    • By using a microfabricated nerve chip with integrated electrodes through which peripheral nerves can generate and make electrical connects, it should be possible to control a remote prosthesis by processing the detected nerve signals. In this study different artificial neural networks have been employed for classification of such complex patterns of signals. The signals were obtained from four electrodes detecting muscle activity in a rat hindlimb as a consequence of applied stimulus to the rat right hindpaw. These signals recorded at four different sites resembles a situation of a nerve chip with four electrodes, which implies that we might be able to use the same strategy when analyzing data from a four-electrode chip to obtain information from the nervous system. In this paper we will address the usefulness of different network topologies for analyzing measured in-vivo data from an implanted perforated nerve chip.

    • Background: Primary immunodeficiency diseases (PIDDs) are clinically and genetically heterogeneous disorders thus far associated with mutations in more than 300 genes. The clinical phenotypes derived from distinct genotypes can overlap. Genetic etiology can be a prognostic indicator of disease severity and can influence treatment decisions. Objective: We sought to investigate the ability of whole-exome screening methods to detect disease-causing variants in patients with PIDDs. Methods: Patients with PIDDs from 278 families from 22 countries were investigated by using whole-exome sequencing. Computational copy number variant (CNV) prediction pipelines and an exome-tiling chromosomal microarray were also applied to identify intragenic CNVs. Analytic approaches initially focused on 475 known or candidate PIDD genes but were nonexclusive and further tailored based on clinical data, family history, and immunophenotyping. Results: A likely molecular diagnosis was achieved in 110 (40%) unrelated probands. Clinical diagnosis was revised in about half (60/ 110) and management was directly altered in nearly a quarter (26/ 110) of families based on molecular findings. Twelve PIDD-causing CNVs were detected, including 7 smaller than 30 Kb that would not have been detected with conventional diagnostic CNV arrays. Conclusion: This high-throughput genomic approach enabled detection of disease-related variants in unexpected genes; permitted detection of low-grade constitutional, somatic, and revertant mosaicism; and provided evidence of a mutational burden in mixed PIDD immunophenotypes.

    • BACKGROUND: Single nucleotide polymorphisms (SNPs) in the promoter region of CRYAB gene have been associated with in multiple sclerosis. CRYAB gene, which encodes alpha B-crystallin (a member of small heat shock protein), was reported as a potential autoimmune target. In this study we investigated whether SNPs in the promoter region of CRYAB gene were also important in the etiology of Type 1 diabetes (T1D).METHODS: Genotyping of SNPs in the promoter region of CRYAB gene was performed in a Swedish cohort containing 444 T1D patients and 350 healthy controls. Three SNPs were included in this study: CRYAB-652 A>G (rs762550), -650 C>G (rs2234702) and -249 C > G (rs14133). Two SNPs (CRYAB-652 and -650) were not included in previous genome wide association studies.RESULTS: CRYAB-650 (rs2234702)*C allele was significantly more frequent in patients than in controls (OR = 1.48, Pc = 0.03). CRYAB-650*C allele was associated with IAA positivity (OR = 8.17, Pc < 0.0001) and IA-2A positivity (OR = 2.14, Pc = 0.005) in T1D patients. This association with IAA was amplified by high-risk HLA carrier state (OR = 10.6, P < 0.0001). No association was found between CRYAB-650 and other autoantibody positivity (GADA and ICA). CRYAB haplotypes were also associated with IAA and IA-2A positivity (highest OR = 2.07 and 2.11, respectively), these associations remain in high HLA-risk T1D patients.CONCLUSIONS: CRYAB-650 was associated with T1D in the Swedish cohort we studied. CRYAB-650*C allele might confers susceptibility to the development of T1D. CRYAB-650 was also associated with the development of IAA-positivity in T1D patients, especially in those carrying T1D high-risk HLA haplotypes.

    • A glass-mat-reinforced thermoplastic (GMT) composite material has been fabricated from randomly oriented continuous glass fibers embedded in a polypropylene matrix. The mechanical constitution of this composite has been characterized by using a linear viscoelastic micromechanically based material model. This material model has subsequently been implemented to several finite-element computer codes for analysis of structural components fabricated from polypropylene GMT. In this paper several example problems have been studied in order to determine the applicability of this modeling approach to predicting time-dependent deformations due to creep in GMT components. These example problems have been solved by utilizing two commercially available codes: ABAQUS, and ANSYS. Furthermore, results obtained with the codes have been compared to both analytic and experimental results, with varying degrees of success. The paper details these results for each of the example problems considered herein. © 2001 Elsevier Science Ltd. All rights reserved.
