    • This contribution reports results from the gas-phase oxidation of 4-bromo-4'-chlorobiphenyl (4,4'-BCB) in order to fathom the formation of toxic species produced during the combustion of mixed halogenated aromatics. In their own right, mixed polybrominated/polychlorinated biphenyls (PXBs) represent a new class of environmental contaminants, recently detected in food and human tissues. Gas chromatography-quadrupole mass spectrometry (GC-QMS), gas chromatography-quadrupole time of flight mass spectrometry (GC-QTOFMS), Fourier transform infrared spectroscopy (FTIR) and ion chromatography (IC) served to analyse the volatile and semi-volatile organic compounds (V/SVOCs), including mixed halogenated dibenzo-p-dioxins and dibenzofurans (PXDD/Fs), and gaseous products including HBr/HCl. The selection of non-ortho substituted PXB as a model species yields a large number of halogenated compounds, including monochloro- and monobromobenzene and higher halogenated benzenes and naphthalenes and derivatives of halogenated benzenes (such as 1-chloro-4-ethynylbenzene). We also detect small amounts of chlorinated and mixed halogenated dibenzofurans. The present study provides insights into the formation of several classes of halogenated and mixed-halogenated pollutants in combustion processes involving both bromine and chlorine sources, such as those of brominated flame retardants and PVC plastics.

  • Globally consistent influences of seasonal precipitation limit grassland biomass response to elevated CO2

Rising atmospheric carbon dioxide concentration should stimulate biomass production directly via biochemical stimulation of carbon assimilation, and indirectly via water savings caused by increased plant water-use efficiency. Because of these water savings, the CO 2 fertilization effect (CFE) should be stronger at drier sites, yet large differences among experiments in grassland biomass response to elevated CO 2 appear to be unrelated to annual precipitation, preventing useful generalizations. Here, we show that, as predicted, the impact of elevated CO 2 on biomass production in 19 globally distributed temperate grassland experiments reduces as mean precipitation in seasons other than spring increases, but that it rises unexpectedly as mean spring precipitation increases. Moreover, because sites with high spring precipitation also tend to have high precipitation at other times, these effects of spring and non-spring precipitation on the CO 2 response offset each other, constraining the response of ecosystem productivity to rising CO 2 . This explains why previous analyses were unable to discern a reliable trend between site dryness and the CFE. Thus, the CFE in temperate grasslands worldwide will be constrained by their natural rainfall seasonality such that the stimulation of biomass by rising CO 2 could be substantially less than anticipated.

  • Genome-wide identification of DNA methylation QTLs in whole blood highlights pathways for cardiovascular disease

    • Identifying methylation quantitative trait loci (meQTLs) and integrating them with disease-associated variants from genome-wide association studies (GWAS) may illuminate functional mechanisms underlying genetic variant-disease associations. Here, we perform GWAS of >415 thousand CpG methylation sites in whole blood from 4170 individuals and map 4.7 million cis- and 630 thousand trans-meQTL variants targeting >120 thousand CpGs. Independent replication is performed in 1347 participants from two studies. By linking cis-meQTL variants with GWAS results for cardiovascular disease (CVD) traits, we identify 92 putatively causal CpGs for CVD traits by Mendelian randomization analysis. Further integrating gene expression data reveals evidence of cis CpG-transcript pairs causally linked to CVD. In addition, we identify 22 trans-meQTL hotspots each targeting more than 30 CpGs and find that trans-meQTL hotspots appear to act in cis on expression of nearby transcriptional regulatory genes. Our findings provide a powerful meQTL resource and shed light on DNA methylation involvement in human diseases.

    • Batteries represent the dominant means for storing electrical energy, but many battery chemistries create waste streams that are difficult to manage, and most possess toxic components that limit their use in biomedical applications. Batteries constructed from materials capable of complete, harmless resorption into the environment or into living organisms after a desired period of operation bypass these disadvantages. However, previously reported eco/bioresorbable batteries offer low operating voltages and modest energy densities. Here, we introduce a magnesium-iodine chemistry and dual (ionic liquid/aqueous) electrolyte to overcome these limitations, enabling significant improvements in voltage, areal capacity, areal energy, areal power, volumetric energy, and volumetric power densities over any alternative. Systematic studies reveal key materials and design considerations. Demonstrations of this technology include power supplies for cardiac pacemakers, wireless environmental monitors, and thermal sensors/actuators. These results suggest strong potential for applications where commercial battery alternatives pose risks to the environment or the human body.

  • No symphony without bassoon and piccolo : Changes in synaptic active zone proteins in Huntington's disease

    • Prominent features of HD neuropathology are the intranuclear and cytoplasmic inclusions of huntingtin and striatal and cortical neuronal cell death. Recently, synaptic defects have been reported on HD-related studies, including impairment of neurotransmitter release and alterations of synaptic components. However, the definite characteristics of synapse dysfunction and the underlying mechanisms remain largely unknown. We studied the gene expression levels and patterns of a number of proteins forming the cytoskeletal matrix of the presynaptic active zones in HD transgenic mice (R6/1), in hippocampal neuronal cultures overexpressing mutant huntingtin and in postmortem brain tissues of HD patients. To investigate the interactions between huntingtin and active proteins, we performed confocal microscopic imaging and immunoprecipitation in mouse and HEK 293 cell line models. The mRNA and protein levels of Bassoon were reduced in mouse and cell culture models of HD and in brain tissues of patients with HD. Moreover, a striking re-distribution of a complex of proteins including Bassoon, Piccolo and Munc 13-1 from the cytoplasm and synapses into intranuclear huntingtin aggregates with loss of active zone proteins and dendritic spines. This re-localization was age-dependent and coincided with the formation of huntingtin aggregates. Using co-immunoprecipitation, we demonstrated that huntingtin interacts with Bassoon, and that this interaction is likely mediated by a third linking protein. Three structural proteins involved in neurotransmitter release in the presynaptic active zones of neurons are altered in expression and that the proteins are redistributed from their normal functional site into mutant huntingtin aggregates.

Many subsequent association studies confirmed this finding and implicated this gene in schizophrenia. However, the exact nature of the role of ANK3 in the pathophysiology remains elusive. In particular, the specific isoforms involved and the nature of the imbalance are unknown.

  • Probabilistic integrated flexible regions of multi-energy industrial parks : Conceptualization and characterization

    • Multi-energy industrial parks, composed of the district energy supply system and terminal industrial loads, are dominant energy consumers with over 50% occupation of total energy consumption. Considering the energy conversion in the district energy supply system and adjustment of production subtasks in terminal industrial loads, the industrial parks could provide considerable flexibility for utility multi-energy systems to deal with contingencies. However, industrial parks may have multiple uncertainties originating from the production subtasks' variability, distributed renewable energy's fluctuation, and random failures of equipment. These uncertainties could make the flexibility assessment of industrial parks unreliable, which may further exacerbate utility system risks. To address the problem, this paper innovatively proposes the concept of probabilistic integrated flexible regions and corresponding characterization approaches, which can effectively describe the credible multi-energy adjustment ability of industrial parks. First, the energy-material flows in the industrial park are modeled considering multiple uncertainties while subject to constraints such as operating limits of equipment, energy and material balancing, and production targets. Then, the modeling and calculation methods of probabilistic integrated flexible regions are given. These regions are labeled with certain probability requirements and visualized in the input multi-energy space. Using a real-world test system that produces air conditioning equipment in China, the results show that the proposed model can effectively reflect the probability characteristics of the integrated flexibility under multiple uncertainties. For this test system, the area of a highly credible integrated flexibility, represented by the probabilistic integrated flexible region with 99% probability, is about half of that obtained by the deterministic model. That is, half of the integrated flexibility evaluated by the deterministic method in the test system may not be practically utilized considering the impacts of uncertainties. Therefore, the proposed method could provide a credible and comprehensive evaluation of the multi-energy industrial park's flexibility considering multiple uncertainties.
