  • Evaluation of feedback methods for improved detection of hindlimb lameness in horses among riding instructors and trainers

    • Lameness, a wellknown issue in sport horses, impedes performance and impairs welfare. Early detection of lameness is essential for horses to receive needed treatment, but detection of hindlimb lameness is challenging. Riding instructors and trainers observe horses in motion in their daily work and could contribute to more efficient lameness detection. In this cross-sectional and prospective study, we evaluated the ability of riding instructors and trainers to assess hindlimb lameness. We also evaluated different feedback methods for improved lameness detection. For the cross-sectional part, n = 64 riding instructors and trainers of varying level and n = 23 high-level trainers were shown 13 videos of trotting horses, lameness degree: 0-3.5 (test 1) and tasked with classifying the horses as sound, left hindlimb lame, or right hindlimb lame. For the prospective part, the riding instructors and trainers of varying levels were randomly allocated to three different groups (a, b, c) and given 14 days of feedback-based, computer-aided training in identifying hindlimb lameness, where they assessed 13 videos (of which three were repeated from test 1) of horses trotting in a straight line. Participants in groups a-c received different feedback after each video (group a: correct answer and re-viewing of video at full and 65% speed; group b: correct answer, re-viewing of video at full and 65% speed, narrator providing explanations; group c: correct answer and re-viewing of video at full speed). After computer-aided training, the participants were again subjected to the video test (test 2). Participants also provided background information regarding level of training etcetera. Effects of participants' background on results were analyzed using analysis of variance, and effects of the different feedback methods were analyzed using generalized estimation equations. On test 1, 44% (group a), 48% (b), 46% (c), and 47% (high-level trainers) of horses were correctly classified. Group a participants significantly improved their test score, both with (p < 0.0001) and without (p = 0.0086) inclusion of repeated videos. For group c, significant improvement was only seen with inclusion of repeated videos (p = 0.041). For group b, no significant improvement was seen (p = 0.51). Although test 2 scores were low, computer-aided training may be useful for improving hindlimb lameness detection.

    • The aggregation of the amyloid β peptide (Aβ) into amyloid fibrils is a defining characteristic of Alzheimer’s disease. Because of the complexity of this aggregation process, effective therapeutic inhibitors will need to target the specific microscopic steps that lead to the production of neurotoxic species. We introduce a strategy for generating fibril-specific antibodies that selectively suppress fibril-dependent secondary nucleation of the 42-residue form of Aβ (Aβ42). We target this step because it has been shown to produce the majority of neurotoxic species during aggregation of Aβ42. Starting from large phage display libraries of single-chain antibody fragments (scFvs), the three-stage approach that we describe includes (i) selection of scFvs with high affinity for Aβ42 fibrils after removal of scFvs that bind Aβ42 in its monomeric form; (ii) ranking, by surface plasmon resonance affinity measurements, of the resulting candidate scFvs that bind to the Aβ42 fibrils; and (iii) kinetic screening and analysis to find the scFvs that inhibit selectively the fibril-catalyzed secondary nucleation process in Aβ42 aggregation. By applying this approach, we have identified four scFvs that inhibit specifically the fibril-dependent secondary nucleation process. Our method also makes it possible to discard antibodies that inhibit elongation, an important factor because the suppression of elongation does not target directly the production of toxic oligomers and may even lead to its increase. On the basis of our results, we suggest that the method described here could form the basis for rationally designed immunotherapy strategies to combat Alzheimer’s and related neurodegenerative diseases.

    • This paper presents findings from real life situations, a longitudinal single case study on the role of natural environments in nature-based rehabilitation (NBR) for individuals with stress-related mental disorders, at the Alnarp Rehabilitation Garden in Sweden. A sample of 43 former clients voluntarily participated in semi-structured interview, and the data were analyzed according to interpretative phenomenological analysis (IPA). Three main superordinate themes were identified as the three phases of NBR-Prelude, Recuperating and Empowerment-explaining and illuminating the role of the natural environments in each phase. An explanatory model of NBR in this context is presented including the three phases of NBR, IRP supportive occupations and a pyramid of supporting environments. A new component of supportive environments was identified and herby named, Social quietness, an important component facilitating personal and intimate engagement with the natural environments.

    • This paper explores environmental education research in primary school. The longitudinal study started in 2003, with 29 children nine years of age, in a city in southern Sweden. The teacher works with projects in science and technology to stimulate the pupils’ interest and participation in environmental education. Semi-structured interviews with the young pupils have been carried out. In order to analyse the classroom communication between the teacher and the children and also among the children, videotaped sequences from the lessons are collected, in which the Socratic dialogue is practiced. Stimulated recall as a method is also used to find out the teacher’s reactions during the lessons. In order to catch environmental details as well as a holistic perspective, the Earth’s system model is used in the analysis, e.g. the lithosphere, the atmosphere and the hydrosphere as well as the biosphere and the technosphere. It was observed that the children are able to argue about possibilities to change different daily routines in different ways, which reduce disturbances to the environment. Some pupils can see the connection between the increasing greenhouse effect and pollution from the cars. Others can see relations between increasing temperature and melting polar caps. In stimulated recall the teacher points out possible connections in the Earth system and in environmental learning. This material is one part of an ongoing longitudinal doctoral study and the videotaped sequences have been caught from year 2003 to 2006. The interviews with the children have been taped every spring from year 2003 to 2006.
