    • In this paper, by virtue of one-dimensional ZnO nanorods and two-dimensional graphene film hybrid structures, both the enhanced current spreading and enhanced light extraction were realized at the same time. A 1 nm/1 nm Ni/Au layer was used as an interlayer between graphene and pGaN to form ohmic contact, which makes the device have a good forward conduction properties. Through the comparison of the two groups of making ZnO nanorods or not, it was found that the 30% light extraction efficiency of the device was improved by using the ZnO nanorods. By analysis key parameters of two groups such as the turn-on voltage, work voltage and reverse leakage current, it was proved that the method for preparing surface nano structure by hydrothermal method self-organization growth ZnO nanorods applied in GaN LEDs has no influence to device's electrical properties. The hybrid structure application in GaN LED, make an achievement of a good ohmic contact, no use of ITO and enhancement of light extraction at the same time, meanwhile it does not change the device structure, introduce additional process, worsen the electrical properties.

    • Point-of-care (POC) diagnostics is playing an increasingly important role in public health, environmental monitoring, and food safety analysis. Smartphones, alone or in conjunction with add-on devices, have shown great capability of data collection, analysis, display, and transmission, making them popular in POC diagnostics. In this article, the state-ofthe- art advances in smartphone-based POC diagnostic technologies and their applications in the past few years are outlined, ranging from in vivo tests that use smartphone’s built-in/external sensors to detect biological signals to in vitro tests that involves complicated biochemical reactions. Novel techniques are illustrated by a number of attractive examples, followed by a brief discussion of the smartphone’s role in telemedicine. The challenges and perspectives of smartphonebased POC diagnostics are also provided.

  • Mendelian randomization and clinical trial evidence supports TYK2 inhibition as a therapeutic target for autoimmune diseases

    • Background: To explore the associations of genetically proxied TYK2 inhibition with a wide range of disease outcomes and biomarkers to identify therapeutic repurposing opportunities, adverse effects, and biomarkers of efficacy.Methods: The loss-of-function missense variant rs34536443 in TYK2 gene was used as a genetic instrument to proxy the effect of TYK2 inhibition. A phenome-wide Mendelian randomization (MR) study was conducted to explore the associations of genetically-proxied TYK2 inhibition with 1473 disease outcomes in UK Biobank (N = 339,197). Identified associations were examined for replication in FinnGen (N = 260,405). We further performed tissue -specific gene expression MR, colocalization analyses, and MR with 247 blood biomarkers. A systematic review of randomized controlled trials (RCTs) on TYK2 inhibitor was performed to complement the genetic evidence.Findings: PheWAS-MR found that genetically-proxied TYK2 inhibition was associated with lower risk of a wide range of autoimmune diseases. The associations with hypothyroidism and psoriasis were confirmed in MR analysis of tissue-specific TYK2 gene expression and the associations with systemic lupus erythematosus, psoriasis, and rheumatoid arthritis were observed in colocalization analysis. There were nominal associations of genetically-proxied TYK2 inhibition with increased risk of prostate and breast cancer but not in tissue-specific expression MR or colocalization analyses. Thirty-seven blood biomarkers were associated with the TYK2 loss-of-function mutation. Evidence from RCTs confirmed the effectiveness of TYK2 inhibitors on plaque psoriasis and reported several adverse effects.Interpretation: This study supports TYK2 inhibitor as a potential treatment for psoriasis and several other autoim-mune diseases. Increased pharmacovigilance is warranted in relation to the potential adverse effects.

  • Plasma proteins and onset of type 2 diabetes and diabetic complications : Proteome-wide Mendelian randomization and colocalization analyses

    • We conduct proteome-wide Mendelian randomization and colocalization analyses to decipher the associa-tions of blood proteins with the risk of type 2 diabetes and diabetic complications. Genetic data on plasma proteome are obtained from 54,306 UK Biobank participants and 35,559 Icelanders. Summary-level data on type 2 diabetes are obtained from the DIAGRAM (DIAbetes Genetics Replication And Meta-analysis con-sortium) consortium (74,124 cases) and FinnGen study (33,043 cases). Data on 10 diabetic complications are obtained from FinnGen and corresponding studies. Among 1,886 proteins, genetically predicted levels of 47 plasma proteins are associated with type 2 diabetes. Eleven of these proteins have strong support of colocalization. Seventeen proteins are associated with at least one diabetic complication, although a few have colocalization support. HLA-DRA, AGER, HSPA1A, and HSPA1B are associated with most microvas-cular complications. This study reveals causal proteins for the onset of type 2 diabetes and diabetic compli-cations, which enhances the understanding of molecular etiology and development of therapeutics.

    • Metal-organic frameworks (MOFs) based on Ti-oxo clusters (Ti-MOFs) represent a naturally self-assembled superlattice of TiO2 nanoparticles separated by designable organic linkers as antenna chromophores, epitomizing a promising platform for solar energy conversion. However, despite the vast, diverse, and well-developed Ti-cluster chemistry, only a scarce number of Ti-MOFs have been documented. The synthetic conditions of most Ti-based clusters are incompatible with those required for MOF crystallization, which has severely limited the development of Ti-MOFs. This challenge has been met herein by the discovery of the [Ti8Zr2O12(COO)(16)] cluster as a nearly ideal building unit for photoactive MOFs. A family of isoreticular photoactive MOFs were assembled, and their orbital alignments were fine-tuned by rational functionalization of organic linkers under computational guidance. These MOFs demonstrate high porosity, excellent chemical stability, tunable photoresponse, and good activity toward photocatalytic hydrogen evolution reactions. The discovery of the [Ti8Zr2O12(COO)(16)] cluster and the facile construction of photoactive MOFs from this cluster shall pave the way for the development of future Ti-MOF-based photocatalysts.   • Innovations in science, technology, engineering, and policy (iSTEP) for addressing environmental issues towards sustainable development

    • Sustainable development depends on the integration of the economy, society, and environment. Yet, escalating environmental challenges pose threats to both society and the economy. Despite progress in addressing environmental issues to promote sustainability, knowledge gaps in scientific research, technological advancement, engineering practice, and policy development persist. In this review, we aim to narrow these gaps by proposing innovation-based solutions and refining existing paradigms. Reviewing past research and actions, we first elucidate the evolution of sustainability science and the essence of sustainable development and its assessment. Secondly, we summarize current major environmental issues, including global warming and climate change, biodiversity loss, land degradation and desertification, and environmental pollution, as well as their relationships with sustainability and the achievement of Sustainable Development Goals (SDGs). Subsequently, this review critically evaluates the role of innovations in science, technology, engineering, and policy (iSTEP) and their synergies in advancing sustainability and SDGs. While their sequential relationships may vary based on specific contexts or sustainability scenarios within the iSTEP framework, each component reinforces the others, fostering continuous improvement. Finally, this review offers recommendations and future perspectives for formulating sustainability roadmaps. Recommendations include fostering a vision of sustainability, promoting interdisciplinary collaboration, and encouraging transboundary cooperation among stakeholders for future sustainability endeavors.

    • Birds are the most species-rich class of tetrapod vertebrates and have wide relevance across many research fields. We explored bird macroevolution using full genomes from 48 avian species representing all major extant clades. The avian genome is principally characterized by its constrained size, which predominantly arose because of lineage-specific erosion of repetitive elements, large segmental deletions, and gene loss. Avian genomes furthermore show a remarkably high degree of evolutionary stasis at the levels of nucleotide sequence, gene synteny, and chromosomal structure. Despite this pattern of conservation, we detected many non-neutral evolutionary changes in protein-coding genes and noncoding regions. These analyses reveal that pan-avian genomic diversity covaries with adaptations to different lifestyles and convergent evolution of traits.

    • A high-performance flexible sodium-ion full battery (FSIFB) is developed by using binder-free composite film (BFCF) electrodes without using conductive carbon and current collectors. Hard carbon fibers decorated with different electrochemical active materials are used as the supporting framework and 3D conductive network of FSIFBs for the first time. Different pre-sodiated anodes and the electrolyte additives are designed for well-matched FSIFBs. Using a porous Na3V2(PO4)(3) coated hard carbon fiber film with a mass loading of 2.34 mg cm(-2) as the cathode and a pre-sodiated graphene/SiC/hard carbon fiber film with a mass loading of 1.50 mg cm(-2) as the anode, an optimized FSIFB is designed. It delivers high output voltage (3.34 V), high energy density (234.1 W h kg(-1) at a high-current rate of 0.5 A g(-1)), ultralong cyclability (over 2905 cycles at 0.5 A g(-1) and 1000 cycles at 5 A g(-1)), and high coulombic efficiency (approaching 100%), which surpasses those of all FSIFBs reported so far. Furthermore, this FSIFB still maintains good electrochemical attributes even at serious bending states in water. The models of the solid electrolyte interphase behavior on the surface of electrodes in the FSIFB are studied by using EIS, and a reaction mechanism and an equivalent electrical circuit are proposed. We also provide the videos of the preparation process for a pouch-type FSIFB to demonstrate its simple operability and potential applications.

    • To develop battery-supercapacitor hybrid devices with high energy and power densities, we propose a rational design of a quaternary hybrid superstructure by using a high-energy biotemplate. This new superstructure is composed of stable fullerene C-60 nanocages, electroactive Na4FeO3, high-energy Li3V2(PO4)(3) and soft carbon as well as tubular ordered mesoporous channels. This design takes advantage of the unique properties of each component, resulting in nanocomposites with synergistic effects to improve the charge transfer and energy storage. We found that this quaternary hybrid electrode has both high energy and power densities as well as a long cycling life in a Li/Na mixed-ion electrolyte, outperforming a multitude of other battery-supercapacitor hybrid devices reported thus far. The charge storage mechanisms of this hybrid superstructure are proposed for optimizing the electrode design.

  • Auto-suppression of Tet dioxygenases protects the mouse oocyte genome from oxidative demethylation

    • DNA cytosine methylation plays a vital role in repressing retrotransposons, and such derepression is linked with developmental failure, tumorigenesis and aging. DNA methylation patterns are formed by precisely regulated actions of DNA methylation writers (DNA methyltransferases) and erasers (TET, ten-eleven translocation dioxygenases). However, the mechanisms underlying target-specific oxidation of 5mC by TET dioxygenases remain largely unexplored. Here we show that a large low-complexity domain (LCD), located in the catalytic part of Tet enzymes, negatively regulates the dioxygenase activity. Recombinant Tet3 lacking LCD is shown to be hyperactive in converting 5mC into oxidized species in vitro. Endogenous expression of the hyperactive Tet3 mutant in mouse oocytes results in genome-wide 5mC oxidation. Notably, the occurrence of aberrant 5mC oxidation correlates with a consequent loss of the repressive histone mark H3K9me3 at ERVK retrotransposons. The erosion of both 5mC and H3K9me3 causes ERVK derepression along with upregulation of their neighboring genes, potentially leading to the impairment of oocyte development. These findings suggest that Tet dioxygenases use an intrinsic auto-regulatory mechanism to tightly regulate their enzymatic activity, thus achieving spatiotemporal specificity of methylome reprogramming, and highlight the importance of methylome integrity for development. Here the authors show that TET dioxygenases, the erasers of DNA methylation, use a self-limiting mechanism via their LCD domain to ensure adaptable methylome status and protect the genome from excessive oxidative methylation.
