  • Multiple sclerosis risk variants regulate gene expression in innate and adaptive immune cells

    • At least 200 single-nucleotide polymorphisms (SNPs) are associated with multiple sclerosis (MS) risk. A key function that could mediate SNP-encoded MS risk is their regulatory effects on gene expression. We performed microarrays using RNA extracted from purified immune cell types from 73 untreated MS cases and 97 healthy controls and then performed Cis expression quantitative trait loci mapping studies using additive linear models. We describe MS risk expression quantitative trait loci associations for 129 distinct genes. By extending these models to include an interaction term between genotype and phenotype, we identify MS risk SNPs with opposing effects on gene expression in cases compared with controls, namely, rs2256814 MYT1 in CD4 cells (q = 0.05) and rs12087340 RF00136 in monocyte cells (q = 0.04). The rs703842 SNP was also associated with a differential effect size on the expression of the METTL21B gene in CD8 cells of MS cases relative to controls (q = 0.03). Our study provides a detailed map of MS risk loci that function by regulating gene expression in cell types relevant to MS.

    • We exposed bulk SiC and films of SiC and B4C to single 25 fs long free-electron-laser pulses with wavelengths between 13.5 and 32 nm. The materials are candidates for x-ray free-electron laser optics. We found that the threshold for surface-damage of the bulk SiC samples exceeds the fluence required for thermal melting at all wavelengths. The damage threshold of the film sample shows a strong wavelength dependence. For wavelengths of 13.5 and 21.7 nm, the damage threshold is equal to or exceeds the melting threshold, whereas at 32 nm the damage threshold falls below the melting threshold.

    • Diagnostic criteria for complex medical conditions caused by a multitude of both genetic and environmental factors should be descriptive and avoid any attribution of causality. Furthermore, the wording used to describe a disorder should be evidence-based and avoid stigmatization of the affected individuals. Both terminology and categorizations should be readily comprehensible for healthcare professionals and guide clinical decision making. Uncertainties with respect to diagnostic issues and their implications may be addressed to direct future clinical research. In this context, the European Association of the Study of Obesity (EASO) considers it an important endeavor to review the current ICD-11 Beta Draft for the definition of overweight and obesity and to propose a substantial revision. We aim to provide an overview of the key issues that we deem relevant for the discussion of the diagnostic criteria. We first discuss the current ICD-10 criteria and those proposed in the ICD 11 Beta Draft. We conclude with our own proposal for diagnostic criteria, which we believe will improve the assessment of patients with obesity in a clinically meaningful way. (C) 2017 The Author(s) Published by S.     • Amazonia's floodplain system is the largest and most biodiverse on Earth. Although forests are crucial to the ecological integrity of floodplains, our understanding of their species composition and how this may differ from surrounding forest types is still far too limited, particularly as changing inundation regimes begin to reshape floodplain tree communities and the critical ecosystem functions they underpin. Here we address this gap by taking a spatially explicit look at Amazonia-wide patterns of tree-species turnover and ecological specialization of the region's floodplain forests. We show that the majority of Amazonian tree species can inhabit floodplains, and about a sixth of Amazonian tree diversity is ecologically specialized on floodplains. The degree of specialization in floodplain communities is driven by regional flood patterns, with the most compositionally differentiated floodplain forests located centrally within the fluvial network and contingent on the most extraordinary flood magnitudes regionally. Our results provide a spatially explicit view of ecological specialization of floodplain forest communities and expose the need for whole-basin hydrological integrity to protect the Amazon's tree diversity and its function.

  • z-GAL: A NOEMA spectroscopic redshift survey of bright Herschel galaxies: II. Dust properties

    • We present the dust properties of 125 bright Herschel galaxies selected from the z-GAL NOEMA spectroscopic redshift survey. All the galaxies have precise spectroscopic redshifts in the range 1.3 < z < 5.4. The large instantaneous bandwidth of NOEMA provides an exquisite sampling of the underlying dust continuum emission at 2 and 3 mm in the observed frame, with flux densities in at least four sidebands for each source. Together with the available Herschel 250, 350, and 500 μm and SCUBA-2 850 μm flux densities, the spectral energy distribution (SED) of each source can be analyzed from the far-infrared to the millimeter, with a fine sampling of the Rayleigh-Jeans tail. This wealth of data provides a solid basis to derive robust dust properties, in particular the dust emissivity index (β) and the dust temperature (Tdust). In order to demonstrate our ability to constrain the dust properties, we used a flux-generated mock catalog and analyzed the results under the assumption of an optically thin and optically thick modified black body emission. The robustness of the SED sampling for the z-GAL sources is highlighted by the mock analysis that showed high accuracy in estimating the continuum dust properties. These findings provided the basis for our detailed analysis of the z-GAL continuum data. We report a range of dust emissivities with β ∼1.5 -3 estimated up to high precision with relative uncertainties that vary in the range 7% 15%, and an average of 2.2 ± 0.3. We find dust temperatures varying from 20 to 50 K with an average of Tdust., ∼30 K for the optically thin case and Tdust., ∼38 K in the optically thick case. For all the sources, we estimate the dust masses and apparent infrared luminosities (based on the optically thin approach). An inverse correlation is found between Tdust and β with β Tdust-0.69, which is similar to what is seen in the local Universe. Finally, we report an increasing trend in the dust temperature as a function of redshift at a rate of 6.5 ±0.5 K/z for this 500 μm-selected sample. Based on this study, future prospects are outlined to further explore the evolution of dust temperature across cosmic time.

  • The discovery of a z = 0.7092 OH megamaser with the MIGHTEE survey

    • We present the discovery of the most distant OH megamaser (OHM) to be observed in the main lines, using data from the MeerKAT International Giga-Hertz Tiered Extragalactic Exploration (MIGHTEE) survey. At a newly measured redshift of z = 0.7092, the system has strong emission in both the 1665 MHz (L ≈ 2500 L-) and 1667 MHz (L ≈ 4.5 × 104 L-) transitions, with both narrow and broad components. We interpret the broad line as a high-velocity-dispersion component of the 1667 MHz transition, with velocity v ∼330 km s-1 with respect to the systemic velocity. The host galaxy has a stellar mass of M = 2.95 × 1010 M- and a star formation rate of SFR = 371 M- yr-1, placing it ∼1.5 dex above the main sequence for star-forming galaxies at this redshift, and can be classified as an ultraluminous infrared galaxy. Alongside the optical imaging data, which exhibit evidence for a tidal tail, this suggests that the OHM arises from a system that is currently undergoing a merger, which is stimulating star formation and providing the necessary conditions for pumping the OH molecule to saturation. The OHM is likely to be lensed, with a magnification factor of ∼2.5, and perhaps more if the maser emitting region is compact and suitably offset relative to the centroid of its host galaxy's optical light. This discovery demonstrates that spectral line mapping with the new generation of radio interferometers may provide important information on the cosmic merger history of galaxies.

  • Comparative genomics reveals mobile pathogenicity chromosomes in Fusarium.

    • Fusarium species are among the most important phytopathogenic and toxigenic fungi. To understand the molecular underpinnings of pathogenicity in the genus Fusarium, we compared the genomes of three phenotypically diverse species: Fusarium graminearum, Fusarium verticillioides and Fusarium oxysporum f. sp. lycopersici. Our analysis revealed lineage-specific (LS) genomic regions in F. oxysporum that include four entire chromosomes and account for more than one-quarter of the genome. LS regions are rich in transposons and genes with distinct evolutionary profiles but related to pathogenicity, indicative of horizontal acquisition. Experimentally, we demonstrate the transfer of two LS chromosomes between strains of F. oxysporum, converting a non-pathogenic strain into a pathogen. Transfer of LS chromosomes between otherwise genetically isolated strains explains the polyphyletic origin of host specificity and the emergence of new pathogenic lineages in F. oxysporum. These findings put the evolution of fungal pathogenicity into a new perspective.
