  • Identifying therapeutic targets for cancer among 2074 circulating proteins and risk of nine cancers
  • 2024
  • Ingår i: Nature Communications. - : Springer Nature. - 2041-1723. ; 15:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Circulating proteins can reveal key pathways to cancer and identify therapeutic targets for cancer prevention. We investigate 2,074 circulating proteins and risk of nine common cancers (bladder, breast, endometrium, head and neck, lung, ovary, pancreas, kidney, and malignant non-melanoma) using cis protein Mendelian randomisation and colocalization. We conduct additional analyses to identify adverse side-effects of altering risk proteins and map cancer risk proteins to drug targets. Here we find 40 proteins associated with common cancers, such as PLAUR and risk of breast cancer [odds ratio per standard deviation increment: 2.27, 1.88-2.74], and with high-mortality cancers, such as CTRB1 and pancreatic cancer [0.79, 0.73-0.85]. We also identify potential adverse effects of protein-altering interventions to reduce cancer risk, such as hypertension. Additionally, we report 18 proteins associated with cancer risk that map to existing drugs and 15 that are not currently under clinical investigation. In sum, we identify protein-cancer links that improve our understanding of cancer aetiology. We also demonstrate that the wider consequence of any protein-altering intervention on well-being and morbidity is required to interpret any utility of proteins as potential future targets for therapeutic prevention.

  • Comparison of prognostic models to predict the occurrence of colorectal cancer in asymptomatic individuals : a systematic literature review and external validation in the EPIC and UK Biobank prospective cohort studies
  • 2019
  • Ingår i: Gut. - : BMJ Publishing Group Ltd. - 0017-5749 .- 1468-3288. ; 68, s. 672-683
  • Tidskriftsartikel (refereegranskat)abstract
    • OBJECTIVE: To systematically identify and validate published colorectal cancer risk prediction models that do not require invasive testing in two large population-based prospective cohorts.DESIGN: Models were identified through an update of a published systematic review and validated in the European Prospective Investigation into Cancer and Nutrition (EPIC) and the UK Biobank. The performance of the models to predict the occurrence of colorectal cancer within 5 or 10 years after study enrolment was assessed by discrimination (C-statistic) and calibration (plots of observed vs predicted probability).RESULTS: The systematic review and its update identified 16 models from 8 publications (8 colorectal, 5 colon and 3 rectal). The number of participants included in each model validation ranged from 41 587 to 396 515, and the number of cases ranged from 115 to 1781. Eligible and ineligible participants across the models were largely comparable. Calibration of the models, where assessable, was very good and further improved by recalibration. The C-statistics of the models were largely similar between validation cohorts with the highest values achieved being 0.70 (95% CI 0.68 to 0.72) in the UK Biobank and 0.71 (95% CI 0.67 to 0.74) in EPIC.CONCLUSION: Several of these non-invasive models exhibited good calibration and discrimination within both external validation populations and are therefore potentially suitable candidates for the facilitation of risk stratification in population-based colorectal screening programmes. Future work should both evaluate this potential, through modelling and impact studies, and ascertain if further enhancement in their performance can be obtained.

  • Association analyses of 249,796 individuals reveal 18 new loci associated with body mass index
  • 2010
  • Ingår i: Nature Genetics. - : Springer Science and Business Media LLC. - 1061-4036 .- 1546-1718. ; 42:11, s. 937-948
  • Tidskriftsartikel (refereegranskat)abstract
    • Obesity is globally prevalent and highly heritable, but its underlying genetic factors remain largely elusive. To identify genetic loci for obesity susceptibility, we examined associations between body mass index and ~2.8 million SNPs in up to 123,865 individuals with targeted follow up of 42 SNPs in up to 125,931 additional individuals. We confirmed 14 known obesity susceptibility loci and identified 18 new loci associated with body mass index (P < 5 × 10−8), one of which includes a copy number variant near GPRC5B. Some loci (at MC4R, POMC, SH2B1 and BDNF) map near key hypothalamic regulators of energy balance, and one of these loci is near GIPR, an incretin receptor. Furthermore, genes in other newly associated loci may provide new insights into human body weight regulation.

  • General and abdominal obesity and risk of esophageal and gastric adenocarcinoma in the European Prospective Investigation into Cancer and Nutrition
  • 2015
  • Ingår i: International Journal of Cancer. - : Wiley. - 0020-7136 .- 1097-0215. ; 137:3, s. 646-657
  • Tidskriftsartikel (refereegranskat)abstract
    • General obesity, as reflected by BMI, is an established risk factor for esophageal adenocarcinoma (EAC), a suspected risk factor for gastric cardia adenocarcinoma (GCC) and appears unrelated to gastric non-cardia adenocarcinoma (GNCC). How abdominal obesity, as commonly measured by waist circumference (WC), relates to these cancers remains largely unexplored. Using measured anthropometric data from 391,456 individuals from the European Prospective Investigation into Cancer and Nutrition (EPIC) study and 11 years of follow-up, we comprehensively assessed the association of anthropometric measures with risk of EAC, GCC and GNCC using multivariable proportional hazards regression. One hundred twenty-four incident EAC, 193 GCC and 224 GNCC were accrued. After mutual adjustment, BMI was unrelated to EAC, while WC showed a strong positive association (highest vs. lowest quintile HR=1.19; 95% CI, 0.63-2.22 and HR=3.76; 1.72-8.22, respectively). Hip circumference (HC) was inversely related to EAC after controlling for WC, while WC remained positively associated (HR=0.35; 0.18-0.68, and HR=4.10; 1.94-8.63, respectively). BMI was not associated with GCC or GNCC. WC was related to higher risks of GCC after adjustment for BMI and more strongly after adjustment for HC (highest vs. lowest quintile HR=1.91; 1.09-3.37, and HR=2.23; 1.28-3.90, respectively). Our study demonstrates that abdominal, rather than general, obesity is an indisputable risk factor for EAC and also provides evidence for a protective effect of gluteofemoral (subcutaneous) adipose tissue in EAC. Our study further shows that general obesity is not a risk factor for GCC and GNCC, while the role of abdominal obesity in GCC needs further investigation. What's new? While mainly general obesity, as measured by body mass index, has been investigated in relation to gastric and esophageal cancer, the effect of a large waist on these cancer sites is unknown. In this article, the authors report results of extensive analysis of measured anthropometry, including measures of general (BMI) and abdominal obesity (waist circumference), collected by the European Prospective Investigation into Cancer and Nutrition (EPIC). They show that general obesity is not a risk factor for esophageal and gastric cancer, while waist circumference strongly increases risk of esophageal cancer and may potentially be related to gastric cardia cancer.

  • Snapshot of the Hymenopteran fauna of Stora Karlsö
  • 2017
  • Ingår i: Entomologisk Tidskrift. - Uppsala. - 0013-886X. ; 138, s. 71-91
  • Recension (refereegranskat)abstract
    • Stora Karlsö is a small island close to Gotland in the Baltic Sea of which the Hymenopteranfauna has not been extensively studied before. In August 2014, a team of eight personscarried out an inventory of Hymenoptera, mainly the parasitoid wasps and sawflies, onthe island. Sampling was done with Malaise traps for a period of 22 days, complementedwith vegetation sweeping, branch shaking and opportunistic handpicking during a five daysojourn. As a result, about 200 species of parasitoid wasps and 14 sawflies are reported forStora Karlsö for the first time. Eleven species are reported as new to Sweden: The sawflyAthalia cornubiae Benson, 1931, the gasteruptiid Gasteruption opacum (Tournier, 1877),the diapriid Spilomicrus rufitarsis (Kieffer, 1911), the eulophid Entedonomphale bulgaricaBoyadzhiev & Triapitsyn, 2007, the braconids Bracon rozneri Papp, 1998 and Gnamptodondecoris (Förster, 1862), and the ichneumonids Bathythrix maculata (Hellén, 1957),Heterischnus filiformis (Gravenhorst, 1829), Lissonota picticoxis Schmiedeknecht, 1900,Mesochorus tipularius Gravenhorst, 1829, Ophion brevicornis Morley, 1915, and Plectochorusiwatensis (Uchida, 1928). Also the gasteruptiid Gasteruption opacum (Tournier,1877) is reported new to Sweden based on a record from inventory by NJ in 2013. Thisdemonstrates how the knowledge of Swedish biodiversity can be substantially augmentedby a short and intensive collecting expedition. We strongly recommend that other places inthe country be subjected to similar efforts.

  • Components of the metabolic syndrome and colorectal cancer risk : a prospective study
  • 2008
  • Ingår i: International Journal of Obesity. - : Springer Science and Business Media LLC. - 0307-0565 .- 1476-5497. ; 32:2, s. 304-314
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: To examine the relation of well-known factors of the metabolic syndrome (MetS) as well as related circulating factors, with risk of colorectal cancer. Methods: We performed a case control study of 306 colorectal cancer cases and 595 matched controls nested in the Northern Sweden Health and Disease Cohort. Levels of C-peptide, glycated haemoglobin (HbA1c), leptin and adiponectin were measured in cryopreserved samples. Body mass index (BMI), systolic and diastolic blood pressure and fasting and post-load plasma glucose, had been measured in a subcohort. Conditional logistic regression was used to calculate odds ratios (OR) of disease, including risk assessments for the MetS factors: obesity (BMI>30 kg m-2), hypertension (blood pressure 140/90 mmHg or use of anti-hypertensive drugs) and hyperglycaemia (fasting glucose 6.1 mmol l-1 or post-load glucose in capillary plasma 8.9 mmol l-1). Results: None of the studied variables were significantly associated with risk across quartiles. Presence of obesity, hypertension and hyperglycaemia significantly increased the risk of colorectal cancer; OR for three vs null factors was 2.57 (95% Confidence Interval [CI] 1.20–5.52; P trend=0.0021), as compared to a 30 to 70% increased risk for the factors in single. Similarly, top decile levels of C-peptide, HbA1c and leptin/adiponectin ratio were associated with an increased risk; ORs for top vs deciles 1–9 were 1.56 (95% CI 0.93–2.62; P=0.090), 1.83 (95% CI 1.00–3.36; P=0.051) and 1.50 (95% CI 0.83–2.71; P=0.18), respectively. Conclusions: Our study support the view that components of the MetS increase risk of colorectal cancer, and further suggests that only very high levels of metabolic factors confer an increased risk.

  • Interbead interactions within oligonucleotide functionalized ferrofluids suitable for magnetic biosensor applications
  • 2007
  • Ingår i: Journal of Physics D. - : IOP Publishing. - 0022-3727 .- 1361-6463. ; 40:5, s. 1320-1330
  • Tidskriftsartikel (refereegranskat)abstract
    • Interactions in oligonucleotide functionalized ferrofluids were investigated by measurements of frequency dependent complex magnetization in a superconducting quantum interference device. The ferrofluids consisted of aqueous suspensions of single-stranded oligonucleotide functionalized 130 nm sized magnetic beads, suitable for further use in magnetic biosensor applications based on changes in the Brownian relaxation behaviour of oligonucleotide-coated beads. The interbead interactions were found to depend strongly on the surface coverage by oligonucleotide molecules. At low surface coverage, aggregates of beads within the ferrofluid were formed, most likely due to interbead SS-crosslinking reactions. When the surface coverage increased, the entropic interbead repulsion arising from the thermal motion of oligonucleotide molecules began to stabilize the ferrofluid by preventing aggregation, and the crosslinking probability decreased. At a surface coverage, of ~40 oligonucleotides per bead, determined by a radioactive labelling analysis, an optimal configuration was obtained for which the ferrofluid behaved as consisting of almost non-interacting beads. At higher oligonucleotide functionalization degrees, the high coverage induced less thermal motion flexibility of the oligonucleotide chains which decreased the entropic repulsion effect. This in turn led to a higher crosslinking probability, as well as an increase in the hydrodynamic size of the beads. Thus, the ferrofluid sample of nearly non-interacting nature, in which an optimal degree of stabilization and the highest Brownian relaxation frequency are achieved in the low surface coverage region, may be considered as the most appropriate one for further use in magnetic biosensor applications, from both a functional and an economic point of view.

  • Increased cortical porosity in women with hip fracture
  • 2017
  • Ingår i: Journal of Internal Medicine. - : Wiley. - 0954-6820. ; 281:5, s. 496-506
  • Tidskriftsartikel (refereegranskat)abstract
    • Background. Hip fractures cause increased mortality and disability and consume enormous healthcare resources. Only 46% of hip fracture patients have osteoporosis at the total hip according to dual-energy X-ray absorptiometry (DXA) measurement. Cortical porosity increases with ageing and is believed to be important for bone strength. Objective. To investigate whether older women with hip fracture have higher cortical porosity than controls, and if so whether this difference is independent of clinical risk factors and areal bone mineral density (aBMD). Methods. From an ongoing population-based study, we identified 46 women with a prevalent X-rayverified hip fracture and 361 control subjects without any fractures. aBMD was measured with DXA. High-resolution peripheral quantitative computed tomography was used to measure bone microstructure at the standard (ultradistal) site and at 14% (distal) of the tibial length. Results. Women with a previous hip fracture had lower aBMD at the femoral neck (-11.8%) and total hip (-14.6%) as well as higher cortical porosity at the ultradistal (32.1%) and distal (29.3%) tibia compared with controls. In multivariable logistic regression analysis, with adjustment for covariates (age, height, weight, smoking, calcium intake, physical activity, walk time, oral glucocorticoids, parental hip fracture, rheumatoid arthritis, previous fall, current bisphosphonate treatment and femoral neck aBMD), cortical porosity at the ultradistal [odds ratio per standard deviation increase (95% confidence interval) 2.61 (1.77-3.85)] and distal [1.57 (1.12-2.20)] sites was associated with prevalent hip fracture. Conclusion. Cortical porosity was associated with prevalent hip fracture in older women independently of femoral neck aBMD and clinical risk factors.
