  • Evaluating the impact of alternative phenotype definitions on incidence rates across a global data network.
  • 2023
  • Ingår i: JAMIA open. - 2574-2531. ; 6:4
  • Tidskriftsartikel (refereegranskat)abstract
    • Developing accurate phenotype definitions is critical in obtaining reliable and reproducible background rates in safety research. This study aims to illustrate the differences in background incidence rates by comparing definitions for a given outcome. We used 16 data sources to systematically generate and evaluate outcomes for 13 adverse events and their overall background rates. We examined the effect of different modifications (inpatient setting, standardization of code set, and code set changes) to the computable phenotype on background incidence rates.Rate ratios (RRs) of the incidence rates from each computable phenotype definition varied across outcomes, with inpatient restriction showing the highest variation from 1 to 11.93. Standardization of code set RRs ranges from 1 to 1.64, and code set changes range from 1 to 2.52.The modification that has the highest impact is requiring inpatient place of service, leading to at least a 2-fold higher incidence rate in the base definition. Standardization showed almost no change when using source code variations. The strength of the effect in the inpatient restriction is highly dependent on the outcome. Changing definitions from broad to narrow showed the most variability by age/gender/database across phenotypes and less than a 2-fold increase in rate compared to the base definition.Characterization of outcomes across a network of databases yields insights into sensitivity and specificity trade-offs when definitions are altered. Outcomes should be thoroughly evaluated prior to use for background rates for their plausibility for use across a global network.

  • High-precision mass measurements for the isobaric multiplet mass equation at A = 52
  • 2017
  • Ingår i: Journal of Physics G: Nuclear and Particle Physics. - : IOP Publishing. - 0954-3899 .- 1361-6471. ; 44:6
  • Tidskriftsartikel (refereegranskat)abstract
    • Masses of 52Co, 52Com, 52Fe, 52Fem, and 52Mn have been measured with the JYFLTRAP double Penning trap mass spectrometer. The isobaric multiplet mass equation for the T = 2 quintet at A = 52 has been studied employing the new mass values. No significant breakdown (beyond the level) of the quadratic form of the IMME was observed (). The cubic coefficient was 6.0(32) keV (). The excitation energies for the isomer and the T = 2 isobaric analog state in 52Co have been determined to be 374(13) keV and 2922(13) keV, respectively. The measured mass values for 52Co and 52Com are 29(10) keV and 16(15) keV higher, respectively, than obtained in a recent storage-ring experiment, and significantly lower than predicted by extrapolations. Consequently, this has an impact on the proton separation energies for 52Co and 53Ni relevant for the astrophysical rapid proton capture process. The Q value for the proton decay from the isomer in 53Co has been determined with an unprecedented precision, keV.

  • Integrated ecological–economic fisheries models—Evaluation, review and challenges for implementation
  • 2018
  • Ingår i: Fish and Fisheries. - : Wiley. - 1467-2960 .- 1467-2979. ; 19:1, s. 1-29
  • Forskningsöversikt (refereegranskat)abstract
    • Marine ecosystems evolve under many interconnected and area-specific pressures. To fulfil society's intensifying and diversifying needs while ensuring ecologically sustainable development, more effective marine spatial planning and broader-scope management of marine resources is necessary. Integrated ecological–economic fisheries models (IEEFMs) of marine systems are needed to evaluate impacts and sustainability of potential management actions and understand, and anticipate ecological, economic and social dynamics at a range of scales from local to national and regional. To make these models most effective, it is important to determine how model characteristics and methods of communicating results influence the model implementation, the nature of the advice that can be provided and the impact on decisions taken by managers. This article presents a global review and comparative evaluation of 35 IEEFMs applied to marine fisheries and marine ecosystem resources to identify the characteristics that determine their usefulness, effectiveness and implementation. The focus is on fully integrated models that allow for feedbacks between ecological and human processes although not all the models reviewed achieve that. Modellers must invest more time to make models user friendly and to participate in management fora where models and model results can be explained and discussed. Such involvement is beneficial to all parties, leading to improvement of models and more effective implementation of advice, but demands substantial resources which must be built into the governance process. It takes time to develop effective processes for using IEEFMs requiring a long-term commitment to integrating multidisciplinary modelling advice into management decision-making.

  • Molecular pathways in patients with systemic lupus erythematosus revealed by gene-centred DNA sequencing
  • 2021
  • Ingår i: Annals of the Rheumatic Diseases. - : BMJ. - 0003-4967 .- 1468-2060. ; 80:1, s. 109-117
  • Tidskriftsartikel (refereegranskat)abstract
    • Objectives: Systemic lupus erythematosus (SLE) is an autoimmune disease with extensive heterogeneity in disease presentation between patients, which is likely due to an underlying molecular diversity. Here, we aimed at elucidating the genetic aetiology of SLE from the immunity pathway level to the single variant level, and stratify patients with SLE into distinguishable molecular subgroups, which could inform treatment choices in SLE. Methods: We undertook a pathway-centred approach, using sequencing of immunological pathway genes. Altogether 1832 candidate genes were analysed in 958 Swedish patients with SLE and 1026 healthy individuals. Aggregate and single variant association testing was performed, and we generated pathway polygenic risk scores (PRS). Results: We identified two main independent pathways involved in SLE susceptibility: T lymphocyte differentiation and innate immunity, characterised by HLA and interferon, respectively. Pathway PRS defined pathways in individual patients, who on average were positive for seven pathways. We found that SLE organ damage was more pronounced in patients positive for the T or B cell receptor signalling pathways. Further, pathway PRS-based clustering allowed stratification of patients into four groups with different risk score profiles. Studying sets of genes with priors for involvement in SLE, we observed an aggregate common variant contribution to SLE at genes previously reported for monogenic SLE as well as at interferonopathy genes. Conclusions: Our results show that pathway risk scores have the potential to stratify patients with SLE beyond clinical manifestations into molecular subsets, which may have implications for clinical follow-up and therapy selection.

  • Elucidating the nature of the proton radioactivity and branching ratio on the first proton emitter discovered 53mCo
  • 2023
  • Ingår i: Nature Communications. - 2041-1723. ; 14
  • Tidskriftsartikel (refereegranskat)abstract
    • The observation of a weak proton-emission branch in the decay of the 3174keV 53mCo isomeric state marked the discovery of proton radioactivity in atomic nuclei in 1970. Here we show, based on the partial half-lives and the decay energies of the possible proton-emission branches, that the exceptionally high angular momentum barriers, lp = 9 and lp = 7, play a key role in hindering the proton radioactivity from 53mCo, making them very challenging to observe and calculate. Indeed, experiments had to wait decades for significant advances in accelerator facilities and multi-faceted state-of-the-art decay stations to gain full access to all observables. Combining data taken with the TASISpec decay station at the Accelerator Laboratory of the University of Jyväskylä, Finland, and the ACTAR TPC device on LISE3 at GANIL, France, we measured their branching ratios as bp1 = 1.3(1)% and bp2 = 0.025(4)%. These results were compared to cutting-edge shell-model and barrier penetration calculations. This description reproduces the order of magnitude of the branching ratios and partial half-lives, despite their very small spectroscopic factors.

  • Automated Aerodynamic Optimization of an Aggressive S-Shaped Intermediate Compressor Duct
  • 2018
  • Ingår i: ASME Turbo Expo 2018: Turbomachinery Technical Conference and Exposition. - : ASME International. ; , s. GT2018-75184, V0-2DT46A002
  • Konferensbidrag (refereegranskat)abstract
    • As bypass-ratio in modern aero engines is continuously increasing over the last decades, the radial offset between low pressure compressor (LPC) and high pressure compressor (HPC), which needs to be overcome by the connecting s-shaped intermediate compressor duct (ICD), is getting higher. Due to performance and weight saving aspects the design of shorter and therefore more aggressive ducts has become an important research topic. In this paper an already aggressive design (with respect to current aero engines) of an ICD with integrated outlet guide vane (OGV) is used as a baseline for an aerodynamic optimization. The aim is to shorten the duct even further while maintaining it separation free. The optimization is broken down into two steps. In the first optimization-step the baseline design is shortened to a feasible extent while keeping weak aerodynamic restrictions. The resulting highly aggressive duct (intermediate design), which is shortened by 19 % in axial length with respect to the baseline, shows separation tendencies of low momentum fluid in the strut/hub region. For the second step, the length of the optimized duct design is frozen. By implementing new design features in the process of the optimizer, this optimization-step aims to eliminate separation and to reduce separation tendencies caused by the aggressive shortening. In particular, these features are: a nonaxisymmetric endwall contouring and parametrization of the strut and the OGV to allow for changes in lift and turning in both blade designs. By comparison of the three designs: Baseline, intermediate (separating flow) and final design, it can be shown, that it is possible to decrease length of the already aggressive baseline design even further, when adding a nonaxisymmetric endwall contouring and changes in blade shape of the strut and OGV. Flow separation can be eliminated while losses are kept low. With a more aggressive and therefore shorter duct the engine length and weight can be reduced. This in turn leads to lighter aircrafts, less fuel consumption and lower CO2 and NOx emissions.

  • Internalization of secreted antigen-targeted antibodies by the neonatal Fc receptor for precision imaging of the androgen receptor axis
  • 2016
  • Ingår i: Science Translational Medicine. - : American Association for the Advancement of Science (AAAS). - 1946-6234 .- 1946-6242. ; 8:367
  • Tidskriftsartikel (refereegranskat)abstract
    • Targeting the androgen receptor (AR) pathway prolongs survival in patients with prostate cancer, but resistance rapidly develops. Understanding this resistance is confounded by a lack of noninvasive means to assess AR activity in vivo. We report intracellular accumulation of a secreted antigen-targeted antibody (SATA) that can be used to characterize disease, guide therapy, and monitor response. AR-regulated human kallikrein-related peptidase 2 (free hK2) is a prostate tissue-specific antigen produced in prostate cancer and androgen-stimulated breast cancer cells. Fluorescent and radio conjugates of 11B6, an antibody targeting free hK2, are internalized and noninvasively report AR pathway activity in metastatic and genetically engineered models of cancer development and treatment. Uptake is mediated by a mechanism involving the neonatal Fc receptor. Humanized 11B6, which has undergone toxicological tests in nonhuman primates, has the potential to improve patient management in these cancers. Furthermore, cellspecific SATA uptake may have a broader use for molecularly guided diagnosis and therapy in other cancers.
