    • GATA-binding protein 3 (GATA3) is a transcription factor that is crucial to mammary gland morphogenesis and differentiation of progenitor cells, and has been suggested to have a tumor suppressor function. The rs570613 single nucleotide polymorphism (SNP) in intron 4 of GATA3 was previously found to be associated with a reduction in breast cancer risk in the Cancer Genetic Markers of Susceptibility project and in pooled analysis of two case-control studies from Norway and Poland (P trend = 0.004), with some evidence for a stronger association with estrogen receptor (ER) negative tumours [Garcia-Closas M et al. (2007) Cancer Epidemiol Biomarkers Prev 16:2269-2275]. We genotyped GATA3 rs570613 in 6,388 cases and 4,995 controls from the Breast Cancer Association Consortium (BCAC) and 5,617 BRCA1 and BRCA2 carriers from the Consortium of Investigators of Modifiers of BRCA1/2 (CIMBA). We found no association between this SNP and breast cancer risk in BCAC cases overall (ORper-allele = 1.00, 95% CI 0.94-1.05), in ER negative BCAC cases (ORper-allele = 1.02, 95% CI 0.91-1.13), in BRCA1 mutation carriers RRper-allele = 0.99, 95% CI 0.90-1.09) or BRCA2 mutation carriers (RRper-allele = 0.93, 95% CI 0.80-1.07). We conclude that there is no evidence that either GATA3 rs570613, or any variant in strong linkage disequilibrium with it, is associated with breast cancer risk in women.

  • Genome-wide meta-analysis of 241,258 adults accounting for smoking behaviour identifies novel loci for obesity traits

    • Few genome-wide association studies (GWAS) account for environmental exposures, like smoking, potentially impacting the overall trait variance when investigating the genetic contribution to obesity-related traits. Here, we use GWAS data from 51,080 current smokers and 190,178 nonsmokers (87% European descent) to identify loci influencing BMI and central adiposity, measured as waist circumference and waist-to-hip ratio both adjusted for BMI. We identify 23 novel genetic loci, and 9 loci with convincing evidence of gene-smoking interaction (GxSMK) on obesity-related traits. We show consistent direction of effect for all identified loci and significance for 18 novel and for 5 interaction loci in an independent study sample. These loci highlight novel biological functions, including response to oxidative stress, addictive behaviour, and regulatory functions emphasizing the importance of accounting for environment in genetic analyses. Our results suggest that tobacco smoking may alter the genetic susceptibility to overall adiposity and body fat distribution.

  • Sleep oscillation-specific associations with Alzheimer's disease CSF biomarkers: novel roles for sleep spindles and tau

    • BackgroundBased on associations between sleep spindles, cognition, and sleep-dependent memory processing, here we evaluated potential relationships between levels of CSF A(42), P-tau, and T-tau with sleep spindle density and other biophysical properties of sleep spindles in a sample of cognitively normal elderly individuals.MethodsOne-night in-lab nocturnal polysomnography (NPSG) and morning to early afternoon CSF collection were performed to measure CSF A(42), P-tau and T-tau. Seven days of actigraphy were collected to assess habitual total sleep time.ResultsSpindle density during NREM stage 2 (N2) sleep was negatively correlated with CSF A(42), P-tau and T-tau. From the three, CSF T-tau was the most significantly associated with spindle density, after adjusting for age, sex and ApoE4. Spindle duration, count and fast spindle density were also negatively correlated with T-tau levels. Sleep duration and other measures of sleep quality were not correlated with spindle characteristics and did not modify the associations between sleep spindle characteristics and the CSF biomarkers of AD.ConclusionsReduced spindles during N2 sleep may represent an early dysfunction related to tau, possibly reflecting axonal damage or altered neuronal tau secretion, rendering it a potentially novel biomarker for early neuronal dysfunction. Given their putative role in memory consolidation and neuroplasticity, sleep spindles may represent a mechanism by which tau impairs memory consolidation, as well as a possible target for therapeutic interventions in cognitive decline.

  • Electronic structure, optical properties, and photocatalytic activities of LaFeO 3-NaTaO 3 solid solution

    • A solid solution photocatalyst, Na 1-xLa xFe 1-xTa xO 3 (x up to 0.06), was prepared by the conventional solid-state method. The photophysical properties of the samples were studied by various experimental techniques and the electronic structures were investigated by using screened hybrid density functional (HSE06) calculations. The solid solution photocatalyst showed absorption of visible light extending up to 450 nm. Upon loading of platinum nanoparticles cocatalyst, the photocatalytic hydrogen evolution of 0.81 μ·mol·h -1·g -1 was obtained for 2% doping of LaFeO 3 in NaTaO 3, under visible radiation (λ > 390 nm; 20% methanol solution). The photocatalytic properties of the solid solution were found to be better than Fe doped NaTaO 3 compounds on account of the suitable band structure. The electronic structure analysis revealed that, in the case of Fe doping at the Ta site, unoccupied electronic states in between the band gap appear that are responsible for the visible-light absorption. However, in the case of La and Fe codoping (passivated doping) the mid-gap electronic states are completely filled, which makes the band structure suitable for the visible-light photocatalysis. The present solid solution of perovskites (LaFeO 3 and NaTaO 3) sheds light on the interesting photophysical properties and photocatalytic activities which could be beneficial for the photocatalysts derived from these compounds.

  • Core-collapse supernova subtypes in luminous infrared galaxies

    • The fraction of core-collapse supernovae (CCSNe) occurring in the central regions of galaxies is not well constrained at present. This is partly because large-scale transient surveys operate at optical wavelengths, making it challenging to detect transient sources that occur in regions susceptible to high extinction factors. Here we present the discovery and follow-up observations of two CCSNe that occurred in the luminous infrared galaxy (LIRG) NGC 3256. The first, SN 2018ec, was discovered using the ESO HAWK-I /GRAAL adaptive optics seeing enhancer, and was classified as a Type Ic with a host galaxy extinction of AV = 2:1+0:3 0:1 mag. The second, AT 2018cux, was discovered during the course of follow-up observations of SN 2018ec, and is consistent with a subluminous Type IIP classification with an AV = 2:1 +/- 0:4 mag of host extinction. A third CCSN, PSN J10275082 4354034 in NGC 3256, was previously reported in 2014, and we recovered the source in late-time archival Hubble Space Telescope imaging. Based on template light curve fitting, we favour a Type IIn classification for it with modest host galaxy extinction of AV = 0:3+0:4 0:3 mag. We also extend our study with follow-up data of the recent Type IIb SN 2019lqo and Type Ib SN 2020fkb that occurred in the LIRG system Arp 299 with host extinctions of AV = 2:1 +0:1 0 :3 and AV = 0:4 +0:1 0 :2 mag, respectively. Motivated by the above, we inspected, for the first time, a sample of 29 CCSNe located within a projected distance of 2.5 kpc from the host galaxy nuclei in a sample of 16 LIRGs. We find, if star formation within these galaxies is modelled assuming a global starburst episode and normal IMF, that there is evidence of a correlation between the starburst age and the CCSN subtype. We infer that the two subgroups of 14 H-poor (Type IIb /Ib /Ic /Ibn) and 15 H-rich (Type II /IIn) CCSNe have di fferent underlying progenitor age distributions, with the H-poor progenitors being younger at 3 sigma significance. However, we note that the currently available sample sizes of CCSNe and host LIRGs are small, and the statistical comparisons between subgroups do not take into account possible systematic or model errors related to the estimated starburst ages.

    • Background Healthy life expectancy (HALE) and disability-adjusted life-years (DALYs) provide summary measures of health across geographies and time that can inform assessments of epidemiological patterns and health system performance, help to prioritise investments in research and development, and monitor progress toward the Sustainable Development Goals (SDGs). We aimed to provide updated HALE and DALYs for geographies worldwide and evaluate how disease burden changes with development. Methods We used results from the Global Burden of Diseases, Injuries, and Risk Factors Study 2015 (GBD 2015) for all-cause mortality, cause-specific mortality, and non-fatal disease burden to derive HALE and DALYs by sex for 195 countries and territories from 1990 to 2015. We calculated DALYs by summing years of life lost (YLLs) and years of life lived with disability (YLDs) for each geography, age group, sex, and year. We estimated HALE using the Sullivan method, which draws from age-specific death rates and YLDs per capita. We then assessed how observed levels of DALYs and HALE differed from expected trends calculated with the Socio-demographic Index (SDI), a composite indicator constructed from measures of income per capita, average years of schooling, and total fertility rate. Findings Total global DALYs remained largely unchanged from 1990 to 2015, with decreases in communicable, neonatal, maternal, and nutritional (Group 1) disease DALYs off set by increased DALYs due to non-communicable diseases (NCDs). Much of this epidemiological transition was caused by changes in population growth and ageing, but it was accelerated by widespread improvements in SDI that also correlated strongly with the increasing importance of NCDs. Both total DALYs and age-standardised DALY rates due to most Group 1 causes significantly decreased by 2015, and although total burden climbed for the majority of NCDs, age-standardised DALY rates due to NCDs declined. Nonetheless, age-standardised DALY rates due to several high-burden NCDs (including osteoarthritis, drug use disorders, depression, diabetes, congenital birth defects, and skin, oral, and sense organ diseases) either increased or remained unchanged, leading to increases in their relative ranking in many geographies. From 2005 to 2015, HALE at birth increased by an average of 2.9 years (95% uncertainty interval 2.9-3.0) for men and 3.5 years (3.4-3.7) for women, while HALE at age 65 years improved by 0.85 years (0.78-0.92) and 1.2 years (1.1-1.3), respectively. Rising SDI was associated with consistently higher HALE and a somewhat smaller proportion of life spent with functional health loss; however, rising SDI was related to increases in total disability. Many countries and territories in central America and eastern sub-Saharan Africa had increasingly lower rates of disease burden than expected given their SDI. At the same time, a subset of geographies recorded a growing gap between observed and expected levels of DALYs, a trend driven mainly by rising burden due to war, interpersonal violence, and various NCDs. Interpretation Health is improving globally, but this means more populations are spending more time with functional health loss, an absolute expansion of morbidity. The proportion of life spent in ill health decreases somewhat with increasing SDI, a relative compression of morbidity, which supports continued efforts to elevate personal income, improve education, and limit fertility. Our analysis of DALYs and HALE and their relationship to SDI represents a robust framework on which to benchmark geography-specific health performance and SDG progress. Country-specific drivers of disease burden, particularly for causes with higher-than-expected DALYs, should inform financial and research investments, prevention efforts, health policies, and health system improvement initiatives for all countries along the development continuum.

    • Meshless methods are a promising new field in computational electromagnetics. Instead of relying on an explicit mesh topology, a numerical solution is computed on an unstructured set of collocation nodes. This allows to model fine geometrical details with high accuracy and facilitates the adaptation of node distributions for optimization or refinement purposes. The radial point interpolation method (RPIM) is a meshless method based on radial basis functions. In this paper, the current state of the RPIM in electromagnetics is reviewed. The localized RPIM scheme is summarized, and the interpolation accuracy is discussed in dependence of important parameters. A time-domain implementation is presented, and important time iteration aspects are reviewed. New formulations for perfectly matched layers and waveguide ports are introduced. An unconditionally stable RPIM scheme is summarized, and its advantages for hybridization with the classical RPIM scheme are discussed in a practical example. The capabilities of an adaptive time-domain refinement strategy based on the experiences on a frequency-domain solver are discussed.

  • Discharge Estimates for Ungauged Rivers Flowing over Complex High-Mountainous Regions based Solely on Remote Sensing-Derived Datasets

    • Reliable information about river discharge plays a key role in sustainably managing water resources and better understanding of hydrological systems. Therefore, river discharge estimation using remote sensing techniques is an ongoing research goal, especially in small, headwater catchments which are mostly ungauged due to environmental or financial limitations. Here, a novel method for river discharge estimation based entirely on remote sensing-derived parameters is presented. The model inputs include average river width, estimated from Landsat imagery by using the modified normalized difference water index (MNDWI) approach; average depth and velocity, based on empirical equations with inputs from remote sensing; channel slope from a high resolution shuttle radar topography mission digital elevation model (SRTM DEM); and channel roughness coefficient via further analysis and classification of Landsat images with support of previously published values. The discharge of the Lhasa River was then estimated based on these derived parameters and by using either the Manning equation (Model 1) or Bjerklie equation (Model 2). In general, both of the two models tend to overestimate discharge at moderate and high flows, and underestimate discharge at low flows. The overall performances of both models at the Lhasa gauge were satisfactory: comparisons with the observations yielded Nash–Sutcliffe efficiency coefficient (NSE) and R2 values ≥ 0.886. Both models also performed well at the upper gauge (Tanggya) of the Lhasa River (NSE ≥ 0.950) indicating the transferability of the methodology to river cross-sections with different morphologies, thus demonstrating the potential to quantify streamflow entirely from remote sensing data in poorly-gauged or ungauged rivers on the Tibetan Plateau.
