  • TXNIP regulates peripheral glucose metabolism in humans

    • Background Type 2 diabetes mellitus ( T2DM) is characterized by defects in insulin secretion and action. Impaired glucose uptake in skeletal muscle is believed to be one of the earliest features in the natural history of T2DM, although underlying mechanisms remain obscure. Methods and Findings We combined human insulin/glucose clamp physiological studies with genome-wide expression profiling to identify thioredoxin interacting protein ( TXNIP) as a gene whose expression is powerfully suppressed by insulin yet stimulated by glucose. In healthy individuals, its expression was inversely correlated to total body measures of glucose uptake. Forced expression of TXNIP in cultured adipocytes significantly reduced glucose uptake, while silencing with RNA interference in adipocytes and in skeletal muscle enhanced glucose uptake, confirming that the gene product is also a regulator of glucose uptake. TXNIP expression is consistently elevated in the muscle of prediabetics and diabetics, although in a panel of 4,450 Scandinavian individuals, we found no evidence for association between common genetic variation in the TXNIP gene and T2DM. Conclusions TXNIP regulates both insulin-dependent and insulin- independent pathways of glucose uptake in human skeletal muscle. Combined with recent studies that have implicated TXNIP in pancreatic beta-cell glucose toxicity, our data suggest that TXNIP might play a key role in defective glucose homeostasis preceding overt T2DM.

    • There are several applications and studies in fundamental research, which require the detection of VUV light at cryogenic temperatures. For these applications, we have developed and successfully tested special designs of gaseous detectors with solid photocathodes able to operate at low temperatures: sealed gaseous detectors with MgF2 windows and windowless detectors. We have experimentally demonstrated that both primary and secondary (due to the avalanche multiplication inside liquids) scintillation light can be recorded by photosensitive gaseous detectors. The results of this work may allow one to significantly improve the operation of some noble liquid TPCs.

  • Proton halo effects in the 8B+64Zn collision around the Coulomb barrier

    • The 8B+64Zn reaction at 38.5 MeV has been studied at HIE-ISOLDE CERN to investigate proton halo effect on the reaction dynamics. For the first time it was used the only existing post-accelerated 8B beam. The measured elastic scattering angular distribution showed a small suppression of the Coulomb-nuclear interference peak, opposite to what observed for the one-neutron halo nucleus 11Be on the same target where a large suppression was observed instead. Inclusive angular and energy distributions of breakup fragments were also measured showing that, both, elastic and non-elastic breakup contribute. The presence of the additional Coulomb interactions halo-core and halo-target in 8B makes the reaction dynamics in this proton-halo nucleus different than the neutron-halo case. © 2023 Institute of Physics Publishing.     • BACKGROUND: Chronotherapy is one of the several approaches to increase efficacy and reduce toxicity of chemotherapy. In a phase II study in the second-line in patients with metastatic colorectal cancer (mCRC), we found that chronomodulated XELOX (XELOX(30Chron)) was a well-tolerated regimen with potentially reduced toxicity. PATIENTS AND METHODS: One hundred and forty-one patients with unresectable mCRC were enrolled in a randomized study comparing standard XELOX (XELOX(30)), arm A, and XELOX(30Chron), arm B-both with short-time infusion of oxaliplatin-with the primary aim of reducing overall toxicity. RESULTS: Overall toxicity grade 2-4 was 90% versus 85%, P = 0.47 and grade 3-4 was 31% versus 37%, P = 0.6 in arm A and B, respectively. We found no significant differences in median overall survival (17.6 versus 15.5 months; P = 0.068) and median progression-free survival (8.9 versus 8.8 months; P = 0.7). The incidence of grade 3 neuropathy was 16% in arm A and 19% in arm B (P = 0.7) after a cumulative dose of oxaliplatin of 1000 mg/m(2). CONCLUSION: XELOX(30Chron) does not reduce toxicity or improve efficacy. A 30-min infusion of oxaliplatin is safe and does not increase the severity of chronic neuropathy.
