    • Prostate cancer (PCa) is the most common malignancy affecting men in the western world. Although radical prostatectomy and radiation therapy can successfully treat PCa in the majority of patients, up to ~30% will experience local recurrence or metastatic disease. Prostate carcinogenesis and progression is typically an androgen-dependent process. For this reason, therapies for recurrent PCa target androgen biosynthesis and androgen receptor function. Such androgen deprivation therapies (ADT) are effective initially, but the duration of response is typically ≤24 months. Although ADT and taxane-based chemotherapy have delivered survival benefits, metastatic PCa remains incurable. Therefore, it is essential to establish the cellular and molecular mechanisms that enable localized PCas to invade and disseminate. It has long been accepted that metastases require angiogenesis. In the present review, we examine the essential role for angiogenesis in PCa metastases, and we focus in particular on the current understanding of the regulation of vascular endothelial growth factor (VEGF) in localized and metastatic PCa. We highlight recent advances in understanding the role of VEGF in regulating the interaction of cancer cells with tumor-associated immune cells during the metastatic process of PCa. We summarize the established mechanisms of transcriptional and post-transcriptional regulation of VEGF in PCa cells and outline the molecular insights obtained from preclinical animal models of PCa. Finally, we summarize the current state of anti-angiogenesis therapies for PCa and consider how existing therapies impact VEGF signaling.

  • The first Brazilian Field Lab fully dedicated to CO2 MMV experiments : from the start-up to the initial results

    • Currently one of the main challenges in CO2 storage research is the development, testing and validation of accurate and efficient Measuring, Monitoring and Verification (MMV) techniques to be deployed at geological sequestration sites that are cost effective yet help minimize risk. This perspective motivated PETROBRAS, the National Oil Major in Brazil, through its R&D investments portfolio to prioritize research projects that would contribute to decreasing the technological gap in the area. The Company's periodic surveys indicated the lack of infrastructure, as well as expertise in CO2 MMV, as two of the most critical issues at the national level. In order to bridge that gap, initial steps were taken in 2010 for the start-up and development of the first CO2 MMV Field Lab in Brazil, fully sponsored by PETROBRAS, with a long term goal of enabling the ranking of the best, most cost-effective MMV technology alternatives to be deployed at commercial large scale CCGS sites scheduled to be installed in the country. In addition to providing basic infrastructure to carry out the CO2 injection and controlled release experiments, the facility was designed for the simultaneous testing of multiple measuring methodologies. Additional benefits of the initiative are the creation of expertise and the acceleration of the know-how in MMV in Brazil, as well as the development of a deeper and more practical knowledge of CO2 dynamics and impacts in a real world, open air scenario. Under the full support of the PETROBRAS R&D Center (CENPES), through its Climate Change Mitigation Technological Program (PROCLIMA), the Brazilian Pilot CO2 MMV Lab was made possible through a joint 4-year research Project, conceived and carried out by PETROBRAS and local academia in Brazil, in close cooperation with international experts. An overview of the Project and the multiple research areas encompassed will be presented, together with the preliminary results of the first CO2 injection campaign, which took place in 2013.   • Microbial Succession under Freeze-Thaw Events and Its Potential for Hydrocarbon Degradation in Nutrient-Amended Antarctic Soil

    • The polar regions have relatively low richness and diversity of plants and animals, and the basis of the entire ecological chain is supported by microbial diversity. In these regions, understanding the microbial response against environmental factors and anthropogenic disturbances is essential to understand patterns better, prevent isolated events, and apply biotechnology strategies. The Antarctic continent has been increasingly affected by anthropogenic contamination, and its constant temperature fluctuations limit the application of clean recovery strategies, such as bioremediation. We evaluated the bacterial response in oil-contaminated soil through a nutrient-amended microcosm experiment using two temperature regimes: (i) 4 degrees C and (ii) a freeze-thaw cycle (FTC) alternating between -20 and 4 degrees C. Bacterial taxa, such as Myxococcales, Chitinophagaceae, and Acidimicrobiales, were strongly related to the FTC. Rhodococcus was positively related to contaminated soils and further stimulated under FTC conditions. Additionally, the nutrient-amended treatment under the FTC regime enhanced bacterial groups with known biodegradation potential and was efficient in removing hydrocarbons of diesel oil. The experimental design, rates of bacterial succession, and level of hydrocarbon transformation can be considered as a baseline for further studies aimed at improving bioremediation strategies in environments affected by FTC regimes.

  • Incidence of chronic obstructive pulmonary disease in a cohort of young adults according to the presence of chronic cough and phlegm

    • Rationale: The few prospective studies aimed at assessing the incidence of chronic obstructive pulmonary disease (COPD) in relation to the presence of chronic cough/phlegm have produced contrasting results. Objectives: To assess the incidence of COPD in a cohort of young adults and to test whether chronic cough/phlegm and dyspnea are independent predictors of COPD. Methods: An international cohort of 5,002 subjects without asthma (ages 20-44 yr) with normal lung function (FEV1/FVC ratio ≥ 70%) from 12 countries was followed from 1991-2002 in the frame of the European Community Respiratory Health Survey II. Incident cases of COPD were those who had an FEV 1/FVC ratio less than 70% at the end of the follow-up, but did not report having had a doctor diagnose asthma during the follow-up. Main Results: The incidence rate of COPD was 2.8 cases/1,000/yr (95% confidence interval [CI], 2.3-3.3). Chronic cough/phlegm was an independent and statistically significant predictor of COPD (incidence rate ratio [IRR], 1.85; 95% CI, 1.17-2.93) after adjusting for smoking habits and other potential confounders, whereas dyspnea was not associated with the disease (IRR = 0.98; 95% CI, 0.64-1.50). Subjects who reported chronic cough/phlegm both at baseline and at the follow-up had a nearly threefold-increased risk of developing COPD with respect to asymptomatic subjects (IRR = 2.88; 95% CI, 1.44-5.79). Conclusions: The incidence of COPD is substantial even in young adults. The presence of chronic cough/phlegm identifies a subgroup of subjects with a high risk of developing COPD, independently of smoking habits.

  • Long-term outcomes in mild/moderate chronic obstructive pulmonary disease in the European community respiratory health survey

    • RATIONALE: Little is known about the long-term outcomes of individuals with mild/moderate chronic obstructive pulmonary disease (COPD) according to spirometric criteria. OBJECTIVES: To test whether nonsmokers and asymptomatic subjects with a spirometric diagnosis of COPD have a steeper decrease in lung function and higher hospitalization rates than subjects without airway obstruction. METHODS: A total of 5,205 subjects without asthma (20-44 years of age) from the general population, with FEV(1) >or= 50% predicted at baseline, were followed for 9 years in the frame of an international cohort study. Percent decrease in FEV(1) (DeltaFEV(1)%) and the annual hospitalization rate for respiratory causes during the follow-up were assessed for each subject. MEASUREMENTS AND MAIN RESULTS: At baseline, 324 (6.2%) subjects had the prebronchodilator FEV(1)/FVC ratio less than the lower limit of normal (LLN-COPD), and 105 (2.0%) subjects had the same ratio less than 0.70 (modified GOLD-COPD). At follow-up, smokers with LLN-COPD (n = 205) had a greater mean DeltaFEV(1)% (1.7%; 95% confidence interval [CI], 0.8-2.7) and a higher hospitalization rate (rate ratio [RR], 2.52; 95% CI, 1.65-3.86) than normal subjects. Similarly, symptomatic subjects with LLN-COPD (n = 104) had DeltaFEV(1)% (2.0%; 95% CI, 0.8-3.3) and the hospitalization rate (RR, 4.18; 95% CI, 2.43-7.21) higher than the reference group. By contrast, nonsmokers and asymptomatic subjects with LLN-COPD had outcomes that were similar or even better than normal subjects. Among subjects with LLN-COPD, the association of symptoms with DeltaFEV(1)% varied according to smoking habits (P = 0.007); it was particularly strong in symptomatic smokers and disappeared in symptomatic nonsmokers. Similar results were found with the modified GOLD classification. CONCLUSIONS: In relatively young populations, COPD is associated with poor long-term outcomes in smokers and in symptomatic subjects only.

  • Risk Factors for Chronic Obstructive Pulmonary Disease in a European Cohort of Young Adults

    • Rationale: Few studies have investigated the factors associated with the early inception of chronic obstructive pulmonary disease (COPD). Objectives: We investigated COPD risk factors in an international cohort of young adults using different spirometric definitions of the disease. Methods. We studied 4,636 subjects without asthma who had prebronchodilator FEV1/FVC measured in the European Community Respiratory Health Survey both in 1991 to 1993 (when they were 20-44 yr old) and in 1999 to 2002. COPD was defined according to the Global Initiative for Chronic Obstructive Lung Disease fixed cut-off criterion (FEV1/FVC < 0.70), and two criteria based on the Quanjer and LuftiBus reference equations (FEV1/FVC less than lower limit of normal). COPD determinants were studied using two-level Poisson regression models. Measurements and Main Results: COPD incidence ranged from 1.85 (lower limit of normal [Quanjer]) to 2.88 (Global Initiative for Chronic Obstructive Lung Disease) cases/1,000/yr. Although about half of the cases had smoked less than 20 pack-years, smoking was the main risk factor for COPD, and it accounted for 29 to 39% of the new cases during the follow-up. Airway hyperresponsiveness was the second strongest risk factor (15-17% of new cases). Other determinants were respiratory infections in childhood and a family history of asthma, whereas the role of sex, age, and of being underweight largely depended on the definition of COPD used. Conclusions: COPD may start early in life. Smoking prevention should be given the highest priority to reduce COPD occurrence. Airway hyperresponsiveness, a family history of asthma, and respiratory infections in childhood are other important determinants of COPD. We suggest the need for a definition of COPD that is not exclusively based on spirometry.

  • Clusters of metabolic risk factors predict cardiovascular events in hypertension with target-organ damage: the LIFE study

    • The relation of metabolic syndrome (MetS) with cardiovascular outcome may be less evident when preclinical cardiovascular disease is present. We explored, in a post hoc analysis, whether MetS predicts cardiovascular events in hypertensive patients with electrocardiographic left ventricular hypertrophy (ECG-LVH) in the Losartan Intervention For Endpoint (LIFE) reduction in hypertension study. MetS was defined by >or=2 risk factors plus hypertension: body mass index >or=30 kg/m(2), high-density lipoprotein (HDL)-cholesterol <1.0/1.3 mmol/l (<40/50 mg/dl) (men/women), glucose >or=6.1 mmol/l (>or=110 mg/dl) fasting or >or=7.8 mmol/l (>or=140 mg/dl) nonfasting or diabetes. Cardiovascular death and the primary composite end point (CEP) of cardiovascular death, stroke and myocardial infarction were examined. In MetS (1,591 (19.3%) of 8,243 eligible patients), low HDL-cholesterol (72%), obesity (77%) and impaired glucose (73%) were similarly prevalent, with higher blood pressure, serum creatinine and Cornell product, but lower Sokolow-Lyon voltage (all P<0.001). After adjusting for baseline covariates, hazard ratios for CEPs and cardiovascular death (4.8+/-1.1 years follow-up) were 1.47 (95% confidence interval (CI), 1.27-1.71)- and 1.73 (95% CI, 1.38-2.17)-fold higher with MetS (both P<0.0001), and were only marginally reduced when further adjusted for diabetes, obesity, low HDL-cholesterol, non-HDL-cholesterol, pulse pressure and in-treatment systolic blood pressure and heart rate. Thus, MetS is associated with increased cardiovascular events in hypertensive patients with ECG-LVH, independently of single cardiovascular risk factors.

  • Glycolaldehyde in Perseus young solar analogs

The earliest evolutionary stages of low-mass protostars are characterised by the so-called hot-corino stage, when the newly born star heats the surrounding material and enrich the gas chemically. Studying this evolutionary phase of solar protostars may help understand the evolution of prebiotic complex molecules in the development of planetary systems. Aims. In this paper we focus on the occurrence of glycolaldehyde (HCOCH 2 OH) in young solar analogs by performing the first homogeneous and unbiased study of this molecule in the Class 0 protostars of the nearby Perseus star forming region. Methods. We obtained sub-arcsec angular resolution maps at 1.3 mm and 1.4 mm of glycolaldehyde emission lines using the IRAM Plateau de Bure (PdB) interferometer in the framework of the CALYPSO IRAM large program. Results. Glycolaldehyde has been detected towards 3 Class 0 and 1 Class I protostars out of the 13 continuum sources targeted in Perseus: NGC1333-IRAS2A1, NGC1333-IRAS4A2, NGC1333-IRAS4B1, and SVS13-A. The NGC1333 star forming region looks particularly glycolaldehyde rich, with a rate of occurrence up to 60%. The glycolaldehyde spatial distribution overlaps with the continuum one, tracing the inner 100 au around the protostar. A large number of lines (up to 18), with upper-level energies E u from 37 K up to 375 K has been detected. We derived column densities ≥10 15 cm -2 and rotational temperatures T rot between 115 K and 236 K, imaging for the first time hot-corinos around NGC1333-IRAS4B1 and SVS13-A. Conclusions. In multiple systems glycolaldehyde emission is detected only in one component. The case of the SVS13-A+B and IRAS4-A1+A2 systems support that the detection of glycolaldehyde (at least in the present Perseus sample) indicates older protostars (i.e. SVS13-A and IRAS4-A2), evolved enough to develop the hot-corino region (i.e. 100 K in the inner 100 au). However, only two systems do not allow us to firmly conclude whether the primary factor leading to the detection of glycolaldehyde emission is the environments hosting the protostars, evolution (e.g. low value of L submm /L int ), or accretion luminosity (high L int ).

  • Mea Culpa! The Role of Guilt in the Work-Life Interface and Satisfaction of Women Entrepreneur

    • The purpose of this study is to analyze the role of mediator of Guilt (in both directions: Family Interference with Work (FIW) and Work Interference with Family (WIF)) in the relationship between Conflict, Job and Life Satisfaction, also investigating the role of Enrichment as moderator. Using PROCESS Macro, the hypothesized models are tested on a sample of 161 women entrepreneurs. Both the mediating role of guilt and the moderating role of enrichment were analyzed through models of mediation and moderate mediation. Results from the analysis support the hypothesized models. Guilt FIW and Guilt WIF mediate the relationship between work Conflict and Job satisfaction, as well the relationship between Conflict and Life satisfaction, and at the same time, Enrichment moderated the mediating processes by which the Conflict affects Job and Life satisfaction via Guilt FIW and Guilt WIF. This study is one of the few that takes into consideration both Conflict and Enrichment in a sample of women entrepreneurs and examines Guilt, which many times presents itself as an “invisible” factor in studies on the work–family interface. © 2022 by the authors.

    • Metal-organic composites are of great interest for a wide range of applications. The control of their structure remains a challenge, one of the problems being a complex interplay of covalent and supramolecular interactions. This paper describes the self-assembly, thermal stability and phase transitions of ordered structures of silver atoms and thiol molecules spanning from the molecular to the mesoscopic scale. Building blocks of molecularly defined clusters formed from 44 silver atoms, each particle coated by a monolayer of 30 thiol ligands, are used as ideal building blocks. By changing solvent and temperature it is possible to tune the self-assembled 3D crystals of pristine nanoparticles or, conversely, 2D layered structures, with alternated stacks of Ag atoms and thiol monolayers. The study investigates morphological, chemical and structural stability of these materials between 25 and 300 °C in situ and ex situ at the nanoscale by combining optical and electronic spectroscopic and scattering techniques, scanning probe microscopies and density-functional theory (DFT) calculations. The proposed wet-chemistry approach is relatively cheap, easy to implement, and scalable, allowing the fabricated materials with tuned properties using the same building blocks.
