  • The Bone Sparing Effects of 2-Methoxyestradiol Are Mediated via Estrogen Receptor-α in Male Mice.
  • 2016
  • Ingår i: Endocrinology. - : The Endocrine Society. - 1945-7170 .- 0013-7227. ; 157:11, s. 4200-4205
  • Tidskriftsartikel (refereegranskat)abstract
    • 2-Methoxyestradiol (2ME2), a metabolite of 17β-estradiol (E2), exerts bone sparing effects in animal models. We hypothesized that the underlying mechanism is back conversion of 2ME2 to E2, which subsequently acts via estrogen receptor (ER)α. We measured serum E2 levels in orchidectomized wild-type (WT) mice treated with 2ME2 66.6 μg/d or placebo. In placebo-treated animals, E2 was below the detection limit. In 2ME2-treated mice, the serum E2 level was 4.97 ± 0.68 pg/mL. This corresponds to the level found in diesterus in cycling female mice. Next, we investigated bone parameters in orchidectomized WT and ERα knockout mice treated with 2ME2 or placebo for 35 days. 2ME2 (6.66 μg/d) preserved trabecular and cortical bone in WT mice. Trabecular volumetric-bone mineral density was 64 ± 20%, and trabecular bone volume/total volume was 60 ± 20% higher in the metaphyseal region of the femur in the 2ME2 group, compared with placebo (P < .01). Both trabecular number and trabecular thickness were increased (P < .01). Cortical bone mineral content in the diaphyseal region of the femur was 31 ± 3% higher in the 2ME2 group, compared with placebo (P < .001). This was due to larger cortical area (P < .001). Three-point bending showed an increased bone strength in WT 2ME2-treated animals compared with placebo (maximum load [Fmax] +19±5% in the 2ME2 group, P < .05). Importantly, no bone parameter was affected by 2ME2 treatment in ERα knockout mice. In conclusion, 2ME2 treatment of orchidectomized mice results in increased serum E2. ERα mediates the bone sparing effects of 2ME2. The likely mediator of this effect is E2 resulting from back conversion of 2ME2.

  • The COMT val158met polymorphism is associated with prevalent fractures in Swedish men.
  • 2008
  • Ingår i: Bone. - : Elsevier BV. - 8756-3282 .- 1873-2763. ; 42:1, s. 107-12
  • Tidskriftsartikel (refereegranskat)abstract
    • INTRODUCTION: Sex steroids are important for growth and maintenance of the skeleton. Catechol-O-methyltransferase (COMT) is an estrogen degrading enzyme. The COMT val158met polymorphism results in a 60-75% difference in enzyme activity between the val (high activity=H) and met (low activity=L) variants. We have previously reported that this polymorphism is associated with bone mineral density (BMD) in young men. The aim of this study was to investigate associations between COMT val158met, BMD and fractures in elderly men. METHODS: Population-based study of Swedish men 75.4, SD 3.2, years of age. Fractures were reported using standardized questionnaires. Fracture and genotype data were available from 2,822 individuals. RESULTS: Total number of individuals with self-reported fracture was 989 (35.0%). Prevalence of >or=1 fracture was 37.2% in COMT(LL), 35.7% in COMT(HL) and 30.4% in COMT(HH) (p<0.05). Early fractures (50 years of age). The OR for fracture of the non-weight bearing skeleton in COMT(HH) compared with COMT(LL+HL) was 0.74 (95% CI 0.59-0.92). No associations between COMT val158met and BMD were found in this cohort of elderly men. CONCLUSIONS: The COMT val158met polymorphism is associated with life time fracture prevalence in elderly Swedish men. This association is mainly driven by early fractures (

  • Near infrared optical projection tomography for assessments of beta-cell mass distribution in diabetes research
  • 2013
  • Ingår i: Journal of Visualized Experiments. - : MyJove Corporation. - 1940-087X. ; 71
  • Tidskriftsartikel (refereegranskat)abstract
    • By adapting OPT to include the capability of imaging in the near infrared (NIR) spectrum, we here illustrate the possibility to image larger bodies of pancreatic tissue, such as the rat pancreas, and to increase the number of channels (cell types) that may be studied in a single specimen. We further describe the implementation of a number of computational tools that provide: 1/ accurate positioning of a specimen's (in our case the pancreas) centre of mass (COM) at the axis of rotation (AR)2; 2/ improved algorithms for post-alignment tuning which prevents geometric distortions during the tomographic reconstruction2 and 3/ a protocol for intensity equalization to increase signal to noise ratios in OPT-based BCM determinations3. In addition, we describe a sample holder that minimizes the risk for unintentional movements of the specimen during image acquisition. Together, these protocols enable assessments of BCM distribution and other features, to be performed throughout the volume of intact pancreata or other organs (e.g. in studies of islet transplantation), with a resolution down to the level of individual islets of Langerhans.

  • Cytokine Autoantibody Screening in the Swedish Addison Registry Identifies Patients With Undiagnosed APS1
  • 2018
  • Ingår i: The Journal of clinical endocrinology and metabolism. - : The Endocrine Society. - 1945-7197 .- 0021-972X. ; 103:1, s. 179-186
  • Tidskriftsartikel (refereegranskat)abstract
    • Context: Autoimmune polyendocrine syndrome type 1 (APS1) is a monogenic disorder that features autoimmune Addison disease as a major component. Although APS1 accounts for only a small fraction of all patients with Addison disease, early identification of these individuals is vital to prevent the potentially lethal complications of APS1.Objective: To determine whether available serological and genetic markers are valuable screening tools for the identification of APS1 among patients diagnosed with Addison disease.Design: We systematically screened 677 patients with Addison disease enrolled in the Swedish Addison Registry for autoantibodies against interleukin-22 and interferon-α4. Autoantibody-positive patients were investigated for clinical manifestations of APS1, additional APS1-specific autoantibodies, and DNA sequence and copy number variations of AIRE.Results: In total, 17 patients (2.5%) displayed autoantibodies against interleukin-22 and/or interferon-α4, of which nine were known APS1 cases. Four patients previously undiagnosed with APS1 fulfilled clinical, genetic, and serological criteria. Hence, we identified four patients with undiagnosed APS1 with this screening procedure. Conclusion: We propose that patients with Addison disease should be routinely screened for cytokine autoantibodies. Clinical or serological support for APS1 should warrant DNA sequencing and copy number analysis of AIRE to enable early diagnosis and prevention of lethal complications.

  • Sustainable development for ICT engineering students : “What’s in it for me”?
  • 2016
  • Konferensbidrag (refereegranskat)abstract
    • The importance of sustainable development (SD) is hardly possible to refute; however, sustainable development has been a relatively peripheral subject in computer-related engineering educations. Sustainability, with its global and potentially all-encompassing connotations, is still seen by many Information and Communication Technology (ICT) students as a topic of little relevance to their future careers. So how can teachers convince these students that sustainability is a topic that can be both relevant and interesting for them? From the point of view of the student; “What’s in it for me?”.In this paper we describe and compare our efforts to plan and teach three introductory courses on SD in three different ICT-related educational programmes at KTH Royal Institute of Technology. The courses were planned separately, but they will be analysed together. We discuss two dimensions that we have found to be imperative in our endeavour to engage our students. The first dimension is to handle the balance between sustainability on a general level versus sustainability as specifically related to ICT. The second dimension is to handle the tension between teaching facts versus an emphasis on students’ reflections and/or practicing skills. We argue that overcoming the challenge of making sustainability relevant to the students is central for successfully teaching these courses.

    • The Swedish EAA System started as a regional initiative and rapidly grew to become a national level activity. An important feature of the system today is its complete integration into the national process for managed introduction and follow-up of new medicines. The system will continue to evolve as a response both to the changing landscape of health innovations and to new policy initiatives at the regional, national and international level.

  • Periodontal Health and Oral Microbiota in Patients with Rheumatoid Arthritis
  • 2019
  • Ingår i: Journal of Clinical Medicine. - : MDPI AG. - 2077-0383. ; 8:5
  • Tidskriftsartikel (refereegranskat)abstract
    • This study aimed to investigate the periodontal health of patients with established rheumatoid arthritis (RA) in relation to oral microbiota, systemic and oral inflammatory mediators, and RA disease activity. Forty patients underwent full-mouth dental/periodontal and rheumatological examination, including collection of blood, saliva, gingival crevicular fluid (GCF) and subgingival plaque. Composition of plaque and saliva microbiota were analysed using 16S rRNA sequencing and levels of inflammatory mediators by multiplex-immunoassay. The majority of the patients (75%) had moderate or severe periodontitis and the rest had no/mild periodontitis. Anti-citrullinated protein antibody (ACPA) positivity was significantly more frequent in the moderate/severe periodontitis (86%) compared to the no/mild group (50%). No significance between groups was observed for RA disease duration or activity, or type of medication. Levels of sCD30/TNFRSF8, IFN-2, IL-19, IL-26, MMP-1, gp130/sIL-6R ss, and sTNF-R1 were significantly higher in serum or GCF, and April/TNFSF13 was significantly higher in serum and saliva samples in moderate/severe periodontitis. The microbial composition in plaque also differed significantly between the two groups. In conclusion, the majority of RA patients had moderate/severe periodontitis and that this severe form of the disease was significantly associated with ACPA positivity, an altered subgingival microbial profile, and increased levels of systemic and oral inflammatory mediators.

  • Different beef breed sires used for crossbreeding with Swedish dairy cows - effects on calving performance and carcass traits
  • 2020
  • Ingår i: Livestock Science. - : Elsevier BV. - 1871-1413 .- 1878-0490. ; 232
  • Tidskriftsartikel (refereegranskat)abstract
    • Improved potential for weight gain and carcass classification traits in calves aimed at slaughter from dairy herds would increase the calves' economic value, and enable an increased beef production. At present, relatively low economic weights are placed on beef traits in the comprehensive Nordic dairy cattle breeding goal. An effective way to improve beef production from dairy herds is therefore to inseminate a larger proportion of dairy heifers and cows with beef bull semen. In the choice of beef bull breed, both effects on calving traits, carcass weight gain and carcass traits need to be considered. The aim of this study was therefore to compare different beef x dairy crosses regarding calving performance and carcass traits. Field data comprising in total close to 1.6 million calving records from Swedish herds, and more than 1.1 million carcass records from commercial slaughter houses, were analyzed using linear models. Differences in least-square means between different breed combinations were estimated for percentage of calving difficulty, stillbirth, daily carcass gain, carcass weight, EUROP conformation and fat classification. The dams were purebred Swedish Red (SR) or Swedish Holstein (SH), and the sires were either of purebred dairy (SR or SH) or of the beef breeds Angus, Hereford, Limousin, Simmental or Charolais. The results showed that crossbreeding dairy cows with beef bulls, especially of late maturing breeds, has potential to considerably improve beef production from dairy herds. Improvement of carcass characteristics and carcass gain were most noticeable in young crossbred bulls, where the highest increase for single breed combinations was 67% for conformation and 17% for carcass gain. Offspring of different beef x dairy combinations displayed varying average carcass gain and characteristics in terms of fatness and conformation in heifers, steers and young bulls, and would fit different types of production systems. Whereas crossbreeding had small or favorable effects on stillbirth, the use of late maturing beef breed sires negatively influenced the calving performance, especially in primiparous dairy cows. By choosing individual beef bulls with high estimated breeding values for calving ease, the percentage of calving difficulty could be reduced, even when using late maturing beef breed sires for crossbreeding.
