  • Dynamic hollow-fibre liquid phase microextraction of dinitrophenols from human plasma: Optimization of an extraction flow system using experimental design methodology

    • The utility of a dynamic hollow-fibre liquid phase microextraction method (optimized using a four-variable experimental design and response surface modelling) for extracting dinitrophenolic compounds from human plasma samples was evaluated. The investigated variables were donor phase salt concentration (10–400 mM), donor phase pH (2–6), acceptor phase pH (7–12), and donor/acceptor phase flow rates (30/7.5 to 70/17.5 μL min−1). Four dinitrophenol pesticides were used as model substances at concentrations of 0.1 μg mL−1 in spiked human plasma samples. Extraction efficiencies ranging from 42 to 77% with RSDs below 9 were achieved with the optimized method. The flow rate and acceptor pH were shown to strongly affect the extraction efficiency for all compounds, while the donor phase pH and salt concentration had minor effects. With a well-defined acceptor phase pH and flow rate the system exhibited high robustness. The limits of quantification for the investigated compounds, using the presented extraction method followed by liquid chromatography/electrospray ionization mass spectrometry in selected ion monitoring mode, ranged from 0.05 to 0.1 μg mL−1 plasma.

  • Dynamic field sampling of airborne organophosphate triesters using solid-phase microextraction under equilibrium and non-equilibrium conditions

    •   A simple setup for dynamic air sampling using a solid-phase microextraction (SPME) device designed for use in the field was evaluated for organophosphate triester vapour under both equilibrium and non-equilibrium conditions. The effects of varying the applied airflows in the sampling device were evaluated in order to optimise the system with respect to the Reynolds number and magnitude of the boundary layer that developed near the surface. Further, the storage stability of the analytes was studied for both capped and uncapped 100-     m m PDMS fibres. Organophosphate triesters are utilized on large scales as flame-retardants and/or plasticizers, for instance in upholstered furniture. In indoor working environments these compounds have become common components in the surrounding air. Measurements were performed in a recently furnished working environment and the concentration of tris(2-choropropyl) phosphate was found to be 7     mg m23 .

    • Estrogen has extensive effects on the immune system. The aim of the present experiments was to compare the effects of 17beta-estradiol (E2) and 4-estren-3alpha,17beta-diol (estren) on T lymphopoiesis and T cell-dependent inflammation. In order to investigate the role of estrogen receptors (ER) in the effects of E2 and estren on the immune system, ER knock-out mice lacking both ERalpha and ERbeta (DERKO) were used. T lymphopoiesis and T cell-dependent inflammation were studied by investigating thymus cellularity, the delayed-type hypersensitivity (DTH) reaction, CD4(+) T cells in spleen and serum levels of interleukin (IL)-6. As expected, the presence of ERs was mandatory for all the effects of E2. In contrast, treatment with estren reduced thymus cellularity in ER knock-out mice, indicating an effect through ER-independent pathways. Interestingly, estren suppressed only DTH, the frequency of CD4(+) T cells in spleen and serum levels of IL-6 in wild-type (WT) mice, but not in mice lacking ERs. Thus, our study is the first to show that estren inhibits T lymphopoiesis via ER-independent pathways, whereas its suppressive effects on inflammation are ER-dependent.

  • Forest edges in boreal landscapes - factors affecting edge influence

    • The boreal forest in Fennoscandia has been subjected to major loss and fragmentation of natural forests due to intensive forestry. This has resulted in that forest edges are now abundant and important landscape features. Edges have documented effects on the structure, function and biodiversity in forests. Edge influence on biodiversity is complex and depends on interactions between many local and regional factors. This thesis focuses on sharp forest edges and their potential to influence biodiversity at the landscape-level. I have developed a method for quantification and characterization of sharp forest edges by interpretation of colour infrared (CIR) aerial photographs in combination with line intersect sampling (LIS) and sample plots. The method was used to estimate density of forest edge in 28 landscapes (each 1600 ha) in northern Sweden, differing in management intensity, landscape composition and geographical location. Forest edges were described in detail using edge, canopy and neighbourhood attributes. By combining these attributes it was possible to classify edges with respect to levels of exposure. A field experiment was conducted to examine the effect of edge contrast on growth of the old forest lichen Usnea longissima. The edge quantification method is accurate and efficient for estimating the length of sharp forest edges on an area basis (edge density, m ha-1) and for collecting detailed attributes of edges and their surroundings. In northern Sweden, the forest edge density is high (54 m ha-1) but varies extensively (12-102 m ha-1) between landscapes. Edge density is strongly correlated with the level of human disturbance and increases towards the southern part of the study area, at lower altitudes were management intensity is highest. Edge orientation, contrast and neighbourhood size shows an immense variation between edges and also varies between edge types. Regenerating edges are generally of higher contrast and face larger neighbourhoods than natural edges. Maintained edges had high contrast but small neighbourhoods. A larger proportion of edges in mature forests are highly exposed to microclimatic edge influence than edges in general. The field experiment revealed that growth of U. longissima was highest near edges where the vegetation on the adjacent area was sheltering, but not shading, the lichen. In the present thesis, I have provided a valuable tool for estimating density of forest edges with potential to yield information on important factors determining edge influence at landscape-level. The large variability in edge density, edge and neighbourhood attributes imply large differences in microclimate anf thus in the potential for ede influence. Management and conservation strategies must incorporate these factors to realistically address edge influence on biota at the landscape-level.

    • BACKGROUND: Dried blood spot samples (DBSS) from newborns are widely used in neonatal screening for selected metabolic diseases and diagnostic possibilities for additional disorders are continuously being evaluated. Primary immunodeficiency disorders comprise a group of more than one hundred diseases, several of which are fatal early in life. Yet, a majority of the patients are not diagnosed due to lack of high-throughput screening methods.METHODOLOGY/PRINCIPAL FINDINGS: We have previously developed a system using reverse phase protein microarrays for analysis of IgA levels in serum samples. In this study, we extended the applicability of the method to include determination of complement component C3 levels in eluates from DBSS collected at birth. Normal levels of C3 were readily detected in 269 DBSS from healthy newborns, while no C3 was detected in sera and DBSS from C3 deficient patients.CONCLUSIONS/SIGNIFICANCE: The findings suggest that patients with deficiencies of specific serum proteins can be identified by analysis of DBSS using reverse phase protein microarrays.

    • The widespread use of scented products causes an increase of allergic contact dermatitis to fragrance compounds in Western countries today. Many fragrance compounds are prone to autoxidation, forming hydroperoxides as their primary oxidation products. Hydroperoxides are known to be strong allergens and to form specific immunogenic complexes. However, the mechanisms for the formation of the immunogenic complexes are largely unknown. We have investigated this mechanism for (5R)-5-isopropenyl-2-methyl-2-cyclohexene-1-hydroperoxide (Lim-2-OOH) by studying the formation of adducts in the reaction between this hydroperoxide and 5,10,15,20-tetraphenyl-21H,23H-porphine iron(III) chloride (Fe(III)TPPCl) in the presence of protected cysteine (NAc-Cys-OMe) or glutathione (GSH). Isolated adducts originate from the addition of the thiol group of NAc-Cys-OMe over the carbon−carbon double bonds of carvone. Furthermore, adducts between NAc-Cys-OMe and carveol as well as between GSH and carvone have been identified. The formation of these adducts most likely proceeds via the radical thiol−ene mechanism. The addition of a terpene moiety to cysteine offers an explanation of the specificity of the immune response to structurally different hydroperoxides. These results also explain the lack of cross-reactivity between carvone and Lim-2-OOH. In conclusion, we propose that immunogenic complexes of olefinic hydroperoxides can be formed via the radical thiol−ene mechanism. These complexes will be specific for the individual olefinic hydroperoxides due to the inclusion of a terpene moiety derived from the hydroperoxide.

  • Formation of composite endothelial cell-mesenchymal stem cell islets : a novel approach to promote islet revascularization

    •   OBJECTIVE: Mesenchymal stem cells (MSCs) contribute to endothelial cell (EC) migration by producing proteases, thereby paving the way into the tissues for ECs. MSCs were added to our previously described composite EC islets as a potential means to improve their capacity for islet angiogenesis. RESEARCH DESIGN AND METHODS: Human islets were coated with primary human bone marrow-derived MSCs and dermal microvascular ECs. The capacity of ECs, with or without MSCs, to adhere to and grow into human islets was analyzed. The survival and functionality of these composite islets were evaluated in a dynamic perifusion assay, and their capacity for angiogenesis in vitro was assessed in a three-dimensional fibrin gel assay. RESULTS: ECs proliferated after culture in MSC-conditioned medium, and MSCs improved the EC coverage threefold compared with EC islets alone. Islet survival in vitro and the functionality of the composite islets after culture were equal to those of control islets. The EC-MSC islets showed a twofold increase in total sprout formation compared with EC islets, and vascular sprouts emanating from the EC-MSC-islet surface showed migration of ECs into the islets and also into the surrounding matrix, either alone or in concert with MSCs. CONCLUSIONS: EC proliferation, sprout formation, and ingrowth of ECs into the islets were enhanced by MSCs. The use of composite EC-MSC islets may have beneficial effects on revascularization and immune regulation. The technique presented allows for pretreatment of donor islets with recipient-derived ECs and MSCs as a means of improving islet engraftment.

  • Formation of Composite Islet Grafts : A novel strategy to promote islet survival and revascularization

    • The islets of Langerhans are small and delicate spheroid organs scattered in the pancreas responsible for insulin production. Transplantation of isolated islets is a beneficial therapy for patients with a severe form of type 1 diabetes. The islets, which normally are richly vascularized in the pancreas, are completely disconnected from the vascular support by the enzymatic digestion during the isolation procedure. Islet viability is affected throughout all steps in this process, from donor death and isolation of islets to culturing and the transplantation process itself. In this thesis a novel strategy to promote islet survival and to re-establish islet vasculature is presented. We show endogenous expression of 51 different genes related to inflammation in cultured islets. Among these genes high expression of MCP-1, MIF, VEGF, thymosin b-10 and IL-8, IL-1β, IL-5R-a, IFN-γ antagonist were found in all donors during the 5- and the 2-day cultures, respectively. Protein expression of these genes can stimulate inflammatory immune responses but also promote tissue repair by attracting curative cells such as endothelial cells (EC) leading to re-establishment of the vasculature. We have established a novel technique by formation of composite islets using EC and mesenchymal stem cells (MSC). EC adhered on the surface of the islets and created a potential blood tolerant surface. The EC-islets showed a degree of protection from the detrimental effects of instant blood-mediated inflammatory reaction (IBMIR) with the major components of IBMIR being decreased in in vitro assays. We combined MSC to the EC-islets with success. The MSC were found to have proliferative effect on EC and the combination of these two cell types on the islets further increased the EC covered surface compared to EC-islets. The EC-MSC-islets in co-culture formed vessel-like structures both into the islets and out to the surrounding matrix. The MSC enhanced the exogenous EC to form vessel-like network in the EC-MSC-islets indicating vascular support by the MSC. The novel strategy and conditions presented herein could alleviate problems related to survival of the islets by promoting revascularization. This would open up a new era in islet transplantation and allow more patients to benefit from this therapy.
