    • The development of chemotherapy resistance is the most vital obstacle to clinical efficacy in gastric cancer (GC). The dysregulation of the Wnt/beta-catenin signaling pathway is critically associated with GC development and chemotherapy resistance. Ferroptosis is a form of regulated cell death, induced by an iron-dependent accumulation of lipid peroxides during chemotherapy. However, whether the Wnt/beta-catenin signaling directly controls resistance to cell death, remains unclear. Here, we show that the activation of the Wnt/beta-catenin signaling attenuates cellular lipid ROS production and subsequently inhibits ferroptosis in GC cells. The beta-catenin/TCF4 transcription complex directly binds to the promoter region of GPX4 and induces its expression, resulting in the suppression of ferroptotic cell death. Concordantly, TCF4 deficiency promotes cisplatin-induced ferroptosis in vitro and in vivo. Thus, we demonstrate that the aberrant activation of the Wnt/beta-catenin signaling confers ferroptosis resistance and suggests a potential therapeutic strategy to enhance chemo-sensitivity for advanced GC patients.

  • Investigation of combustion enhancement by ozone additive in CH(4)/air flames using direct laminar burning velocity measurements and kinetic simulations

    • The effect of ozone additive on the enhancement of the burning velocity for premixed methane-air flames is investigated by both experimental measurements and kinetic simulations. Laminar burning velocities with and without O(3) were directly measured using the Heat Flux method. The O(3) molecules were introduced into the system by a dielectric-barrier-discharge ozone generator installed in the O(2) gas line, which provided prompt control of on/off of the O(3) feed into the system, enabling a precise comparison of the measured burning velocity with and without ozone additives. Noticeable burning velocity enhancement was observed at off-stoichiometric conditions rather than stoichiometric conditions. With 3730 ppm O(3) additive in the oxidizer, experimental data shows similar to 8% burning velocity increase in fuel-rich mixtures and similar to 3.5% burning velocity increase for the stoichiometric mixture. With 7000 ppm ozone additive in the oxidizer, maximum similar to 16% burning velocity increase was observed at fuel-lean conditions while similar to 9.0% was found at fuel-rich conditions. An O(3) kinetic mechanism involving 16 elementary reactions together with the GRI-Mech 3.0 was composed and validated through CHEMKIN calculations, which gives good predictions of the burning velocities with and without O(3) additives. Extra O radicals contributed by O(3) molecules in the pre-heat zone initiate and accelerate the chain-branching reactions and consequently increase the burning velocity.     • Larger thalamic volume has been found in children with obsessive-compulsive disorder (OCD) and children with clinical-level symptoms within the general population. Particular thalamic subregions may drive these differences. The ENIGMA-OCD working group conducted mega- and meta-analyses to study thalamic subregional volume in OCD across the lifespan. Structural T1-weighted brain magnetic resonance imaging (MRI) scans from 2649 OCD patients and 2774 healthy controls across 29 sites (50 datasets) were processed using the FreeSurfer built-in ThalamicNuclei pipeline to extract five thalamic subregions. Volume measures were harmonized for site effects using ComBat before running separate multiple linear regression models for children, adolescents, and adults to estimate volumetric group differences. All analyses were pre-registered (https://osf.io/73dvy) and adjusted for age, sex and intracranial volume. Unmedicated pediatric OCD patients (<12 years) had larger lateral (d = 0.46), pulvinar (d = 0.33), ventral (d = 0.35) and whole thalamus (d = 0.40) volumes at unadjusted p-values <0.05. Adolescent patients showed no volumetric differences. Adult OCD patients compared with controls had smaller volumes across all subregions (anterior, lateral, pulvinar, medial, and ventral) and smaller whole thalamic volume (d = −0.15 to −0.07) after multiple comparisons correction, mostly driven by medicated patients and associated with symptom severity. The anterior thalamus was also significantly smaller in patients after adjusting for thalamus size. Our results suggest that OCD-related thalamic volume differences are global and not driven by particular subregions and that the direction of effects are driven by both age and medication status.

  • Heme oxygenase-1 derived carbon monoxide permits maturation of myeloid cells

    • Critical functions of the immune system are maintained by the ability of myeloid progenitors to differentiate and mature into macrophages. We hypothesized that the cytoprotective gas molecule carbon monoxide (CO), generated endogenously by heme oxygenases (HO), promotes differentiation of progenitors into functional macrophages. Deletion of HO-1, specifically in the myeloid lineage (Lyz-Cre:Hmox1flfl), attenuated the ability of myeloid progenitors to differentiate toward macrophages and decreased the expression of macrophage markers, CD14 and macrophage colony-stimulating factor receptor (MCSFR). We showed that HO-1 and CO induced CD14 expression and efficiently increased expansion and differentiation of myeloid cells into macrophages. Further, CO sensitized myeloid cells to treatment with MCSF at low doses by increasing MCSFR expression, mediated partially through a PI3K-Akt-dependent mechanism. Exposure of mice to CO in a model of marginal bone marrow transplantation significantly improved donor myeloid cell engraftment efficiency, expansion and differentiation, which corresponded to increased serum levels of GM-CSF, IL-1α and MCP-1. Collectively, we conclude that HO-1 and CO in part are critical for myeloid cell differentiation. CO may prove to be a novel therapeutic agent to improve functional recovery of bone marrow cells in patients undergoing irradiation, chemotherapy and/or bone marrow transplantation.

  • JAZF1 ameliorates age and diet-associated hepatic steatosis through SREBP-1c -dependent mechanism

    • JAZF zinc finger 1 (JAZF1) is involved in glucose and lipid metabolisms. However, its role in aging- and nutrient-related hepatic steatosis is unclear. In the current study, we demonstrated that JAZF1 expression was markedly down-regulated in obesity-associated mice and nonalcoholic fatty liver disease (NAFLD) patients. During aging, JAZF1 expression was gradually down-regulated in both C57BL/6 J and JAZF1-Tg mice. In JAZF1-Tg mice, body fat content and hepatosteatosis were protected from HFD-induced steatosis, and accompanied by decreased lipogenesis gene expression. The inhibitory effects of hepatic steatosis in JAZF1-Tg mice, however, were disappeared during aging. In hepatocytes, over-expression of JAZF1 attenuated, while knockdown of JAZF1 enhanced the expression of lipogenesis genes. The over-expressing of JAZF1 in hepatocytes displayed the increased adenosine monophosphate-activated protein kinase (AMPK) phosphorylation and decreased sterol regulatory element-binding protein 1c (SREBP-1c) expression. The roles of JAZF1 were partially attenuated by Compound C. Mechanistically, JAZF1 suppressed SREBP-1c expression through the inhibition of transcriptional activity of liver X receptor response elements (LXREs) in the SREBP-1c promoter. Data illustrate that JAZF1 may have a crucial role in the regulation of age and nutrient-associated hepatosteatosis through an AMPK/SREBP-1c-dependent mechanism.

  • Quantitative Estimation of the Climatic Effects of Carbon Transferred by International Trade

    • Carbon transfer via international trade affects the spatial pattern of global carbon emissions by redistributing emissions related to production of goods and services. It has potential impacts on attribution of the responsibility of various countries for climate change and formulation of carbon-reduction policies. However, the effect of carbon transfer on climate change has not been quantified. Here, we present a quantitative estimate of climatic impacts of carbon transfer based on a simple CO2 Impulse Response Function and three Earth System Models. The results suggest that carbon transfer leads to a migration of CO2 by 0.1-3.9 ppm or 3-9% of the rise in the global atmospheric concentrations from developed countries to developing countries during 1990-2005 and potentially reduces the effectiveness of the Kyoto Protocol by up to 5.3%. However, the induced atmospheric CO2 concentration and climate changes (e.g., in temperature, ocean heat content, and sea-ice) are very small and lie within observed interannual variability. Given continuous growth of transferred carbon emissions and their proportion in global total carbon emissions, the climatic effect of traded carbon is likely to become more significant in the future, highlighting the need to consider carbon transfer in future climate negotiations.

    • Fault diagnosis plays a vital role in the modern industry. In this research, a joint vibration signal analysis and deep learning method for fault diagnosis is proposed. The vibration signal analysis is a well-established technique for condition monitoring, and deep learning has shown its potential in fault diagnosis. In the proposed method, the time-frequency technique, named as S transform, is applied to transfer the vibration signals to images, and then an improved convolutional neural network (CNN) is applied to classify these images. The results show the proposed method has achieved the significant improvement.
