  • The ALMA Survey of 70 μm Dark High-mass Clumps in Early Stages (ASHES). VII. Chemistry of Embedded Dense Cores

    • We present a study of the chemistry toward 294 dense cores in 12 molecular clumps, using data obtained from the ALMA Survey of 70 μm dark High-mass clumps in Early Stages. We identified 97 protostellar cores and 197 prestellar core candidates, based on the detection of outflows and molecular transitions of high upper-energy levels (E u /k > 45 K). The detection rate of the N2D+ emission toward the protostellar cores is 38%, which is higher than 9% for the prestellar cores, indicating that N2D+ does not exclusively trace prestellar cores. The detection rates of the DCO+ emission are 35% for the prestellar cores and 49% for the protostellar cores, which are higher than those for N2D+, implying that DCO+ appears more frequently than N2D+ in both prestellar and protostellar cores. Both the N2D+ and DCO+ abundances appear to decrease from the prestellar to the protostellar stage. The DCN, C2D, and 13CS emission lines are rarely seen in the dense cores of early evolutionary phases. The detection rate of the H2CO emission toward dense cores is 52%, three times higher than that for CH3OH (17%). In addition, the H2CO detection rate, abundance, line intensities, and line widths increase with the core evolutionary status, suggesting that the H2CO line emission is sensitive to protostellar activity.

    • A functional C3-symmetric benzene-1,3,5-tricarboxamide-anchored diarylethene molecule (C3-BTE) is synthesized, which evolves into ordered superstructures with tunable morphology properties both in different solvent and via alternative light stimuli. The formation of rod-like assemblies from C3-BTE is reported and the self-assembly property of C3-BTE can be realized through alternative light stimuli in nonpolar solvents such as toluene.

  • Pathogenic antibody response to glucose-6-phosphate isomerase targets a modified epitope uniquely exposed on joint cartilage

    • ObjectivesTo identify the arthritogenic B cell epitopes of glucose-6-phosphate isomerase (GPI) and their association with rheumatoid arthritis (RA). MethodsIgG response towards a library of GPI peptides in patients with early RA, pre-symptomatic individuals and population controls, as well as in mice, were tested by bead-based multiplex immunoassays and ELISA. Monoclonal IgG were generated, and the binding specificity and affinity were determined by ELISA, gel size exclusion chromatography, surface plasma resonance and X-ray crystallography. Arthritogenicity was investigated by passive transfer experiments. Antigen-specific B cells were identified by peptide tetramer staining. ResultsPeptide GPI(293-307) was the dominant B cell epitope in K/BxN and GPI-immunised mice. We could detect B cells and low levels of IgM antibodies binding the GPI(293-307) epitopes, and high affinity anti-GPI(293-307) IgG antibodies already 7 days after GPI immunisation, immediately before arthritis onset. Transfer of anti-GPI(293-307) IgG antibodies induced arthritis in mice. Moreover, anti-GPI(293-307) IgG antibodies were more frequent in individuals prior to RA onset (19%) than in controls (7.5%). GPI(293-307)-specific antibodies were associated with radiographic joint damage. Crystal structures of the Fab-peptide complex revealed that this epitope is not exposed in native GPI but requires conformational change of the protein in inflamed joint for effective recognition by anti-GPI(293-307) antibodies. ConclusionsWe have identified the major pathogenic B cell epitope of the RA-associated autoantigen GPI, at position 293-307, exposed only on structurally modified GPI on the cartilage surface. B cells to this neo-epitope escape tolerance and could potentially play a role in the pathogenesis of RA.

  • Application of effective water-energy management based on digital twins technology in sustainable cities construction

    • This work aims to effectively resolve the problem of waterlogging in cities and manage water resources in sustainable cities. Digital Twins (DTs) technology was applied to the Urban Drainage System (UDS) and solves the modeling and scheduling problems of emergency drainage through the core model construction method of DTs. Firstly, the components of the UDS that are necessary in the process of building the model were listed according to the entity elements of the five-dimensional (5D) DTs model. Then, this work analyzed the essential data of the DTs of UDS and the Data Collection method according to the data elements of the 5D DTs model. Finally, the Multi-Level Dynamic Priority and Importance Scheduling (MDPIS) algorithm was proposed based on the Fixed Priority Scheduling (FPS) algorithm, which was verified by the simulation experiment. The experimental results indicated that the MDPIS algorithm showed significant performance in the rainfall scene with large fluctuations compared with the FPS algorithm. Specifically, the average improvement ratio was the highest, reaching 49.81%; the overall improvement rate was constant at about 48%. The operation results showed an apparent correlation between the catchment parameters and the overflow loss of the pumping station. The improved MDPIS algorithm can effectively utilize the water storage capacity and drainage capacity of the pumping station and reduce overflow losses during rainfall by dynamically adjusting the priority to solve the problem of urban inland inundation. The DTs-based UDS proposed here can effectively mitigate the overflow loss and improve the working efficiency of the pumping station cluster, promoting the development of Substainable Cities.
