  • Toxicity evaluation of PEDOT/biomolecular composites intended for neural communication electrodes

    • Electrodes coated with the conducting polymer poly(3,4-ethylene dioxythiophene) (PEDOT) possess attractive electrochemical properties for stimulation or recording in the nervous system. Biomolecules, added as counter ions in electropolymerization, could further improve the biomaterial properties, eliminating the need for surfactant counter ions in the process. Such PEDOT/biomolecular composites, using heparin or hyaluronic acid, have previously been investigated electrochemically. In the present study, their biocompatibility is evaluated. An agarose overlay assay using L929 fibroblasts, and elution and direct contact tests on human neuroblastoma SH-SY5Y cells are applied to investigate cytotoxicity in vitro. PEDOT: heparin was further evaluated in vivo through polymer-coated implants in rodent cortex. No cytotoxic response was seen to any of the PEDOT materials tested. The examination of cortical tissue exposed to polymer-coated implants showed extensive glial scarring irrespective of implant material (Pt:polymer or Pt). However, quantification of immunological response, through distance measurements from implant site to closest neuron and counting of ED1+ cell density around implant, was comparable to those of platinum controls. These results indicate that PEDOT: heparin surfaces were non-cytotoxic and show no marked difference in immunological response in cortical tissue compared to pure platinum controls.

  • Wire electronics and woven logic, as a potential technology for neuroelectronic implants

    • New strategies to improve neuron coupling to neuroelectronic implants are needed. In particular, to maintain functional coupling between implant and neurons, foreign body response like encapsulation must me minimized. Apart from modifying materials to mitigate encapsulation it has been shown that with extremely thin structures, encapsulation will be less pronounced. We here utilize wire electrochemical transistors (WECTs) using conducting polymer coated fibers. Monofilaments down to 10 μm can be successfully coated and weaved into complex networks with built in logic functions, so called textile logic. Such systems can control signal patterns at a large number of electrode terminals from a few addressing fibres. Not only is fibre size in the range where less encapsulation is expected but textiles are known to make successful implants because of their soft and flexible mechanical properties. Further, textile fabrication provides versatility and even three dimensional networks are possible. Three possible architectures for neuroelectronic systems are discussed. WECTs are sensitive to dehydration and materials for better durability or improved encapsulation is needed for stable performance in biological environments.

  • Substrate Control of Biotechnical Fedbatch Processes. Robustness and the Role of Adaptivity

    • Results from experiments on laboratory scale fedbatch processes are presented as well as analysis and design of the control system. The main reason for control is to track the drastic growth in feed demand during a cultivation. Variations in the amount and quality of the inocculum makes precalculated dosage schemes of limited value to obtain reproducible cultivation conditions. Two processes have been studied on a laboratory scale, production of bakers' yeast, and production of the enzyme salicylate hydroxylase using a strain of bacteria. Direct measurement was used to monitor the feed demand. A regulator structure is proposed based on an observer for the exponentially growing feed demand. It can be viewed as a modified PID regulator around a dosage scheme, but it is less sensitive to errors in the dosage scheme than conventional PID control. The a priori knowledge of the feed profile is further relaxed by introduction of adaptation of the growth rate parameter. The obtained non-linear control system has a simple structure and stability is garanteed for a wide range of initial values using the technique of Liapunov function. The linearized system is analysed in the frequency domain and the adaptation is shown to have negligible influence on the loop phase margin. The adaptive regulator is tested in simulation against real feed profiles and shows good results.

    • α-Galactosidase activity in Rhodothermus marinus is dependent on the composition of the growth media. A maximum of 46 μU g-1 cell dry weight was obtained using minimal medium with galactooligo- or polysaccharides as single carbon source. An enzyme hydrolysing both high and low molecular weight galacto-saccharides was partly purified from the cell fractions. The molecular weight was 200 kDa (native) and 50 kDa (monomer). It was optimally active at 85°C, with a half-life of 2 h at 75°C, and had a broad pH range (4-8).

    • The extensive use of rubber products, mainly tires, and the difficulties to recycle those products, has resulted in world wide environmental problems. Microbial devulcanisation is a promising way to increase the recycling of rubber materials. One obstacle is that several microorganisms tested for devulcanisation are sensitive to rubber additives. A way to overcome this might be to detoxify the rubber material with fungi prior to the devulcanisation. In this study, 15 species of white-rot and brown-rot fungi have been screened with regard to their capacity to degrade an aromatic model compound in the presence of ground waste tire rubber. The most effective fungus, Resinicium bicolor, was used for detoxification of rubber material. Increase in growth of the desulfurising bacterium Thiobacillus ferrooxidans in presence of the rubber treated with Resinicium bicolor compared to untreated rubber demonstrated that detoxification with fungi is possible.

    • Biotechnological leaching has been proposed as a suitable method for extraction of vanadium from spent catalysts and oil ash. In the biological leaching process, the vanadium(V) can be reduced to vanadium(IV), which is a less toxic and more soluble form of the vanadium. The present investigation showed that Acidithiobacillus ferrooxidans efficiently reduced vanadium(V) in the form of vanadium pentaoxide, to vanadyl(IV) ions, and tolerated high concentrations of vanadium(IV) and vanadium(V). A. ferrooxidans was compared with Acidithiobacillus thiooxidans, which has previously been utilized for vanadium leaching and reduction. Vanadium pentaoxide and sodium vanadate were used as model compounds. The results of this study indicate possibilities to develop an economical and technically feasible process for biotechnological vanadium recovery. (C) 2003 Elsevier Ltd. All rights reserved.
