  • Gender and telomere length : Systematic review and meta-analysis

    • Background: It is widely believed that females have longer telomeres than males, although results from studies have been contradictory. Methods: We carried out a systematic review and meta-analyses to test the hypothesis that in humans, females have longer telomeres than males and that this association becomes stronger with increasing age. Searches were conducted in EMBASE and MEDLINE (by November 2009) and additional datasets were obtained from study investigators. Eligible observational studies measured telomeres for both females and males of any age, had a minimum sample size of 100 and included participants not part of a diseased group. We calculated summary estimates using random-effects meta-analyses. Heterogeneity between studies was investigated using sub-group analysis and meta-regression. Results: Meta-analyses from 36 cohorts (36,230 participants) showed that on average females had longer telomeres than males (standardised difference in telomere length between females and males 0.090, 95% CI 0.015, 0.166; age-adjusted). There was little evidence that these associations varied by age group (p = 1.00) or cell type (p = 0.29). However, the size of this difference did vary by measurement methods, with only Southern blot but neither real-time PCR nor Flow-FISH showing a significant difference. This difference was not associated with random measurement error. Conclusions: Telomere length is longer in females thanmales, although this difference was not universally found in studies that did not use Southern blot methods. Further research on explanations for the methodological differences is required. (C) 2013 Published by Elsevier Inc.

    • The flow fields and heat transfer characteristics in a parallel-plate channel with a transversely placed spiral coil insert were investigated by three-dimensional numerical simulation. The structure of multi-longitudinal-vortices (MLVs) induced by the spiral coil and the effects of MLVs on velocity and temperature fields were studied. The three-dimensional spiral coil induces a series of longitudinal vortices in the channel including leading longitudinal vortex, mainstream longitudinal vortices, near-wall longitudinal vortices, and rear central longitudinal vortex. Transport by the longitudinal vortices can increase the mass exchange between the boundary layer and the mainstream, which speeds up the heat migration from the channel walls and enhances the heat diffusion in the mainstream.

  • Genome-wide association analysis identifies six new loci associated with forced vital capacity

    • Forced vital capacity (FVC), a spirometric measure of pulmonary function, reflects lung volume and is used to diagnose and monitor lung diseases. We performed genome-wide association study meta-analysis of FVC in 52,253 individuals from 26 studies and followed up the top associations in 32,917 additional individuals of European ancestry. We found six new regions associated at genome-wide significance (P < 5 × 10(-8)) with FVC in or near EFEMP1, BMP6, MIR129-2-HSD17B12, PRDM11, WWOX and KCNJ2. Two loci previously associated with spirometric measures (GSTCD and PTCH1) were related to FVC. Newly implicated regions were followed up in samples from African-American, Korean, Chinese and Hispanic individuals. We detected transcripts for all six newly implicated genes in human lung tissue. The new loci may inform mechanisms involved in lung development and the pathogenesis of restrictive lung disease.

  • Supervised Link Prediction Using Multiple Sources

    • Link prediction is a fundamental problem in social network analysis and modern-day commercial applications such as Facebook and Myspace. Most existing research approaches this problem by exploring the topological structure of a social network using only one source of information. However, in many application domains, in addition to the social network of interest, there are a number of auxiliary social networks and/or derived proximity networks available. The contribution of the paper is twofold: (1) a supervised learning framework that can effectively and efficiently learn the dynamics of social networks in the presence of auxiliary networks; (2) a feature design scheme for constructing a rich variety of path-based features using multiple sources, and an effective feature selection strategy based on structured sparsity. Extensive experiments on three real-world collaboration networks show that our model can effectively learn to predict new links using multiple sources, yielding higher prediction accuracy than unsupervised and singlesource supervised models.

  • Replication and Recombination of Herpes Simplex Virus DNA

    • Replication of Herpes simplex virus takes place in the cell nucleus and is carried out by a replisome composed of viral proteins: the UL30/42 DNA polymerase, the UL5,8,52 helicase-primase and the UL29 single-strand DNA binding protein ICP8. The replisome is loaded on origins of replication by an initiator protein OBP/UL9. Virus replication is intimately coupled to recombination and repair, often performed by cellular proteins. Here we review significant new developments: the three-dimensional structures for the DNA polymerase, the polymerase accessory factor and the single-strand DNA binding protein, the reconstitution of a functional replisome in vitro, the elucidation of the mechanism for activation of origins of DNA replication, the identification of cellular proteins actively involved in or responding to viral DNA replication and, finally, the elucidation of requirements for formation of replication foci in the nucleus and effects on protein localization.

    • Nanotopographical features are found to have significant effects on bone behavior. In the presentstudy, nanoporous aluminas with different pore sizes (20, 100 and 200 nm in diameter), were eva-luated for their osteoinductive and drug eluting properties. W20-17 marrow stromal cells wereseeded on nanoporous alumina with and without the addition of BMP-2. Although cell prolifera-tion was not affected by pore size, osteogenic differentiation was 200 nm as compared to 20 and100 nm pores induced higher alkaline phosphatase activity (ALP) and osteocalcin expression le-vels, thus indicating osteoblastic differentiation. Cell morphology revealed that cells cultured on20 nm pores adopted a rounded shape, while larger pores (200 nm) elicited an elongated morpho-logy. Furthermore, ALP expression levels were consistently higher on BMP-2 loaded nanoporousalumina surfaces compared to unloaded surfaces, indicating that not only is nanoporous aluminaosteoinductive, but also has the potential to be used as a drug eluting bone-implant coating.

  • Genome-wide association study of glioma and meta-analysis

    • Gliomas account for approximately 80 % of all primary malignant brain tumors and, despite improvements in clinical care over the last 20 years, remain among the most lethal tumors, underscoring the need for gaining new insights that could translate into clinical advances. Recent genome-wide association studies (GWAS) have identified seven new susceptibility regions. We conducted a new independent GWAS of glioma using 1,856 cases and 4,955 controls (from 14 cohort studies, 3 case-control studies, and 1 population-based case-only study) and found evidence of strong replication for three of the seven previously reported associations at 20q13.33 (RTEL), 5p15.33 (TERT), and 9p21.3 (CDKN2BAS), and consistent association signals for the remaining four at 7p11.2 (EGFR both loci), 8q24.21 (CCDC26) and 11q23.3 (PHLDB1). The direction and magnitude of the signal were consistent for samples from cohort and case-control studies, but the strength of the association was more pronounced for loci rs6010620 (20q,13.33; RTEL) and rs2736100 (5p15.33, TERT) in cohort studies despite the smaller number of cases in this group, likely due to relatively more higher grade tumors being captured in the cohort studies. We further examined the 85 most promising single nucleotide polymorphism (SNP) markers identified in our study in three replication sets (5,015 cases and 11,601 controls), but no new markers reached genome-wide significance. Our findings suggest that larger studies focusing on novel approaches as well as specific tumor subtypes or subgroups will be required to identify additional common susceptibility loci for glioma risk.
