    • BACKGROUND Both genetic and lifestyle factors contribute to individual-level risk of coronary artery disease. The extent to which increased genetic risk can be offset by a healthy lifestyle is unknown. METHODS Using a polygenic score of DNA sequence polymorphisms, we quantified genetic risk for coronary artery disease in three prospective cohorts - 7814 participants in the Atherosclerosis Risk in Communities (ARIC) study, 21,222 in the Women's Genome Health Study (WGHS), and 22,389 in the Malmö Diet and Cancer Study (MDCS) - And in 4260 participants in the cross-sectional BioImage Study for whom genotype and covariate data were available. We also determined adherence to a healthy lifestyle among the participants using a scoring system consisting of four factors: no current smoking, no obesity, regular physical activity, and a healthy diet. RESULTS The relative risk of incident coronary events was 91% higher among participants at high genetic risk (top quintile of polygenic scores) than among those at low genetic risk (bottom quintile of polygenic scores) (hazard ratio, 1.91; 95% confidence interval [CI], 1.75 to 2.09). A favorable lifestyle (defined as at least three of the four healthy lifestyle factors) was associated with a substantially lower risk of coronary events than an unfavorable lifestyle (defined as no or only one healthy lifestyle factor), regardless of the genetic risk category. Among participants at high genetic risk, a favorable lifestyle was associated with a 46% lower relative risk of coronary events than an unfavorable lifestyle (hazard ratio, 0.54; 95% CI, 0.47 to 0.63). This finding corresponded to a reduction in the standardized 10-year incidence of coronary events from 10.7% for an unfavorable lifestyle to 5.1% for a favorable lifestyle in ARIC, from 4.6% to 2.0% in WGHS, and from 8.2% to 5.3% in MDCS. In the BioImage Study, a favorable lifestyle was associated with significantly less coronary-artery calcification within each genetic risk category. CONCLUSIONS Across four studies involving 55,685 participants, genetic and lifestyle factors were independently associated with susceptibility to coronary artery disease. Among participants at high genetic risk, a favorable lifestyle was associated with a nearly 50% lower relative risk of coronary artery disease than was an unfavorable lifestyle.

    • Consumption of fruits and vegetables is associated with a lower overall mortality. The aim of this study was to identify causes of death through which this association is established. More than 450,000 participants from the European Prospective Investigation into Cancer and Nutrition study were included, of which 25,682 were reported deceased after 13 years of follow-up. Information on lifestyle, diet and vital status was collected through questionnaires and population registries. Hazard ratios (HR) with 95 % confidence intervals (95 % CI) for death from specific causes were calculated from Cox regression models, adjusted for potential confounders. Participants reporting consumption of more than 569 g/day of fruits and vegetables had lower risks of death from diseases of the circulatory (HR for upper fourth 0.85, 95 % CI 0.77-0.93), respiratory (HR for upper fourth 0.73, 95 % CI 0.59-0.91) and digestive system (HR for upper fourth 0.60, 95 % CI 0.46-0.79) when compared with participants consuming less than 249 g/day. In contrast, a positive association with death from diseases of the nervous system was observed. Inverse associations were generally observed for vegetable, but not for fruit consumption. Associations were more pronounced for raw vegetable consumption, when compared with cooked vegetable consumption. Raw vegetable consumption was additionally inversely associated with death from neoplasms and mental and behavioral disorders. The lower risk of death associated with a higher consumption of fruits and vegetables may be derived from inverse associations with diseases of the circulatory, respiratory and digestive system, and may depend on the preparation of vegetables and lifestyle factors. © 2014 Springer Science+Business Media Dordrecht.

    • One of the principal goals of glycoprotein research is to correlate glycan structure and function. Such correlation is necessary in order for one to understand the mechanisms whereby glycoprotein structure elaborates the functions of myriad proteins. The accurate comparison of glycoforms and quantification of glycosites are essential steps in this direction. Mass spectrometry has emerged as a powerful analytical technique in the field of glycoprotein characterization. Its sensitivity, high dynamic range, and mass accuracy provide both quantitative and sequence/structural information. As part of the 2012 ABRF Glycoprotein Research Group study, we explored the use of mass spectrometry and ancillary methodologies to characterize the glycoforms of two sources of human prostate specific antigen (PSA). PSA is used as a tumor marker for prostate cancer, with increasing blood levels used to distinguish between normal and cancer states. The glycans on PSA are believed to be biantennary N-linked, and it has been observed that prostate cancer tissues and cell lines contain more antennae than their benign counterparts. Thus, the ability to quantify differences in glycosylation associated with cancer has the potential to positively impact the use of PSA as a biomarker. We studied standard peptide-based proteomics/glycomics methodologies, including LC-MS/MS for peptide/glycopeptide sequencing and label-free approaches for differential quantification. We performed an interlaboratory study to determine the ability of different laboratories to correctly characterize the differences between glycoforms from two different sources using mass spectrometry methods. We used clustering analysis and ancillary statistical data treatment on the data sets submitted by participating laboratories to obtain a consensus of the glycoforms and abundances.     • We report on SRG/eROSITA, ZTF, ASAS-SN, Las Cumbres, NEOWISE-R, and Swift XRT/UVOT observations of the unique ongoing event AT 2019avd, located in the nucleus of a previously inactive galaxy at z = 0.029. eROSITA first observed AT 2019avd on 2020-04-28 during its first all sky survey, when it was detected as an ultra-soft X-ray source (kT ~ 85 eV) that was ≳90 times brighter in the 0.2−2 keV band than a previous 3σ upper flux detection limit (with no archival X-ray detection at this position). The ZTF optical light curve in the ~450 days preceding the eROSITA detection is double peaked, and the eROSITA detection coincides with the rise of the second peak. Follow-up optical spectroscopy shows the emergence of a Bowen fluorescence feature and high-ionisation coronal lines ([Fe X] 6375 Å, [Fe XIV] 5303 Å), along with persistent broad Balmer emission lines (FWHM ~ 1400 km s−1). Whilst the X-ray properties make AT 2019avd a promising tidal disruption event (TDE) candidate, the optical properties are atypical for optically selected TDEs. We discuss potential alternative origins that could explain the observed properties of AT 2019avd, such as a stellar binary TDE candidate, or a TDE involving a super massive black hole binary.

    • Calcite veins in Paleoproterozoic granitoids on the Baltic Shield are the focus of this study. These veins are distinguished by their monomineralic character, unusual thickness and closeness to Neoproterozoic dolerite dykes and therefore have drawn attention. The aim of this study was to define the source of these veins and to unravel their isotopic and chemical nature by carrying out fine-scale studies. Seven calcite veins covering a depth interval of 50-420m below the ground surface and composed of breccias or crack-sealed fillings typically expressing syntaxial growth were sampled and analysed for a variety of physicochemical variables: homogenization temperature (T-h) and salinity of fluid inclusions, and stable isotopes (Sr-87/Sr-86, C-13/C-12, O-18/O-16), trace-element concentrations (Fe, Mn, Mg, Sr, rare earth elements) and cathodoluminescence (CL) of the solid phase. The fluid-inclusion data show that the calcites were precipitated mainly from relatively low-temperature (T-h=73-106 degrees C) brines (13.4-24.5wt.% CaCl2), and the Sr-87/Sr-86 is more radiogenic than expected for Rb-poor minerals precipitated from Neoproterozoic fluids. These features, together with the distribution of C-13 and O-18 values, provide evidence that the calcite veins are not genetic with the nearby Neoproterozoic dolerite dykes, but are of Paleozoic age and were precipitated from warm brines expressing a rather large variability in salinity. Whereas the isotopic and chemical variables express rather constant average values among the individual veins, they vary considerably on fine-scale across individual veins. This has implications for understanding processes causing calcite-rich veins to form and capture trace metals in crystalline bedrock settings.

  • Magnetic and Rotational Evolution of rho CrB from Asteroseismology with TESS

    • During the first half of main-sequence lifetimes, the evolution of rotation and magnetic activity in solar-type stars appears to be strongly coupled. Recent observations suggest that rotation rates evolve much more slowly beyond middle age, while stellar activity continues to decline. We aim to characterize this midlife transition by combining archival stellar activity data from the Mount Wilson Observatory with asteroseismology from the Transiting Exoplanet Survey Satellite (TESS). For two stars on opposite sides of the transition (88 Leo and rho CrB), we independently assess the mean activity levels and rotation periods previously reported in the literature. For the less active star (rho CrB), we detect solar-like oscillations from TESS photometry, and we obtain precise stellar properties from asteroseismic modeling. We derive updated X-ray luminosities for both stars to estimate their mass-loss rates, and we use previously published constraints on magnetic morphology to model the evolutionary change in magnetic braking torque. We then attempt to match the observations with rotational evolution models, assuming either standard spin-down or weakened magnetic braking. We conclude that the asteroseismic age of rho CrB is consistent with the expected evolution of its mean activity level and that weakened braking models can more readily explain its relatively fast rotation rate. Future spectropolarimetric observations across a range of spectral types promise to further characterize the shift in magnetic morphology that apparently drives this midlife transition in solar-type stars.
