  • Clinical relevance of breast cancer-related genes as potential biomarkers for oral squamous cell carcinoma

    • Background: Squamous cell carcinoma of the oral cavity (OSCC) is a common cancer form with relatively low 5-year survival rates, due partially to late detection and lack of complementary molecular markers as targets for treatment. Molecular profiling of head and neck cancer has revealed biological similarities with basal-like breast and lung carcinoma. Recently, we showed that 16 genes were consistently altered in invasive breast tumors displaying varying degrees of aggressiveness. Methods: To extend our findings from breast cancer to another cancer type with similar characteristics, we performed an integrative analysis of transcriptomic and proteomic data to evaluate the prognostic significance of the 16 putative breast cancer-related biomarkers in OSCC using independent microarray datasets and immunohistochemistry. Predictive models for disease-specific (DSS) and/or overall survival (OS) were calculated for each marker using Cox proportional hazards models. Results: We found that CBX2, SCUBE2, and STK32B protein expression were associated with important clinicopathological features for OSCC (peritumoral inflammatory infiltration, metastatic spread to the cervical lymph nodes, and tumor size). Consequently, SCUBE2 and STK32B are involved in the hedgehog signaling pathway which plays a pivotal role in metastasis and angiogenesis in cancer. In addition, CNTNAP2 and S100A8 protein expression were correlated with DSS and OS, respectively. Conclusions: Taken together, these candidates and the hedgehog signaling pathway may be putative targets for drug development and clinical management of OSCC patients.

    • Genetic and epigenetic (DNA methylation, histone modifications, microRNA expression) crosstalk promotes inactivation of tumor suppressor genes or activation of oncogenes by gene loss/hypermethylation or duplications/hypomethylation, respectively. The 8p11-p12 chromosomal region is a hotspot for genomic aberrations (chromosomal rearrangements, amplifications and deletions) in several cancer forms, including breast carcinoma where amplification has been associated with increased proliferation rates and reduced patient survival. Here, an integrative genomics screen (DNA copy number, transcriptional and DNA methylation profiling) performed in 229 primary invasive breast carcinomas identified substantial coamplification of the 8p11-p12 genomic region and the MYC oncogene (8q24.21), as well as aberrant methylation and transcriptional patterns for several genes spanning the 8q12.1-q24.22 genomic region (ENPP2, FABP5, IMPAD1, NDRG1, PLEKHF2, RRM2B, SQLE, TAF2, TATDN1, TRPS1, VPS13B). Taken together, our findings suggest that MYC activity and aberrant DNA methylation may also have a pivotal role in the aggressive tumor phenotype frequently observed in breast carcinomas harboring 8p11-p12 regional amplification.

    • Genomic instability contributes to the neoplastic phenotype by deregulating key cancer-related genes, which in turn can have a detrimental effect on patient outcome. DNA amplification of the 8p11-p12 genomic region has clinical and biological implications in multiple malignancies, including breast carcinoma where the amplicon has been associated with tumor progression and poor prognosis. However, oncogenes driving increased cancer-related death and recurrent genetic features associated with the 8p11-p12 amplicon remain to be identified. In this study, DNA copy number and transcriptome profiling data for 229 primary invasive breast carcinomas (corresponding to 185 patients) were evaluated in conjunction with clinicopathological features to identify putative oncogenes in 8p11-p12 amplified samples. Illumina paired-end whole transcriptome sequencing and whole-genome SNP genotyping were subsequently performed on 23 samples showing high-level regional 8p11-p12 amplification to characterize recurrent genetic variants (SNPs and indels), expressed gene fusions, gene expression profiles and allelic imbalances. We now show previously undescribed chromothripsis-like patterns spanning the 8p11-p12 genomic region and allele-specific DNA amplification events. In addition, recurrent amplification-specific genetic features were identified, including genetic variants in the HIST1H1E and UQCRHL genes and fusion transcripts containing MALAT1 non-coding RNA, which is known to be a prognostic indicator for breast cancer and stimulated by estrogen. In summary, these findings highlight novel candidate targets for improved treatment of 8p11-p12 amplified breast carcinomas.

    • Soft magnetic composite (SMC) is considered as an alternative solution for core materials of inductors used in LCL filters in power converter applications. Performance of SMC inductors in damping a range of harmonics caused by voltage switching is an interesting topic. The method used in this paper is to test the inductors together with a 70 kW PWM inverter to examine the filtering effects for different harmonics and the harmonic losses as well as the temperature rise of the SMC inductors. The study is conducted with different inductor currents and switching frequencies (5 kHz, 10 kHz and 15 kHz). The harmonic spectra of voltages and currents as well as the harmonic losses calculated based on the measured values. A FE model is built to study distributions of magnetic fluxes, losses and temperature. The model is verified based on comparisons of computed and experimental results.

  • Physical restraint use in institutional care of old people in Sweden between 2000 and 2007

    • Background Physical restraint use is common in institutional care for old people and mainly used to prevent falls, despite that the preventive effect has been questioned. The aim was to investigate the use of physical restraints in Sweden, between 2000 and 2007. Method Data were collected from two comparable census surveys conducted in all institutional care units for old people in 2000 (n=3,804) and 2007 (n=2,970). Information on residents’ characteristics, physical restraint use and falls was collected by means of the Multi-Dimensional Dementia Assessment Scale (MDDAS). Results In 2000 16.2% (95% confidence interval (CI) 15.2-17.4) of the residents were restrained compared to 18.4% (95% CI 17.0-19.8) in 2007 (P = 0.018). Adjusting for resident’s characteristics showed that residents in 2007 were 1.2 (95% CI 1.0-1.5) times more likely to be physically restrained, relative to the residents in 2000. In 2007 the residents had been restrained longer, and a higher proportion were restrained for an unknown reason. Conclusion Physical restraint use increased between 2000 and 2007, residents were restrained for a longer time and a greater number were restrained for an unknown reason. It is possible that the changes in physical restraint use might be dependent on factors other than the residents’ characteristics.

The aim of the study was to investigate the use of physical restraints in Sweden in 2000 and 2007. Methods: Data were collected from two comparable census surveys conducted in all institutional care units for old people in 2000 (n = 3,669) and 2007 (n = 2,914). Information on residents' characteristics and physical restraint use was collected using the Multi-Dimensional Dementia Assessment Scale (MDDAS). Results: In 2000 16.0% (95% confidence interval (CI) 14.8%-17.2%) of the residents were restrained compared to 18.2% (95% CI 16.8%-19.6%) in 2007 (p = 0.017). Adjusting for residents' characteristics showed that residents in 2007 were more likely to be physically restrained, relative to the residents in 2000 (odds ratio (OR) 1.031, 95% CI 1.005-1.058, p = 0.017). In 2007 the residents had been restrained longer, and a higher proportion were restrained for unknown reasons. Conclusions: Physical restraint use is still common. Moreover, the findings of this study suggest a small increase (OR 1.031) in the prevalence of physical restraint use from 2000 to 2007 adjusted for residents' characteristics.

  • Unraveling the ground-state structure of BaZrO3 by neutron scattering experiments and first-principle calculations

    • The all-inorganic perovskite barium zirconate, BaZrO3, is a widely used material in a range of different technological applications. However, fundamental questions surrounding the crystal structure of BaZrO3, especially in regard to its ground-state structure, remain. While diffraction techniques indicate a cubic structure all the way down to T = 0 K, several first-principles phonon calculation studies based on density functional theory indicate an imaginary (unstable) phonon mode due to the appearance of an antiferrodistortive transition associated with rigid rotations of ZrO6 octahedra. The first-principles calculations are highly sensitive to the choice of exchange-correlation functional and, using six well-established functional approximations, we show that a correct description about the ground-state structure of BaZrO3 requires the use of hybrid functionals. The ground-state structure of BaZrO3 is found to be cubic, which is corroborated by experimental results obtained from neutron powder diffraction, inelastic neutron scattering, and neutron Compton scattering experiments.

  • Utilization of the natural variation in wood properties–a comparison between pilot plant and mill scale trials

    • Four different pulpwood classes of Norway spruce (Picea abies (L.) Karst). were tested in mill scale production of TMP at Hallsta Papermill. The objectives of the trial were to evaluate both rather extreme and more traditional roundwood classes in a full-scale TMP-plant with respect to pulp properties and energy requirements. The experiment was partly based on the results from the proceeding pilot plant trial within the EuroFiber project. The results generally confirmed the findings in the pilot plant trial. Young wood with a high proportion of juvenile wood gave a pulp with attractive optical properties, but somewhat lower tear index and higher energy consumption. The other extreme, sawmill chips from old trees of heavy dimensions, required lower energy input and gave higher tear index, but inferior optical properties. The two more traditional classes gave pulps with properties between the two extremes.
