  • The JCMT BISTRO Survey: A Spiral Magnetic Field in a Hub-filament Structure, Monoceros R2

    • We present and analyze observations of polarized dust emission at 850 μm toward the central 1 × 1 pc hub-filament structure of Monoceros R2 (Mon R2). The data are obtained with SCUBA-2/POL-2 on the James Clerk Maxwell Telescope (JCMT) as part of the B-fields in Star-forming Region Observations survey. The orientations of the magnetic field follow the spiral structure of Mon R2, which are well described by an axisymmetric magnetic field model. We estimate the turbulent component of the magnetic field using the angle difference between our observations and the best-fit model of the underlying large-scale mean magnetic field. This estimate is used to calculate the magnetic field strength using the Davis–Chandrasekhar–Fermi method, for which we also obtain the distribution of volume density and velocity dispersion using a column density map derived from Herschel data and the C18O (J = 3 - 2) data taken with HARP on the JCMT, respectively. We make maps of magnetic field strengths and mass-to-flux ratios, finding that magnetic field strengths vary from 0.02 to 3.64 mG with a mean value of 1.0 ± 0.06 mG, and the mean critical mass-to-flux ratio is 0.47 ± 0.02. Additionally, the mean Alfvén Mach number is 0.35 ± 0.01. This suggests that, in Mon R2, the magnetic fields provide resistance against large-scale gravitational collapse, and the magnetic pressure exceeds the turbulent pressure. We also investigate the properties of each filament in Mon R2. Most of the filaments are aligned along the magnetic field direction and are magnetically subcritical.

    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

  • Auction-based cloud service allocation and sharing for logistics product service system

    • Sustainability is increasingly viewed as a desired goal of social development. The logistics industry without exception is aware of the importance of sustainable development. To achieve this goal, the logistics industry is leveraging cutting-edge technologies, such as product service systems (PSS) and cloud manufacturing (CMfg), to design logistics product service systems (LPSS). In LPSS, public logistics resources are allocated to customers and private logistics resources are shared between customers. However, logistics resource allocation and sharing services have been impeded by a lack of efficient methods. In this context, this paper proposes an auction-based cloud service allocation and sharing method for LPSS. Firstly, LPSS is defined and elaborated based on the adoption of PSS in the logistics industry. Secondly, multi-unit Vickery (MV) auctions and one-sided Vickrey-Clarke-Groves (O-VCG) combinatorial auctions are proposed to address logistics resource allocation and sharing problems respectively in LPSS. Two auctions are introduced specifically, and their relevant properties are investigated, including incentive compatibility, allocative efficiency, budget balance, and individual rationality. Thirdly, computational studies are conducted to examine the performance of two auctions. The results reveal that MV auctions can efficiently allocate public logistics resources through ensuring the utility of logistics service providers under dynamic supply and demand. Additionally, O-VCG auctions can effectively integrate and share idle private logistics resources, which promotes sustainability in the logistics industry. Through integrating MV auctions with O-VCG auctions, the utility of logistics service providers can be increased.

  • Genome-wide annotation of protein-coding genes in pig

    • Background: There is a need for functional genome-wide annotation of the protein-coding genes to get a deeper understanding of mammalian biology. Here, a new annotation strategy is introduced based on dimensionality reduction and density-based clustering of whole-body co-expression patterns. This strategy has been used to explore the gene expression landscape in pig, and we present a whole-body map of all protein-coding genes in all major pig tissues and organs. Results: An open-access pig expression map (www.rnaatlas.org ) is presented based on the expression of 350 samples across 98 well-defined pig tissues divided into 44 tissue groups. A new UMAP-based classification scheme is introduced, in which all protein-coding genes are stratified into tissue expression clusters based on body-wide expression profiles. The distribution and tissue specificity of all 22,342 protein-coding pig genes are presented. Conclusions: Here, we present a new genome-wide annotation strategy based on dimensionality reduction and density-based clustering. A genome-wide resource of the transcriptome map across all major tissues and organs in pig is presented, and the data is available as an open-access resource (www.rnaatlas.org), including a comparison to the expression of human orthologs.

  • The JCMT BISTRO Survey: Studying the Complex Magnetic Field of L43

    • We present observations of polarized dust emission at 850 mu m from the L43 molecular cloud, which sits in the Ophiuchus cloud complex. The data were taken using SCUBA-2/POL-2 on the James Clerk Maxwell Telescope as a part of the BISTRO large program. L43 is a dense (N-H2 similar to 10(22) - 10(23) cm(-2)) complex molecular cloud with a submillimeter-bright starless core and two protostellar sources. There appears to be an evolutionary gradient along the isolated filament that L43 is embedded within, with the most evolved source closest to the Sco OB2 association. One of the protostars drives a CO outflow that has created a cavity to the southeast. We see a magnetic field that appears to be aligned with the cavity walls of the outflow, suggesting interaction with the outflow. We also find a magnetic field strength of up to similar to 160 +/- 30 mu G in the main starless core and up to similar to 90 +/- 40 mu G in the more diffuse, extended region. These field strengths give magnetically super- and subcritical values, respectively, and both are found to be roughly trans-Alfvenic. We also present a new method of data reduction for these denser but fainter objects like starless cores.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

  • B-fields in Star-forming Region Observations (BISTRO): Magnetic Fields in the Filamentary Structures of Serpens Main

    • We present 850 mu m polarimetric observations toward the Serpens Main molecular cloud obtained using the POL-2 polarimeter on the James Clerk Maxwell Telescope as part of the B-fields In STar-forming Region Observations survey. These observations probe the magnetic field morphology of the Serpens Main molecular cloud on about 6000 au scales, which consists of cores and six filaments with different physical properties such as density and star formation activity. Using the histogram of relative orientation (HRO) technique, we find that magnetic fields are parallel to filaments in less-dense filamentary structures where NH2 < 0.93 x 10(22) cm(-2) (magnetic fields perpendicular to density gradients), while they are perpendicular to filaments (magnetic fields parallel to density gradients) in dense filamentary structures with star formation activity. Moreover, applying the HRO technique to denser core regions, we find that magnetic field orientations change to become perpendicular to density gradients again at NH2 approximate to 4.6 x 10(22) NH2 approximate to 16 x 10(22) cm(-2), magnetic fields change back to being parallel to density gradients once again, which can be understood to be due to magnetic fields being dragged in by infalling material. In addition, we estimate the magnetic field strengths of the filaments (B-POS = 60-300 mu G)) using the Davis-Chandrasekhar-Fermi method and discuss whether the filaments are gravitationally unstable based on magnetic field and turbulence energy densities.

    • We have investigated the performance of advection schemes (focusing mainlyon Total Variation Diminishing, TVD, schemes)applied in modelling diesel sprays, and assessed their influence on liquidspray penetration and fuel vapour distribution. Here,we compare sprays simulated using several types of TVD schemes (Superbee,MUSCL, limited Linear, and UMIST) – andstandard upwind and linear schemes as references – in conjunction withthree different turbulence models (standard, RNG andLaunder-Sharma k-ε models), to non-reacting diesel sprays observed inthe Sandia high-pressure, high-temperature constant-volume vessel. The OpenFOAM CFD code was used for all of the simulationsdescribed. In addition to comparing thesimulations to experimental data, we provide overall assessments of theperformance and utility of the TVD schemes in multi-dimensional diesel modelling.

    • Three-dimensional simulations of ignition and combustion of a dieselspray were conducted. The primary goal of the work was to compare twodifferent CFD codes: OpenFOAM, an object-oriented C++ based code, andKIVA-3V. The spray is modelled by the Eulerian-Lagrangian approach inboth codes, with several common submodels. Some important sub-modelsimplemented include \emph{inter alia} aKelvin-Helmholtz/Rayleigh-Taylor (KH/RT) model for spray break-up, animproved spray collision model, and a Partially Stirred Reactor (PaSR)model for turbulence-chemistry interaction. Both CFD codes solve thechemical reaction equations in a fully coupled manner. A cubic-shaped Cartesianmesh was used in the KIVA-3V simulations, while a polyhedral meshincluding a combination of hexagonal and prism-shaped cells wasconstructed for the OpenFOAM computations.The effects of high EGR and ambient temperature on the ignition and flamelift-off processes of a diesel spray were investigated. Sandia experimentsconducted in a high-pressure and high-temperature constant-volume vessel werechosen for the simulations and validations. A single spray was injected intothe vessel, and EGR was mimicked by reducing the oxygen concentration. Thediesel reference fuel (n-heptane) was considered. For the study, a medium-sizemechanism involving 83 species and 338 reactions was employed. The mechanismwas validated using the CHEMKIN II package and the reaction rate constantswere adjusted on the basis of measurements of auto-ignition delays ofn-heptane/air mixtures in shock-tube experiments (with equivalence ratiosranging from 0.2 to 0.4 at 50 bar, and from 0.5 to 2.0 at 13.5 bar and 41.0bar), laminar flame speeds (1 atm and 3 atm), and flame structure inburner-stabilized premixed flames (1 atm).The simulations demonstrate that both CFD codes are capable of spray ignitionand combustion studies, though both show stronggrid-dependence. The numerical results show that the ignition delay,flame lift-off and combustion temperature of the spray are stronglyinfluenced by EGR and ambient gas temperature. These predictions arein agreement with measurements. Nevertheless, differences are observedbetween the results predicted by OpenFOAM and those from KIVA-3V, forexample, the flame predicted by the former is thinner and longer than that by thelatter, which requires further investigation.

    • Directed differentiation of human induced pluripotent stem cells (iPSCs) into retinal pigmented epithelium (RPE) holds great promise in cell replacement therapy for patients suffering from degenerative eye diseases, including age-related macular degeneration (AMD). In this study, we generated iPSCs from human dermal fibroblasts (HDFs) by electroporation with episomal plasmid vectors encoding OCT4, SOX2, KLF4, L-MYC together with p53 suppression. Intriguingly, cell reprogramming resulted in a metastable transcriptional activation and selective demethylation of neural and retinal specification-associated genes, such as OTX2, RX1 and SIX3. In contrast, RPE progenitor genes were transcriptionally silent in HDFs and descendant iPSCs. Overexpression of OCT4 and SOX2 directly stimulated the expression of OTX2, RX1 and SIX3 in HDFs and iPSCs. Luciferase and chromatin immunoprecipitation (ChIP) assays further identified an OCT4- and two SOX2-binding sites located in the proximal promoter of OTX2. Histone acetylation and methylation on the local promoter also participated in the reactivation of OTX2. The transcriptional conversion of RX1 and SIX3 genes partially attributed to DNA demethylation. Subsequently, iPSCs were induced into the RPE cells displaying the characteristics of polygonal shapes and pigments, and expressing typical RPE cell markers. Taken together, our results establish readily efficient and safe protocols to produce iPSCs and iPSC-derived RPE cells, and underline that the reactivation of anterior neural transcription factor OTX2, eye field transcription factor RX1 and SIX3 in iPSCs is a feature of pluripotency acquisition and predetermines the potential of RPE differentiation.
