  • Kinetics of functional beta cell mass decay in a diphtheria toxin receptor mouse model of diabetes

    • Functional beta cell mass is an essential biomarker for the diagnosis and staging of diabetes. It has however proven technically challenging to study this parameter during diabetes progression. Here we have detailed the kinetics of the rapid decline in functional beta cell mass in the RIP-DTR mouse, a model of hyperglycemia resulting from diphtheria toxin induced beta cell ablation. A novel combination of imaging modalities was employed to study the pattern of beta cell destruction. Optical projection tomography of the pancreas and longitudinal in vivo confocal microscopy of islets transplanted into the anterior chamber of the eye allowed to investigate kinetics and tomographic location of beta cell mass decay in individual islets as well as at the entire islet population level. The correlation between beta cell mass and function was determined by complementary in vivo and ex vivo characterizations, demonstrating that beta cell function and glucose tolerance were impaired within the first two days following treatment when more than 50% of beta cell mass was remaining. Our results illustrate the importance of acquiring quantitative functional and morphological parameters to assess the functional status of the endocrine pancreas.

  • Radial variation of heat transport in L-mode JET discharges

    • In this paper, we analyze heat transport in the JET tokamak using data from its high resolution ECE diagnostic and analyses based on the transfer entropy (TE). The analysis reveals that heat transport is not smooth and continuous, but is characterized by 'trapping regions' separated by `minor transport barriers'. Meat may 'jump over' these barriers and when the heating power is raised, this 'jumping' behavior becomes more prominent. To check that our results are relevant for global heat transport, we deduced an effective diffusion coefficient from the TE results. Both its value and overall radial variation are consistent with heat diffusivities reported in literature. The detailed radial structure of the effective diffusion coefficient was shown to be linked to the mentioned minor transport barriers.

  • High-Definition DIC Imaging Uncovers Transient Stages of Pathogen Infection Cycles on the Surface of Human Adult Stem Cell-Derived Intestinal Epithelium

    • Interactions between individual pathogenic microbes and host tissues involve fast and dynamic processes that ultimately impact the outcome of infection. Using live-cell microscopy, these dynamics can be visualized to study, e.g., microbe motility, binding and invasion of host cells, and intrahost-cell survival. Such methodology typically employs confocal imaging of fluorescent tags in tumor-derived cell line infections on glass. This allows high-definition imaging but poorly reflects the host tissue’s physiological architecture and may result in artifacts. We developed a method for live-cell imaging of microbial infection dynamics on human adult stem cell-derived intestinal epithelial cell (IEC) layers. These IEC layers are grown in apical imaging chambers, optimized for physiological cell arrangement and fast, but gentle, differential interference contrast (DIC) imaging. This allows subsecond visualization of both microbial and epithelial surface ultrastructure at high resolution without using fluorescent reporters. We employed this technology to probe the behavior of two model pathogens, Salmonella enterica serovar Typhimurium and Giardia intestinalis, at the intestinal epithelial surface. Our results reveal pathogen-specific swimming patterns on the epithelium and show that Salmonella lingers on the IEC surface for prolonged periods before host cell invasion, while Giardia uses circular swimming with intermittent attachments to scout for stable adhesion sites. The method even permits tracking of individual Giardia flagella, demonstrating that active flagellar beating and attachment to the IEC surface are not mutually exclusive. This work describes a generalizable and relatively inexpensive approach to resolving dynamic pathogen-IEC layer interactions, applicable even to genetically nontractable microorganisms.

    • A synthetic diagnostic has been developed for the JET lost alpha scintillator probe, based on the ASCOT fast ion orbit following code and the AFSI fusion source code. The synthetic diagnostic models the velocity space distribution of lost fusion products in the scintillator probe. Validation with experimental measurements is presented, where the synthetic diagnostic is shown to predict the gyroradius and pitch angle of lost DD protons and tritons. Additionally, the synthetic diagnostic reproduces relative differences in total loss rates in multiple phases of the discharge, which can be used as a basis for total loss rate predictions.

  • Synthetic NPA diagnostic for energetic particles in JET plasmas

    • Neutral particle analysis (NPA) is one of the few methods for diagnosing fast ions inside a plasma by measuring neutral atom fluxes emitted due to charge exchange reactions. The JET tokamak features an NPA diagnostic which measures neutral atom fluxes and energy spectra simultaneously for hydrogen, deuterium and tritium species. A synthetic NPA diagnostic has been developed and used to interpret these measurements to diagnose energetic particles in JET plasmas with neutral beam injection (NBI) heating. The synthetic NPA diagnostic performs a Monte Carlo calculation of the neutral atom fluxes in a realistic geometry. The 4D fast ion distributions, representing NBI ions, were simulated using the Monte Carlo orbit-following code ASCOT. Neutral atom density profiles were calculated using the FRANTIC neutral code in the JINTRAC modelling suite. Additionally, for rapid analysis, a scan of neutral profiles was precalculated with FRANTIC for a range of typical plasma parameters. These were taken from the JETPEAK database, which includes a comprehensive set of data from the flat-top phases of nearly all discharges in recent JET campaigns. The synthetic diagnostic was applied to various JET plasmas in the recent hydrogen campaign where different hydrogen/deuterium mixtures and NBI configurations were used. The simulated neutral fluxes from the fast ion distributions were found to agree with the measured fluxes, reproducing the slowing-down profiles for different beam isotopes and energies and quantitatively estimating the fraction of hydrogen and deuterium fast ions.

    • Neutron activation technique has been widely used for the monitoring of neutron fluence at the Joint European Torus (JET) whereas it is foreseen to be employed at future fusion plants, such as ITER and DEMO. Neutron activation provides a robust tool for the measurement of neutron fluence in the complex environment encountered in a tokamak. However, activation experiments previously performed at JET showed that the activation foils used need to be calibrated in a real fusion environment in order to provide accurate neutron fluence data. Triggered by this challenge, an improved neutron activation method for the evaluation of neutron fluence at fusion devices has been developed. Activation assemblies similar to those used at JET were irradiated under 14 MeV neutrons at the Frascati Neutron Generator (FNG) reference neutron field. The data obtained from the calibration experiment were applied for the analysis of activation foil measurements performed during the implemented JET Deuterium-Deuterium (D-D) campaign. The activation results were compared against thermoluminescence measurements and a satisfactory agreement was observed. The proposed method provides confidence on the use of activation technique for the precise estimation of neutron fluence at fusion devices and enables its successful implementation in the forthcoming JET Deuterium-Tritium (D-T) campaign.

    • Real-time (RT) disruption prediction (DP) is essential to trigger mitigation actions that avoid irreversible damage to the devices. This paper deals with disruption mitigation alarms and performs the RT implementation of a probabilistic predictor. The RT implementation has been carried out with a fast controller with DAQ FPGA-based data acquisition devices corresponding to ITER catalogue (in particular, a reconfigurable Input/Output platform has been used). Up to three input signals have been used and relevant information for the prediction is extracted from the temporal and the frequency domains. The signals are read from the JET database. Then D/A conversions are carried out and used as inputs to the real time system. In this way, the whole process of digitization, data analysis and prediction is performed. The computation time for each prediction takes less than 200 mu s.

  • Long-Term Outcome of Mustard/Senning Correction for Transposition of the Great Arteries in Sweden and Denmark

    • BACKGROUND: The atrial switch operation, the Mustard or Senning operation, for the transposition of the great arteries (TGA) was introduced in the late 1950s and was the preferred surgery for TGA until the early 1990s. The Mustard and Senning operation involves extensive surgery in the atria and leaves the right ventricle as the systemic ventricle. The Mustard and Senning cohort is now well into adulthood and we begin to see the long-term outcome. METHODS AND RESULTS: All the 6 surgical centers that performed Mustard and Senning operations in Sweden and Denmark identified all operated TGA patients. Information about death was obtained in late 2007 and early 2008 from the Danish and Swedish Centralised Civil Register by using the patients' unique national Civil Registration Numbers. Four hundred sixty-eight patients undergoing the atrial switch operation were identified. Perioperative 30-day mortality was 20%, and 60% were alive after 30 years of follow-up. Perioperative mortality was significantly increased by the presence of a ventricular septal defect, left ventricular outflow obstruction, surgery early in the Mustard and Senning era. However, only pacemaker implantation is predictive of long-term outcome (hazard ratio, 1.90; 95% confidence interval, 1.05-3.46, P=0.04), once the TGA patient has survived the perioperative period. The risk of reoperation was correlated to the presence of associated defects and where the first Mustard/Senning operation was performed. CONCLUSIONS: The long-term survival of patients with Mustard and Senning correction for TGA appears to be primarily determined by factors in the right ventricle and tricuspid valve and not the timing of or the type of surgery in childhood. Cardiac function necessitating the implantation of a pacemaker is associated with an increase in mortality.
