  • Evaluation of the Chemcatcher and DGT passive samplers for monitoring metals with highly fluctuating water concentrations
  • 2007
  • Ingår i: Journal of Environmental Monitoring. ; 9:7, s. 672-681
  • Tidskriftsartikel (refereegranskat)abstract
    • Passive sampling devices accumulate chems. continuously from water and can provide time weighted av. (TWA) concns. of pollutants over the exposure period. Hence, they offer a no. of advantages over other conventional monitoring techniques such as spot or grab sampling. The diffusive gradient in thin film (DGT) and the Chemcatcher passive samplers can be used to provide TWA concns. of labile metals, but the approaches to their calibration differ. DGT uses diffusion coeffs. of metals in the hydrogel layer, whereas Chemcatcher uses metal specific uptake rates, with both sets of values obtained under controlled lab. conditions with const. aq. metal concns. However, little is known of how such samplers respond to fluctuating concns. We evaluated the responsiveness of these two passive sampling devices to rapidly changing concns. of Cd, Cu, Ni, Pb and Zn in natural freshwater, over a relatively short deployment time. Maximum metal concns. in water were varied between 70 and 140 mg L-1. Expts. were carried out in a tank with a rotating carousel system and filled with Meuse River water, allowing a degree of control over exptl. conditions while using natural river water. Fluctuating concns. were obtained by stepwise addn. of std. solns. of the metals. The reliability and accuracy of the TWA concns. measured by the samplers were assessed by comparison with concns. of the metals in spot samples of water taken regularly over the deployment period. The spot samples of water were either unfiltered (total), filtered (0.45 mm) or ultrafiltered (5 kDa). Predictive speciation modeling using the visual MINTEQ program was also undertaken. There was reasonable agreement between the TWA concns. of Cd and Ni obtained with Chemcatcher and DGT and the total Cd and Ni concns. measured in repeated unfiltered spot samples. For elements (i.e. Cu, Pb, Zn) that assoc. to a significant degree with suspended solids, colloids or dissolved org. carbon, or form complexes with large org. ligands, optimum agreement was with the filtered or ultrafiltered fractions and with the predicted inorg. and inorg.-fulvic acid assocd. fractions. While Chemcatcher-based TWA concn. ranges for Cu and Zn were in best agreement with the total filtered fraction, there was lack of agreement for Pb. The combined use of DGT devices with open pore (OP) and restricted pore (RP) gels allowed the labile fraction of metal assocd. with large org. ligands or DOC to be differentiated and quantified, since this is available to DGT OP but unable to diffuse into the DGT RP. This evaluation of the two sampling devices clearly demonstrated their ability to react reliably to transient peaks in concn. of metal pollutants in water and indicated where future efforts are needed to improve calibration data. Such samplers may prove valuable in responding to the monitoring requirements of the European Union's Water Framework Directive. [on SciFinder (R)]

  • Strategic monitoring for the European Water Framework Directive
  • 2006
  • Ingår i: TrAC - Trends in Analytical Chemistry. - : Elsevier BV. - 0165-9936. ; 25:7, s. 704-715
  • Tidskriftsartikel (refereegranskat)abstract
    • This article first reviews the principal monitoring requirements of the Water Framework Directive (WFD) of the European Union (EU) and assesses how contaminant monitoring may fit into a risk-assessment approach. In this context, we show the limited ability of conventional trace-contaminant-monitoring methods to fulfil all of the WFD requirements. We then clearly define and exemplify the roles and the functions of a new set of monitoring tools, using three case studies based on datasets that we obtained during a field trial in the River Meuse as part of the Screening methods for Water data InFormaTion (SWIFT-WFD) project in support of implementing the WFD.

  • Targeting the Main Protease (Mpro, nsp5) by Growth of Fragment Scaffolds Exploiting Structure-Based Methodologies
  • 2024
  • Ingår i: ACS Chemical Biology. - : American Chemical Society (ACS). - 1554-8929 .- 1554-8937. ; 19:2, s. 563-574
  • Tidskriftsartikel (refereegranskat)abstract
    • The main protease Mpro, nsp5, of SARS-CoV-2 (SCoV2) is one of its most attractive drug targets. Here, we report primary screening data using nuclear magnetic resonance spectroscopy (NMR) of four different libraries and detailed follow-up synthesis on the promising uracil-containing fragment Z604 derived from these libraries. Z604 shows time-dependent binding. Its inhibitory effect is sensitive to reducing conditions. Starting with Z604, we synthesized and characterized 13 compounds designed by fragment growth strategies. Each compound was characterized by NMR and/or activity assays to investigate their interaction with Mpro. These investigations resulted in the four-armed compound 35b that binds directly to Mpro. 35b could be cocrystallized with Mpro revealing its noncovalent binding mode, which fills all four active site subpockets. Herein, we describe the NMR-derived fragment-to-hit pipeline and its application for the development of promising starting points for inhibitors of the main protease of SCoV2.

  • Preexisting autoantibodies to type I IFNs underlie critical COVID-19 pneumonia in patients with APS-1.
  • 2021
  • Ingår i: The Journal of experimental medicine. - 1540-9538. ; 218:7
  • Tidskriftsartikel (refereegranskat)abstract
    • Patients with biallelic loss-of-function variants of AIRE suffer from autoimmune polyendocrine syndrome type-1 (APS-1) and produce a broad range of autoantibodies (auto-Abs), including circulating auto-Abs neutralizing most type I interferons (IFNs). These auto-Abs were recently reported to account for at least 10% of cases of life-threatening COVID-19 pneumonia in the general population. We report 22 APS-1 patients from 21 kindreds in seven countries, aged between 8 and 48 yr and infected with SARS-CoV-2 since February 2020. The 21 patients tested had auto-Abs neutralizing IFN-α subtypes and/or IFN-ω; one had anti-IFN-β and another anti-IFN-ε, but none had anti-IFN-κ. Strikingly, 19 patients (86%) were hospitalized for COVID-19 pneumonia, including 15 (68%) admitted to an intensive care unit, 11 (50%) who required mechanical ventilation, and four (18%) who died. Ambulatory disease in three patients (14%) was possibly accounted for by prior or early specific interventions. Preexisting auto-Abs neutralizing type I IFNs in APS-1 patients confer a very high risk of life-threatening COVID-19 pneumonia at any age.

  • Husbandry conditions of spotted ratfish (Hydrolagus colliei, Chimaeriformes) in aquaria for successful embryonic development and long-term survival of juveniles
  • 2024
  • Ingår i: Zoo Biology. - 0733-3188 .- 1098-2361. ; 43:2, s. 188-198
  • Tidskriftsartikel (refereegranskat)abstract
    • The spotted ratfish Hydrolagus colliei is the most common holocephalan species exhibited in aquaria worldwide for introducing deep-sea environments and raising awareness of their conservation. However, little is known about the biology of H. colliei. Current practices in aquaria allow long-term survival of sexually mature H. colliei specimens; however, this species struggles to complete a reproductive cycle in captivity mostly because embryos do not reach the hatchling stage. The aquarists of Planet Ocean Montpellier (POM, France) have bred H. colliei for 15 years and recorded parameters suitable for this species' successful embryonic and post-embryonic development. POM aquarists now regularly record egg-laying events of H. colliei and use four tanks to incubate eggs and raise neonates, late hatchlings, early and intermediate juveniles, subadults, and sexually mature specimens. In this work we provide the first long-term biometric data on H. colliei from the hatchling to the subadult stage. We also report the biotic and abiotic parameters sufficient to breed H. colliei in aquaria. We finally describe the methods used to facilitate individual monitoring of specimens along the ontogeny and several pathologies identified in this species, their putative causes, and the corresponding treatments. This work highlights the importance of ex situ research and points to the valuable outcomes of collaborative efforts between aquaria and academia in deciphering the biology of species whose study in the wild remains challenging.

  • PDRs4All : A JWST Early Release Science Program on Radiative Feedback from Massive Stars
  • 2022
  • Ingår i: Publications of the Astronomical Society of the Pacific. - : IOP Publishing. - 0004-6280 .- 1538-3873. ; 134:1035
  • Tidskriftsartikel (refereegranskat)abstract
    • Massive stars disrupt their natal molecular cloud material through radiative and mechanical feedback processes. These processes have profound effects on the evolution of interstellar matter in our Galaxy and throughout the universe, from the era of vigorous star formation at redshifts of 1-3 to the present day. The dominant feedback processes can be probed by observations of the Photo-Dissociation Regions (PDRs) where the far-ultraviolet photons of massive stars create warm regions of gas and dust in the neutral atomic and molecular gas. PDR emission provides a unique tool to study in detail the physical and chemical processes that are relevant for most of the mass in inter- and circumstellar media including diffuse clouds, proto-planetary disks, and molecular cloud surfaces, globules, planetary nebulae, and star-forming regions. PDR emission dominates the infrared (IR) spectra of star-forming galaxies. Most of the Galactic and extragalactic observations obtained with the James Webb Space Telescope (JWST) will therefore arise in PDR emission. In this paper we present an Early Release Science program using the MIRI, NIRSpec, and NIRCam instruments dedicated to the observations of an emblematic and nearby PDR: the Orion Bar. These early JWST observations will provide template data sets designed to identify key PDR characteristics in JWST observations. These data will serve to benchmark PDR models and extend them into the JWST era. We also present the Science-Enabling products that we will provide to the community. These template data sets and Science-Enabling products will guide the preparation of future proposals on star-forming regions in our Galaxy and beyond and will facilitate data analysis and interpretation of forthcoming JWST observations.

  • Light-ion production in the interaction of 175 MeV neutrons with iron and bismuth
  • 2011
  • Ingår i: Journal of Korean Physical Society.
  • Konferensbidrag (refereegranskat)abstract
    • We have measured double-differential (angle and energy) cross sections for light-ion (p, d, t, 3He and α) production in the interaction of quasi-monoenergetic 175 MeV neutrons with iron and bismuth. Measurements have been performed at the The Svedberg Laboratory, Uppsala (Sweden), using the Medley setup which allows low-energy thresholds and wide energy and angular ranges. Medley is a spectrometer system consisting of eight three-element telescopes placed at angles from 20◦ to 160◦, in steps of 20◦. Each telescope is composed of two silicon surface barrier detectors and a CsI(Tl) scintillator, to perform particle identification, fully stop the produced light-ions and measure their kinetic energy. The time-of-flight was used to reduce the contribution from the low energy tail in the accepted incident neutron spectrum. We report double-differential production cross sections for protons, deuterons, tritons, 3He and α particles and compare them with model calculations with TALYS-1.2.

  • Medley spectrometer for light ions in neutron-induced reactions at 175 MeV
  • 2011
  • Ingår i: Nuclear Instruments and Methods in Physics Research Section A. - : Elsevier BV. - 0168-9002 .- 1872-9576. ; 646:1, s. 100-107
  • Tidskriftsartikel (refereegranskat)abstract
    • We have upgraded the Medley spectrometer to measure neutron-induced double differential cross-sections for light-ion production, with neutron energies up to 175 MeV. Measurements were performed at the quasi-monoenergetic neutron beam line at the The Svedberg Laboratory, in Uppsala (Sweden). Medley is a spectrometer system composed of eight three-element telescope detectors; Delta E-E technique is used for particle identification and total kinetic energy measurement. We have improved particle separation and installed new detectors that fully stop ions with kinetic energy up to 170 MeV. To reduce the contribution of the low energy tail in the quasi-monoenergetic neutron spectrum, we have applied time-of-flight techniques and investigated the results for measurements with 175 MeV neutrons. We have investigated the response function of the CsI(Tl) scintillators and describe a method for efficiency correction. We have studied the signal-to-background ratio of the new configuration of the spectrometer and of the collimation system. A method to obtain absolute cross-section normalization, separating the peak neutrons from the low-energy tail, is proposed. Finally we have characterized the 175 MeV quasi-monoenergetic neutron spectrum with the Medley spectrometer. This upgraded configuration has been used in 2007 and 2009 for light-ion production measurements at 175 MeV from C, Si, O, Fe, Bi and U.

  • Multiple Miscarriages Are Associated with the Risk of Ovarian Cancer: Results from the European Prospective Investigation into Cancer and Nutrition
  • 2012
  • Ingår i: PLoS ONE. - : Public Library of Science (PLoS). - 1932-6203. ; 7:5
  • Tidskriftsartikel (refereegranskat)abstract
    • While the risk of ovarian cancer clearly reduces with each full-term pregnancy, the effect of incomplete pregnancies is unclear. We investigated whether incomplete pregnancies (miscarriages and induced abortions) are associated with risk of epithelial ovarian cancer. This observational study was carried out in female participants of the European Prospective Investigation into Cancer and Nutrition (EPIC). A total of 274,442 women were followed from 1992 until 2010. The baseline questionnaire elicited information on miscarriages and induced abortions, reproductive history, and lifestyle-related factors. During a median follow-up of 11.5 years, 1,035 women were diagnosed with incident epithelial ovarian cancer. Despite the lack of an overall association (ever vs. never), risk of ovarian cancer was higher among women with multiple incomplete pregnancies (HR >= 4vs.0: 1.74, 95% CI: 1.20-2.70; number of cases in this category: n = 23). This association was particularly evident for multiple miscarriages (HR >= 4vs.0: 1.99, 95% CI: 1.06-3.73; number of cases in this category: n = 10), with no significant association for multiple induced abortions (HR >= 4vs.0: 1.46, 95% CI: 0.68-3.14; number of cases in this category: n = 7). Our findings suggest that multiple miscarriages are associated with an increased risk of epithelial ovarian cancer, possibly through a shared cluster of etiological factors or a common underlying pathology. These findings should be interpreted with caution as this is the first study to show this association and given the small number of cases in the highest exposure categories.
