  • Measuring attrition resistance of oxygen carrier particles for chemical looping combustion with a customized jet cup

    • A customized jet cup for measuring attrition resistance of oxygen carrier particles for chemical looping combustion has been constructed and used to evaluate 25 different material samples, all of which previously have been subject to continuous operation in chemical looping reactors at Chalmers University of Technology. The effect of continuous operation has been assessed by comparing attrition behavior of fresh particles with that of used ones. It is concluded that the correlation between the jet cup tests and operational experience is robust, and that there is always considerable difference in attrition resistance between fresh and used particles of the same batch. Composite materials with NiO or Fe2O3 as active phase and Al2O3-, NiAl2O4- or MgAl2O4-based support and materials based on the CaMnO3-δ perovskite structure typically had high attrition resistance, which improved further following operation with fuel. Combined (FexMn1-x)2O3 oxides and all materials containing smaller or larger amounts of either CuO or ZrO2 experienced reduced attrition resistance during operation with fuel, and usually also had low attrition resistance to begin with. Fresh particles of the commonly used oxygen carrier ilmenite had reasonably high attrition resistance, while ilmenite that had been subject to chemical looping combustion of natural gas showed higher rate of attrition. No strong correlation between the commonly used crushing strength index and attrition resistance measured with jet cup could be established, but it was clear that particles with a crushing strength above 2N were much more likely to have high attrition resistance compared to softer particles. As compared to crushing strength, the jet cup testing was better correlated to attrition in actual operation.

    • It is challenging to measure dietary exposure with techniques that are both accurate and applicable to free-living individuals. We performed a cross-over intervention, with 24 healthy individuals, to capture the acute metabolic response of a cereal breakfast (CB) and an egg and ham breakfast (EHB). Fasting and postprandial urine samples were analyzed using H-1 nuclear magnetic resonance (NMR) spectroscopy and multivariate data analysis. Metabolic profiles were distinguished in relation to ingestion of either CB or EHB. Phosphocreatine/creatine and citrate were identified at higher concentrations after consumption of EHB. Beverage consumption (i.e., tea or coffee) could clearly be seen in the data. 2-furoylglycine and 5-hydroxymethyl-2-furoic acid - potential biomarkers for coffee consumption were identified at higher concentrations in coffee drinkers. Thus H-1 NMR urine metabolomics is applicable in the characterization of acute metabolic fingerprints from meal consumption and in the identification of metabolites that may serve as potential biomarkers.   • Nutritional metabolomics: Postprandial response of meals relating to vegan, lacto-ovo vegetarian, and omnivore diets

    • Metabolomics provide an unbiased tool for exploring the modulation of the human metabolome in response to food intake. This study applied metabolomics to capture the postprandial metabolic response to breakfast meals corresponding to vegan (VE), lacto ovo-vegetarian (LOV), and omnivore (OM) diets. In a cross over design 32 healthy volunteers (16 men and 16 females) consumed breakfast meals in a randomized order during three consecutive days. Fasting and 3 h postprandial serum samples were collected and then subjected to metabolite profiling using1 H-nuclear magnetic resonance (NMR) spectroscopy. Changes in concentration of identified and discriminating metabolites, between fasting and postprandial state, were compared across meals. Betaine, choline, and creatine displayed higher concentration in the OM breakfast, while 3-hydroxyisobutyrate, carnitine, proline, and tyrosine showed an increase for the LOV and unidentified free fatty acids displayed a higher concentration after the VE breakfast. Using1 H NMR metabolomics it was possible to detect and distinguish the metabolic response of three different breakfast meals corresponding to vegan, lacto-ovo vegetarian, and omnivore diets in serum. © 2018 by the authors.   • The H-1 NMR serum metabolomics response to a two meal challenge: a cross-over dietary intervention study in healthy human volunteers

    • Background: Metabolomics represents a powerful tool for exploring modulation of the human metabolome in response to food intake. However, the choice of multivariate statistical approach is not always evident, especially for complex experimental designs with repeated measurements per individual. Here we have investigated the serum metabolic responses to two breakfast meals: an egg and ham based breakfast and a cereal based breakfast using three different multivariate approaches based on the Projections to Latent Structures framework. Methods: In a cross over design, 24 healthy volunteers ate the egg and ham breakfast and cereal breakfast on four occasions each. Postprandial serum samples were subjected to metabolite profiling using H-1 nuclear magnetic resonance spectroscopy and metabolites were identified using 2D nuclear magnetic resonance spectroscopy. Metabolic profiles were analyzed using Orthogonal Projections to Latent Structures with Discriminant Analysis and Effect Projections and ANOVA-decomposed Projections to Latent Structures. Results: The Orthogonal Projections to Latent Structures with Discriminant Analysis model correctly classified 92 and 90% of the samples from the cereal breakfast and egg and ham breakfast, respectively, but confounded dietary effects with inter-personal variability. Orthogonal Projections to Latent Structures with Effect Projections removed inter-personal variability and performed perfect classification between breakfasts, however at the expense of comparing means of respective breakfasts instead of all samples. ANOVA-decomposed Projections to Latent Structures managed to remove inter-personal variability and predicted 99% of all individual samples correctly. Proline, tyrosine, and N-acetylated amino acids were found in higher concentration after consumption of the cereal breakfast while creatine, methanol, and isoleucine were found in higher concentration after the egg and ham breakfast. Conclusions: Our results demonstrate that the choice of statistical method will influence the results and adequate methods need to be employed to manage sample dependency and repeated measurements in cross-over studies. In addition, H-1 nuclear magnetic resonance serum metabolomics could reproducibly characterize postprandial metabolic profiles and identify discriminatory metabolites largely reflecting dietary composition.

  • From Here to Sustainability – Is the Lisbon/Göteborg agenda delivering?

    • Executive Summary The European Councils held in Lisbon (2000) and in Göteborg (2001) gave the Union a new direction by establishing a long term strategy with sustainable development as the overarching objective. Sustainable development means, in this context, goals for economic, social and environmental policy, which are both mutually consistent and capable of delivering enhanced economic growth. To assure progress towards an agreed range of targets, the open method of coordination (OMC) has been adopted as the process for the implementation of the strategy. The strategy for sustainable development is a long-term one and, although the deadline originally set for the Lisbon agenda was 2010, it is clear that sustainable development has a much longer time-horizon and also that there is a global dimension to sustainable development, not just an EU one. In the run up to the mid-term review of the Lisbon strategy, this report by the European Panel for Sustainable Development, EPSD, offers an assessment of the EU approach to sustainable development. The report is based on official documents, research reports and background reports prepared by researchers from different disciplines. It concentrates on the EU-15 Member States, because the ten new members that acceded to the EU in May 2004 have not (yet!) been subject to the same commitments in relation to sustainable development. However, in future work by the EPSD, it is anticipated that the coverage will be extended to embrace all 25 Member States. The report starts with a discussion on the political process, followed by an examination of the economic, social and environmental dimensions of the strategy, of the potential of new technologies, and of the results delivered by the Member States. The final chapters include discussions on impact assessment and the global dimension of sustainable development. The focus of the report is on: − The integration of the three dimensions of sustai nable development and the policies that affect them into one coherent strategy − The implementation of the strategy through the open method of co-ordination The main messages of the report are that it is vital to: • Maintain the original commitment to sustainable development as the overarching objective of the Lisbon strategy and improve the co-ordination between the three pillars of the strategy: the economic, social and environmental dimensions [...]

    • Huntington's disease is a severe progressive neurodegenerative disorder caused by a CAG-expansion in the IT15 gene, which encodes huntingtin. The disease primarily affects the neostriatum and cerebral cortex and also associates with increased incidence of diabetes. Here, we show that mutant huntingtin disrupts intracellular transport and insulin secretion by direct interference with microtubular beta-tubulin. We demonstrate that mutant huntingtin impairs glucose-stimulated insulin secretion in insulin-producing beta-cells, without altering stored levels of insulin. Using VSVG-YFP, we show that mutant huntingtin retards post-Golgi transport. Moreover, we demonstrate that the speed of insulin vesicle trafficking is reduced. Using immunoprecipitation of mutant and wild-type huntingtin in combination with mass spectrometry, we reveal an enhanced and aberrant interaction between mutant huntingtin and beta-tubulin, implying the underlying mechanism of impaired intracellular transport. Thus, our findings have revealed a novel pathogenetic process by which mutant huntingtin may disrupt hormone exocytosis from beta-cells and possibly impair vesicular transport in any cell that expresses the pathogenic protein.

  • Unexpected early extinction of the European pond turtle (Emys orbicularis) in Sweden and climatic impact on its Holocene range

    • Using ancient DNA sequences of subfossil European pond turtles (Emys orbicularis) from Britain, Central and North Europe and accelerator mass spectrometry radiocarbon dating for turtle remains from most Swedish sites, we provide evidence for a Holocene range expansion of the pond turtle from the southeastern Balkans into Britain, Central Europe and Scandinavia, according to the 'grasshopper pattern' of Hewitt. Northeastern Europe and adjacent Asia were colonized from another refuge located further east. With increasing annual mean temperatures, pond turtles reached southern Sweden approximately 9800 years ago. Until approximately 5500 years ago, rising temperatures facilitated a further range expansion up to ostergotland, Sweden (approximately 58 degrees 30'N). However, around 5500 years ago pond turtle records suddenly terminate in Sweden, some 1500 years before the Holocene thermal maximum ended in Scandinavia and distinctly earlier than previously thought. This extinction coincides with a temporary cooling oscillation during the Holocene thermal maximum and is likely related to lower summer temperatures deteriorating reproductive success. Although climatic conditions improved later again, recolonization of Sweden from southern source populations was prevented by the Holocene submergence of the previous land connection via the Danish Straits that occurred approximately 8500 years ago.

  • Rapid depletion of B lymphocytes by ultra-low-dose rituximab delivered intrathecally

    • Objective: We are conducting an open-label phase 1b study on the efficacy of intrathecal (IT) administration of rituximab, provided via an Ommaya reservoir, for the treatment of progressive multiple sclerosis (PMS). The objective of this initial study was to monitor B lymphocytes in peripheral blood (PB) and CSF from the first 10 patients 1 year posttreatment.Methods: Dose titration was performed with daily escalation from 1 mg to 25 mg IT rituximab (n=3). Lymphocyte subpopulations were monitored daily during dose escalation in PB by flow cytometry and subsequently every 3 months for 1 year, after a total dose of 3 x 25 mg. PB B-lymphocyte subpopulations for the remaining patients (n = 7) were monitored at regular intervals. CSF lymphocyte subpopulations for all patients were monitored by flow cytometry every 2-3 months.Results: The PB B-lymphocyte count dropped rapidly after the first 2 injections (total dose of 3.5 mg IT rituximab) to undetectable levels. Three 25-mg doses given once per week depleted peripheral B lymphocytes entirely for the following 3-6 month period.Conclusions: Monoclonal antibodies seem to rapidly redistribute to the peripheral compartment following IT injection. Ultra-low doses of rituximab given IT are sufficient to cause complete depletion of peripheral B lymphocytes, indicating that low-dose IT treatment has the potential to be effective in both the CNS and systemic compartments.Classification of evidence: This study provides Class IV evidence that for patients with PMS, rituximab provided via an Ommaya reservoir depletes peripheral blood B lymphocytes.
