  • Life Cycle Impacts of Road Infrastructure : Assessment of energy use and greenhouse gas emissions

    • Road infrastructure is essential in the development of human society, but has both negative and positive impacts. Large amounts of money and natural resources are spent each year on its construction, operation and maintenance. Obviously, there is potentially significantenvironmental impact associated with these activities. Thus the need for integration of life cycle environmental impacts of road infrastructure into transport planning is currently being widely recognised on international and national level. However certain issues, such as energy use and greenhouse gas (GHG) emissions from the construction, maintenance and operation of road infrastructure, are rarely considered during the current transport planning process in Sweden and most other countries.This thesis examined energy use and GHG emissions for the whole life cycle (construction, operation, maintenance and end-of-life) of road infrastructure, with the aim of improving transport planning on both strategic and project level. Life Cycle Assessment (LCA) was applied to two selected case studies: LCA of a road tunnel and LCA of three methods for asphalt recycling and reuse: hot in-plant, hot in-place and reuse as unbound material. The impact categories selected for analysis were Cumulative Energy Demand (CED) and Global Warming Potential (GWP). Other methods used in the research included interviews and a literature review.The results of the first case study indicated that the operational phase of the tunnel contributed the highest share of CED and GWP throughout the tunnel’s life cycle. Construction of concrete tunnels had much higher CED and GWP per lane-metre than construction of rocktunnels. The results of the second case study showed that hot in-place recycling of asphalt gave slightly more net savings of GWP and CED than hot in-plant recycling. Asphalt reuse was less environmentally beneficial than either of these alternatives, resulting in no net savings of GWP and minor net savings of CED. Main sources of data uncertainty identified in the two case-studies included prediction of future electricity mix and inventory data for asphalt concrete.This thesis contributes to methodological development which will be useful to future infrastructure LCAs in terms of inventory data collection. It presents estimated amounts of energy use and GHG emissions associated with road infrastructure, on the example of roadtunnel and asphalt recycling. Operation of road infrastructure and production of construction materials are identified as the main priorities for decreasing GHG emissions and energy use during the life cycle of road infrastructure. It was concluded that the potential exists for significant decreases in GHG emissions and energy use associated with the road transport system if the entire life cycle of road infrastructure is taken into consideration from the very start of the policy-making process.

    • Quantitative ultrasound of the heel captures heel bone properties that independently predict fracture risk and, with bone mineral density (BMD) assessed by X-ray (DXA), may be convenient alternatives for evaluating osteoporosis and fracture risk. We performed a meta-analysis of genome-wide association (GWA) studies to assess the genetic determinants of heel broadband ultrasound attenuation (BUA; n = 14 260), velocity of sound (VOS; n = 15 514) and BMD (n = 4566) in 13 discovery cohorts. Independent replication involved seven cohorts with GWA data (in silico n = 11 452) and new genotyping in 15 cohorts (de novo n = 24 902). In combined random effects, meta-analysis of the discovery and replication cohorts, nine single nucleotide polymorphisms (SNPs) had genome-wide significant (P < 5 x 10(-8)) associations with heel bone properties. Alongside SNPs within or near previously identified osteoporosis susceptibility genes including ESR1 (6q25.1: rs4869739, rs3020331, rs2982552), SPTBN1 (2p16.2: rs11898505), RSPO3 (6q22.33: rs7741021), WNT16 (7q31.31: rs2908007), DKK1 (10q21.1: rs7902708) and GPATCH1 (19q13.11: rs10416265), we identified a new locus on chromosome 11q14.2 (rs597319 close to TMEM135, a gene recently linked to osteoblastogenesis and longevity) significantly associated with both BUA and VOS (P < 8.23 x 10(-14)). In meta-analyses involving 25 cohorts with up to 14 985 fracture cases, six of 10 SNPs associated with heel bone properties at P < 5 x 10(-6) also had the expected direction of association with any fracture (P < 0.05), including three SNPs with P < 0.005: 6q22.33 (rs7741021), 7q31.31 (rs2908007) and 10q21.1 (rs7902708). In conclusion, this GWA study reveals the effect of several genes common to central DXA-derived BMD and heel ultrasound/DXA measures and points to a new genetic locus with potential implications for better understanding of osteoporosis pathophysiology.

  • Throughput of the WCDMA Random Access Channel

    • The uplink random access channel will be a key part of the 3rd generation wireless systems as it will be used for initial contact with base stations as well as for packet data transmission. In this paper, the throughput of preamble power ramping with 16 orthogonal channels currently adopted as the UTRA standard for random access is evaluated. It is shown that the tight power restrictions necessary in a CDMA system leads to degraded throughput at high loads. A modification to the scheme that significantly increases throughput for high loads is suggested.

    • Pili have only been discovered in the major Gram-positive pathogens in the past decade and they have been found to play an important role in colonisation and virulence. Pili have been shown to have many important functions including attachment to host tissues, mediating bacterial aggregation, biofilm formation and binding to proteins in the extracellular matrix. In this study, sortase-dependent pili have been found to be expressed on the surface of Finegoldia magna ALB8. F. magna is a Gram-positive anaerobic coccus that, primarily, is a commensal of the skin and mucous membranes, but has also been isolated from various clinical infection sites and is associated with soft-tissue abscesses, wound infections and bone and prosthetic joint infections. In this study, F. magna ALB8 was found to harbour three sortases at the pilus locus, two of which bear high similarity to class C sortases in Streptococcus pneumoniae. Two putative sortase-dependent pili proteins were found in the locus, with one being identified as the major pilus subunit, Fmp1 (F. magna pilus subunit 1), due to its high similarity to other major pilus proteins in prominent Gram-positive pathogens. The presence of sortase-dependent pili was confirmed experimentally through recombinant production of Fmp1 and production of antiserum. The Fmp1 antiserum was used in Western blot to show the presence of a high molecular weight protein ladder, characteristic of the presence of pili, in trypsin released cell wall surface proteins from F. magna. The presence of sortase-dependent pili was visually confirmed by transmission electron microscopy, which showed the binding of gold labelled anti-Fmp1 to individual pilus proteins along the pilus. Furthermore, pili could also be found to bind and interact with keratinocytes in the epidermal layer of human skin, suggesting an adhesive role for pili on F. magna. Our work represents the first description of pilus structures in F. magna. This discovery further elucidates F. magna physiology and allows for additional analysis of host-bacterial interactions in future studies.

    • A new control structure is proposed to improve user experience of wireless Internet. Information on radio bandwidth and queue length available in the radio network, close to the base station, is used in a proxy that resides between the Internet and the cellular system. The control algorithm in the proxy sets the window size according to event-triggered information on radio bandwidth changes and time-triggered information on the queue length at the wireless link. The mechanism is compared to TCP Reno in two simulation scenarios. The first scenario models a dedicated channel with stepwise changes in the bandwidth, while the second scenario models the High-speed Downlink Shared Channel recently introduced by 3GPP. The proposed mechanism significantly reduces the amount of buffer space needed in the radio network, and it also gives modest improvements to user response time and link utilization. Reduced buffering is particularly beneficial for third-party end-to-end real-time services such as voice, video, and online gaming.

  • Age-dependent impact of the major common genetic risk factor for COVID-19 on severity and mortality

    • BACKGROUND. There is considerable variability in COVID-19 outcomes among younger adults, and some of this variation may be due to genetic predisposition. METHODS. We combined individual level data from 13,888 COVID-19 patients (n = 7185 hospitalized) from 17 cohorts in 9 countries to assess the association of the major common COVID-19 genetic risk factor (chromosome 3 locus tagged by rs10490770) with mortality, COVID-19-related complications, and laboratory values. We next performed metaanalyses using FinnGen and the Columbia University COVID-19 Biobank. RESULTS. We found that rs10490770 risk allele carriers experienced an increased risk of all-cause mortality (HR, 1.4; 95% CI, 1.2-1.7). Risk allele carriers had increased odds of several COVID-19 complications: severe respiratory failure (OR, 2.1; 95% CI, 1.6-2.6), venous thromboembolism (OR, 1.7; 95% CI, 1.2-2.4), and hepatic injury (OR, 1.5; 95% CI, 1.2-2.0). Risk allele carriers age 60 years and younger had higher odds of death or severe respiratory failure (OR, 2.7; 95% CI, 1.8-3.9) compared with those of more than 60 years (OR, 1.5; 95% CI, 1.2-1.8; interaction, P = 0.038). Among individuals 60 years and younger who died or experienced severe respiratory failure, 32.3% were risk-variant carriers compared with 13.9% of those not experiencing these outcomes. This risk variant improved the prediction of death or severe respiratory failure similarly to, or better than, most established clinical risk factors. CONCLUSIONS. The major common COVID-19 genetic risk factor is associated with increased risks of morbidity and mortality, which are more pronounced among individuals 60 years or younger. The effect was similar in magnitude and more common than most established clinical risk factors, suggesting potential implications for future clinical risk management.

We also aimed to examine comorbidity between GAD and other psychiatric diagnoses, such as depression.
