  • Organ-specific metabolic pathways distinguish prediabetes, type 2 diabetes, and normal tissues

    • Environmental and genetic factors cause defects in pancreatic islets driving type 2 diabetes (T2D) together with the progression of multi-tissue insulin resistance. Mass spectrometry proteomics on samples from five key metabolic tissues of a cross-sectional cohort of 43 multi-organ donors provides deep coverage of their proteomes. Enrichment analysis of Gene Ontology terms provides a tissue-specific map of altered biological processes across healthy, prediabetes (PD), and T2D subjects. We find widespread alterations in several relevant biological pathways, including increase in hemostasis in pancreatic islets of PD, increase in the complement cascade in liver and pancreatic islets of PD, and elevation in cholesterol biosynthesis in liver of T2D. Our findings point to inflammatory, immune, and vascular alterations in pancreatic islets in PD that are hypotheses to be tested for potential contributions to hormonal perturbations such as impaired insulin and increased glucagon production. This multi-tissue proteomic map suggests tissue-specific metabolic dysregulations in T2D. © 2022 The Author(s)

  • Genetic regulation of IL1RL1 methylation and IL1RL1-a protein levels in asthma

    • Interleukin-1 receptor–like 1 (IL1RL1) is an important asthma gene. (Epi)genetic regulation of IL1RL1 protein expression has not been established. We assessed the association between IL1RL1 single nucleotide polymorphisms (SNPs), IL1RL1 methylation and serum IL1RL1-a protein levels, and aimed to identify causal pathways in asthma.Associations of IL1RL1 SNPs with asthma were determined in the Dutch Asthma Genome-wide Association Study cohort and three European birth cohorts, BAMSE (Children/Barn, Allergy, Milieu, Stockholm, an Epidemiological survey), INMA (Infancia y Medio Ambiente) and PIAMA (Prevention and Incidence of Asthma and Mite Allergy), participating in the Mechanisms of the Development of Allergy study. We performed blood DNA IL1RL1 methylation quantitative trait locus (QTL) analysis (n=496) and (epi)genome-wide protein QTL analysis on serum IL1RL1-a levels (n=1462). We investigated the association of IL1RL1 CpG methylation with asthma (n=632) and IL1RL1-a levels (n=548), with subsequent causal inference testing. Finally, we determined the association of IL1RL1-a levels with asthma and its clinical characteristics (n=1101).IL1RL1 asthma-risk SNPs strongly associated with IL1RL1 methylation (rs1420101; p=3.7×10−16) and serum IL1RL1-a levels (p=2.8×10−56). IL1RL1 methylation was not associated with asthma or IL1RL1-a levels. IL1RL1-a levels negatively correlated with blood eosinophil counts, whereas there was no association between IL1RL1-a levels and asthma.In conclusion, asthma-associated IL1RL1 SNPs strongly regulate IL1RL1 methylation and serum IL1RL1-a levels, yet neither these IL1RL1-methylation CpG sites nor IL1RL1-a levels are associated with asthma.

    • The mechanical behavior of a Fe80Zr10Cr10 alloy has been studied at ambient and high temperature. This Fe 80Zr10Cr10 alloy, whoose microstructure is formed by alternate lamellae of Laves phase and ferrite, constitutes a very simple example of an in situ CMA phase composite. The role of the Laves phase type was investigated in a previous study while the present work focuses on the influence of the microstructure length scale owing to a series of alloys cast at different cooling rates that display microstructures with Laves phase lamellae width ranging from ∼50 nm to ∼150 nm. Room temperature compression tests have revealed a very high strength (up to 2 GPa) combined with a very high ductility (up to 35%). Both strength and ductility increase with reduction of the lamella width. High temperature compression tests have shown that a high strength (900 MPa) is maintained up to 873 K. Microstructural study of the deformed samples suggests that the confinement of dislocations in the ferrite lamellae is responsible for strengthening at both ambient and high temperature. The microstructure scale in addition to CMA phase structural features stands then as a key parameter for optimization of mechanical properties of CMA in situ composites. © 2014 National Institute for Materials Science.

  • Unlocking the unsustainable rice-wheat system of Indian Punjab : Assessing alternatives to crop-residue burning from a systems perspective

    • Crop residue burning in Indian Punjab emits particulate matter with detrimental impacts on health, climate and that threaten agricultural production. Though legal and technological barriers to residue burning exist – and alternatives considered more profitable to farmers – residue burning continues. We review black carbon (BC) emissions from residue burning in Punjab, analyse social-ecological processes driving residue burning, and rice and wheat value-chains. Our aims are to a) understand system feedbacks driving agricultural practices in Punjab; b) identify systemic effects of alternatives to residue burning and c) identify companies and financial actors investing in agricultural production in Punjab. We find feedbacks locking the system into crop residue burning. The Government of India has greatest financial leverage and risk in the current system. Corporate stakeholders have little financial incentive to enact change, but sufficient stakes in the value chains to influence change. Agricultural policy changes are necessary to reduce harmful impacts of current practices, but insufficient to bringing about sustainability. Transformative changes will require crop diversification, circular business models and green financing. Intermediating financial institutions setting sustainability conditions on loans could leverage these changes. Sustainability requires the systems perspective we provide, to reconnect production with demand and with supporting environmental conditions.

  • Dynamics of fungal diversity and interactions with environmental elements in response to wheat straw biochar amended poultry manure composting

    • The fungal dynamics and its correlation with physicochemical and gaseous emission were investigated using metagenomics and Heat map illustrator (HEMI). Five different concentrations of wheat straw biochar (WSB) were applied to poultry manure (PM) and composted for 50 days; those without the WSB treatment were used as a control. The results revealed the dominant phyla to be Chytridiomycota, Mucoromycota, Ascomycota and Basidiomycota, while Batrachochytrium, Rhizophagus, Mucor, and Puccinia were the superior genera. In particular, the diversity of Chytridiomycota and Ascomycota was more abundant among all of the treatments. Overall, the diversity of the fungal species was correspondent, but relative abundance varied significantly among all of the composts. Principle Coordinate Analysis (PCoA) and Non-Metric Multi- Dimensional Scaling (NMDS) indicated that different concentrations of WSB applied treatments have significantly distinct fungal communities. In addition, correlation analyses of fungal interactions with environmental elements via HEMI also indicate a clear difference among the treatments. Ultimately, the relative abundance of fungal composition significantly influenced the PM compost treated by the WSB.

    • Biorefinery perceived as impeccable encouraging perspective of generating multiple bio-products from organic solid waste through integration of various relevant techniques. With the improved attractiveness of circular bio-economy as a greatly potential approach in view of social soaring energy demands and biodiversity conservation, biorefinery contributed as sustainable strategic mechanism for emerging circular bio-economy. Present study outlined a comprehensive review of the status for organic solid waste derived from agriculture, industry and urban toward to available biorefinery approach. In addition, evaluated techno-economic assessment and proposed the current bottleneck challenges and future perspective for sustainability of circular bio-economy in China. The results demonstrated that the diversity kinds of solid waste played an essential role in implement of biorefinery in circular bio-economy. Achieving sustainable circular bio-economy requires coordinated development of economic, environment and social. There is still a great gap between actual performance with expectation of biorefinery in bio-economy, comprehensive investigations and depth exploration were critical for sustainability.

  • Oxygen-Driven Growth Regulation and Defect Passivation in Chemical Vapor Deposited MoS 2 Monolayers

    • Due to the lowest formation energies, sulfur vacancies are inevitable in the vapor-phase chemical vapor deposition (CVD) of MoS2, which act as deep donors and induce midgap defect states, making the material intrinsically n-type. The postgrowth oxygen passivation of such defects has been the subject of a large number of recent studies because passivation of defects augments the photoluminescence quantum yield by several orders. In this study, by introducing an SiO2/Si wafer in close proximity to the growth substrate, we were able to supply trace oxygen in situ during the growth while simultaneously enabling chemisorption of oxygen at defect sites on the basal plane of large-area MoS2 monolayers. Low-temperature photoluminescence spectroscopy allowed us to distinguish clearly the nature of oxygen bonding in defective MoS2 grown with and without the trace oxygen. Chemisorption of oxygen enabled elimination of defect-related bound exciton emission at the near band edge transition of MoS2, leading to about 300% enhancement in the photoluminescence. We observed unusual splitting of the first-order A1g Raman mode in monolayer MoS2 films when the sulfur vacancies are not compensated by oxygen. The present study provides new experimental evidence to better distinguish between chemisorption and physisorption of oxygen and may serve as an effective way to tune the optical properties of van der Waals crystals during the large-area CVD process.
