    • The multifunctionality of forest systems calls for appropriately complex modelling approaches to capture social and ecosystem dynamics. Using a social-ecological systems framework, we review the functionality of 31 existing agent-based models applied to managed forests. Several applications include advanced cognitive and emotional decision-making, crucial for understanding complex sustainability challenges. However, far from all demonstrate representation of key elements in a social-ecological system like direct interactions, and dynamic representations of social and ecological processes. We conclude that agent-based approaches are adequately complex for simulating both social and ecological subsystems, but highlight three main avenues for further development: i) robust methodological standards for calibration and validation of agent-based approaches; ii) modelling of agent learning, adaptive governance and feedback loops; iii) coupling to ecological models such as dynamic vegetation models or species distribution models. We round-off by providing a set of questions to support social-ecological systems modelling choices.

The workshop was initiated by researchers at the University of Waterloo with support from the Ontario Genomics Institute (OGI), Natural Sciences and Engineering Research Council of Canada (NSERC) and the University of Waterloo.

    • The 4th Ovarian Cancer Consensus Conference of the Gynecologic Cancer InterGroup was held in Vancouver, Canada, in June 2010. Representatives of 23 cooperative research groups studying gynecologic cancers gathered to establish international consensus on issues critical to the conduct of large randomized trials. Group C, 1 of the 3 discussion groups, examined recurrent ovarian cancer, and we report the consensus reached regarding 4 questions. These included the following: (1) What is the role of cytoreductive surgery for recurrent ovarian cancer? (2) How do we define distinct patient populations in need of specific therapeutic approaches? (3) Should end points for trials with recurrent disease vary from those of first-line trials? (4) Is CA-125 progression alone sufficient for entry/eligibility into clinical trials?

  • Survival following allogeneic transplant in patients with myelofibrosis

    • Allogeneic hematopoietic cell transplantation (HCT) is the only curative therapy for myelofibrosis (MF). In this large multicenter retrospective study, overall survival (OS) in MF patients treated with allogeneic HCT (551 patients) and without HCT (non-HCT) (1377 patients) was analyzed with Cox proportional hazards model. Survival analysis stratified by the Dynamic International Prognostic Scoring System (DIPSS) revealed that the first year of treatment arm assignment, due to upfront risk of transplant-related mortality (TRM), HCT was associated with inferior OS compared with non-HCT (non-HCT vs HCT: DIPSS intermediate 1 [Int-1]: hazard ratio [HR] = 0.26, P < .0001; DIPSS-Int-2 and higher: HR, 0.39, P < .0001). Similarly, in the DIPSS low-risk MF group, due to upfront TRM risk, OS was superior with non-HCT therapies compared with HCT in the first-year post treatment arm assignment (HR, 0.16, P = .006). However, after 1 year, OS was not significantly different (HR, 1.38, P = .451). Beyond 1 year of treatment arm assignment, an OS advantage with HCT therapy in Int-1 and higher DIPSS score patients was observed (non-HCT vs HCT: DIPSS-Int-1: HR, 2.64, P < .0001; DIPSS-Int-2 and higher: HR, 2.55, P < .0001). In conclusion, long-term OS advantage with HCT was observed for patients with Int-1 or higher risk MF, but at the cost of early TRM. The magnitude of OS benefit with HCT increased as DIPSS risk score increased and became apparent with longer follow-up.

    • I jordbrukslandskapet produceras en mängd ekosystemtjänster, där mat, foder och fiber kanske är de mest uppenbara. För att dessa försörjande tjänster skall kunna produceras på ett långsiktigt hållbart sätt krävs även en rad så kallade intermediära ekosystemtjänster, som kretslopp av näringsämnen, pollinering, naturlig kontroll av skadeinsekter och reglering av vattenflöden. Även om dessa ekosystemtjänster bidrar till lantbrukarnas lönsamhet, behöver det inte löna sig för den enskilda lantbrukaren att sköta dem. Därför behövs effektiva styrmedel – riktade eller generella – som ger lantbrukare tydliga incitament att gynna biologisk mångfald och ekosystemtjänster. Sådana styrmedel behöver dessutom nå lantbrukare inom områden som idag enbart nås i begränsad utsträckning. För att åstadkomma detta är inte bara ersättningssystemet viktigt, utan även samverkan som tar fasta på jordbrukarnas egna drivkrafter och lokala förutsättningar. Multi-level governance är en modell för samverkan där beslutsfattande på olika nivåer matchas med ekologiska principer för att öka miljöpolitikens effektivitet.

  • Evaluating the large-scale hydrological cycle response within the Pliocene Model Intercomparison Project Phase 2 (PlioMIP2) ensemble

    • The mid-Pliocene (∼3 Ma) is one of the most recent warm periods with high CO2 concentrations in the atmosphere and resulting high temperatures, and it is often cited as an analog for near-term future climate change. Here, we apply a moisture budget analysis to investigate the response of the large-scale hydrological cycle at low latitudes within a 13-model ensemble from the Pliocene Model Intercomparison Project Phase 2 (PlioMIP2). The results show that increased atmospheric moisture content within the mid-Pliocene ensemble (due to the thermodynamic effect) results in wetter conditions over the deep tropics, i.e., the Pacific intertropical convergence zone (ITCZ) and the Maritime Continent, and drier conditions over the subtropics. Note that the dynamic effect plays a more important role than the thermodynamic effect in regional precipitation minus evaporation (PmE) changes (i.e., northward ITCZ shift and wetter northern Indian Ocean). The thermodynamic effect is offset to some extent by a dynamic effect involving a northward shift of the Hadley circulation that dries the deep tropics and moistens the subtropics in the Northern Hemisphere (i.e., the subtropical Pacific). From the perspective of Earth's energy budget, the enhanced southward cross-equatorial atmospheric transport (0.22 PW), induced by the hemispheric asymmetries of the atmospheric energy, favors an approximately 1∘ northward shift of the ITCZ. The shift of the ITCZ reorganizes atmospheric circulation, favoring a northward shift of the Hadley circulation. In addition, the Walker circulation consistently shifts westward within PlioMIP2 models, leading to wetter conditions over the northern Indian Ocean. The PlioMIP2 ensemble highlights that an imbalance of interhemispheric atmospheric energy during the mid-Pliocene could have led to changes in the dynamic effect, offsetting the thermodynamic effect and, hence, altering mid-Pliocene hydroclimate.

    • Intensive agriculture is often bad for wildlife. Does this imply that a goal to boost wildlife on agricultural land is best met through a general reduction in intensity? We argue that such an approach may not be optimal, since cost functions for provision of wildlife on agricultural land may be non-convex, due to fixed costs associated with such provision. This implies that, even when farms are identical, it may be preferable to split them into groups of high providers and low providers. We test our hypothesis in a study of the optimal management of mown grasslands in southern Sweden, where the two products are silage and successful reproduction of ground-nesting birds, and the variable controlled by the farmer is the date of the first mowing. We show that the optimal solution is likely to involve some farmers maintaining profit-maximizing practices while other-identical-farmers delay their first mowing significantly. The superiority of such split solutions may have major implications for agricultural policy.

    • The Pliocene epoch has great potential to improve our understanding of the long-term climatic and environmental consequences of an atmospheric CO2 concentration near similar to 400 parts per million by volume. Here we present the large-scale features of Pliocene climate as simulated by a new ensemble of climate models of varying complexity and spatial resolution based on new reconstructions of boundary conditions (the Pliocene Model Intercomparison Project Phase 2; PlioMIP2). As a global annual average, modelled surface air temperatures increase by between 1.7 and 5.2 degrees C relative to the pre-industrial era with a multi-model mean value of 3.2 degrees C. Annual mean total precipitation rates increase by 7 % (range: 2 %-13 %). On average, surface air temperature (SAT) increases by 4.3 degrees C over land and 2.8 degrees C over the oceans. There is a clear pattern of polar amplification with warming polewards of 60 degrees N and 60 degrees S exceeding the global mean warming by a factor of 2.3. In the Atlantic and Pacific oceans, meridional temperature gradients are reduced, while tropical zonal gradients remain largely unchanged. There is a statistically significant relationship between a model's climate response associated with a doubling in CO2 (equilibrium climate sensitivity; ECS) and its simulated Pliocene surface temperature response. The mean ensemble Earth system response to a doubling of CO2 (including ice sheet feedbacks) is 67 % greater than ECS; this is larger than the increase of 47 % obtained from the PlioMIP1 ensemble. Proxy-derived estimates of Pliocene sea surface temperatures are used to assess model estimates of ECS and give an ECS range of 2.6-4.8 degrees C. This result is in general accord with the ECS range presented by previous Intergovernmental Panel on Climate Change (IPCC) Assessment Reports.

    • Ekosystemtjänster kan bidra till att synliggöra värden i naturen som människor är beroende av för sin överlevnad och sin välfärd. Att utveckla markanvändning samt integrera värdet av ekosystemtjänster i lantbrukares produktionsbeslut är nödvändigt för att möta en ökad efterfrågan av jordbruksprodukter samtidigt som de svenska miljömålen ska uppnås.I projektet har verktyg utvecklats tillsammans med mottagare, vilket säkrar relevans och att värdet av ekosystemtjänster kan kommuniceras till rätt nivå för beslut; lokal, regional och nationell. Värderingsmodellerna (verktygen) har anpassats till lokal nivå (gård), regional nivå (Skåne) och nationell nivå (Sverige) för att kunna stödja beslut om skötsel av jordbruksmark från gård till myndighet för effektiv produktion av ekosystemtjänster.I rapporten undersöks hur värdet på ekosystemtjänster och naturkapital kan användas för att göra väl grundade beslut är viktigt för att bidra till bättre underlag för beslut om jordbrukspolicy. Studien visar på ett konkret sätt hur begreppet ekosystemtjänster kan komma till praktisk nytta. Den här rapporten är ett resultat från ett av sju projekt inom forskningssatsningen Värdet av ekosystemtjänster och har författats av forskare vid Lunds Universitet och SLU (Sveriges Lantbruksuniversitet). Projektet finansierades av Naturvårdsverkets miljöforskningsanslag.

    • Ever since Stephen Pagets 1889 hypothesis, metastatic organotropism has remained one of cancers greatest mysteries. Here we demonstrate that exosomes from mouse and human lung-, liver-and brain-tropic tumour cells fuse preferentially with resident cells at their predicted destination, namely lung fibroblasts and epithelial cells, liver Kupffer cells and brain endothelial cells. We show that tumour-derived exosomes uptaken by organ-specific cells prepare the pre-metastatic niche. Treatment with exosomes from lung-tropic models redirected the metastasis of bone-tropic tumour cells. Exosome proteomics revealed distinct integrin expression patterns, in which the exosomal integrins alpha(6)beta(4) and alpha(6)beta(1) were associated with lung metastasis, while exosomal integrin alpha(v)beta(5) was linked to liver metastasis. Targeting the integrins alpha(6)beta(4) and alpha(v)beta(5) decreased exosome uptake, as well as lung and liver metastasis, respectively. We demonstrate that exosome integrin uptake by resident cells activates Src phosphorylation and pro-inflammatory S100 gene expression. Finally, our clinical data indicate that exosomal integrins could be used to predict organ-specific metastasis.
