammatory response to Mycobacterium tuberculosis infection in human macrophages
  • 2014
  • Ingår i: PLOS ONE. - : PLOS. - 1932-6203. ; 9:7
  • Tidskriftsartikel (refereegranskat)abstract
    • Macrophages in the lung are the primary cells being infected by Mycobacterium tuberculosis (Mtb) during the initial manifestation of tuberculosis. Since the adaptive immune response to Mtb is delayed, innate immune cells such as macrophages and neutrophils mount the early immune protection against this intracellular pathogen. Neutrophils are short-lived cells and removal of apoptotic cells by resident macrophages is a key event in the resolution of inflammation and tissue repair. Since anti-inflammatory activity is not compatible with effective immunity to intracellular pathogens, we therefore investigated how uptake of apoptotic neutrophils modulates the function of Mtb-activated human macrophages. We show that Mtb infection exerts a potent proinflammatory activation of human macrophages with enhanced gene activation and release of proinflammatory cytokines and that this response was augmented by apoptotic neutrophils. The enhanced macrophage response is linked to apoptotic neutrophil-driven activation of the NLRP3 inflammasome and subsequent IL-1β signalling. We also demonstrate that apoptotic neutrophils not only modulate the inflammatory response, but also enhance the capacity of infected macrophages to control intracellular growth of virulent Mtb. Taken together, these results suggest a novel role for apoptotic neutrophils in the modulation of the macrophage-dependent inflammatory response contributing to the early control of Mtb infection.

  • Bokkapitel (övrigt vetenskapligt/konstnärligt)abstract
    • Kungahälla was the southernmost town in medieval Norway. The site, now in Sweden, is about 2 kmwest of today's Kungälv. Kungahälla occupies a significant place in the oldest written sources, the Norse sagas, where we have a glimpse of the important role that the town played in early medieval Norwegian society. Opinions have been divided as to how far back this role can he taken, but there is no certain written information before the first half of the twelfth century. The written sources nevertheless tell us that royal influence over the town was considerable. Kungahälla is first mentioned as one of six civitates in Norway in 1130, by the English historian Ordericus Vitalis. Kungahalla was the site of the first monastery to be founded in Bohuslan, Kastellekioster, an Augustinian house in all probability founded in the 1160s under the auspices of Archbishop Eystein. In the latter half of the thirteenth century a Franciscan friary was also built, mentioned for the first time in 1272. An important structure is the fortress on Ragnhildsholmen in Nordre Älv, completed by 1257 At the end of the nineteenth century, Wilhelm Berg investigated the ruins of the RagnhiIdsholmen fortress and a few years later the Augustinian friary, as well as parts of the town. A small-scale investigation of the Augustinian friary also took place in 1942. In the 1950s there were excavations of the Franciscan friary, when parts of a churchyard withthe remains of a surrounding wall were documented. In addition, parts of yet another churchyard with its wall remains were investigated in 1958. There was no concerted picture of the archaeological history of the place until publication no. 29 of the Medieval Town Project. During the 1980s the possibility of conducting new archaeological investigations within the area ofthe old town arose once again. After trial digs in 1985, a programme was drawn up for a project, the main aim of which was to determine the character of Kungahälla in the period up to the thineenth-century expansion. It is reasonable to regard the investigations of Kungahälla as part of the broad range of urban investigations geared to the Early Middle Ages in Scandinavia. What makes the Kungahälla Project particularly interesting is, of course, the geographical location of the town, along with the problems associated with the concept of urbanization that the archaeological findings have clearly demonstrated. There are several circumstances in the early development which, in our opinion, make it very interesting to formulate a more ambiguous concept of urbanization than scholars have previously workedwith. Ordericus mentions a civitas in the mid-1130s, but what did it look like, and what functions did it have? Was it a town in whatever sense we mean by the concept of town? Or is it the case that urbanization can stand for a much broader process, or be a narrow part of a much broader process for which we should use some other term? In the book we present the results of the investigations, also including a couple of more analytical sections (chapters 9 and 10) which sum up the findings and try to put them in a broader context, while also providing a point of departure for continued discussion.

  • Endothelial dysfunction in growth hormone transgenic mice
  • 2006
  • Ingår i: Clinical Science. - 0143-5221 .- 1470-8736. ; 110:2, s. 217-25
  • Tidskriftsartikel (refereegranskat)abstract
    • Acromegaly [overproduction of GH (growth hormone)] is associated with cardiovascular disease. Transgenic mice overexpressing bGH (bovine GH) develop hypertension and hypercholesterolaemia and could be a model for cardiovascular disease in acromegaly. The aims of the present study were to investigate the effects of excess GH on vascular function and to test whether oxidative stress affects endothelial function in bGH transgenic mice. We studied the ACh (acetylcholine)-induced relaxation response in aortic and carotid rings of young (9-11 weeks) and aged (22-24 weeks) female bGH transgenic mice and littermate control mice, without and with the addition of a free radical scavenger {MnTBAP [Mn(III)tetrakis(4-benzoic acid)porphyrin chloride]}. We also measured mRNA levels of eNOS (endothelial nitric oxide synthase) and EC-SOD (extracellular superoxide dismutase). Intracellular superoxide anion production in the vascular wall was estimated using a dihydroethidium probe. Carotid arteries from bGH transgenic mice had an impaired ACh-induced relaxation response (young, 46 +/- 7% compared with 69 +/- 8%; aged, 52 +/- 5% compared with 80 +/- 3%; P < 0.05), whereas endothelial function in aorta was intact in young but impaired in aged bGH transgenic mice. Endothelial dysfunction was corrected by addition of MnTBAP in carotid arteries from young mice and in aortas from aged mice; however, MnTBAP did not correct endothelial dysfunction in carotid arteries from aged bGH transgenic mice. There was no difference in intracellular superoxide anion production between bGH transgenic mice and control mice, whereas mRNA expression of EC-SOD and eNOS was increased in aortas from young bGH transgenic mice compared with control mice (P < 0.05). We interpret these data to suggest that bGH overexpression is associated with a time- and vessel-specific deterioration in endothelial function, initially caused by increased oxidative stress and later by other alterations in vascular function.

  • Dielectric loss determination of fine residual waste electrical and electronic equipment for understanding of heat development during microwave pyrolysis
  • 2013
  • Ingår i: Journal of Analytical and Applied Pyrolysis. - : Elsevier BV. - 0165-2370. ; 103, s. 142-148
  • Tidskriftsartikel (refereegranskat)abstract
    • Waste electrical and electronic equipment (WEEE) contains rare and valuable metals which are important to recycle, but it also has high organic content. The conventional methods used today for treatment of WEEE cannot recycle 100%; they generate valuable fine residual fractions. For further processing of these residues, microwave pyrolysis has shown to be promising since it reduces the organic content and liberate enclosed metal pieces. However, to fully control the process, the dielectric properties of the materials need to be known but are difficult to determine due to the complex composition and structure of WEEE. This paper aims to describe a suitable procedure for determination of dielectric loss in six WEEE residual fine fractions and to correlate the results to heating behaviour during microwave pyrolysis. Three fractions are "dusts" (light, medium and heavy dust) and three fractions are called "particulate materials" and contain smaller particles (0-7 mm, 0-20 mm and 7-12 mm). The method chosen was Vector Network Analyzer (VNA) using waveguide WR430 equipped with polymer foam to contain the samples which consisted of 1 dl of WEEE fine fraction. The scattering parameters were measured and dielectric loss in the materials was calculated. The measurements were performed in frequency region 1.3-2.7 GHz to include the industrial frequency 2.45 GHz. Differences between dielectric losses for the materials were determined by ANOVA. It was shown that the scattering parameter measurements were rather stable for the dust materials and the losses could be determined. For the larger sized particulate materials a lot of reflection was observed during the measurements, due to fine metal wires in the material. The losses could be determined after removing these wires. The previously observed higher heating rate and shorter time for mass reduction of dusts could not be explained by differences in dielectric loss. However, since VNA measurements indicate that metal wires in the material disturb the field; presence of wires in the particulate materials might be one cause for slower heating rate and observed temperature drops during pyrolysis.
