  • Immune response and barrier dysfunction-related proteomic signatures in preclinical phase of Crohn's disease highlight earliest events of pathogenesis

    • OBJECTIVE: The measure of serum proteome in the preclinical state of Crohn's disease (CD) may provide insight into biological pathways involved in CD pathogenesis. We aimed to assess associations of serum proteins with future CD onset and with other biomarkers predicting CD risk in a healthy at-risk cohort.DESIGN: In a nested case-control study within the Crohn's and Colitis Canada Genetics Environment Microbial Project (CCC-GEM) cohort, which prospectively follows healthy first-degree relatives (FDRs), subjects who developed CD (n=71) were matched with four FDRs remaining healthy (n=284). Using samples at recruitment, serum protein profiles using the Olink Proximity Extension Assay platform was assessed for association with future development of CD and with other baseline biomarkers as follows: serum antimicrobial antibodies (AS: positive antibody sum) (Prometheus); faecal calprotectin (FCP); gut barrier function using the fractional excretion of lactulose-to-mannitol ratio (LMR) assay.RESULTS: We identified 25 of 446 serum proteins significantly associated with future development of CD. C-X-C motif chemokine 9 (CXCL9) had the highest OR with future risk of CD (OR=2.07 per SD, 95% CI 1.58 to 2.73, q=7.9e-5), whereas matrix extracellular phosphoglycoprotein had the lowest OR (OR 0.44, 95% CI 0.29 to 0.66, q=0.02). Notably, CXCL9 was the only analyte significantly associated with all other CD-risk biomarkers with consistent direction of effect (FCP: OR=2.21; LMR: OR=1.67; AS: OR=1.59) (q<0.05 for all).CONCLUSION: We identified serum proteomic signatures associated with future CD development, reflecting potential early biological processes of immune and barrier dysfunction.

  • A novel whole blood gene expression signature for asthma, dermatitis, and rhinitis multimorbidity in children and adolescents

    • Background: Allergic diseases often occur in combination (multimorbidity). Human blood transcriptome studies have not addressed multimorbidity. Large-scale gene expression data were combined to retrieve biomarkers and signaling pathways to disentangle allergic multimorbidity phenotypes.Methods: Integrated transcriptomic analysis was conducted in 1233 participants with a discovery phase using gene expression data (Human Transcriptome Array 2.0) from whole blood of 786 children from three European birth cohorts (MeDALL), and a replication phase using RNA Sequencing data from an independent cohort (EVA-PR, n = 447). Allergic diseases (asthma, atopic dermatitis, rhinitis) were considered as single disease or multimorbidity (at least two diseases), and compared with no disease.Results: Fifty genes were differentially expressed in allergic diseases. Thirty-two were not previously described in allergy. Eight genes were consistently overexpressed in all types of multimorbidity for asthma, dermatitis, and rhinitis (CLC, EMR4P, IL5RA, FRRS1, HRH4, SLC29A1, SIGLEC8, IL1RL1). All genes were replicated the in EVA-PR cohort. RT-qPCR validated the overexpression of selected genes. In MeDALL, 27 genes were differentially expressed in rhinitis alone, but none was significant for asthma or dermatitis alone. The multimorbidity signature was enriched in eosinophil-associated immune response and signal transduction. Protein-protein interaction network analysis identified IL5/JAK/STAT and IL33/ST2/IRAK/TRAF as key signaling pathways in multimorbid diseases. Synergistic effect of multimorbidity on gene expression levels was found.Conclusion: A signature of eight genes identifies multimorbidity for asthma, rhinitis, and dermatitis. Our results have clinical and mechanistic implications, and suggest that multimorbidity should be considered differently than allergic diseases occurring alone.

  • Genome architecture and selective signals compensatorily shape plastic response to a new environment

    • Transcriptional plasticity interacts with natural selection in complex ways and is crucial for the survival of species under rapid climate change. How 3D genome architecture affects transcriptional plasticity and its interaction with genetic adaptation are unclear. We transplanted estuarine oysters to a new environment and found that genes located in active chromatin regions exhibited greater transcriptional plasticity, and changes in these regions were negatively correlated with selective signals. This indicates a trade-off be- tween 3D active regions and selective signals in shaping plastic responses to a new environment. Specifically, a mutation, lincRNA, and changes in the accessibility of a distal enhancer potentially affect its interaction with the ManIIa gene, which regulates the muscle function and survival of oysters. Our findings reveal that 3D genome architecture compensates for the role of genetic adaptation in environmental response to new environments and provide insights into synergetic genetic and epigenetic interactions critical for fitness-related trait and survival in a model marine species.

  • Fabrication and characterization of bioactive zirconia-based nanocrystalline glass-ceramics for dental abutment

    • Zirconia-based ceramics are becoming a popular biomaterial in dental implantology due to their natural silver-white color, excellent mechanical properties, and good biocompatibility. However, zirconia-based ceramics are biologically inert, which limits their ability to integrate with the surrounding human tissues. To solve this problem, the bioactive elements of calcium (Ca) and phosphorus (P) were doped in high-strength ZrO2-SiO2 nanocrystalline glass-ceramics (NCGCs) to overcome the biological inertness of ZrO2-based ceramics. XRD results showed that tetragonal zirconia (t-ZrO2) and monoclinic zirconia (m-ZrO2) were the only two crystalline phases after spark plasma sintering. Ca and P dopants acted as destabilizer of t-ZrO2, enhancing its transformability to m-ZrO2 during sintering. The amount of t-ZrO2 exerted significant effects on the average flexural strength of the NCGCs. The NCGC with 45 mol% ZrO2 were composed of 64.5 vol% t-ZrO2 and 35.5 vol% m-ZrO2 after sintering at 1230 degrees C. And, the average flexural strength and Vickers hardness of the NCGC was 615 MPa and 1049 HV, respectively. In comparison, the NCGC with 65 mol% ZrO2 were composed of 12.6 vol% t-ZrO2 and 87.4 vol% m-ZrO2 after sintering at 1150 degrees C. The average flexural strength and Vickers hardness of the NCGC was 293 MPa and 839 HV, respectively. Interestingly, the NCGCs exhibited a plastic deformation behavior during flexural strength test, which was different from traditional brittle ceramics. The ion release results demonstrated that Ca2+ and Si4+ ions kept on releasing from the surface of the material. The formation of hydroxyapatite in the in-vitro apatite formation test indicated that the NCGCs had good biological activity. The doped ZrO2-based NCGCs combined moderate strength and good bioactivity. Hence, the NCGCs show promising potential to be used in sub-gingival regions, such as dental abutments.

  • Non-fullerene acceptors with branched side chains and improved molecular packing to exceed 18% efficiency in organic solar cells

    • Molecular design of acceptor and donor molecules has enabled major progress in organic photovoltaics. Li et al. show that branched alkyl chains in non-fullerene acceptors allow favourable morphology in the active layer, enabling a certified device efficiency of 17.9%. Molecular design of non-fullerene acceptors is of vital importance for high-efficiency organic solar cells. The branched alkyl chain modification is often regarded as a counter-intuitive approach, as it may introduce an undesirable steric hindrance that reduces charge transport in non-fullerene acceptors. Here we show the design and synthesis of a highly efficient non-fullerene acceptor family by substituting the beta position of the thiophene unit on a Y6-based dithienothiophen[3,2-b]-pyrrolobenzothiadiazole core with branched alkyl chains. It was found that such a modification to a different alkyl chain length could completely change the molecular packing behaviour of non-fullerene acceptors, leading to improved structural order and charge transport in thin films. An unprecedented efficiency of 18.32% (certified value of 17.9%) with a fill factor of 81.5% is achieved for single-junction organic solar cells. This work reveals the importance of the branched alkyl chain topology in tuning the molecular packing and blend morphology, which leads to improved organic photovoltaic performance.

  • A Review of Relative Pollen Productivity Estimates From Temperate China for Pollen-Based Quantitative Reconstruction of Past Plant Cover

    • Model-based quantitative reconstruction of past plant cover in Europe has shown great potential for: (i) testing hypotheses related to Holocene vegetation dynamics, biodiversity, and their relationships with climate and land use; (ii) studying long term interactions between climate and land use. Similar model-based quantitative reconstruction of plant cover in China has been restricted due to the lack of standardized datasets of existing estimates of relative pollen productivity (RPP). This study presents the first synthesis of all RPP values available to date for 39 major plant taxa from temperate China and proposes standardized RPP datasets that can be used for model-based quantitative reconstructions of past plant cover using fossil pollen records for the region. We review 11 RPP studies in temperate China based on modern pollen and related vegetation data around the pollen samples. The study areas include meadow, steppe and desert vegetation, various woodland types, and cultural landscapes. We evaluate the strategies of each study in terms of selection of study areas and distribution of study sites; pollen- and vegetation-data collection in field; vegetation-data collection from satellite images and vegetation maps; and data analysis. We compare all available RPP estimates, select values based on precise rules and calculate mean RPP estimates. We propose two standardized RPP datasets for 31 (Alt1) and 29 (Alt2) plant taxa. The ranking of mean RPPs (Alt-2) relative to Poaceae (= 1) for eight major taxa is: Artemisia (21) > Pinus (18.4) > Betula (12.5) > Castanea (11.5) > Elaeagnaceae (8.8) > Juglans (7.5) > Compositae (4.5) > Amaranthaceae/Chenopodiaceae (4). We conclude that although RPPs are comparable between Europe and China for some genera and families, they can differ very significantly, e.g., Artemisia, Compositae, and Amaranthaceae/Chenopodiaceae. For some taxa, we present the first RPP estimates e.g. Castanea, Elaeagnaceae, and Juglans. The proposed standardized RPP datasets are essential for model-based reconstructions of past plant cover using fossil pollen records from temperate China.

    • Advances in thin film light-emitting devices have fueled the rapid growth of a new class of solid-state lighting devices, featuring low fabrication cost, high quantum efficiency, and broadband spectrum coverage, etc. In contrast to the conventional inorganic semiconductors that rely on lattice matched high crystalline quality substrate, solution processable thin films eliminate the dependence on the substrate, which is highly desired for the ease and versatility of integrations with foreign medium. By taking this advantage, this work developed an ultracompact solution to control the directionality of thin film emitters using integrated dielectric metasurface through one step spin-coating process. As a proof of concept, directional emissions from perovskite nanocrystal thin film, including collimated light emissions and two-dimensional beam steering, are experimentally demonstrated. Notably, our approach, where light emitters were integrated on the back side of substrate after the fabrication of metasurface, judiciously avoids any potential degradation of material optical quality caused by the multi-step nanofabrication. Therefore, it can serve as a generalized scheme to engage the advantageous properties of dielectric metasurface, including the compactness, high efficiency, and beam controllability with the emerging thin film light-emitting diodes (LEDs), which is applicable to a wide range of solution processable materials, including organic light-emitting diodes, quantum-dot light emitting diodes, polymer LEDs, and perovskite LEDs, opening up new pathways to develop low-cost and ultra-compact solid state light sources with versatile beams characteristics.   • Growth of High Material Quality Group III-Antimonide Semiconductor Nanowires by a Naturally Cooling Process

    • We report on a simple but powerful approach to grow high material quality InSb and GaSb nanowires in a commonly used tube furnace setup. The approach employs a process of stable heating at a high temperature and then cooling down naturally to room temperature with the nanowire growth occurred effectively during the naturally cooling step. As-grown nanowires are analyzed using a scanning electron microscope and a transmission electron microscope equipped with an energy-dispersive X-ray spectroscopy setup. It is shown that the grown nanowires are several micrometers in lengths and are zincblende InSb and GaSb crystals. The FET devices are also fabricated with the grown nanowires and investigated. It is shown that the grown nanowires show good, desired electrical properties and should have potential applications in the future nanoelectronics and infrared optoelectronics.
