    • The ground-state decay of 26Al(0+) (T 1/2=1.05× 106) has a shorter life-time than the Universe. The presence of this element in the Galaxy was measured via g-ray spectroscopy, showing that the nucleosynthesis of this element is an ongoing process in stars. The proton-capture reaction 26Si(p,γ) 27P competes with the production of 26Al(0+) by β-decay. Coulomb dissociation of 27P has been suggested as an indirect method to measure radiative-proton capture when the direct reaction is not feasible. Such an experiment was performed at GSI with a secondary 27P beam produced by fragmenting a 36Ar primary beam at 500 A MeV.   • Central acceptance testing for camera technologies for the cherenkov telescope array

    • The Cherenkov Telescope Array (CTA) is an international initiative to build the next generation ground based very-high energy gamma-ray observatory. It will consist of telescopes of three different sizes, employing several different technologies for the cameras that detect the Cherenkov light from the observed air showers. In order to ensure the compliance of each camera technology with CTA requirements, CTA will perform central acceptance testing of each camera technology. To assist with this, the Camera Test Facilities (CTF) work package is developing a detailed test program covering the most important performance, stability, and durability requirements, including setting up the necessary equipment. Performance testing will include a wide range of tests like signal amplitude, time resolution, dead-time determination, trigger efficiency, performance testing under temperature and humidity variations and several others. These tests can be performed on fully-integrated cameras using a portable setup at the camera construction sites. In addition, two different setups for performance tests on camera sub-units are being built, which can provide early feedback for camera development. Stability and durability tests will include the long-term functionality of movable parts, water tightness of the camera housing, temperature and humidity cycling, resistance to vibrations during transport or due to possible earthquakes, UV-resistance of materials and several others. Some durability tests will need to be contracted out because they will need dedicated equipment not currently available within CTA. The planned test procedures and the current status of the test facilities will be presented.

  • The long road to elimination : malaria mortality in a South African population cohort over 21 years

    • Background: Malaria elimination is on global agendas following successful transmission reductions. Nevertheless moving from low to zero transmission is challenging. South Africa has an elimination target of 2018, which may or may not be realised in its hypoendemic areas.Methods: The Agincourt Health and Demographic Surveillance System has monitored population health in north-eastern South Africa since 1992. Malaria deaths were analysed against individual factors, socioeconomic status, labour migration and weather over a 21-year period, eliciting trends over time and associations with covariates.Results: Of 13 251 registered deaths over 1.58 million person-years, 1.2% were attributed to malaria. Malaria mortality rates increased from 1992 to 2013, while mean daily maximum temperature rose by 1.5 °C. Travel to endemic Mozambique became easier, and malaria mortality increased in higher socioeconomic groups. Overall, malaria mortality was significantly associated with age, socioeconomic status, labour migration and employment, yearly rainfall and higher rainfall/temperature shortly before death.Conclusions: Malaria persists as a small but important cause of death in this semi-rural South African population. Detailed longitudinal population data were crucial for these analyses. The findings highlight practical political, socioeconomic and environmental difficulties that may also be encountered elsewhere in moving from low-transmission scenarios to malaria elimination.

    • Context and purpose: Individual participant data-level meta-regression (IPD) analysis is superior to meta-regression based on aggregate data in determining Dietary Reference Values (DRV) for vitamin D. Using data from randomized controlled trials (RCTs) with vitamin D3-fortified foods, we undertook an IPD analysis of the response of winter serum 25-hydroxyvitamin (25(OH)D) to total vitamin D intake among children and adults and derived DRV for vitamin D.Methods: IPD analysis using data from 1429 participants (ages 2–89 years) in 11 RCTs with vitamin D-fortified foods identified via a systematic review and predefined eligibility criteria. Outcome measures were vitamin D DRV estimates across a range of serum 25(OH)D thresholds using unadjusted and adjusted models.Results: Our IPD-derived estimates of vitamin D intakes required to maintain 97.5% of winter 25(OH)D concentrations ≥ 25 and ≥ 30 nmol/L are 6 and 12 µg/day, respectively (unadjusted model). The intake estimates to maintain 90%, 95% and 97.5% of concentrations ≥ 50 nmol/L are 33.4, 57.5 and 92.3 µg/day, respectively (unadjusted) and 17.0, 28.1 and 43.6 µg/day, respectively (adjusted for mean values for baseline serum 25(OH)D, age and BMI).Conclusions: IPD-derived vitamin D intakes required to maintain 90%, 95% and 97.5% of winter 25(OH)D concentrations ≥ 50 nmol/L are much higher than those derived from standard meta-regression based on aggregate data, due to the inability of the latter to capture between person-variability. Our IPD provides further evidence that using food-based approaches to achieve an intake of 12 µg/day could prevent vitamin D deficiency (i.e., serum 25(OH)D < 30 nmol/L) in the general population.

  • Gold-modified paper as microfluidic substrates with reduced biofouling in potentiometric ion sensing

    • Microfluidic sampling media based on paper and its modifications with either gold nanoparticles or sputtered gold were evaluated for potentiometric determination of Na+, K+, and Cl– ions in clinically relevant samples. The measurements were conducted in comparison to other commonly considered microfluidic substrates, i.e. sponge, polyester textile, and polyamide textile. Ion determination was done by using solid-contact ion-selective electrodes based on plasticized PVC membranes for Na+, K+, and Cl– ions and utilizing PEDOT(PSS) or PEDOT(Cl) as the ion-to-electron transducer. The solid-contact ion-selective electrodes and a solid-state reference electrode were placed directly on the substrate into which the sample solution was wicked. Transport of bovine serum albumin (BSA) through the paper substrate was studied by ellipsometry. Modification of the paper substrates by gold nanoparticles (AuNPs) was found to slow down the transport of BSA through the paper, when compared with unmodified paper substrates and when compared with all the other alternative sampling matrices studied. The retention of BSA obtained with AuNP-modified paper substrates significantly improved the accuracy of the potentiometric ion determinations in sweat, saliva, artificial tears, and artificial serum. The potentiometric results were validated by inductively coupled plasma optical emission spectrometry (ICP-OES) and ion chromatography (IC). The study indicates that modification of paper by AuNPs is a feasible approach to reduce biofouling of sensors that are used in the paper-based analysis of clinically relevant samples. © 2021 Elsevier B.V.

  • Experimental phasing opportunities for macromolecular crystallography at very long wavelengths

    • Despite recent advances in cryo-electron microscopy and artificial intelligence-based model predictions, a significant fraction of structure determinations by macromolecular crystallography still requires experimental phasing, usually by means of single-wavelength anomalous diffraction (SAD) techniques. Most synchrotron beamlines provide highly brilliant beams of X-rays of between 0.7 and 2 Å wavelength. Use of longer wavelengths to access the absorption edges of biologically important lighter atoms such as calcium, potassium, chlorine, sulfur and phosphorus for native-SAD phasing is attractive but technically highly challenging. The long-wavelength beamline I23 at Diamond Light Source overcomes these limitations and extends the accessible wavelength range to λ = 5.9 Å. Here we report 22 macromolecular structures solved in this extended wavelength range, using anomalous scattering from a range of elements which demonstrate the routine feasibility of lighter atom phasing. We suggest that, in light of its advantages, long-wavelength crystallography is a compelling option for experimental phasing.

    • Constitutional mismatch repair deficiency (CMMRD) syndrome is a rare and aggressive cancer predisposition syndrome. Because a scarcity of data on this condition contributes to management challenges and poor outcomes, we aimed to describe the clinical spectrum, cancer biology, and impact of genetics on patient survival in CMMRD.In this cohort study, we collected cross-sectional and longitudinal data on all patients with CMMRD, with no age limits, registered with the International Replication Repair Deficiency Consortium (IRRDC) across more than 50 countries. Clinical data were extracted from the IRRDC database, medical records, and physician-completed case record forms. The primary objective was to describe the clinical features, cancer spectrum, and biology of the condition. Secondary objectives included estimations of cancer incidence and of the impact of the specific mismatch-repair gene and genotype on cancer onset and survival, including after cancer surveillance and immunotherapy interventions.We analysed data from 201 patients (103 males, 98 females) enrolled between June 5, 2007 and Sept 9, 2022. Median age at diagnosis of CMMRD or a related cancer was 8·9 years (IQR 5·9-12·6), and median follow-up from diagnosis was 7·2 years (3·6-14·8). Endogamy among minorities and closed communities contributed to high homozygosity within countries with low consanguinity. Frequent dermatological manifestations (117 [93%] of 126 patients with complete data) led to a clinical overlap with neurofibromatosis type 1 (35 [28%] of 126). 339 cancers were reported in 194 (97%) of 201 patients. The cumulative cancer incidence by age 18 years was 90% (95% CI 80-99). Median time between cancer diagnoses for patients with more than one cancer was 1·9 years (IQR 0·8-3·9). Neoplasms developed in 15 organs and included early-onset adult cancers. CNS tumours were the most frequent (173 [51%] cancers), followed by gastrointestinal (75 [22%]), haematological (61 [18%]), and other cancer types (30 [9%]). Patients with CNS tumours had the poorest overall survival rates (39% [95% CI 30-52] at 10 years from diagnosis; log-rank p<0·0001 across four cancer types), followed by those with haematological cancers (67% [55-82]), gastrointestinal cancers (89% [81-97]), and other solid tumours (96% [88-100]). All cancers showed high mutation and microsatellite indel burdens, and pathognomonic mutational signatures. MLH1 or MSH2 variants caused earlier cancer onset than PMS2 or MSH6 variants, and inferior survival (overall survival at age 15 years 63% [95% CI 55-73] for PMS2, 49% [35-68] for MSH6, 19% [6-66] for MLH1, and 0% for MSH2; p<0·0001). Frameshift or truncating variants within the same gene caused earlier cancers and inferior outcomes compared with missense variants (p<0·0001). The greater deleterious effects of MLH1 and MSH2 variants as compared with PMS2 and MSH6 variants persisted despite overall improvements in survival after surveillance or immune checkpoint inhibitor interventions.The very high cancer burden and unique genomic landscape of CMMRD highlight the benefit of comprehensive assays in timely diagnosis and precision approaches toward surveillance and immunotherapy.     • We present a three-stage analysis of genome-wide SNP data in 1,222 German individuals with myocardial infarction and 1,298 controls, in silico replication in three additional genome-wide datasets of coronary artery disease (CAD) and subsequent replication in similar to 25,000 subjects. We identified one new CAD risk locus on 3q22.3 in MRAS (P = 7.44 x 10(-13); OR = 1.15, 95% CI = 1.11-1.19), and suggestive association with a locus on 12q24.31 near HNF1A-C12orf43 (P = 4.81 x 10(-7); OR = 1.08, 95% CI = 1.05-1.11).
