  • Genetic studies of body mass index yield new insights for obesity biology.
  • 2015
  • Ingår i: Nature. - : Springer Science and Business Media LLC. - 0028-0836 .- 1476-4687. ; 518:7538, s. 197-401
  • Tidskriftsartikel (refereegranskat)abstract
    • Obesity is heritable and predisposes to many diseases. To understand the genetic basis of obesity better, here we conduct a genome-wide association study and Metabochip meta-analysis of body mass index (BMI), a measure commonly used to define obesity and assess adiposity, in up to 339,224 individuals. This analysis identifies 97 BMI-associated loci (P < 5 × 10(-8)), 56 of which are novel. Five loci demonstrate clear evidence of several independent association signals, and many loci have significant effects on other metabolic phenotypes. The 97 loci account for ∼2.7% of BMI variation, and genome-wide estimates suggest that common variation accounts for >20% of BMI variation. Pathway analyses provide strong support for a role of the central nervous system in obesity susceptibility and implicate new genes and pathways, including those related to synaptic function, glutamate signalling, insulin secretion/action, energy metabolism, lipid biology and adipogenesis.

  • Kinh Settlers in Viet Nam’s Northern Highlands : Natural Resources Management in a Cultural Context
  • 2004
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This study deals with the Kinh (or Viet) majority people who have migrated from the lowland Red River Delta to the mountainous areas of northern Viet Nam, and their adjustment to a new social and physical environment. Its aim is to analyse the social and cultural consequences for these migrants when settling in communities populated with people who belong to the national ethnic minorities (the Tày, the Giay and the Ngan peoples).Focus is on impacts in new interactive situations. The case is a special one in that it focuses on majority people's adaptation to minorities, and to a lesser extent vice versa. The Kinhs' view of how a "civilised" landscape ought to look like and how to utilise the natural resources therein demonstrated to be a central theme when discussing restructuring of the migrants' livelihood. This fact indicates the cultural dimension in the exploitation of the natural landscape and the reconstruction of the subsistence system. In the process of adaptation to a new social environment (as well as to a new physical one), social interactions between the Kinh and the ethnic minorities have proven to be important steps towards integration. One factor that turned out to be decisive in the integration process is the harmonising of life cycle ceremonies (especially weddings and funerals) between the Kinh and the minorities.New knowledge is accumulated locally, based on pooled experience. The study concerns how new knowledge on natural resources management is formed through a mixture of the migrants' knowledge from the Red River Delta and the minorities' knowledge of the local area. With a background in the delta area the Kinh brought the old knowledge of advanced wet rice production with them when migrating to the highlands. The facts show that the influence on the subsistence system has not been a one-way flow. That is, not only has the Kinh changed the minorities' agriculture system, but also the minorities' systems have had an impact on the Kinhs' system so that it now is more adapted to the conditions in the highlands.

  • Technology Dynamics in Multi-Technology Industries : Selection and Variety Creation through the Lens of Path Dependency and Path Generation
  • 2014
  • Licentiatavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • This thesis studies technological development in multi-technology industries, or industries characterized by co-existence of several technological alternatives that are used in the same or largely overlapping applications and markets. Although a lot of industries can be considered as multi-technological, they are understudied in the current research. Therefore, thesis purpose is to explain the processes of variety creation and selection in such industries. These two processes, considered in evolutionary economics literature as central for understanding technology dynamics, have specific characteristics in multi-technology industries and require a separate conceptualization. As theoretical tools to consider the processes of selection and variety creation, the concepts of path dependency and path generation are chosen, respectively. To account for the specifics of the selection process, the framework of multiple paths and path interaction is suggested. This framework acknowledges the existence of persistence and self-reinforcing mechanisms at the level of narrowly defined technologies as well as at the overall industry level. The specifics of the process of variety creation are found to be defined by the influence from the process of selection. Therefore, variety creation is studied by analyzing how path dependency affects path generation. Three types of such influence are suggested: negative, neutral and positive.Using the suggested concepts, the empirical case of the multi-technology lighting industry is considered with the help of patents analysis as well as the study of secondary data sources. As a result, several specific characteristics of the processes of selection and variety creation are revealed and explained. The process of selection is found to be present at two levels: the level of narrowly defined technologies and the overall industry level. While the former level is well in line with traditional conceptualizations of selection mechanisms, it is the latter one that represents the specifics of multi-technology industries. The overall industry-level selection is based on partial overlaps between technologies and helps to explain why selection is not fully based on technology competition, but allows for positive interaction between alternatives. For the process of variety creation, it is found to be influenced by the process of selection in three different ways. First, negative influence is found in the form of barriers imposed on the process of new technology development. Second, neutral influence is observed in the way how the established technology shaped some of the characteristics of the new ones. This type of influence explains the cumulative nature of variety creation in multi-technology industries. Third, positive influence is seen in the form of opportunities provided for new technology development, which also explains the on-going and industry-internal character of variety creation. In sum, both processes are found to be different from traditional conceptualizations, and the interplay between them helps to sustain the multi-technological character of such industries.

  • Decentralizing hydraulic society : Actor responses to institutional arrangements in Vietnam
  • 2013
  • Doktorsavhandling (övrigt vetenskapligt/konstnärligt)abstract
    • Irrigation decentralization has been launched worldwide with high expectations of improved governance, efficiency, and productivity through  democratic processes. However, there is widespread recognition that decentralization is, in reality, unlikely to bring about these positive outcomes. Poor implementation is widely blamed for these failures, but the current study argues that this is only partially true. Decentralization is usually treated as a technical way of power transfer that can be evaluated through quantitative  indicators. Much attention goes to searching solutions to constraints during implementation while less emphasis is on understanding of contexts, processes and the consequences of institutional changes. The common approach mainly focuses on local water users’ organizations but gives insufficient attention to the cross-level interactions between involved actors, their responses to reforms, and power relationships. This approach answers the question: what should be done, but not: why do constraints/limitations occur and why are such contradictory results obtained? In this way the approach itself diffuses some of the challenges in reality and the root of problems leading to surprise consequences and failures during  implementation. In this study, two irrigation systems in Vietnam were selected to examine the emerging decentralization processes, actors’ responses to the institutional changes, as well as the main options, challenges and constraints to decentralization. My ambition is also to further understand why constraints occur and have counterproductive effects. A mixed methods approach combining qualitative and quantitative methods, and cross-case analysis are applied. The database was built systematically, based on previous experience in this field, and participatory fieldwork. Decentralization is regarded as a power-laden process involving not just local organizations, but also state agencies, local authorities at different levels, state irrigation companies (IDMCs), and farmers. The study describes how institutional arrangements for decentralization have effects on the dynamics and relations of power between actors in multi-level governance, resulting in impacts on autonomy, accountability, participation, and incentives of involved actors. The analysis shows that the legal framework and several well-intended efforts for decentralization have been launched. However, outcomes in terms of power transfer and actors’ performance are not those expected. The current institutional arrangements have created major constraints: (i) imbalanced power relations, (ii) new resource allocation mechanisms creating dependency among actors, (iii) passiveness, vested interest and privilege within state agencies and IDMCs, and (iv) free-riding behavior and distrust among farmers. Prevailing conditions defy policy intentions by finding a new version of a centralized power structure at provincial level, in which power of state agencies and IDMCs are reinforced. Unexpected effects emerge on the autonomy, accountability, participation, and incentives of all involved actors. Decentralization of irrigation systems is a complex process, not merely a technical transfer of power. It is hard to achieve even with policy in place. All the complexities and implications of irrigation need to be thoroughly considered, with the dynamics in society included. Evaluations of decentralization based on production and economic terms are not enough. Impacts of institutional arrangements on power relations between actors, on their incentives, maneuver room in multi-level governance, and ability to comprehend and influence processes, need to be assessed thoroughly. Essential roles remain for central government when irrigation management is decentralized.

  • Atmospheric composition in the European Arctic and 30 years of the Zeppelin Observatory, Ny-Ålesund
  • 2022
  • Ingår i: Atmospheric Chemistry And Physics. - : Copernicus GmbH. - 1680-7316 .- 1680-7324. ; 22:5, s. 3321-3369
  • Tidskriftsartikel (refereegranskat)abstract
    • The Zeppelin Observatory (78.90∘ N, 11.88∘ E) is located on Zeppelin Mountain at 472 m a.s.l. on Spitsbergen, the largest island of the Svalbard archipelago. Established in 1989, the observatory is part of Ny-Ålesund Research Station and an important atmospheric measurement site, one of only a few in the high Arctic, and a part of several European and global monitoring programmes and research infrastructures, notably the European Monitoring and Evaluation Programme (EMEP); the Arctic Monitoring and Assessment Programme (AMAP); the Global Atmosphere Watch (GAW); the Aerosol, Clouds and Trace Gases Research Infrastructure (ACTRIS); the Advanced Global Atmospheric Gases Experiment (AGAGE) network; and the Integrated Carbon Observation System (ICOS). The observatory is jointly operated by the Norwegian Polar Institute (NPI), Stockholm University, and the Norwegian Institute for Air Research (NILU). Here we detail the establishment of the Zeppelin Observatory including historical measurements of atmospheric composition in the European Arctic leading to its construction. We present a history of the measurements at the observatory and review the current state of the European Arctic atmosphere, including results from trends in greenhouse gases, chlorofluorocarbons (CFCs) and hydrochlorofluorocarbons (HCFCs), other traces gases, persistent organic pollutants (POPs) and heavy metals, aerosols and Arctic haze, and atmospheric transport phenomena, and provide an outline of future research directions.

  • A principal component meta-analysis on multiple anthropometric traits identifies novel loci for body shape
  • 2016
  • Ingår i: Nature Communications. - : Springer Science and Business Media LLC. - 2041-1723. ; 7
  • Tidskriftsartikel (refereegranskat)abstract
    • Large consortia have revealed hundreds of genetic loci associated with anthropometric traits, one trait at a time. We examined whether genetic variants affect body shape as a composite phenotype that is represented by a combination of anthropometric traits. We developed an approach that calculates averaged PCs (AvPCs) representing body shape derived from six anthropometric traits (body mass index, height, weight, waist and hip circumference, waist-to-hip ratio). The first four AvPCs explain >99% of the variability, are heritable, and associate with cardiometabolic outcomes. We performed genome-wide association analyses for each body shape composite phenotype across 65 studies and meta-analysed summary statistics. We identify six novel loci: LEMD2 and CD47 for AvPC1, RPS6KA5/C14orf159 and GANAB for AvPC3, and ARL15 and ANP32 for AvPC4. Our findings highlight the value of using multiple traits to define complex phenotypes for discovery, which are not captured by single-trait analyses, and may shed light onto new pathways.

  • The African Food Crisis: Lessons from the Asian Green Revolution.
  • 2005
  • Samlingsverk (redaktörskap) (övrigt vetenskapligt/konstnärligt)abstract
    • This book is the outcome of a three-year project coordinated by a group of Swedish researchers and with collaborating scholars from Africa and Asia. It provides a comparative study between Asian agricultural development during the Green Revolution in food production and the current problematic agricultural situation in sub-Saharan Africa. Case studies of eight African and eight Asian countries (focusing on the early part of the Green Revolution) were carried out. Based on these studies the book presents a causal and explanatory model of Asian green revolutions and discusses why it has occurred in Asia, but not in Africa.

  • Protein-altering variants associated with body mass index implicate pathways that control energy intake and expenditure in obesity
  • 2018
  • Ingår i: Nature Genetics. - : Nature Publishing Group. - 1061-4036 .- 1546-1718. ; 50:1, s. 26-41
  • Tidskriftsartikel (refereegranskat)abstract
    • Genome-wide association studies (GWAS) have identified >250 loci for body mass index (BMI), implicating pathways related to neuronal biology. Most GWAS loci represent clusters of common, noncoding variants from which pinpointing causal genes remains challenging. Here we combined data from 718,734 individuals to discover rare and low-frequency (minor allele frequency (MAF) < 5%) coding variants associated with BMI. We identified 14 coding variants in 13 genes, of which 8 variants were in genes (ZBTB7B, ACHE, RAPGEF3, RAB21, ZFHX3, ENTPD6, ZFR2 and ZNF169) newly implicated in human obesity, 2 variants were in genes (MC4R and KSR2) previously observed to be mutated in extreme obesity and 2 variants were in GIPR. The effect sizes of rare variants are similar to 10 times larger than those of common variants, with the largest effect observed in carriers of an MC4R mutation introducing a stop codon (p.Tyr35Ter, MAF = 0.01%), who weighed similar to 7 kg more than non-carriers. Pathway analyses based on the variants associated with BMI confirm enrichment of neuronal genes and provide new evidence for adipocyte and energy expenditure biology, widening the potential of genetically supported therapeutic targets in obesity.
