  • Fps1p controls the accumulation and release of the compatible solute glycerol in yeast osmoregulation.

    • The accumulation of compatible solutes, such as glycerol, in the yeast Saccharomyces cerevisiae, is a ubiquitous mechanism in cellular osmoregulation. Here, we demonstrate that yeast cells control glycerol accumulation in part via a regulated, Fps1p-mediated export of glycerol. Fps1p is a member of the MIP family of channel proteins most closely related to the bacterial glycerol facilitators. The protein is localized in the plasma membrane. The physiological role of Fps1p appears to be glycerol export rather than uptake. Fps1 delta mutants are sensitive to hypo-osmotic shock, demonstrating that osmolyte export is required for recovery from a sudden drop in external osmolarity. In wild-type cells, the glycerol transport rate is decreased by hyperosmotic shock and increased by hypo-osmotic shock on a subminute time scale. This regulation seems to be independent of the known yeast osmosensing HOG and PKC signalling pathways. Mutants lacking the unique hydrophilic N-terminal domain of Fps1p, or certain parts thereof, fail to reduce the glycerol transport rate after a hyperosmotic shock. Yeast cells carrying these constructs constitutively release glycerol and show a dominant hyperosmosensitivity, but compensate for glycerol loss after prolonged incubation by glycerol overproduction. Fps1p may be an example of a more widespread class of regulators of osmoadaptation, which control the cellular content and release of compatible solutes.

  • Heavy Metals and Metalloids As a Cause for Protein Misfolding and Aggregation

    • While the toxicity of metals and metalloids, like arsenic, cadmium, mercury, lead and chromium, is undisputed, the underlying molecular mechanisms are not entirely clear. General consensus holds that proteins are the prime targets; heavy metals interfere with the physiological activity of specific, particularly susceptible proteins, either by forming a complex with functional side chain groups or by displacing essential metal ions in metalloproteins. Recent studies have revealed an additional mode of metal action targeted at proteins in a non-native state; certain heavy metals and metalloids have been found to inhibit the in vitro refolding of chemically denatured proteins, to interfere with protein folding in vivo and to cause aggregation of nascent proteins in living cells. Apparently, unfolded proteins with motile backbone and side chains are considerably more prone to engage in stable, pluridentate metal complexes than native proteins with their well-defined 3D structure. By interfering with the folding process, heavy metal ions and metalloids profoundly affect protein homeostasis and cell viability. This review describes how heavy metals impede protein folding and promote protein aggregation, how cells regulate quality control systems to protect themselves from metal toxicity and how metals might contribute to protein misfolding disorders.

    • We have recently developed a new modeling capability to embed the implicit particle-in-cell (PIC) model iPIC3D into the Block-Adaptive-Tree-Solarwind-Roe-Upwind-Scheme magnetohydrodynamic (MHD) model. The MHD with embedded PIC domains (MHD-EPIC) algorithm is a two-way coupled kinetic-fluid model. As one of the very first applications of the MHD-EPIC algorithm, we simulate the interaction between Jupiter's magnetospheric plasma and Ganymede's magnetosphere. We compare the MHD-EPIC simulations with pure Hall MHD simulations and compare both model results with Galileo observations to assess the importance of kinetic effects in controlling the configuration and dynamics of Ganymede's magnetosphere. We find that the Hall MHD and MHD-EPIC solutions are qualitatively similar, but there are significant quantitative differences. In particular, the density and pressure inside the magnetosphere show different distributions. For our baseline grid resolution the PIC solution is more dynamic than the Hall MHD simulation and it compares significantly better with the Galileo magnetic measurements than the Hall MHD solution. The power spectra of the observed and simulated magnetic field fluctuations agree extremely well for the MHD-EPIC model. The MHD-EPIC simulation also produced a few flux transfer events (FTEs) that have magnetic signatures very similar to an observed event. The simulation shows that the FTEs often exhibit complex 3-D structures with their orientations changing substantially between the equatorial plane and the Galileo trajectory, which explains the magnetic signatures observed during the magnetopause crossings. The computational cost of the MHD-EPIC simulation was only about 4 times more than that of the Hall MHD simulation. Key Points

Integrated modeling (IM) of present experiments and future tokamak reactors requires the provision of computational resources and numerical tools capable of simulating multiscale spatial phenomena as well as fast transient events and relatively slow plasma evolution within a reasonably short computational time. Recent progress in the implementation of the new computational resources for fusion applications in Europe based on modern supercomputer technologies (supercomputer MARCONI-FUSION), in the optimization and speedup of the EU fusion-related first-principle codes, and in the development of a basis for physics codes/modules integration into a centrally maintained suite of IM tools achieved within the EUROfusion Consortium is presented. Physics phenomena that can now be reasonably modelled in various areas (core turbulence and magnetic reconnection, edge and scrape-off layer physics, radio-frequency heating and current drive, magnetohydrodynamic model, reflectometry simulations) following successful code optimizations and parallelization are briefly described. Development activities in support to IM are summarized. They include support to (1) the local deployment of the IM infrastructure and access to experimental data at various host sites, (2) the management of releases for sophisticated IM workflows involving a large number of components, and (3) the performance optimization of complex IM workflows.

    • Models for long‐term glass alteration are required to satisfy performance predictions of vitrified nuclear waste in various disposal scenarios. Durability parameters are usually extracted from short‐term laboratory tests, and sometimes checked with long‐term natural experiments on glasses, termed analogues. In this paper, a unique potential ancient glass analogue from Sweden is discussed. The hillfort glass found at Broborg represents a unique case study as a vitrified waste glass analogue to compare to Low Activity Waste glass to be emplaced in near surface conditions at Hanford (Washington State). Glasses at Broborg have similar and dissimilar compositions to LAW glass, allowing the testing of long‐term alteration of different glass chemistries. Additionally, the environmental history of the site is reasonably well documented. Initial investigations on previously collected samples established methodologies for handling and characterizing these artifacts by laboratory methods while preserving their alteration layers and cultural context. Evidence of possible biologically influenced glass alteration, and differential alteration in the two types of glass found at the Broborg site is presented.
