  • alphaB-crystallin promotes tumor angiogenesis by increasing vascular survival during tube morphogenesis

    • Selective targeting of endothelial cells in tumor vessels requires delineation of key molecular events in formation and survival of blood vessels within the tumor microenvironment. To this end, proteins transiently up-regulated during vessel morphogenesis were screened for their potential as targets in antiangiogenic tumor therapy. The molecular chaperone alpha B-crystallin was identified as specifically induced with regard to expression level, modification by serine phosphorylation, and subcellular localization during tubular morphogenesis of endothelial cells. Small interfering RNA-mediated knockdown of alpha B-crystallin expression did not affect endothelial proliferation but led to attenuated tubular morphogenesis, early activation of proapoptotic caspase-3, and increased apoptosis. alpha B-crystallin was expressed in a subset of human tumor vessels but not in normal capillaries. Tumors grown in alpha B-crystallin(-/-) mice were significantly less vascularized than wild-type tumors and displayed increased areas of apoptosis/necrosis. Importantly, tumor vessels in alpha B-crystallin(-/-) mice were leaky and showed signs of caspase-3 activation and extensive apoptosis. Ultrastructural analyses showed defective vessels partially devoid of endothelial lining. These data strongly implicate alpha B-crystallin as an important regulator of tubular morphogenesis and survival of endothelial cell during tumor angiogenesis. Hereby we identify the small heat shock protein family as a novel class of anglogenic modulators.

  • Changes in design precipitation over the Nordic-Baltic region as given by convection-permitting climate simulations

    • The increased risk of flooding due to global warming and subsequent heavy rainfall events in the Nordic-Baltic region, call for recommendations directed at long-term planning. One example of such recommendations are climate change allowances. These are often based on expected changes in design precipitation as given by climate model simulations, and are used as a buffer on top of current design values to avoid a future increased damage potential as a consequence of climate change. We here compute expected changes in precipitation design values, so-called climate factors (CFs), for the Nordic-Baltic region, based on 3 km convection permitting simulations from the Nordic Convection Permitting Climate Projections project. These have the advantage of explicitly resolving convection, which has been shown to be the main contributor to increased rainfall. We assess the dependence of the CFs on rainfall duration, return period, and geographical location, focusing on the summer (convective) season, short durations and the high emission scenario RCP8.5. We also compare these CFs to those computed from a non-convection permitting regional climate model ensemble. We found higher CFs for the longer return period, with only few exceptions, and distinctly higher CFs going from daily to sub-daily durations. However, the different simulations give conflicting results for very short-duration rainfall (<3 h). The huge difference in the climate sensitivity of driving GCMs dominates the magnitude of estimated return levels. Our analysis is shaped by the high computational costs of running convection permitting models, resulting in a very limited ensemble (3 members) representing a single emission scenario (RCP8.5). Therefore, we believe that combining results from convection permitting simulations with a larger ensemble (21 members) of non-convection permitting simulations adds value to the assessment of robust climate change allowances for heavy precipitation in the Nordic-Baltic region.

  • Treatment of idiopathic rhinitis with kinetic oscillations : a multi-centre randomized controlled study

    • Conclusions: The potential effects of KOS are still uncertain regarding the most effective air pressure to be used as well as the physiological effects on the nasal mucosa. The results of the study do not support a convincing treatment effect by KOS on IR.Objectives: Idiopathic rhinitis (IR) is a common disorder, affecting ∼10-20% of the population. A new method for treating IR, Kinetic Oscillation Stimulation (KOS), has been reported to have beneficial effects on total vasomotor symptom scores (TVRSS). The primary objective with this study was to evaluate if a mean pressure of 65 mbar, pressure amplitude of 100 mbar, and 68Hz treatment with KOS had a positive effect on total vasomotor symptom scores (TVRSS), as compared with a mean pressure of 65 mbar, pressure amplitude of 4 mbar, and 68Hz treatment in patients with idiopathic rhinitis.Methods: Two hundred and seven patients were randomized (Full Analysis Set, FAS) in the study, including five visits and lasting for approximate to 25 weeks. All patients had two treatment episodes, and all patients had at least one treatment, meant as active, with high amplitude pressure for 10min in each nostril. Group 1 had two such treatments, and Group 2 had one treatment with low amplitude pressure, initially meant as placebo, on one occasion. Because of numerical improvements in these two groups, a new control group, Group 3, was introduced. They had one new control treatment where the balloon was inserted into the nose, without any air inflation and without oscillations.Results: KOS treatment with high amplitude pressure did not have significant beneficial effects as compared to low amplitude pressure on TVRSS. Numerical improvements in TVRSS and SNOT 22 were found when comparing high and low amplitude pressure treatments with uninflated balloon treatment. However, this part of the study was initially single-blinded, and these results were secondary objectives.

  • Adenosine stress myocardial perfusion detected with CT compared with attenuation-corrected SPECT

    • Purpose: To asses adenosine stress myocardial perfusion by cardiac CT and compare with simultaneously performed attenuation corrected SPECT.Methods: 11 patients, 9 men and 2 women >2months post primary PCI, with manifest myocardial damage and remaining stenoses in the coronary circulation, were studied with myocardial perfusion CT under vasodilatory stress. The investigation started with a topogram followed by a testbolus of iodine whereafter the coronary artery study was performed in sequence mode. Adenosine was then infused for at least five minutes at the standard rate of 140ug/kg/min. After three minutes, 6 MBq/kg of 99mTc-tetrofosmin was injected immediately followed by 80ml iodine contrast. The wash-in of iodine was monitored by CT scanning of a 7cm long cardiac volume segment every other second for 22s. One hour after the CT scan, myocardial SPECT was performed. Scanning required the patients to tolerate breath holding for 22s, have a heart rate <80/min and body weight <85kg, and their kidney function should allow 140ml 370mg iodine contrast to be given.Results: All 11 patients tolerated the full adenosine infusion and scanning was successful. One patient could not be analyzed due to noisy images. In two patients, the limited scanning volume did not cover the entire base of the heart. Three patients had no defect on SPECT. Patients with a defect had on average myocardial blood flow 80ml/100ml tissue/min in the defect area and 142ml in the segments with the highest perfusion, while patients without defect had 98 and 141ml, respectively.Conclusion: Peak myocardial perfusion may be determined with CT under adenosine stress and compared with attenuation corrected SPECT. Initial experience shows that the method is sensitive to timing of bolus, to noisy images and results may diverge from those obtained with SPECT.

    • We developed nanoparticles with tailored magnetic properties for sensitive detection of biomolecules directly in biological samples in a single step. Thermally blocked nanoparticles obtained by thermal hydrolysis are mixed with sample solutions and the variation of the magnetic relaxation due to surface binding is used to detect the presence of biomolecules. The binding events significantly increase the hydrodynamic volume of nanoparticles, thus changing their Brownian relaxation frequency which is measured by a specifically developed AC-susceptometer.The system was tested for the presence of Brucella antibodies in serum samples from infected cows and the surface of the nanoparticles was functionalized with lipopolysaccarides (LPS) from Brucella abortus. The hydrodynamic volume of functionalized particles increased by 25-35% as a result of the binding of the antibodies, as measured by changes in the susceptibility in an alternating magnetic field. The method has shown high sensitivity, with detection limit of 7 nmol·L-1 in serum without any pre-treatment of the biological samples.The detection method is very sensitive, cost-efficient and versatile, giving a direct indication if the animal is infected or not, making it suitable for point-of care applications. The functionalization of tailored magnetic nanoparticles can be modified to suit numerous homogenous assays for a wide range of applications.

    • Many problems in computer vision can be turned into a large-scale boolean optimization problem, which is in general NP-hard. In this paper, we further develop one of the most successful approaches, namely roof duality, for approximately solving such problems for higher-order models. Two new methods that can be applied independently or in combination are investigated. The first one is based on constructing relaxations using generators of the submodular function cone. In the second method, it is shown that the roof dual bound can be applied in an iterated way in order to obtain a tighter relaxation. We also provide experimental results that demonstrate better performance with respect to the state-of-the-art, both in terms of improved bounds and the number of optimally assigned variables.

    • Recent advances in single-cell transcriptomics are ideally placed to unravel intratumoral heterogeneity and selective resistance of cancer stem cell (SC) subpopulations to molecularly targeted cancer therapies. However, current single-cell RNA-sequencing approaches lack the sensitivity required to reliably detect somatic mutations. We developed a method that combines high-sensitivity mutation detection with whole-transcriptome analysis of the same single cell. We applied this technique to analyze more than 2,000 SCs from patients with chronic myeloid leukemia (CML) throughout the disease course, revealing heterogeneity of CML-SCs, including the identification of a subgroup of CML-SCs with a distinct molecular signature that selectively persisted during prolonged therapy. Analysis of nonleukemic SCs from patients with CML also provided new insights into cell-extrinsic disruption of hematopoiesis in CML associated with clinical outcome. Furthermore, we used this single-cell approach to identify a blast-crisis-specific SC population, which was also present in a subclone of CML-SCs during the chronic phase in a patient who subsequently developed blast crisis. This approach, which might be broadly applied to any malignancy, illustrates how single-cell analysis can identify subpopulations of therapy-resistant SCs that are not apparent through cell-population analysis.

  • High-speed 850-nm VCSELs for 40 Gb/s transmission

    • We have explored the possibility to extend the data transmission rate for standard 850-nm GaAs-based VCSELs beyond the 10 Gbit/s limit of today's commercially available directly-modulated devices. By sophisticated tailoring of the design for high-speed performance we demonstrate that 10 Gb/s is far from the upper limit. For example, the thermal conductivity of the bottom mirror is improved by the use of binary compounds, and the electrical parasitics are kept at a minimum by incorporating a large diameter double layered oxide aperture in the design. We also show that the intrinsic high speed performance is significantly improved by replacing the traditional GaAs QWs with strained InGaAs QWs in the active region. The best overall performance is achieved for a device with a 9 μm diameter oxide aperture, having in a threshold current of 0.6 mA, a maximum output power of 9 mW, a thermal resistance of 1.9 °C/mW, and a differential resistance of 80 Ω.   • A zero-temperature study of vortex mobility in two-dimensional vortex glass models

    • Three different vortex glass models are studied by examining the energy barrier against vortex motion cross the system. In the two-dimensional gauge glass this energy barrier is found to increase logarithmically with system size which opens up for the existence of low-temperature phase with zero resistivity. Associated with the large-energy barriers is breaking of ergodicity which explains why the well-established results from equilibrium studies could fail. The behavior of the more realistic random pinning model is however different with decreasing energy barriers and no finite critical temperature.
