    • Dye-sensitized interfaces in photocatalytic and solar cells systems are significantly affected by the choice of electrolyte solvent. In the present work, the interface between the hydrophobic Ru-complex Z907, a commonly used dye in molecular solar cells, and TiO2 was investigated with ambient pressure photoelectron spectroscopy (AP-PES) to study the effect of water atmosphere on the chemical and electronic structure of the dye/TiO2 interface. Both laboratory-based Al K alpha as well as synchrotron-based ambient pressure measurements using hard X-ray (AP-HAXPES) were used. AP-HAXPES data were collected at pressures of up to 25 mbar (i.e., the vapor pressure of water at room temperature) showing the presence of an adsorbed water overlayer on the sample surface. Adopting a quantitative AP-HAXPES analysis methodology indicates a stable stoichiometry in the presence of the water atmosphere. However, solvation effects due to the presence of water were observed both in the valence band region and for the S 1s core level and the results were compared with DFT calculations of the dye-water complex.

  • Comparison of PFASs contamination in the freshwater and terrestrial environments by analysis of eggs from osprey (Pandion haliaetus), tawny owl (Strix aluco), and common kestrel (Falco tinnunculus)

    • The level of PFAS (per- and polyfluorinated alkyl substances) contamination in freshwater and terrestrial Swedish environments in 2013/2014 was assessed by analyzing a range of perfluorinated alkyl acids, fluorotelomer acids, sulfonamides, sulfonamidoethanols and polyfluoralkyl phosphate diesters (diPAPs) in predator bird eggs. Stable isotopes ((13)C and (15)N) were analyzed to elucidate the dietary source. The tawny owl (Strix aluco, n=10) and common kestrel (Falco tinnunculus, n=40), two terrestrial species, and the osprey (Pandion haliaetus, n=30), a freshwater specie were included. In addition, a temporal trend (1997-2001, 2008-2009, 2013) in osprey was studied as well. The PFAS profile was dominated by perfluorooctane sulfonic acid (PFOS) in eggs from osprey and tawny owl, while for common kestrel perfluorinated carboxylic acids (∑PFCA) exceeded the level of PFOS. PFOS concentration in osprey eggs remained at the same level between 1997 and 2001 and 2013. For the long-chained PFCAs, there were a significant increase in concentrations in osprey eggs between 1997 and 2001 and 2008-2009. The levels of PFOS and PFCAs were about 10 and five times higher, respectively, in osprey compared to tawny owl and common kestrel. Evidence of direct exposure from PFCA precursor compounds to birds in both freshwater and terrestrial environment was observed. Low levels of diPAPs were detected in a few samples of osprey (<0.02-2.4ng/g) and common kestrel (<0.02-0.16ng/g) eggs, and 6:2 FTSA was detected in a majority of the osprey eggs (<6.3-52ng/g). One saturated telomer acid (7:3 FTCA), which is a transformation marker from precursor exposure, was detected in all species (<0.24-2.7ng/g). The (15)N data showed higher levels in osprey eggs compared to tawny owl and common kestrel, indicating that they feed on a 2-3 times higher trophic level. We conclude that ospreys are continuously exposed to PFAS at levels where adverse toxic effects have been observed in birds.

  • Perfluoroalkyl substances (PFASs) in food and water from Faroe Islands

    • Diet and drinking water are suggested to be major exposure pathways for perfluoroalkyl substances (PFASs). In this study, food items and water from Faroe Islands sampled in 2011/2012 were analyzed for 11 perfluoroalkyl carboxylic acids (PFCAs) and 4 perfluoroalkane sulfonic acids (PFSAs). The food samples included milk, yoghurt, crSme fraiche, potatoes, fish, and fish feed, and the water samples included surface water and purified drinking water. In total, nine PFCAs and four PFSAs were detected. Generally, the levels of PFAS were in the lower picogram per gram range. Perfluorobutanoic acid was a major contributor to the total PFASs concentration in water samples and had a mean concentration of 750 pg/L. Perfluoroundecanoic acid (PFUnDA) was predominating in milk and wild fish with mean concentrations of 170 pg/g. Perfluorooctane sulfonic acid (PFOS) was most frequently detected in food items followed by PFUnDA, perfluorononanoic acid, and perfluorooctanoic acid (PFOA). Levels of PFUnDA and PFOA exceeded those of PFOS in milk and fish samples. Prevalence of long-chain PFCAs in Faroese food items and water is confirming earlier observations of their increase in Arctic biota. Predominance of short-chain and long-chain homologues indicates exposure from PFOS and PFOA replacement compounds.

  • Deciphering colorectal cancer genetics through multi-omic analysis of 100,204 cases and 154,587 controls of European and east Asian ancestries

    • Colorectal cancer (CRC) is a leading cause of mortality worldwide. We conducted a genome-wide association study meta-analysis of 100,204 CRC cases and 154,587 controls of European and east Asian ancestry, identifying 205 independent risk associations, of which 50 were unreported. We performed integrative genomic, transcriptomic and methylomic analyses across large bowel mucosa and other tissues. Transcriptome- and methylome-wide association studies revealed an additional 53 risk associations. We identified 155 high-confidence effector genes functionally linked to CRC risk, many of which had no previously established role in CRC. These have multiple different functions and specifically indicate that variation in normal colorectal homeostasis, proliferation, cell adhesion, migration, immunity and microbial interactions determines CRC risk. Crosstissue analyses indicated that over a third of effector genes most probably act outside the colonic mucosa. Our findings provide insights into colorectal oncogenesis and highlight potential targets across tissues for new CRC treatment and chemoprevention strategies.

  • An Interactive Visualization Tool for Collaborative Construction Logistics Planning—Creating a Sustainable Project Vicinity

    • The intensity of urban development is presently high, creating a construction boom. The number of transports per project is a major consideration in urban goods transport and emissions from a project. Presently, the stakeholders take part in a “blame game” in assigning fault for the emissions from construction transport and the disturbances to society in the vicinity of construction sites. Incorporation of logistics into urban planning requires an increased understanding of the interaction between construction transport flows and urban land use, and the inclusion of different stakeholders. The purpose of the study is to support collaborative planning of construction transport in urban planning, and specifically to explore how a planning tool based on interactive visualization could be designed. An action research process has generated two prototypes of an interactive visualization tool for collaborative planning of construction transport. The prototype facilitates a “shared deliberation space” by identifying alternatives and assessing predicted consequences, which supports a collaborative urban planning process. Based on the research conducted, we claim that the responsibility of construction transport planning should be taken by the municipality, i.e., the urban planning and traffic planning functions. © 2022 by the authors.

    • Störningsfri stad har kombinerat kunskaper inom Trafikplanering, Visualisering, Bygglogistik, Sensorer och Offentlig planering och styrning. Projektet har letts av Linköpings universitet, Institutionen för Teknik och Naturvetenskap och finansierats av Vinnova, IHS. Projektet har löpt mellan slutet av 2018 och 2021 ut. Projektdeltagare har varit VTI, RISE, Norrköpings kommun, Linköpings kommun, Stockholm stad, Uppsala kommun, Trafikverket Projekt Ostlänken, LE Lundbergs Fastigheter, Wiklunds Åkeri, Wellsec, SanktKors, Byggföretagen, Telenor och Logistikia.Projektet har utvecklat en verktygsprototyp som stöttar kollaborativ planering av byggtrafik och stadsplanering genom att visualisera pågående byggprojekts sammanlagda mängd transporter och var i staden (i form av en kartvy) det uppstår störningar i form av förseningar pga dessa. Verktyget bygger på en MATSim trafikmodell (i nuläget Norrköping) inkluderande både persontransporter (modellerat baserat på mobilnätsdata) och byggtrafik (modellerat baserat på data från bygglogistiklösningar). Med hjälp av verktyget kan olika scenarier för att minska störningarna av byggtransporter testas. För att stötta användandet av verktyget har en kollaborativ planeringsprocess som visar på hur de olika aktörerna involverade i planeringen av byggtransporter och stadsutveckling behöver interagera med varandra utvecklats. Projektet också deltagit i utvecklingen av en sensorbox (partiklar, luftfuktighet, temperatur och ljud) samt slangmätningsmetodik för att öka kunskapen om byggtransporter. Baserat på de resultat som projektet har behovsägarna har påbörjat arbeten med att bättre hantera de störningar som uppstår pga byggprojekt i staden.

    • Background Internet-delivered self-help programmes with added guidance have shown efficacy in social anxiety disorder, unguided self-help has been insufficiently studied. Aims To evaluate the efficacy of guided and unguided self-help social anxiety disorder. Method Participants followed a cognitive-behavioural self-help programme in the form of either pure bibliotherapy or an internet-based treatment with therapist guidance and online group discussions. A subsequent trial was conducted to evaluate treatment specificity. Participants (n=235) were randomised to one of three conditions in the first trial, or one of four conditions in the second. Results Pure bibliotherapy and the internet-based treatment were better than waiting list on measures of social anxiety, general anxiety, depression and quality of life. The internet-based therapy had the highest effect sizes, but directly comparable effects were noted for bibliotherapy augmented with online group discussions. Gains were well maintained a year later. Conclusions Unguided self-help through bibliotherapy can produce enduring improvement for individuals with social anxiety disorder.

    • We performed fine mapping of 39 established type 2 diabetes (T2D) loci in 27,206 cases and 57,574 controls of European ancestry. We identified 49 distinct association signals at these loci, including five mapping in or near KCNQ1. 'Credible sets' of the variants most likely to drive each distinct signal mapped predominantly to noncoding sequence, implying that association with T2D is mediated through gene regulation. Credible set variants were enriched for overlap with FOXA2 chromatin immunoprecipitation binding sites in human islet and liver cells, including at MTNR1B, where fine mapping implicated rs10830963 as driving T2D association. We confirmed that the T2D risk allele for this SNP increases FOXA2-bound enhancer activity in islet- and liver-derived cells. We observed allele-specific differences in NEUROD1 binding in islet-derived cells, consistent with evidence that the T2D risk allele increases islet MTNR1B expression. Our study demonstrates how integration of genetic and genomic information can define molecular mechanisms through which variants underlying association signals exert their effects on disease.

    • Background: There are potential conflicts between authorities and companies to fund new premium priced drugs especially where there are effectiveness, safety and/or budget concerns. Dabigatran, a new oral anticoagulant for the prevention of stroke in patients with non-valvular atrial fibrillation (AF), exemplifies this issue. Whilst new effective treatments are needed, there are issues in the elderly with dabigatran due to variable drug concentrations, no known antidote and dependence on renal elimination. Published studies showed dabigatran to be cost-effective but there are budget concerns given the prevalence of AF. These concerns resulted in extensive activities pre- to post-launch to manage its introduction. Objective: To (i) review authority activities across countries, (ii) use the findings to develop new models to better manage the entry of new drugs, and (iii) review the implications based on post-launch activities. Methodology: (i) Descriptive review and appraisal of activities regarding dabigatran, (ii) development of guidance for key stakeholder groups through an iterative process, (iii) refining guidance following post launch studies. Results: Plethora of activities to manage dabigatran including extensive pre-launch activities, risk sharing arrangements, prescribing restrictions and monitoring of prescribing post launch. Reimbursement has been denied in some countries due to concerns with its budget impact and/or excessive bleeding. Development of a new model and future guidance is proposed to better manage the entry of new drugs, centering on three pillars of pre-, pen-, and post-launch activities. Post-launch activities include increasing use of patient registries to monitor the safety and effectiveness of new drugs in clinical practice. Conclusion: Models for introducing new drugs are essential to optimize their prescribing especially where concerns. Without such models, new drugs may be withdrawn prematurely and/or struggle for funding.
