  • Ancient human genomes suggest three ancestral populations for present-day Europeans

    • We sequenced the genomes of a similar to 7,000-year-old farmer from Germany and eight similar to 8,000-year-old hunter-gatherers from Luxembourg and Sweden. We analysed these and other ancient genomes(1-4) with 2,345 contemporary humans to show that most present-day Europeans derive from at least three highly differentiated populations: west European hunter-gatherers, who contributed ancestry to all Europeans but not to Near Easterners; ancient north Eurasians related to Upper Palaeolithic Siberians(3), who contributed to both Europeans and Near Easterners; and early European farmers, who were mainly of Near Eastern origin but also harboured west European hunter-gatherer related ancestry. We model these populations' deep relationships and show that early European farmers had similar to 44% ancestry from a 'basal Eurasian' population that split before the diversification of other non-African lineages.

  • Altered C-tactile processing in human dynamic tactile allodynia

    • Human unmyelinated (C) tactile afferents signal the pleasantness of gentle skin stroking on hairy (nonglabrous) skin. After neuronal injury, that same type of touch can elicit unpleasant sensations: tactile allodynia. The prevailing pathophysiological explanation is a spinal cord sensitization, triggered by nerve injury, which enables Aβ afferents to access pain pathways. However, a recent mouse knockout study demonstrates that C-tactile afferents are necessary for allodynia to develop, suggesting a role for not only Aβ but also C-tactile afferent signaling. To examine the contribution of C-tactile afferents to the allodynic condition in humans, we applied the heat/capsaicin model of tactile allodynia in 43 healthy subjects and in 2 sensory neuronopathy patients lacking Aβ afferents. Healthy subjects reported tactile-evoked pain, whereas the patients did not. Instead, patients reported their C-touch percept (faint sensation of pleasant touch) to be significantly weaker in the allodynic zone compared to untreated skin. Functional magnetic resonance imaging in 18 healthy subjects and in 1 scanned patient indicated that stroking in the allodynic and control zones evoked different responses in the primary cortical receiving area for thin fiber signaling, the posterior insular cortex. In addition, reduced activation in the medial prefrontal cortices, key areas for C-tactile hedonic processing, was identified. These findings suggest that dynamic tactile allodynia is associated with reduced C-tactile mediated hedonic touch processing. Nevertheless, because the patients did not develop allodynic pain, this seems dependent on Aβ signaling, at least under these experimental conditions.

  • Testing source-sink theory : the spill-over of mussel recruits beyond marine protected areas

    • Source-sink theory has contributed to our understanding of the function of protected areas, particularly due to their role as population sources. Marine reserves are a preferred management tool for the conservation of natural populations, creating areas of good quality habitat and thus improving population connectivity by enhancing larval supply and recruitment among shores. Despite recent advances in the study of protected areas in the context of the source-sink theory, rigorous and empirical testing of marine reserves as metapopulation sources for the adjacent areas remain largely unexplored. We investigated the role of marine reserves as population sources, whether there was spill-over beyond the reserve boundaries and if so, whether spill-over was directional. We measured percentage cover and recruitment of mussels (Perna perna) at two reserves and two comparably sized exploited control areas on the south-east coast of South Africa where unprotected populations are severely affected by artisanal exploitation. Adult abundances were enhanced within reserves, but decreased towards their edges. We predicted that recruitment would mirror adult abundances and show directionality, with northern shores having greater recruitment following the prevalent northward flow of near-shore currents. There were, however, no correlations between adult abundances and recruitment for any months or shores, and no clear spatial patterns in recruitment (i.e. similar patterns occurred at reserves and controls). The results emphasise that, while reserves may act as important refuges by protecting adult abundances, their influence on promoting recovery of near-by exploited shores through larval spill-over may be overestimated.

  • Methodological aspects of environmental assessment of buildings

    • The built environment contributes extensively to the overall environmental impact of society. An increasing number of tools have been developed worldwide for comprehensive environmental assessment and rating of buildings in order to make the building sector more sustainable. These tools are expected to drive and facilitate future environmental improvements and market transformation in the sector. This thesis explores different methodological aspects in tool development using experiences from two large Swedish projects, the EcoEffect and ByggaBo tools, which were developed with a high level of stakeholder participation in order to be of practical use in the building sector.   The methodological aspects explored and discussed here include an approach for systematic selection of assessment aspects (energy use, indoor air quality, etc.) in tools (Paper 3), and a systematic procedure for selecting practical indicators using theoretical (e.g. validity/environmental relevance) and practical (e.g. costs) criteria (Papers 2 and 3). An approach for simple communication of complex results is presented with examples from 26 multi-family buildings (Paper 4). This approach allows a building’s ‘environmental efficiency’ to be presented in one diagram, without weighting the two distinct assessment areas energy use and indoor environmental quality. Paper 5 discusses the contextual issue of internal use of environmental indicators in property management organisations through reviews of environmental performance evaluation and organisation theory literature and comparisons with actual case studies. The EcoEffect (Paper 1) and the ByggaBo tools are also compared and summarised.   The case studies of real buildings and experiences from the EcoEffect and ByggaBo projects allowed data collection, calculation procedures and different practical applications of such tools to be evaluated. Poor data availability sometimes limits assessments, and improved internal routines and database developments in the building sector would allow more reliable environmental assessments.   Reviews of numerous indicators in Paper 3 (and 2) and literature revealed that environmental relevance was not a key aspect when current environmental performance indicators and building rating tools were constructed. This thesis suggests that environmental relevance and systematic procedures be prioritised in order to provide robust and trustworthy tools for environmental assessment of buildings.  Recommendations, some of which are generally applicable to other environmental assessments, include selection of environmentally relevant indicators, systematic procedures for handling theoretical and practical considerations in tool development, aggregation and weighting methods, use of a life cycle perspective and inclusion of performance-based rather than feature-based indicators. Since it is likely that the information these tools provide will increasingly be used by authorities, building users, economic incentive providers such as banks, etc., the methodological developments suggested here to strengthen tool rigour are important for future tool development processes.

    • Disorder broadening of core-level binding energies is a general effect observed in random alloys, and identifies an opportunity for studying specific local environments experimentally. Here we study it in an archetypical system: face-centered-cubic Cu50Au50. While the disorder broadening is clearly detectable at Au, at Cu it is below the detection limit. We supplement experiments by a theoretical study where we model the alloy by a large supercell constructed as a special quasirandom structure and calculate binding-energy shifts at all sites in the supercell. Theory shows that the suppression of the disorder broadening at Cu results from a delicate balance between the influence of local chemical environment and inhomogeneous lattice distortions on the site-resolved core-level shifts. Surprisingly, even larger relaxation-induced shifts are observed at Au sites.
