    • The article shows how a grammar-didactic model designed for an L1 context can be used for grammar learning as well as language and writing development in an L2 con-text. Because classrooms in Sweden, Denmark and Norway are becoming increasingly heterogenous, teachers must be able to develop the language of pupils with differing linguistic predispositions. The model that is being proposed is designed around explicit teaching with the table for sentence structure as point of departure (see Appendix 1). The purpose of the model is to act as a tool for language development, while simul-taneously contributing to the consolidation of grammatical knowledge. In the article, results from an intervention study are presented, where the model has been used for grammar teaching and process-writing in a class of L2 pupils. The pupils’ grammar learning, as it manifests in their results on the grammar exam as well as in their handlingof the teacher’s comments on their texts, is examined using the concepts appropriation, internalization and implementation. The pupils’ exam results show to which extent the pupils have appropriated grammatical knowledge, while their handling of the text-comments show if and how they have internalized and potentially implemented the grammatical knowledge. In the analysis, the pupils’ exam results and texts are compared, focusing on clausal hierarchy, inversion and V2 as well as agreement and definiteness in nominal phrases – areas that have previously been shown to be central for language development while at the same time problematic when learning grammar. The analyses indicate that the pupils have appropriated the grammatical knowledge to differing extents and that some are on their way toward an internalization of the knowledge. One conclusion is that, while this way of working requires great effort by teachers and pupils, the formal grammatical knowledge and the practical working with texts appear to benefit each other.

    • Mars Orbiting Plasma Surveyor (MOPS) is a microsatellite mission focused on studies of the near -Mars environment and the planet - solar wind interaction. The recent findings by the ESA Mars Express mission further highlighted the complexity of the processes taking place at the planet resulting from the solar wind interaction that strongly affect the planet's atmosphere. However, despite many previous Martian missions carrying different types of space plasma experiments, a comprehensive investigation including simultaneous measurements of particles, fields, and waves has never been performed. We consider a spinning spacecraft of a wet mass of 76.1 kg with a 9.7 kg payload, which can “hitchhike” on another platform until Mars orbit insertion, and then be released into a suitable orbit. The spacecraft design is based on the experience gained in very successful Swedish space plasma missions, Viking, Freja, Astrid-1, and Astrid-2. In the present mission design, the MOPS spacecraft is equipped with its own 1m high gain antenna for direct communication with the Earth. The payload includes a wave experiment with wire booms, magnetometer with a rigid boom, Langmuir probes, electron and ion energy spectrometers and an ion mass analyzer. An energetic neutral atom imager and an UV photometer may complete the core payload. One of the proposed scenarios is piggy - backing on the Russian Phobos - Grunt mission to be launched to Mars in 2011.

    • Small molecule neurotransmitters are essential for the function of the nervous system, and neurotransmitter imbalances are often connected to neurological disorders. The ability to quantify such imbalances is important to provide insights into the biochemical mechanisms underlying the disorder. This proof-of-principle study presents online quantification of small molecule neurotransmitters, specifically acetylcholine, γ-aminobutyric acid (GABA) and glutamate, in rat brain tissue sections using nanospray desorption electrospray ionization (nano-DESI) mass spectrometry imaging. By incorporating deuterated internal standards in the nano-DESI solvent we show identification, accurate mapping, and quantification of these small neurotransmitters in rat brain tissue without introducing any additional sample preparation steps. We find that GABA is about twice as abundant in the medial septum-diagonal band complex (MSDB) as in the cortex, while glutamate is about twice as abundant in the cortex as compared to the MSDB. The study shows that nano-DESI is well suited for imaging of small molecule neurotransmitters in health and disease.

    • Early detection of pancreatic ductal adenocarcinoma (PDAC) is challenging, and late diagnosis partly explains the low 5-year survival. Novel and sensitive biomarkers are needed to enable early PDAC detection and improve patient outcomes. Tissue polypeptide specific antigen (TPS) has been studied as a biomarker in PDAC diagnostics, and it has previously been shown to reflect clinical status better than the ‘golden standard’ biomarker carbohydrate antigen 19-9 (CA 19-9) that is most widely used in the clinical setting. In this cross-sectional case-control study using pre-diagnostic plasma samples, we aim to evaluate the potential of TPS as a biomarker for early PDAC detection. Furthermore, in a subset of individuals with multiple samples available at different time points before diagnosis, a longitudinal analysis was used. We assessed plasma TPS levels using enzyme-linked immunosorbent assay (ELISA) in 267 pre-diagnostic PDAC plasma samples taken up to 18.8 years before clinical PDAC diagnosis and in 320 matched healthy controls. TPS levels were also assessed in 25 samples at PDAC diagnosis. Circulating TPS levels were low both in pre-diagnostic samples of future PDAC patients and in healthy controls, whereas TPS levels at PDAC diagnosis were significantly increased (odds ratio 1.03; 95% confidence interval: 1.01–1.05) in a logistic regression model adjusted for age. In conclusion, TPS levels increase late in PDAC progression and hold no potential as a biomarker for early detection.

  • Large Scale Library Generation for High Throughput Sequencing Authors and Affiliations

    • Background: Large efforts have recently been made to automatethe sample preparation protocols for massively parallel sequencing in order to match the increasing instrument throughput. Still, the size selection through agarose gel electrophoresis separation is a labor-intensive bottleneck of these protocols. Methodology/Principal Findings: In this study a method for automatic library preparation and size selection on a liquid handling robot is presented. The method utilizes selective precipitation of certain sizes of DNA molecules on to paramagnetic beads for cleanup and selection after standard enzymatic reactions. Conclusions/Significance: The method is used to generate libraries for de novo and re-sequencing on the Illumina HiSeq 2000 instrument with a throughput of 12 samples per instrument in approximately 4 hours. The resulting output data show quality scores and pass filter rates comparable to manually prepared samples. The sample size distribution can be adjusted for each application, and are suitable for all high throughput DNA processing protocols seeking to control size intervals.
