  • Does FLT3 Mutation Impact Survival After Hematopoietic Stem Cell Transplantation for Acute Myeloid Leukemia? : A Center for International Blood and Marrow Transplant Research (CIBMTR) Analysis

    • BACKGROUND: Patients with FMS like tyrosine kinase 3 (FLT3)-mutated acute myeloid leukemia (AML) have a poor prognosis and are referred for early allogeneic hematopoietic stem cell transplantation (HCT). METHODS: Data from the Center for International Blood and Marrow Transplant Research (CIBMTR) were used to evaluate 511 adult patients with de novo AML who underwent HCT during 2008 through 2011 to determine whether FLT3 mutations had an impact on HCT outcomes. RESULTS: In total, 158 patients (31%) had FLT3 mutations. Univariate and multivariate analyses revealed an increased risk of relapse at 3 years in the FLT3 mutated group compared with the wild-type (WT) group (38% [95% confidence interval (CI), 30%-45%] vs 28% [95% CI, 24%-33%]; P = .04; relative risk, 1.60 [95% CI, 1.15-2.22]; P = .0048). However, FLT3 mutation status was not significantly associated with nonrelapse mortality, leukemia-free survival, or overall survival. Although more patients in the FLT3 mutated group died from relapsed primary disease compared with those in the WT group (60% vs 46%), the 3-year overall survival rate was comparable for the 2 groups (mutated group: 49%; 95% CI, 40%-57%; WT group: 55%, 95% CI, 50%-60%; P = .20). CONCLUSIONS: The current data indicate that FLT3 mutation status did not adversely impact overall survival after HCT, and about 50% of patients with this mutation who underwent HCT were long-term survivors.

  • Polyphenylsilole multilayers - an insight from X-ray electron spectroscopy and density functional theory

    • We present a combined investigation by means of X-ray photoelectron spectroscopy (XPS) and near-edge X-ray absorption fine-structure (NEXAFS) spectroscopy of condensed multilayers of two polyphenylsiloles, namely hexaphenylsilole (HPS) and tetraphenylsilole (TPS). Both compounds exhibit very similar spectroscopic signatures, whose interpretation is aided by density functional theory (DFT) calculations. High-resolution XPS spectra of the Si 2p and C 1s core levels of these multilayers indicate a positively charged silicon ion flanked by two negatively charged adjacent carbon atoms in the silole core of both molecules. This result is corroborated quantitatively by DFT calculations on isolated HPS (TPS) molecules, which show a natural bond orbital partial charge of + 1.67 e (+1.58 e) on the silicon and -0.34 e (-0.58 e) on the two neighbouring carbon atoms in the silole ring. These charges are conserved in direct contact with a Cu(111) substrate for films of submonolayer coverage, as evidenced by the Si 2p XPS data. The C K-edge NEXAFS spectra of HPS and TPS multilayers exhibit distinct and differing features. Their main characteristics reappear in the simulated spectra and are assigned to the different inequivalent carbon species in the molecule. The angle-dependent measurements hardly reveal any dichroism, i.e., the molecular p-systems are not uniformly oriented parallel or perpendicular with respect to the surface. Changes in the growth conditions of TPS, i.e., a reduction of the substrate temperature from 240 K to 80 K during deposition, lead to a broadening of both XPS and NEXAFS signatures, as well as an upward shift of the Si 2p and C 1s binding energies, indicative of a less ordered growth mode at low temperature.

  • A Parameter Selection Method for Wind Turbine Health Management through SCADA Data

    • Wind turbine anomaly or failure detection using machine learning techniques through supervisory control and data acquisition (SCADA) system is drawing wide attention from academic and industry While parameter selection is important for modelling a wind turbine's condition, only a few papers have been published focusing on this issue and in those papers interconnections among sub-components in a wind turbine are used to address this problem. However, merely the interconnections for decision making sometimes is too general to provide a parameter list considering the differences of each SCADA dataset. In this paper, a method is proposed to provide more detailed suggestions on parameter selection based on mutual information. First, the copula is proven to be capable of simplifying the estimation of mutual information. Then an empirical copula-based mutual information estimation method (ECMI) is introduced for application. After that, a real SCADA dataset is adopted to test the method, and the results show the effectiveness of the ECMI in providing parameter selection suggestions when physical knowledge is not accurate enough.

    • The Rosetta spacecraft spent similar to 2 years orbiting comet 67P/Churyumov-Gerasimenko, most of it at distances that allowed surface characterization and monitoring at submeter scales. From December 2014 to June 2016, numerous localized changes were observed, which we attribute to cometary-specific weathering, erosion, and transient events driven by exposure to sunlight and other processes. While the localized changes suggest compositional or physical heterogeneity, their scale has not resulted in substantial alterations to the comet's landscape. This suggests that most of the major landforms were created early in the comet's current orbital configuration. They may even date from earlier if the comet had a larger volatile inventory, particularly of CO or CO2 ices, or contained amorphous ice, which could have triggered activity at greater distances from the Sun.

    • Background Human reproduction depends on the fusion of a mature oocyte with a sperm cell to form a fertilized egg. The genetic events that lead to the arrest of human oocyte maturation are unknown. Methods We sequenced the exomes of five members of a four-generation family, three of whom had infertility due to oocyte meiosis I arrest. We performed Sanger sequencing of a candidate gene, TUBB8, in DNA samples from these members, additional family members, and members of 23 other affected families. The expression of TUBB8 and all other β-tubulin isotypes was assessed in human oocytes, early embryos, sperm cells, and several somatic tissues by means of a quantitative reverse-transcriptase-polymerase-chain-reaction assay. We evaluated the effect of the TUBB8 mutations on the assembly of the heterodimer consisting of one α-tubulin polypeptide and one β-tubulin polypeptide (α/β-tubulin heterodimer) in vitro, on microtubule architecture in HeLa cells, on microtubule dynamics in yeast cells, and on spindle assembly in mouse and human oocytes. Results We identified seven mutations in the primate-specific gene TUBB8 that were responsible for oocyte meiosis I arrest in 7 of the 24 families. TUBB8 expression is unique to oocytes and the early embryo, in which this gene accounts for almost all the expressed β-tubulin. The mutations affect chaperone-dependent folding and assembly of the α/β-tubulin heterodimer, disrupt microtubule behavior on expression in cultured cells, alter microtubule dynamics in vivo, and cause catastrophic spindle-assembly defects and maturation arrest on expression in mouse and human oocytes. Conclusions TUBB8 mutations have dominant-negative effects that disrupt microtubule behavior and oocyte meiotic spindle assembly and maturation, causing female infertility.     • Herein, we reported the designed synthesis of three isostructural three-dimensional covalent organic frameworks (3D COFs) with -H, -Me, or -F substituents, which have similar crystallinity and topology. Their crystal structures were determined by continuous rotation electron diffraction (cRED), and all three 3D COFs were found to adopt a fivefold interpenetrated pts topology. More importantly, the resolution of these cRED datasets reached up to 0.9-1.0 angstrom, enabling the localization of all non-hydrogen atomic positions in a COF framework directly by 3D ED techniques for the first time. In addition, the precise control of the pore environments through the use of different functional groups led to different selectivities for CO2 over N-2. We have thus confirmed that polycrystalline COFs can be definitely studied to the atomic level as other materials, and this study should also inspire the design and synthesis of 3D COFs with tailored pore environments for interesting applications.

    • The combination of conventional transition-metal-catalyzed coupling (2e(-) process) and photoredox catalysis (1e(-) process) has emerged as a powerful approach to catalyze difficult cross-coupling reactions under mild reaction conditions. Reported is a palladium carbodicarbene (CDC) complex that mediates both a Suzuki-Miyaura coupling and photoredox catalysis for C-N bond formation upon visible-light irradiation. These two catalytic pathways can be combined to promote both conventional transition-metal-catalyzed coupling and photoredox catalysis to mediate C-H arylation under ambient conditions with a single catalyst in an efficient one-pot process.

    • The surface-enhanced Raman scattering (SERS) spectroscopy and inelastic electron tunneling spectroscopy (IETS) are employed to study trans-1,2-bis (4-pyridyl) ethylene (BPE)/gold system. Both junction and complex forms are considered for the SERS simulations. It is predicted that the peak at 1581 cm-1 is more intense in the junction forms than that in the complex forms. Time dependent density functional theory calculations show that the relative intensity is mainly controlled by the excitation energy derivative respect to the normal modes, and the total intensity is governed by the excitation energy of the excited states. The CH bending modes dominate the IET spectra when BPE adsorbed on the flat gold surfaces. While, the pyridyl ring deformation modes are more active when BPE adsorbed on the edge of the gold clusters. For BPE adsorbed on the tip of gold clusters, the pyridyl ring and CC stretching modes show significant contribution to the IET spectra.

  • The Mediation Role of Self-Esteem for Self-Stigma on Quality of Life for People With Schizophrenia : A Retrospectively Longitudinal Study

    • Background: Among patients with schizophrenia, there is evidence of a negative association between self-stigma and subjective quality of life (SQoL), and self-esteem was an important mediator in the association. We attempted to use a longitudinal study to investigate the aforementioned mediation on a sample with schizophrenia.Methods: We used longitudinal data retrieved from medical records of a psychiatric centre between June 2014 and December 2015. In the data, we retrieved information of self-stigma using the Self-Stigma Scale - Short; SQoL, using the WHO questionnaire on the Quality of Life - Short Form; and self-esteem, using the Rosenberg Self-Esteem Scale. All the measures were evaluated five times. Linear mixed-effect models accompanied by Sobel tests were used to tackle the mediating effects.Results: Data from 74 patients (57 males) with schizophrenia were eligible for analysis; their mean (SD) age was 39.53 (10.67); mean age of onset was 22.95 (8.38). Self-esteem was a mediator for patients in physical (p = .039), psychological (p = .003), and social SQoL (p = .004), but not in environment SQoL (p = .051).Conclusion: Based on our findings, mental health professionals could tailor different programs to patients with schizophrenia, such as self-stigma reduction and self-esteem improvement programs. However, treatment as a whole should be sensitive to both self-stigma and self-esteem. Also, we should consider individuals' health and wellbeing from social perspectives of disability rather than the medical model of disability emphasising symptoms and medications.

  • Use of software-defined radio receivers in two-way satellite time and frequency transfers for UTC computation

    • Two-way satellite time and frequency transfer (TWSTFT) is a primary technique for the generation of coordinated universal time (UTC). About 20 timing laboratories around the world continuously operate TWSTFT using satellite time and ranging equipment (SATRE19) modems for remote time and frequency comparisons in this context. The precision of the SATRE TWSTFT as observed today is limited by an apparent daily variation pattern (diurnal) in the TWSTFT results. The observed peak-to-peak variation have been found as high as 2 ns in some cases. Investigations into the origins of the diurnals have so far provided no complete understanding about the cause of the diurnals. One major contributor to the diurnals, however, could be related to properties of the receive part in the modem. In 2014 and 2015, it was demonstrated that bypassing the receive part and the use of software-defined radio (SDR) receivers in TWSTFT ground stations (SDR TWSTFT) instead could considerably reduce both the diurnals and the measurement noise. In 2016, the International Bureau of Weights and Measures (BIPM) and the Consultative Committee for Time and Frequency (CCTF) working group (WG) on TWSTFT launched a pilot study on the application of SDR receivers in the TWSTFT network for UTC computation. The first results of the pilot study were reported to the CCTF WG on TWSTFT annual meeting in May 2017, demonstrating that SDR TWSTFT shows superior performance compared to that of SATRE TWSTFT for practically all links between participating stations. In particular, for continental TWSTFT links, in which the strongest diurnals appear, the use of SDR TWSTFT results in a significant suppression of the diurnals by a factor of between two and three. For the very long inter-continental links, e.g. the Europe-to-USA links where the diurnals are less pronounced, SDR TWSTFT achieved a smaller but still significant gain of 30%. These findings are supported by an evaluation of some of the links with an alternate technique based on GPS signals (GPS IPPP) as reported in this paper. Stimulated by these results, the WG on TWSTFT prepared a recommendation for the 21st CCTF meeting, which proposed the introduction of SDR TWSTFT in UTC generation. With CCTF approval of the recommendation, a roadmap was developed for the implementation of SDR TWSTFT in UTC generation. In accordance with the roadmap, most of the stations that participated in the pilot study have updated the SDR TWSTFT settings to facilitate the use of SDR TWSTFT data in UTC generation. In addition, the BIPM conducted a final evaluation to validate the long-term stability of SDR TWSTFT links, made test runs using the BIPM standard software for the calculation of UTC, now including SDR TWSTFT data, and started to calculate SDR TWSTFT time links as backup from October 2017. The use of SDR TWSTFT in UTC generation will begin in 2018.
