  • Human Gut Faecalibacterium prausnitzii Deploys a Highly Efficient Conserved System To Cross-Feed on beta-Mannan-Derived Oligosaccharides

    • beta-Mannans are hemicelluloses that are abundant in modern diets as components in seed endosperms and common additives in processed food. Currently, the collective understanding of beta-mannan saccharification in the human colon is limited to a few keystone species, which presumably liberate low-molecular-weight mannooligosaccharide fragments that become directly available to the surrounding microbial community. Here, we show that a dominant butyrate producer in the human gut, Faecalibacterium prausnitzii, is able to acquire and degrade various beta-mannooligosaccharides (beta-MOS), which are derived by the primary mannanolytic activity of neighboring gut microbiota. Detailed biochemical analyses of selected protein components from their two beta-MOS utilization loci (F. prausnitzii beta-MOS utilization loci [FpMULs]) supported a concerted model whereby the imported beta-MOS are stepwise disassembled intracellularly by highly adapted enzymes. Coculturing experiments of F. prausnitzii with the primary degraders Bacteroides ovatus and Roseburia intestinalis on polymeric beta-mannan resulted in syntrophic growth, thus confirming the high efficiency of the FpMULs' uptake system. Genomic comparison with human F. prausnitzii strains and analyses of 2,441 public human metagenomes revealed that FpMULs are highly conserved and distributed worldwide. Together, our results provide a significant advance in the knowledge of beta-mannan metabolism and the degree to which its degradation is mediated by cross-feeding interactions between prominent beneficial microbes in the human gut. IMPORTANCE Commensal butyrate-producing bacteria belonging to the Firmicutes phylum are abundant in the human gut and are crucial for maintaining health. Currently, insight is lacking into how they target otherwise indigestible dietary fibers and into the trophic interactions they establish with other glycan degraders in the competitive gut environment. By combining cultivation, genomic, and detailed biochemical analyses, this work reveals the mechanism enabling F. prausnitzii, as a model Ruminococcaceae within Firmicutes, to cross-feed and access beta-mannan-derived oligosaccharides released in the gut ecosystem by the action of primary degraders. A comprehensive survey of human gut metagenomes shows that FpMULs are ubiquitous in human populations globally, highlighting the importance of microbial metabolism of beta-mannans/beta-MOS as a common dietary component. Our findings provide a mechanistic understanding of the beta-MOS utilization capability by F. prausnitzii that may be exploited to select dietary formulations specifically boosting this beneficial symbiont, and thus butyrate production, in the gut.

    • Humans have co-evolved with a dense community of microbial symbionts that inhabit the lower intestine. In the colon, secreted mucus creates a barrier that separates these microorganisms from the intestinal epithelium(1). Some gut bacteria are able to utilize mucin glycoproteins, the main mucus component, as a nutrient source. However, it remains unclear which bacterial enzymes initiate degradation of the complex O-glycans found in mucins. In the distal colon, these glycans are heavily sulfated, but specific sulfatases that are active on colonic mucins have not been identified. Here we show that sulfatases are essential to the utilization of distal colonic mucin O-glycans by the human gut symbiont Bacteroides thetaiotaomicron. We characterized the activity of 12 different sulfatases produced by this species, showing that they are collectively active on all known sulfate linkages in O-glycans. Crystal structures of three enzymes provide mechanistic insight into the molecular basis of substrate specificity. Unexpectedly, we found that a single sulfatase is essential for utilization of sulfated O-glycans in vitro and also has a major role in vivo. Our results provide insight into the mechanisms of mucin degradation by a prominent group of gut bacteria, an important process for both normal microbial gut colonization(2) and diseases such as inflammatory bowel disease(3). A single sulfatase produced by a bacterium found in the human colon is essential for degradation of sulfated O-glycans in secreted mucus.

  • CASCADE - The Circum-Arctic Sediment CArbon DatabasE

    • Biogeochemical cycling in the semi-enclosed Arctic Ocean is strongly influenced by land-ocean transport of carbon and other elements and is vulnerable to environmental and climate changes. Sediments of the Arctic Ocean are an important part of biogeochemical cycling in the Arctic and provide the opportunity to study present and historical input and the fate of organic matter (e.g., through permafrost thawing). Comprehensive sedimentary records are required to compare differences between the Arctic regions and to study Arctic biogeochemical budgets. To this end, the Circum-Arctic Sediment CArbon DatabasE (CASCADE) was established to curate data primarily on concentrations of organic carbon (OC) and OC isotopes (delta C-13, Delta C-14) yet also on total N (TN) as well as terrigenous biomarkers and other sediment geochemical and physical properties. This new database builds on the published literature and earlier unpublished records through an extensive international community collaboration. This paper describes the establishment, structure and current status of CASCADE. The first public version includes OC concentrations in surface sediments at 4244 oceanographic stations including 2317 with TN concentrations, 1555 with delta C-13-OC values and 268 with Delta C-14-OC values and 653 records with quantified terrigenous biomarkers (high-molecular-weight n-alkanes, n-alkanoic acids and lignin phenols). CASCADE also includes data from 326 sediment cores, retrieved by shallow box or multi-coring, deep gravity/piston coring, or sea-bottom drilling. The comprehensive dataset reveals large-scale features of both OC content and OC sources between the shelf sea recipients. This offers insight into release of pre-aged terrigenous OC to the East Siberian Arctic shelf and younger terrigenous OC to the Kara Sea. Circum-Arctic sediments thereby reveal patterns of terrestrial OC remobilization and provide clues about thawing of permafrost. CASCADE enables synoptic analysis of OC in Arctic Ocean sediments and facilitates a wide array of future empirical and modeling studies of the Arctic carbon cycle. The database is openly and freely available online (https://doi.org/10.17043/cascade; Martens et al., 2021), is provided in various machine-readable data formats (data tables, GIS shapefile, GIS raster), and also provides ways for contributing data for future CASCADE versions. We will continuously update CASCADE with newly published and contributed data over the foreseeable future as part of the database management of the Bolin Centre for Climate Research at Stockholm University.

  • Remediating reduced memory specificity in bipolar disorder : A case study using a Computerized Memory Specificity Training

    • OBJECTIVES: Reduced autobiographical memory specificity (rAMS) is a vulnerability factor found across unipolar depression (UD), posttraumatic stress disorder (PTSD), eating disorder, schizophrenia, and bipolar disorder (BD). A group delivered psychological therapy training called Memory Specificity Training (MeST) remediates rAMS in UD and PTSD, with additional downstream effects on related psychological processes and symptoms. Its impact in BD is unknown. In this case study, we examined the impact of a computerized version of MeST (c-MeST) on improving AMS and related symptoms and processes in participant with rapid cycling type I BD.METHOD: An experimental case study with an ABA design was used. During baseline (14 days, Phase A), the training phase (nine sessions across 17 days, Phase B), and a 1-month follow-up (Phase A), memory specificity, depressive symptoms, and related processes and symptoms were repeatedly measured.RESULTS: Memory specificity increased significantly after the participant completed c-MeST. Session-to-session scores indicated that AMS improved most from the in-person baseline assessment to the first online session. All other measures of processes and symptoms deteriorated during the training phase but regressed to baseline during follow-up.CONCLUSION: Memory specificity was improved as indicated by increased AMS from pre-intervention measurement to 1-month follow-up. Other improvements in symptoms were not observed. Rather, some related maladaptive psychological processes and symptoms worsened during the training phase and regressed to baseline during follow-up.

    • Three-dimensional electromagnetic calculations for loaded transverse electromagnetic (TEM) transmission cells are presented. Based on those calculations a prediction of the perturbation of the standard uniform field in the TEM-cell, due to the scattering by inhomogeneous structures placed inside the cell cavity, is given. The influence of the dimensions of a lossy structure and its position in the TEM-cell on its absorption of the electromagnetic fields is presented. Knowing the perturbation of the uniform field is important for good interpretation of the biological experimental results.
