    • Understanding the responses of tundra systemsto global change has global implications. Most tundraregions lack sustained environmental monitoring and oneof the only ways to document multi-decadal change is toresample historic research sites. The International PolarYear (IPY) provided a unique opportunity for such researchthrough the Back to the Future (BTF) project (IPY project#512). This article synthesizes the results from 13 paperswithin this Ambio Special Issue. Abiotic changes includeglacial recession in the Altai Mountains, Russia; increasedsnow depth and hardness, permafrost warming, andincreased growing season length in sub-arctic Sweden;drying of ponds in Greenland; increased nutrient availabilityin Alaskan tundra ponds, and warming at mostlocations studied. Biotic changes ranged from relativelyminor plant community change at two sites in Greenland tomoderate change in the Yukon, and to dramatic increasesin shrub and tree density on Herschel Island, and in subarcticSweden. The population of geese tripled at one sitein northeast Greenland where biomass in non-grazed plotsdoubled. A model parameterized using results from a BTFstudy forecasts substantial declines in all snowbeds andincreases in shrub tundra on Niwot Ridge, Colorado overthe next century. In general, results support and provideimproved capacities for validating experimental manipulation,remote sensing, and modeling studies.

  • The XXL Survey: XLIII. The quasar radio loudness dichotomy exposed via radio luminosity functions obtained by combining results from COSMOS and XXL-S X-ray selected quasars

    • We studied a sample of 274 radio and X-ray selected quasars (XQSOs) detected in the COSMOS and XXL-S radio surveys at 3 GHz and 2.1 GHz, respectively. This sample was identified by adopting a conservative threshold in X-ray luminosity, LX [2-10 keV] ≥ 1044 erg s-1, selecting only the most powerful quasars. A number of previous studies on the origin of radio emission in type-1 quasars have focused on the radio loudness distributions, some claiming to have found evidence for bimodality, pointing toward the existence of two physically different mechanisms for the radio emission. Using available multiwavelength data, we examined various criteria for the selection of radio-loud (RL) and radio-quiet (RQ) XQSOs and found that the number of RL/RQ XQSOs changes significantly depending on the chosen criterion. This discrepancy arises due to the different criteria tracing different physical processes and due to the fact that our sample was selected from flux-limited radio and X-ray surveys. Another approach to study the origin of radio emission in XQSOs is via their radio luminosity functions (RLF). We constructed the XQSO 1.4 GHz RLFs in six redshift bins at 0:5 ≤ z ≤ 3:75. The lower-1.4 GHz luminosity end shows a higher normalization than expected only from AGN contribution in all studied redshift bins. We found that the so-called "bump"is mostly dominated by emission due to star-forming processes within the host galaxies of XQSOs. As expected, AGN-related radio emission is the dominant contribution at the higher-luminosity end of RLF. To study the evolution of the XQSO RLF, we used a combination of analytic forms from the literature to constrain the "bump"due to star formation and the higher-luminosity AGN part of the RLF. We defined two 1.4 GHz luminosity thresholds, Lth;SF and Lth;AGN, below and above which more than 80% of sources contributing to the RLF are dominated by star formation and AGN-related activity, respectively. The two thresholds evolve with redshift, which is most likely driven by the strong evolution of star formation rates of the XQSO host galaxies. We found that both the lower and higher luminosity ends evolve significantly in density, while their luminosity evolution parameters are consistent with being constant. We found that the lower-luminosity end evolves both in density and luminosity, while the higher-luminosity end evolves significantly only in density. Our results expose the dichotomy of the origin of radio emission: while the higher-luminosity end of the XQSO RLF is dominated by AGN activity, the lower-luminosity end is dominated by the star formation-related processes.

  • Cartography of opportunistic pathogens and antibiotic resistance genes in a tertiary hospital environment

    • Although disinfection is key to infection control, the colonization patterns and resistomes of hospital-environment microbes remain underexplored. We report the first extensive genomic characterization of microbiomes, pathogens and antibiotic resistance cassettes in a tertiary-care hospital, from repeated sampling (up to 1.5 years apart) of 179 sites associated with 45 beds. Deep shotgun metagenomics unveiled distinct ecological niches of microbes and antibiotic resistance genes characterized by biofilm-forming and human-microbiome-influenced environments with corresponding patterns of spatiotemporal divergence. Quasi-metagenomics with nanopore sequencing provided thousands of high-contiguity genomes, phage and plasmid sequences (>60% novel), enabling characterization of resistome and mobilome diversity and dynamic architectures in hospital environments. Phylogenetics identified multidrug-resistant strains as being widely distributed and stably colonizing across sites. Comparisons with clinical isolates indicated that such microbes can persist in hospitals for extended periods (>8 years), to opportunistically infect patients. These findings highlight the importance of characterizing antibiotic resistance reservoirs in hospitals and establish the feasibility of systematic surveys to target resources for preventing infections. Spatiotemporal characterization of microbial diversity and antibiotic resistance in a tertiary-care hospital reveals broad distribution and persistence of antibiotic-resistant organisms that could cause opportunistic infections in a healthcare setting.

  • Low-energy Coulomb excitation of Sr 96,98 beams

    • The structure of neutron-rich Sr96,98 nuclei was investigated by low-energy safe Coulomb excitation of radioactive beams at the REX-ISOLDE facility, CERN, with the MINIBALL spectrometer. A rich set of transitional and diagonal E2 matrix elements, including those for non-yrast structures, has been extracted from the differential Coulomb-excitation cross sections. The results support the scenario of a shape transition at N=60, giving rise to the coexistence of a highly deformed prolate and a spherical configuration in Sr98, and are compared to predictions from several theoretical calculations. The experimental data suggest a significant contribution of the triaxal degree of freedom in the ground state of both isotopes. In addition, experimental information on low-lying states in Rb98 has been obtained.

    • Neutron-rich ^{96,98}Sr isotopes have been investigated by safe Coulomb excitation of radioactive beams at the REX-ISOLDE facility. Reduced transition probabilities and spectroscopic quadrupole moments have been extracted from the differential Coulomb excitation cross sections. These results allow, for the first time, the drawing of definite conclusions about the shape coexistence of highly deformed prolate and spherical configurations. In particular, a very small mixing between the coexisting states is observed, contrary to other mass regions where strong mixing is present. Experimental results have been compared to beyond-mean-field calculations using the Gogny D1S interaction in a five-dimensional collective Hamiltonian formalism, which reproduce the shape change at N=60.

  • Safety and efficacy of RNAi therapy for transthyretin amyloidosis

    • BACKGROUND: Transthyretin amyloidosis is caused by the deposition of hepatocyte-derived transthyretin amyloid in peripheral nerves and the heart. A therapeutic approach mediated by RNA interference (RNAi) could reduce the production of transthyretin. METHODS: We identified a potent antitransthyretin small interfering RNA, which was encapsulated in two distinct first- and second-generation formulations of lipid nanoparticles, generating ALN-TTR01 and ALN-TTR02, respectively. Each formulation was studied in a single-dose, placebo-controlled phase 1 trial to assess safety and effect on transthyretin levels. We first evaluated ALN-TTR01 (at doses of 0.01 to 1.0 mg per kilogram of body weight) in 32 patients with transthyretin amyloidosis and then evaluated ALN-TTR02 (at doses of 0.01 to 0.5 mg per kilogram) in 17 healthy volunteers. RESULTS: Rapid, dose-dependent, and durable lowering of transthyretin levels was observed in the two trials. At a dose of 1.0 mg per kilogram, ALN-TTR01 suppressed transthyretin, with a mean reduction at day 7 of 38%, as compared with placebo (P=0.01); levels of mutant and nonmutant forms of transthyretin were lowered to a similar extent. For ALN-TTR02, the mean reductions in transthyretin levels at doses of 0.15 to 0.3 mg per kilogram ranged from 82.3 to 86.8%, with reductions of 56.6 to 67.1% at 28 days (P<0.001 for all comparisons). These reductions were shown to be RNAi-mediated. Mild-to-moderate infusion-related reactions occurred in 20.8% and 7.7% of participants receiving ALN-TTR01 and ALN-TTR02, respectively. CONCLUSIONS: ALN-TTR01 and ALN-TTR02 suppressed the production of both mutant and nonmutant forms of transthyretin, establishing proof of concept for RNAi therapy targeting messenger RNA transcribed from a disease-causing gene.     • Frontotemporal dementia (FTD) is typically associated with changes in behaviour, language and movement. However, recent studies have shown that patients can also develop an abnormal response to pain, either heightened or diminished. We aimed to investigate this symptom in mutation carriers within the Genetic FTD Initiative (GENFI).MethodsAbnormal responsiveness to pain was measured in 462 GENFI participants: 281 mutation carriers and 181 mutation-negative controls. Changes in responsiveness to pain were scored as absent (0), questionable or very mild (0.5), mild (1), moderate (2) or severe (3). Mutation carriers were classified into C9orf72 (104), GRN (128) and MAPT (49) groups, and into presymptomatic and symptomatic stages. An ordinal logistic regression model was used to compare groups, adjusting for age and sex. Voxel-based morphometry was performed to identify neuroanatomical correlates of abnormal pain perception.ResultsAltered responsiveness to pain was present to a significantly greater extent in symptomatic C9orf72 expansion carriers than in controls: mean score 0.40 (SD 0.71) vs 0.00 (0.04), reported in 29% vs 1%. No significant differences were seen between the other symptomatic groups and controls, or any of the presymptomatic mutation carriers and controls. Neural correlates of altered pain perception in C9orf72 expansion carriers were the bilateral thalamus and striatum as well as a predominantly right-sided network of regions involving the orbitofrontal cortex, inferomedial temporal lobe and cerebellum.ConclusionChanges in pain perception are a feature of C9orf72 expansion carriers, likely representing a disruption in somatosensory, homeostatic and semantic processing, underpinned by atrophy in a thalamo-cortico-striatal network.

  • z-GAL: A NOEMA spectroscopic redshift survey of bright Herschel galaxies: I. Overview

    • Using the IRAM NOrthern Extended Millimetre Array (NOEMA), we conducted a Large Programme (z-GAL) to measure redshifts for 126 bright galaxies detected in the Herschel Astrophysical Large Area Survey (H-ATLAS), the HerMES Large Mode Survey (HeLMS), and the Herschel Stripe 82 (HerS) Survey. We report reliable spectroscopic redshifts for a total of 124 of the Herschel-selected galaxies. The redshifts are estimated from scans of the 3 and 2-mm bands (and, for one source, the 1-mm band), covering up to 31 GHz in each band, and are based on the detection of at least two emission lines. Together with the Pilot Programme, where 11 sources had their spectroscopic redshifts measured, our survey has derived precise redshifts for 135 bright Herschel-selected galaxies, making it the largest sample of high-z galaxies with robust redshifts to date. Most emission lines detected are from 12CO (mainly from J = 2 1 to 5 4), with some sources seen in [CI] and H2O emission lines. The spectroscopic redshifts are in the range 0.8

  • A global metagenomic map of urban microbiomes and antimicrobial resistance

    • We present a global atlas of 4,728 metagenomic samples from mass-transit systems in 60 cities over 3 years, representing the first systematic, worldwide catalog of the urban microbial ecosystem. This atlas provides an annotated, geospatial profile of microbial strains, functional characteristics, antimicrobial resistance (AMR) markers, and genetic elements, including 10,928 viruses, 1,302 bacteria, 2 archaea, and 838,532 CRISPR arrays not found in reference databases. We identified 4,246 known species of urban microorganisms and a consistent set of 31 species found in 97% of samples that were distinct from human commensal organisms. Profiles of AMR genes varied widely in type and density across cities. Cities showed distinct microbial taxonomic signatures that were driven by climate and geographic differences. These results constitute a high-resolution global metagenomic atlas that enables discovery of organisms and genes, highlights potential public health and forensic applications, and provides a culture-independent view of AMR burden in cities.
