  • Building bridges to operationalise one health : A Sino-Swedish collaboration to tackle antibiotic resistance

    • Antibiotic resistance is a complex global health challenge. The recent Global Action Plan on antimicrobial resistance highlights the importance of adopting One Health approaches that can cross traditional disciplinary boundaries. We report on the early experiences of a multisectoral Sino-Swedish research project that aims to address gaps in our current knowledge and seeks to improve the situation through system-wide interventions. Our research project is investigating antibiotic use and resistance in a rural area of China through a combination of epidemiological, health systems and laboratory investigations. We reflect here on the challenges inherent in conducting long distance cross-disciplinary collaborations, having now completed data and sample collection for a baseline situation analysis. In particular, we recognise the importance of investing in aspects such as effective communication, shared conceptual frameworks and leadership. We suggest that our experiences will be instructive to others planning to develop similar international One Health collaborations.

  • Band-rejection fiber filter and fiber sensor based on a Bragg fiber of transversal resonant structure

    • We propose a novel band-rejection fiber filter based on a Bragg fiber of transversal resonant structure, which can also be used as a fiber sensor. Defect layers are introduced in the periodic high/low index structure in the cladding of the Bragg fiber. Coupling between the core mode and the defect mode results in large confinement loss for some resonant wavelengths inside the band gap of the Bragg fiber. A segment of the Bragg fiber of transversal resonant structure can be used as a band-rejection fiber filter, whose characteristics are mainly determined by the defect layer. The loss peak wavelength of the Bragg fiber is dependent on the refractive index and the thickness of the defect layer which indicates its applications of refractive index and strain sensing.

  • Allosteric inhibitor remotely modulates the conformation of the orthestric pockets in mutant IDH2/R140Q

    • Neomorphic mutation R140Q in the metabolic enzyme isocitrate dehydrogenase 2 (IDH2) is found to be a driver mutation in cancers. Recent studies revealed that allosteric inhibitors could selectively inhibit IDH2/R140Q and induce differentiation of TF-1 erythroleukemia and primary human AML cells. However, the allosteric inhibition mechanism is not very clear. Here, we report the results from computational studies that AGI-6780 binds tightly with the divalent cation binding helices at the homodimer interface and prevents the transition of IDH2/R140Q homodimer to a closed conformation that is required for catalysis, resulting in the decrease of the binding free energy of NADPHs. If the allosteric inhibitor is removed, the original open catalytic center of IDH2/R140Q will gradually reorganize to a quasi-closed conformation and the enzymatic activity might recover. Unlike IDH2/R140Q, AGI-6780 locks one monomer of the wild-type IDH2 in an inactive open conformation and the other in a half-closed conformation, which can be used to explain the selectivity of AGI-6780. Our results suggest that conformational changes are the primary contributors to the inhibitory potency of the allosteric inhibitor. Our study will also facilitate the understanding of the inhibitory and selective mechanisms of AG-221 (a promising allosteric inhibitor that has been approved by FDA) for mutant IDH2.

  • Enhanced efficiency of polymer solar cells by improving molecular aggregation and broadening the absorption spectra

    • Alkylthio-substituted thiophene-based benzo[1,2-b:4,5-b’]dithiophene (BDT) was used to construct PBDTS-DTBT, a medium band gap donor-acceptor (D-A) polymer with 5,6-difluoro-4,7-bis[4-(2-octyldodecyl)thiophene-2-yl]benzo[c] [1,2,5]thiadiazole (DTBT). The incorporation of sulfur atoms into the side chains not only lowered the highest occupied molecular orbital (HOMO) energy level but also improved molecular aggregation and thus afforded lower band gap (1.68 eV) with full width at half maximum (FWHM) of 170 nm in comparison to that of the analogous polymer (PBDT-DTBT) without sulfur atoms in side chains. Therefore, the bulk heterojunction polymer solar cells based on PBDTS-DTBT with 2% diiodooctane (DIO) as processing additive showed a high power conversion efficiency (PCE) of 9.73% with high open-circuit voltage (V OC , 0.93 V), large short-circuit current density (J SC , 14.23 mA/cm 2 ) and high fill factor (FF, 0.735).

  • Momentum-resolved resonant inelastic soft X-ray scattering (qRIXS) endstation at the ALS

    • A momentum resolved resonant inelastic X-ray scattering (qRIXS) experimental station with continuously rotatable spectrometers and parallel detection is designed to operate at different beamlines at synchrotron and free electron laser (FEL) facilities. This endstation, currently located at the Advanced Light Source (ALS), has five emission ports on the experimental chamber for mounting the high-throughput modular soft X-ray spectrometers (MXS) [24]. Coupled to the rotation from the supporting hexapod, the scattered X-rays from 27.5° (forward scattering) to 152.5° (backward scattering) relative to the incident photon beam can be recorded, enabling the momentum-resolved RIXS spectroscopy. The components of this endstation are described in details, and the preliminary RIXS measurements on highly oriented pyrolytic graphite (HOPG) reveal the low energy vibronic excitations from the strong electron-phonon coupling at C K edge around σ* band. The grating upgrade option to enhance the performance at low photon energies is presented and the potential of this spectroscopy is discussed in summary.

  • The offshore wind speed changes in China: an insight into CMIP6 model simulation and future projections

    • Offshore wind speed in China plays a key role in affecting air-sea interactions, coastal tides, and wind energy, but its changes in a warming climate and the associated causes remain unclear. Based on the ERA5 reanalysis and the Coupled Model Intercomparison Project Phase 6 (CMIP6) models, this study evaluates the past and future variations of wind speed at 10 m (WS10) over China's offshore seas in summer and winter. The results show that the CMIP6 multi-model mean performs well in simulating the climatological patterns (1981-2010) of WS10 for both seasons. The trends and leading variabilities in WS10 are also reasonably reproduced in the South China Sea (SCS). In the northern SCS, WS10 has strengthened during both seasons in the recent decades. In contrast, in the East China Sea (ECS), WS10 has increased (decreased) during summer (winter). Further attribution analysis suggests that the forcing of greenhouse gasses (aerosols) may make WS10 stronger (weaker) in the two seas and for both seasons, while natural variability tends to slow down (speed up) WS10 in the SCS and ECS during summer (winter). In addition, according to the CMIP6 model projections under various warming scenarios, WS10 is likely to increase over both the northern SCS and the ECS in summer, while WS10 will increase over the northern SCS but decrease over the ECS in winter. Differences in the projected WS10 changes in the ECS during summer and winter are attributed to the projected intensification (weakening) of the East Asian summer (winter) monsoon circulation.

  • Effective Urban Traffic Monitoring by Vehicular Sensor Networks

    • Traffic monitoring in urban transportation systems can be carried out based on vehicular sensor networks. Probe vehicles (PVs), such as taxis and buses, and floating cars (FCs), such as patrol cars for surveillance, can act as mobile sensors for sensing the urban traffic and send the reports to a traffic-monitoring center (TMC) for traffic estimation. In the TMC, sensing reports are aggregated to form a traffic matrix, which is used to extract traffic information. Since the sensing vehicles cannot cover all the roads all the time, the TMC needs to estimate the unsampled data in the traffic matrix. As this matrix can be approximated to be of low rank, matrix completion (MC) is an effective method to estimate the unsampled data. However, our previous analysis on the real traces of taxis in Shanghai reveals that MC methods do not work well due to the uneven samples of PVs, which is common in urban traffic. To exploit the intrinsic relationship between the unevenness of samples and traffic estimation error, we study the temporal and spatial entropies of samples and successfully define the important criterion, i.e., average entropy of the sampling process. A new sampling rule based on this relationship is proposed to improve the performance of estimation and monitoring.With the sampling rule, two new patrol algorithms are introduced to plan the paths of controllable FCs to proactively participate in trafficmonitoring. By utilizing the patrol algorithms for real-data-set analysis, the estimation error reduces from 35% to about 10%, compared with the random patrol or interpolation method in traffic estimation. Both the validity of the exploited relationship and the effectiveness of the proposed patrol control algorithms are demonstrated.
